Objective : The object of this study was to observe the effects of aqueous extracts of Chunglijagam-tang(CLJGT), which has traditionally been used in Korean medicine for treating various diseases, on streptozotocin(STZ)-induced rat diabetes and related complications: diabetic nephropathy, hepatopathy and hyperlipemia. Methods : CLJGT extracts were orally administered once a day for 28 days at a dosage 50, 100 and 200mg/kg from 21 days after STZ treatment, and the changes on body weights, blood glucose levels, kidney and liver weights, serum BUN(blood urea nitrogen), creatinine, AST(aspartate transaminase), ALT(alanine aminotransferase), HDL(high-density lipoprotein), LDL (low-density lipoprotein), total cholesterol and triglyceride levels were observed with pancreatic malondialdehyde(MDA) and glutathione(GSH) contents. The results were compared with silymarin 100mg/kg. Results : Significant decrease of blood glucose levels, kidney and liver weights, serum BUN, creatinine, AST, ALT, LDL, total cholesterol, triglyceride levels, pancreatic malondialdehyde contents and significant increase of body weights, serum HDL levels, pancreatic glutathione contents were detected in CLJGT extracts 50, 100 and 200mg/kg administered groups as compared to the STZ control group. Conclusion : CLJGT extracts showed favorable effects on the STZ-induced diabetes and related complications mediated by their antioxidant effects as similar to silymarin. Therefore, it is expected that DBEH has potential for use in the management of diabetes and various diabetic complications.

Our objective of current study was to investigate the development of bone and heart in association with diabetes mellitus (DM). DM was induced by administering an intraperitoneal injection of streptozotocin (STZ; 60 mg/kg) to 4-week-old Sprague-Dawley rats. Body weight and blood glucose were monitored, and rats were sacrificed after 2 or 5 weeks. The left ventricle (LV), including the interventricular septum, was weighed, and body weight and tibial bone length were assessed. Young diabetic rats showed reduced growth in terms of tibial length and body weight compared to controls. Moreover, diabetic males showed more significant growth suppression and reduced LV size than diabetic females. Morphometric analysis of tibiae from diabetic rats revealed suppressed bone growth at 2 and 5 weeks, with no difference between genders. STZ-induced diabetes decreased bone growth and retarded pre-pubertal heart development. As a result, diabetes may increase cardiovascular risk factors and lead to eventual heart failure. Therefore, new therapeutic approaches are required for diabetic children exhibiting growth retardation. Heart growth factor, exercise, and cardiopulmonary physical therapy may be required to promote heart development and physiological function.

Objectives : This study was performed to prove the anti-hypertensive and anti-diabetic effect of Mori Ramulus(MR). Methods : The examination was done on the femoral artery occlusion of the rat, and check the blood pressure. In addition, we observed therapueutic effects on diabetes rats induced by streptozotocin. Results : MR MeOH Ex, showed few effect on descending blood pressure in SD rat model. MR CHC13 layer (MRC) showed significant effect on descending blood pressure. MR was effective on the streptozotocin-induced diabetes SD rats. Especially MRC reduced the amount of serum glucose, BUN, triglyceride and insulin level. Conclusions : MRC could induce descending blood pressure in the rat model. Also, it is expected to be effective in diabetes treatment.
Fifteen mongrel dogs (14 male and 1 female) were injected intravenously with 30mg of streptozotocin and 50mg of alloxan monohydrate per kilogram of body weight to induce diabetes mellitus. Before treatment with streptozotocin and alloxan fasting serum glucose concentration was determined every other day or thrice a week (Monday, Wednesday, Friday) for 3 months. Among 15 dogs 4 dogs developed diabetes mellitus and survived more than 9 weeks without injection of insulin. After treatment fasting serum glucose and hemoglobin $A_1$ concentrations of the 4 dogs were determined every other day or thrice a week. Fasting serum glucose concentration increased acutely from 24 hours after treatment and then showed severe fluctuation. Hemoglobin $A_1$ concentration increased gradually until 7~9 weeks after treatment and then showed very slow increase afterwards. Correlation of hemoglobin $A_1$ to fasting glucose concentration was relatively weak(r = 0.10~0.80). Hemoglobin $A_1$ and fasting glucose concentration of preceding 7 week showed very high correlation (r = 0.98~0.99). It was indicated that hemoglobin $A_1$ concentration in chemically induced diabetic dogs reflects mean glucose concentration of preceding 7~9 weeks.
In this study, we investigated the antioxidative effects of So-Dang-Tang (SDT) on streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intraperitoneal injection of STZ (45 mg/kg body weight) into Sprague-Dawley rats. The SDT (200 mg/kg) and the reference drug, glibenclimide (1mg/kg), were orally administered once a day for 28 days in STZ-induced diabetic rats. The activity of antioxidant enzymes, including those of superoxide dismutase (SOD) and catalase, and the levels of glutathione (GSH) and production of malondialdehyde (MDA) were measured in the liver, kidney, and pancreas of diabetic rats. Treatment with SDT in STZ-induced diabetic rats significantly increased the activities of antioxidant enzymes and GSH levels in the liver, kidney, and pancreas when compared to those of the STZ-control group. SDT also significantly decreased lipid peroxidation product and MDA levels in STZ-induced diabetic rats. These results indicate that SDT has an antioxidative action in STZ-induced diabetic rats.
Diabetes mellitus is associated with vascular complications, including an impairment of vascular function and alterations in the reactivity of blood vessels to vasoactive substances in various vasculature. In the present study, the authors have observed endothelin-B ($ET_B$) receptor agonist-induced relaxation in precontracted mesenteric arterial segments from streptozotocin (STZ)-induced diabetic rats, which was not shown from control rats or in other arterial segments from diabetic rats. Accordingly, the goal of this study was to investigate in what way STZ-induced diabetes altered reactivity of the mesenteric arterial bed and to examine the causal relaxation, if any, between this $ET_B$ receptor-mediated relaxation and endothelial paracrine function, especially nitric oxide (NO) production. The relaxation induced by $ET_B$ agonists was not observed in mesenteric arteries without endothelium. The relaxation to $ET_B$ agonists was completely abolished by pretreatment with BQ788, but not by BQ610. $N_{\omega}-nitro-L-arginine$ methyl ester and soluble guanylate cyclase inhibitors, methylene blue or LY83583 significantly attenuated the relaxant responses to $ET_B$ agonists, respectively. When the expression of eNOS and iNOS was evaluated on agarose gel stained with ethidium bromide, the expression of eNOS mRNA in diabetic rats was significantly decreased, but the expression of iNOS was increased compared with control rats. Furthermore, the iNOS-like immunostaining was densely detected in the endothelium and slightly in the arterial smooth muscle of diabetic rats, but not in control rats. These observations suggest that $ET_B$ receptor may not play a role in maintaining mesenteric vascular tone in normal situation. However, the alterations in $ET_B$ receptor sensitivity were found in diabetic rats and lead to the $ET_B$ agonist-induced vasorelaxation, which is closely related to NO production. In the state of increased vascular resistance of diabetic mesenteric vascular bed, enhanced NO production by activation of iNOS could lead to compensatory vasorelaxation to modulate adequate perfusion pressure to splanchnic area.
We have investigated the effects of the solvent fractions from the aleurone layer of Oryza sativa cv. Heugjinmi on plasma glucose and lipid levels in streptozotocin-induced diabetic rats. Sprague-Dawley rats were divided into 6 groups, normal, diabetic control, and four experimental groups. Diabetes mellitus was induced by injection of streptozotocin(45 mg/kg body weight) into tail vein, and fraction of Oryza sativa cv. Heugjinmi was administered orally in experimental rats for 14 days. The body weight, glucose, cholesterol, HDL-cholesterol, triglyceride, free fatty acid level, and aminotransferase activities were analyzed. It was identified that acetone and anthocyanin fractions significantly reduced the levels of glucose, triglyceride, and free fatty acid in plasma of diabetic rats. And HDL-cholesterol levels of acetone and anthocyanin groups were higher than those of diabetic control group. From these data, it can be postulated that acetone and anthocyanin fractions might possess rather potent hypoglycemic and hypotriglyceridemic properties in streptozotocin-induced diabetic rats.
Objectives : In order to evaluate the effect of Samguiyong-tang (SGYT) on diabetes, we prepared two types of Samguiyong-tang (Type-I and -II) which was composed of three kinds of oriental drug such as Ginseng, Angelica gigantis radix and Deer antler. Type I was traditional hot-water extract prepared from three kinds of drug, and Type II was the mixture of ethanol-extract of ginseng and hot-water extract prepared from the other two drugs. Methods : We tested the effects of SGYT on the blood glucose levels in normal rats by the method of glucose tolerance test. And also examined the effects of SGYT on the levels in normal rats or diabetic rats induced by Streptozotocin during 20 days. Results : 1. In the course of oral glucose tolerance test, the blood glucose level decreased by administration of SGYT I or II in normal rats. 2. In the course administration of SGYT during 20 days in normal rats, the blood glucose levels decreased until day 4 by Type I or Type II, but thereafter the level was recovered to the normal. 3. In the course administration of SGYT during 20 days in the diabetic rats induced by streptozotocin, Type I (SGYT) had some effect on the blood glucose levels only at 12 day, and Type II (SGYT) decreased the levels from 6th day and so on, significantly. Conclusions : The results suggested that SGYT II had some decreasing effects on the blood glucose levels in diabetic rats induced by Streptozotocin.

Extract of Acanthopanax senticosus has recently been demonstrated to possess significant antidiabetic potential, in accordance with the traditional use of this plant as an antidiabetic natural health product. The present study evaluated the effects of fermented extract (FE) of this plant on glucose-stimulated insulin secretion, glucose uptake, and streptozotocin-induced type 1 diabetes model. A 3 h pretreatment with FE prevented $IL-1{\beta}$ and $IFN-{\gamma}$ toxicity in isolated rat islets. However, it did not affect insulin-stimulated glucose uptake in C2C12 myotubes. In addition, pretreatment of mice with FE blocked the destruction of streptozotocin-induced islets and the development of type 1 diabetes. FE reduced blood glucose level, increased insulin secretion, and improved glucose tolerance in streptozotocin-treated mice, whereas nonfermented extract (NFE) had moderate effects. Immunohistochemical staining for insulin clearly showed that pretreatment with FE blocked the STZ-induced islets destruction and restored the number of islet cells that secreted insulin to the level of the control. Although the active principles and their mechanisms of action remain to be identified, FE may nevertheless represent a novel complementary therapy and a source of novel therapeutic agents against type 1 diabetes mellitus.
Hypoglycemic effect of Tabebuia avellandae was investigated in the streptozotocin(STZ)-induced diabetic rats. Diabetes was induced in male Sprague-Dawley rats by injections of STZ (45 mg/kg, i.v.). Rats weighing 200-250 g were divided into 6 groups: normal, STZ-control, hexane fr., CHCl$_3$fr., BuOH fr. and $H_2O$ fr. group. Normal and STZ-control rats received 3% Tween 80 only. Four groups of diabetic rats were administered orally at doses of 100, 400, 300 and 400 mg/kg/day of hexane, CHC1$_3$, BuOH and $H_2O$ fr. respectively. Fractions were administered orally to the rats for 7 days after STZ injection. All rats were anesthetized with ether, blood samples were taken by cardiac puncture for clinical chemistry and the rats were killed by exsanguination. Liver, kidney, heart and spleen were removed, weighed and analyzed. We measured glucose, protein, cholesterol and triglyceride levels in the plasma and glycogen, cholesterol and triglyceride levels in liver. The extent of blood glucose decrement in rats administered $H_2O$ fraction was greater than that in the STZ-control rats. The serum cholesterol and triglyceride levels were significantly lowered by administration of $H_2O$ fraction compared with those of STZ-control group. Treatment of rats with Tabebuia avellandae fractions caused decreases in STZ-induced elevation of cholesterol and triglyceride. Liver triglyceride level was significantly lowered hexane and BuOH fraction group compared with STZ-control group. These results suggest that $H_2O$ fraction of Tabebuia avellandae has the hypoglycemic action against STZ-induced diabetic rats.
