Effect of Hematoxylin on Glucose Metabolism in Soleus Muscle of Streptozotocin-Induced Diabetic Rats

  • 천연 색소성분의 하나인 hematoxylin은 Streptozotocin으로 유도한 당뇨병쥐에서 혈당강하 효과를 나타내는 것으로 관찰되었으며, 이때 혈중 insulin치에는 영향을 미치지 않았다. 혈당강하기전 연구의 일환으로 당뇨병쥐의 soleuo muscle에서의 포도당 대사에 미치는 영향을 검토한 결과 in vivo 및 in vitro 실험 모두에서 대사능이 증가되었고 이는 포도당대사 과정에서의 insulin 작용의 강화와 glycgen 합성의 증가에 기인하는 것으로 추정된다.

Hypoglycemic and Hypolipemic Effects of Ixeris dentata in Diabetic Rats

  • Blood glucose and total lipid levels in rats with streptozotocin-induced diabetes were determined after intraperitoneal administration of methanolic extract of Ixeris dentat whole plants and its main component, cynaroside (=luteolin 7-O--$\beta$-D-glucoside). From the data obtained, it was concluded that intraperitoneal administration of the methanolic extract produced a significant hypoglycemic effect. Total blood lipids were also decreased. Cynaroside in the streptozotocin-diabetic rats failed to exhibit hypoglycemic effect but a significant hypolipemic activity was observed. Thus, it is suggested that this methanolic extract may contain one or more hypoglycemic and hypolipemic principles including the main flavone glucoside, cynaroside, which can significantly reduce the levels triglyceride and total cholesterol in streptozotocin-diabetic rats.

  • 본 실험은 streptozotocin(STZ)으로 당뇨병이 유도된 쥐의 혈장 요소질소량, 지질수준 및 간 지질수준, 간 효소 활성에 천연 기능성 소재 화합물이 미치는 영향을 평가하는 것이 목적이었다. 총 콜레스테롤은 기능성 소재를 보충 급여한 실험군에서 유의적으로 낮은 수치(70.69 mg/dL)를 보였으며, 이로 인해 총 콜레스테롤에 대한 HDL 콜레스테롤의 비율이 증가(42.6에서 51.5%로) 하였다. 그러나, SOD, CAT, GSH-Px, GSH 및 LPO 활성에는 변화가 없었으며, 이는 기능성 소재의 보충 급여가 당뇨 쥐의 산화적 스트레스에는 큰 영향을 주지 않음을 의미한다. 보충 급여가 당뇨 쥐의 AST 수치 감소시켰으며, 이는 천연 기능성 소재 화합물 섭취가 간 기능 손상을 입은 STZ 유발 당뇨 쥐에 회복작용을 함을 알 수 있었다.

  • Type 1 diabetes mellitus (T1DM) is an autoimmune disease characterized by extreme insulin deficiency due to an overall reduction in the mass of functional pancreatic ${\beta}$-cells. Several animal models have been used to study T1DM. Amongst these, the mini-pig seems to be the most ideal model for diabetes research, owing to similarities with humans in anatomy and physiology. The purpose of this study was to analyze differentially expressed pancreatic proteins in a streptozotocin (STZ)-induced mini-pig T1DM model. Pancreas proteins from mini-pigs treated with STZ were separated by two-dimensional gel electrophoresis, and 11 protein spots were found to be altered significantly when compared with control mini-pigs. The data in this study utilizing proteomic analysis provide a valuable resource for the further understanding of the T1DM pathomechanism.

Effect of Berberis tinctoria leaf (Berberidaceae) extract on antidiabetic, antihyperlipidemic and antioxidant status in streptozotocin induced diabetes in rats

  • The present study was carried out to investigate the antidiabetic and antioxidant effect of methanolic extract of Berberis tinctoria leaves (MEBT), in streptozotocin induced diabetic rats. Oral administration of MEBT extract (150 mg/kg and 300 mg/kg) for a period of 14 days. Blood glucose levels, body weight food and liquid intake were measured on every $5^{th}$ day over a period of 14 days. In diabetic rats, MEBT at the dose of 150 mg/kg and 300 mg/kg body weight resulted in significant reduction in blood glucose levels. The study was further investigated to determine antioxidant and antihyperlipidemic potential of MEBT in streptozotocin (STZ) induced diabetic rats. These results suggest that the MEBT possess antidiabetic activity and is able to ameliorate biochemical damages in STZ induced diabetic rats and the results were found to be in a dose dependent manner.

Liver Dysfunction and Oxidative Stress in Streptozotocin-Induced Diabetic Rats: Protective Role of Artemisia Turanica

  • Objectives: Oxidative stress plays a central role in diabetes-induced complications. In the present study, the protevtive effect of Artemisia turanica (A. turanica) was evaluated against diabetes-induced liver oxidative stress and dysfunction. Methods: Fifty male Wistar rats were randomly divided into five groups: control, diabetic, diabetic + metformin, diabetic + A. turanica extract, and diabetic + A. turanica extract + metformin. Experimental diabetes was induced by a single-dose (55 mg/kg, intraperitoneally (ip)) injection of streptozotocin (STZ). Metformin (300 mg/kg) and A. turanica extract (70 mg/kg) were orally administrated three days after STZ injection for four weeks. The levels of malondialdehyde (MDA), total thiol content and superoxide dismutase (SOD) and catalase activities were measured in the liver tissue. Serum glucose concentration, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities were also determined. Results: In the diabetic group, serum glucose concentration, serum AST and ALT activities and liver MDA level were significantly higher while tissue total thiol content as well as catalase and SOD activities were lower, compared to the control group. Serum glucose in diabetic rats treated with metformin + A. turanica extract showed a significant decrease compared with the diabetic group. In all the A. turanica extract and metformin treated groups, serum ALT, tissue MDA level, total thiol content and SOD activity significantly improved compared with the diabetic rats. However, treatment of the diabetic rats only with metformin could not significantly change the activities of catalase and AST compared with the diabetic group. Conclusion: These findings suggested that A. turanica extract had a therapeutic effect on liver dysfuncyion and oxidative stress induced by diabetes, that may be probably due to its antioxidant and antiinflammatory effects.

생진양혈탕가미방과 매괴화가 Streptozotocin으로 유발된 고혈당 생쥐의 췌장에 미치는 영향 (Effects of Saengjinyanghyutang-gamibang and Rosa rugosa on the Pancreas in the Streptozotocin Treated Mice for Induction of Diabetes)

  • Objectives : This study has been carried out to investigate the effects of the Saengjinyanghyutang-gamibang and Rosa rugosa extracts on pancreas of the hyperglycemia mice induced with streptozotocin. Methods : We examined immunohistochemistry for insulin and COX-2, ultrastructural changes of acini by electron microscope, and changes of the blood glucose and BUN levels. Results : The ${\beta}-cells$ on Langerhnan's islet were destructed by administration of streptozotocin, so that few insulin-positive cells were observed in the control group. However, a lot of insulin-positive cells were observed in the experimental groups. These cells had recovered from the damage. As a result of COX-2 immunostain, COX-2 expression were highest in the control group other than the Saengjinyanghyutang-gamibang and Rosa rugosa extracts administered groups. As the electron microscopical observation, the centroacinar cells and acini of pancreas were destructed or damaged by administration of streptozotocin in the control group, but these recovered !Tom the damage in the other experimental groups. The levels of serum glucose were decreased remarkably on the Rosa rugosa and Saengjinyanghyutang-gamibang extracts administered groups compared with control group. Conclusions : These results suggest that administration of the Rosa rugosa and Saengjinyanghyultang-gamibang extracts to the mice reduced the damage induced by streptozotocin.

Streptozotocin유발 당뇨쥐에서 세사민과 세사몰린의 항산화효과 (Antioxidant Effects of Sesamin and Sesamolin in Streptozotocin-induced Diabetes Mellitus Rat)

  • 산화적 스트레스는 당뇨병과 관련된 맥관계병변의 전개에 아주 중요한 역할을 한다고 알려져 있다. 따라서 본 실험에서는 streptozotocin(STZ)투여에 의한 당뇨유발쥐를 사용하여 참께함유 항산화성분인 sesamin과 sesamolin의 혈당과 생체내 항산화 방어효소에 미치는 효과에 대하여 알아보고자, 당뇨유발쥐에 0.2% sesamin또는 0.2% sesamolin 함유사료를 3주간 급여하여, 혈당농도, 뇨당농도, 혈청 GOT와 GPT활성, 혈청 및 간조직의 지질과산화물,그리고 GSH함량과 GST활성에 미치는 영향을 조사하였다. STZ투여군의 혈당량은 319.4 $\pm$105.74mg/dL였는데, sesamin급여군은 146.56$\pm$23.99mg/dL, sesamolin급여군은170.74$\pm$105.94mg/dL로 유의하게 감소하였다. 당뇨병쥐의 뇨당농도가 21.16$\pm$13.72 mg/dL였는데, 세사민과 세사몰린 급여시에는 19.52 $\pm$ 8.99 mg/day와 10.93 $\pm$ 7.79 mg/day로 유의하게 감소하였다. STZ투여에 의하여 증가된 GOT와 GPT활성도는 세사민과 세사몰린의 급여에 의하여 GPT활성도만이 유의하게 감소되었다. STZ투여에 의해 증가한 간조직 중의 지질과산화물 생성은 세사민과 세사몰린 급여에 의해 유의하게 감소하였다. STZ투여에 의해 감소된 신장의 GSH함량과 GST활성은 세사민과 세사몰린에 의해 회복되었다. 이상의 결과 세사민과 세사몰린이 신장조직에서 일차적으로 산화된 산화물을 포집하고 GSH를 이용하여 체내의 독성 물질을 전이 분해시키는 작용을 하여 STZ에 의해 유도된 각종 독성을 무독화시켜 신장의 손상을 보호하여 당뇨억제 작용과 함께 항산화 효과를 나타낸 것으로 생각된다.

둥굴레(Polygonatum odoratum)분획물과 비타민 E 투여가 Streptozotocin 유발 당뇨 흰쥐의 혈당과 지질과산화에 미치는 영향 (The Effect of Butanol Fraction of Polygonatum odoratum with Vitamin E on Blood Glucose Levels and Lipid Peroxidations in Streptozotocin-Induced Diabetic Rats)

  • The hypoglycemic effects of butanol(BuOH) fraction of Polygonatum odoratum with vitamin E in streptozotocin(STZ)-induced diabetic rats were investigated. Sprague-Dawley rats weighing 200-230g were devided into five groups, and four groups induced diabetes mellitus by the STZ injection(45mg/kg b.w.) into the tail vein : Normal, diabetic-control, and three diabetic experimental groups(p. odoratum group, P. od-vit. E group and Vit. E group). All groups were fed on a AIN-76 diet, and the experimental groups were orally administered with the BuOH fraction of Polygonatum odoratum(500mg/kg b.w.) and vitamin E(10mg/kg b.w.) for 14 days. The body weight, diet intake and organ weights were monitored. The plasma levels of glucose, insulin, cholesterol, triglyceride, HDL-cholesterol and aspartate and alanine aminotransferase activities were analyzed. The levels of glycogen in liver and muscle, cholesterol in liver were determined. The malondialdehyde(MDA) levels in liver, kidney and lung were assayed. The body weight loss was seen in P. odoratum group, P. od-vit. E group, Vit. E group and diabetic control group, while the loss in P. odoratum group was much less than that in the diabetic control group. The plasma glucose levels were significantly lowered in P. odoratum group compared to diabetic control group. The plasma insulin levels were noticeably higher in P. odoratum and Vit. E groups. The rats in P. odoratum and P. od-vit. E group showed higher liver glycogen levels than in the diabetic control group. The MDA levels in liver, kidney and lung were also significantly reduced in P. od-vit. E and Vit. E groups compared to the diabetic control group. The results suggest that the administration of BuOH fraction of Polygonatum odoratum along with vitamin E reduced blood glucose levels and peroxidative tissue damage in STZ-induced diabetic rats, showing the possibility of preventive and therapeutic use of the wild edible plant to the diabetes mellitus. 방사선조사가 당뇨 백서의 측두하악관절에 미치는 영향 (Effect of irradiation on the temporomandibular joint in streptozotocin-induced diabetic rat)

  • Purpose: To investigate the histopathological changes in the temporomandibular joint in streptozotocin-induced diabetic rat following irradiation. Materials and Methods : Sprague-Dawley rats weighing about 250 gm were divided into three groups: control, diabetic, and diabetic-irradiated groups. Diabetes mellitus was induced in the rats by injecting streptozotocin. Rats in the control group were injected with citrate buffer only. After 5 days, the head and neck region of the rats in diabetic-irradiated group were irradiated with single absorbed dose of 10 Gy. The rats were killed at 1, 3, 7, 14, 21, and 28 days after irradiation. The specimen including the temporomandibular joint were sectioned and observed using a histopathological method. Results : In the diabetic group, severe bone resorption in the mandibular condyle was observed throughout the period of experiment. Necrosis of bone marrow and trabeculae was observed at 28 days after diabetic state. Atrophy and fibrosis in the retrodisca] tissue was gradually progressed during the time of the experiment. In the diabetic-irradiated group, severe bone resorption in the mandibular condyle was observed during the early experimental phases, but regeneration of bone marrow was initiated at ]4 days after diabetic state and irradiation. A]so, calcification of abnormal trabeculae was observed at 28 days after diabetic state and irradiation. The retrodisca] tissue was degenerated in the early experimental phases, but it had been gradually regenerated during the experimental time. Conclusion: This experiment suggests that bone resorption and degeneration in the mandibular condyle are caused by the induction of diabetes, and abnormal bone formation is induced after irradiation in diabetic state.
