항암활성을 지닌 Streptococcus pyogenes의 적정 살균조건에 관한 연구 (제 1보) 살균조건에 미치는 열 처리의 영향 (Studies on the Optimal Conditions of Sterilization for Streptococcus pyogenes (Part 1) Effect of Heat Treatment on Typical Death Rate)

  • 성홍열, 단독, 패혈증의 원인균인 Streptococcus pyogenes중에서 항암효과가 좋은 균주를 선정하여 항암제 생산을 위한 기초실험으로서 열 처리에 따른 적정 살균조건을 검토하여 다음과 같은 결과를 얻었다. Streptococcus pyogenes의 생육 최적온도는 37$^{\circ}C$이었고 대수기에서 평균 세대시간은 20분이었으며, 5$0^{\circ}C$ 이상으로 열 처리하면 생존균이 감소하였다. 균 현탁액을 pH6, 7, 8, 9로 조절한 후, 5$0^{\circ}C$에서 열 처리하였을 때는 대수적으로 사멸하였고, 사멸속도상수 k값은 각각 0.1448, 0.1194, 0.1273, 0.1707(min.$^{-1}$)이었으나 60, 70, 80, 9$0^{\circ}C$로 열 처리하면 10분만에 99%이상이 사멸되었으며 10$0^{\circ}C$에서는 완전히 사멸되었다.

Streptococcus sp. JEJ-6에 의한 가용성 전분으로부터 L-Lactic Acid 생성에 관한 연구 (Study on the Production of L-Latic Acid from Soluble Starch by Streptococcus sp. JEJ-6)

  • The strain producing L-lactic acid from starch was isolated from kimchi. The isolated strain was identified as a homofermentative Streptococcus sp. through its morphological, cultural, biochemical characteristics, and named Streptococcus sp. JEJ-6. Lactic acids are of two types, one L-specific and the other D-specific form in a stereospecific form. Streptococcus sp. JEJ-6 produced selectively L-lactic acid from all of the tested carbon sources. The optimum conditions for the L-lactic acid production from the isolated microorganism were determined. For the maximum yield of L-lactic acid from Streptococcus sp. JEJ-6, the cell should be harvested at the early stationary phase, and the growth temperature, pH, and NaCl concentration should be 37$^{\circ}C$, pH 7.0 and 0.1%, respectively. 4% Soluble starch as substrate and organic nitrogen sources such as peptone and yeast extract should be used for the best yield. The optimum pH of the nicotinamide adenine dinucleotide(NAD)-dependent and NAD-independent lactate dehydrogenase(LDH) activities was pH 8.5 and pH 7.0, respectively.

Bactericidal Effect of the Aos Denti Germ for Denture Cleansing Effervescent Tablet against Oral Microorganisms

  • Human mouth environment is known to include a variety bacteria, including Streptococcus spp., Staphylococcus spp., Actinomyces spp., Lactobacillus spp., Candida spp., Enterobacteriaceae, et al. Human oral microorganisms can cause dental caries, gingivitis, periodontitis, respiratory tract infection, and cardiovascular disease. Thus, right denture cleaning is essential to oral and general human health. The aim of this study was to evaluate the bactericidal effect of a sodium dichloroisocyanurate-based effervescent tablet (Aos Denti Germ, Aos Company, Chungbuk, Korea) against oral microorganisms. A total of 5 species Streptococcus spp. (Streptococcus anginosus, Streptococcus mitis, Streptococcus mutans, Streptococcus oralis, and Streptococcus sobrinus), Actinomyces oris, Candida albicans, and Escherichia coli were used in this study. All strains were exposed to the distilled water prepared with effervescent tablet. After the exposure, the mixture of strains and effervescent tablet was inoculated onto blood agar or MacConkey agar plate and cultured at $36^{\circ}C$. All strains were killed immediately on exposure to effervescent tablet. The results suggested that effervescent tablet could be used as an effective denture cleanser for dental hygiene.

Streptococcus의 유당분해에 대한 자일리톨의 효과 (THE EFFECT OF XYLITOL ON THE LACTOSE FERMENTATION OF STREPTOCOCCUS)

  • 자일리톨은 탄소 5개가 있는 탄수화물로 치아우식증을 억제할 목적으로 사용되는 자당 대체물이다. 본 논문은 구강 중요 세균인 Streptococcus mutans, Streptococcus oralis, Streptococcus Salivarius에서 유당분해에 대한 자일리톨의 작용을 관찰하고자 하였다. 세균에 대한 유당과 자일리톨의 병합효과를 보기 위하여 분광광도계를 이용한 흡광도를 측정하였고 생균수 검사를 실시하였다. Streptococcus mutans 또는 ${\beta}-galactosidase$에 의한 유당분해에 대한 자일리톨 효과를 보기 위하여 thin layer chromatography와 lactose-PTS activity test를 실시하여 다음과 같은 결과를 얻었다. 1. 유당과 자일리톨을 병합 첨가한 배지에 Streptococcus mutans를 배양하면 배양액 흡광도가 배양 8시간에 증가하지 않다가 배양 24시간에 증가하였다. 배양 8시간에서의 생균수도 유당 단독보다 유당과 자일리톨 병합 첨가 시 적게 나타났다. 2. 유당과 자일리톨을 병합 첨가한 배지에 Streptococcus oralis와 Streptococcus salivarius를 배양하면 배양액 흡광도가 배양 8시간에 증가하지 않다가 배양 24시간에 증가하였다. 3. 배지에 유당 단독으로 첨가할 때보다도 유당과 자일리톨을 병합 첨가한 배지에 Streptococcus mutars를 8시간 배양하면 thin-layer chromatography상 남아있는 유당이 많았으나, 배양 24시간에는 모든 유당이 분해되었다. 4. 배지에 유당 단독으로 첨가할 때보다도 유당과 자일리톨을 병합 첨가한 배지에서 Streptococcus mutars의 lactose-PTS 활성도가 낮았다. 5. 배지에 유당 단독으로 첨가할 때보다도 유당과 자일리톨을 병합 첨가한 배지에 ${\beta}-galactosidase$ 작용시 thin-layer chromatography상 유당이 많았다. 이상의 결과는 자일리톨이 Streptococcus의 유당 이용을 억제함을 시사하였다.

Antimicrobial activity of Medicinal Plant Extracts against Streptococcus mutans

  • This study was carried out to research antimicrobial agents from medicinal plants, Lonicera japonica, Pinellia ternata, Dictamnus albus, Cryptotympana pustulata, Pinus densiflora, Bupleurum falcatum, Forsythia saxatilis, Castanea crenata, Hovenia dulcis, Prunus sargentii. The ethanol extracts of 10 medicinal plants were tested for the antimicrobial activity against Streptococcus mutans. The extracts of Pinus densiflora showed significant antimicrobial activity against Streptococcus mutans. These results suggested that the extract from Pinus densiflora could be a candidate for new antimicrobial agents against Streptococcus mutans.

  • For the purpose of developing of anticariogenic agents, P-phenylphenol derivatives were synthesized and determined their antibacterial activities against a cariogenic bacterium, Streptococcus mutans. Among synthetic compounds, 2-nitro-6-bromo-p-phenylphenol showed as potent antibacterial activity as magnolol and honokiol.

  • One bacterial strain, that had made the largest inhibition zone at the antagonism assay and also that lost the inhibition activity by the protease treatment, was isolated from raw milk. That strain was identified as Streptococcus salivarius subsp. thermophilus. The specific growht rate of this strain was maximum at 45$\circ $C. However, at this temperature the strain produced no bacteriocin. The bacteriocin activity was quite stable even at high temperature. Moreover, the activity of the vacteriocin was sensitive to proteases. but not to $\alpha $-amylase, DNase I, or RNase.

  • Streptococcus mutans는 구강 내에 상존하는 치아우식증의 주요 원인균으로서 치면의 피막에 부착 후 glucan을 형성하여 세균의 군락을 이루며, 외부로부터 공급된 자당대사를 통하여 유기산을 생성함으로써 법랑질을 탈회시킨다. 치아우식 활성도의 평가를 위한 단클론항체를 이용한 방법은 진료실에서 빠른 시간 내에 간편하게 타액에 존재하는 Streptococcus mutans의 정량분석이 가능한 방법이다. 이 연구는 3세에서 6세 사이의 어린이 15명을 대상으로 자극성 타액을 채취하여 시판 중인 단클론항체를 이용한 Salivacheck$^{TM}$ Mutans, strip을 이용한 Dentocult$^{(R)}$-SM 그리고 MSB배지 배양법으로서 타액 내 Streptococcus mutans를 측정한 후 그 값을 우식경험치아수와 비교하여 상관관계를 알아보았다. Saliva-check$^{TM}$ Mutans를 이용한 방법은 Dentocult$^{(R)}$-SM과 MSB배지법과 통계학적으로 유의한 상관관계를 보였으나(p<0.05), MSB배지법은 어린이의 우식경험치아수와 통계학적으로 유의한 결과를 나타내지 않았다(p=0.34).

  • Statement of problem: Streptococcus produces energy and forms extracellular polysaccharides by metabolizing sucrose. Insoluble glucan, a kind of extracellular polysaccharide, is the important material of dental plaque. Fructose affects the metabolism of sucrose. Purpose: The purpose of this study was to evaluate the effect of fructose on the metabolism of sucrose in Streptococcus mutans. Materials and methods: To determine the effect of fructose on the formation of artificial plaque by Streptococcus mutans Ingbritt, S. mutans and fructose were placed in beakers containing M17 broth and sucrose. The wires were hung on frameworks inserted into cork stoppers, and then immersed in each of the beakers. After the incubation with gentle shaking, each wire was weighed. To analyze the effect of fructose on the sucrose metabolism by S. mutans or glucosyltransferase, S. mutans and fructose were placed in M17 broth containing sucrose. After the incubation. the remaining sucrose and polymers were analysed by thin layer chromatography. Results: The following results were obtained; 1. When Streptococcus mutans was cultured in the media containing 3% sucrose for 8 hours, the mean weight of formed artificial plaque on the wires was $124.3{\pm}3.0mg$, whereas being reduced to $20.7{\pm}10.2mg$ in the media added with 3% sucrose and 4% fructose(p<0.05) 2. When the control containing glucose was added with sucrose, the optical density of Streptococcus mutans solution cultured for 24 hours was not increased compared with the control, while being increased by adding with fructose. 3. When Streptococcus mutans was incubated in the media added with sucrose and fructose for 8 hours, the number of viable cells was increased compared with the media added with sucrose. 4. The amount of remained sucrose was increased in Streptococcus mutans culture supernatant of media added with sucrose and fructose than with sucrose only. but the amount of produced insoluble glucan was decreased. 5. The amounts of remained sucrose and produced soluble glucan were increased in the culture of glucosyltransferase-contained media added with sucrose and fructose than with sucrose only, but the amount of produced insoluble glucan was decreased . Conclusion: These results indicated that the sucrose metabolism and the production of insoluble glucan were inhibited in Streptococcus mutans by adding fructose in the media containing sucrose.

섬오가피로부터 분리된 acanthoic acid의 항치아우식효과 (Anticariogenic activity of acanthoic acid isolated from Acanthopanax koreanum $N_{AKAI}$ against oral pathogens)

  • 치면세균막내의 미생물 중 Streptococcus mutans $N_{AKAI}$는 치면세균막형성에 가장 중요한 역할을 하는 원인균으로 알려져 있으며, Streptococcus mutans $N_{AKAI}$는 치아우식을 일으키는 주요 원인균으로 주목을 받고 있다. Streptococcus mutans $N_{AKAI}$균에 항균효과를 나타내는 화합물 acanthoic acid (AA, $C_{20}H_{30}O_2$)을 Acanthopanax koreanum $N_{AKAI}$의 클로로포롬 추출물에서 각종 컬럼크로마토그래피법으로 추출분리하여 ms, 1H-NMR 그리고 13C-NMR 등을 이용하여 구조를 동정하였다. AA의 Streptococcus mutans $N_{AKAI}$에 대한 최소억제농도(MIC)와 최소살균농도(MBC)를 조사하였으며, 이 때 각각 2와 $4\:{\mu}g/mL$을 나타냈었다. 또한 다른 천연물에서 분리된 화합물인 tanshinone IIA는 8/16\;${\mu}g/mL$, 녹차의 추출물과 eucalyptoi은 $250/500\;{\mu}g/mL$이었다. 그리고 또 다른 치주질환의 원균인 Streptococcus sobrinus $N_{AKAI}$, Streptococcus sanguis $N_{AKAI}$ 및 Streptococcus gordonii $N_{AKAI}$의 최소억제 농도의 범위는 $4{\sim}32\;{\mu}g/mL$이었다. 우리의 연구결과에서 Acanthopanax koreanum $N_{AKAI}$에서 분리한 acanthoic acid 치주질환 원인균들을 억제하는 항균제로의 가능성을 확인하였다.
