Shell Finite Element for Nonlinear Analysis of Reinforced Concrete Containment Building (철근콘크리트 격납건물의 비선형 해석을 위한 쉘 유한요소)

  • It is absolutely essential that safety assessment of the containment buildings during service life because containment buildings are last barrier to protect radioactive substance due to the accidents. Therefore, this study describes an enhanced degenerated shell finite element(FE) which has been developed for nonlinear FE analysis of reinforced concrete(RC) containment buildings with elasto-plastic material model. For the purpose of the material nonlinear analysis, Drucker-Prager failure criteria is adapted in compression region and material parameters which determine the shape of the failure envelop are derived from biaxial stress tests. Reissner-Mindlin(RM) assumptions are adopted to develop the degenerated shell FE so that transverse shear deformation effects is considered. However, it is found that there are serious defects such as locking phenomena in RM degenerated shell FE since the stiffness matrix has been overestimated in some situations. Therefore, shell formulation is provided in this paper with emphasis on the terms related to the stiffness matrix based on assumed strain method. Finally, the performance of the present shell element to analysis RC containment buildings is tested and demonstrated with several numerical examples. From the numerical tests, the present results show a good agreement with experimental data or other numerical results.

Study on Production of Poly-γ-Glutamic Acid by Bacillus subtilis CH-10 (Bacillus subtilis CH-10에 의한 폴리감마글루탐산의 생산에 관한 연구)

  • A bacterium that produced a large amount of poly-$\gamma$-glutamate (PGA) was isolated from the compost and designated as Bacillus subtilis CH-10. The optimum temperature and pH for PGA production were at $37^{\circ}C$ and 7.5, respectively. The maximum amount of PGA production (18.84 mg/ml) was obtained when it was grown in a medium containing 3% L-glutamate and 5% sucrose at $37^{\circ}C$ with shaking. The result that the L-glutamate significantly induced PGA production indicates that it produces a PGA by the glutamate dependent manner. Some properties of the PGA obtained at different times of cultivation were investigated by SDS-PAGE and ninhydrin analysis. The PGA production was elongated along with cultivation time and maximum amount was achieved at 96 h. Average molecular weight of PGA was estimated to be 1100 kDa by FDNB method.

Genetic Variation in Mutants Induced by Gamma Ray in Hypsizigus marmoreus (느티만가닥버섯에서 감마선에 의한 돌연변이체들의 유전적 변이)

  • This research was carried out to evaluate whether gamma ray is a useful tool for breeding new strains of mushrooms. For this research, 5 mutant groups, 20 strains of Hypsizigus marmoreus, 2 strains of Lyophyllum decastes, and 1 strain of Lyophyllum shimeji were used. Monokaryon spores from one variety of H. marmoreus were irradiated with 50~2,000 Gy of gamma ray. The propriety dose was 50~200 Gy for mutagenesis. Mutant monokaryon mycelia crossed each order to become dikaryon mycelia. The internal transcribed spacer (ITS) regions of rDNA were amplified using PCR, and the products were sequenced. The sequences of the ITS regions (16 partial rDNA, complete ITS1, 5.8 rDNA and partial rDNA) were analyzed by PCR, and strains of H. marmoreus, L. decastes, and L. shimeji were auto-sequenced. The lengths of the sequenced ITSs were 1,052~1,143 nucleotides. Genetic matrices were calculated using Nei-Li's genetic distance coefficient based on ITS sequence. The dissimilarities were 0~3.35% in strains of H. Hypsizigus. In addition, a phylogenetic tree was constructed based on ITS sequences using the neighbor-joining (NJ) method. The phylogenetic tree revealed that 23 strains and 5 mutant groups were divided into 12 clusters; the mutant groups fell into different clusters. These results show that mushroom spores were mutated effectively by gamma ray; therefore, gamma ray could be a useful tool for breeding new strains of mushrooms.

  • The transfer of genetic material into pea tissue was accomplished by using an avirulent strain of Agrobacterium tumefaciens containing the binary vector. The method used for transformation requires non-tissue culture steps as it involves the inoculation of the site of the shoot removed of germinating seeds. The identification of ${\beta}$-glucuronidase activity in the tissues of $T_0$ pea plants indicates that the plant expressible ${\beta}$-glucuronidase gene, contained the T-DNA region from pLPBO2, had been transferred at least into somatic tissues. Putative transformed $T_0$ pea plants were advanced to produce $T_1$ plants which were also assayed for the presence of the transferred ${\beta}$-glucuronidase gene. The presence of the ${\beta}$-glucuronidase gene in DNAs isolated from $T_1$ plant was demonstrated by DNA gel blot hybridization. This analysis revealed that the transformed plants contained ${\beta}$-glucuronidase gene.

An Estimation on Two Stroke Low Speed Diesel Engines' Shaft Fatigue Strength due to Torsional Vibrations in Time Domain (시간영역에서 과도 비틀림 진동에 의한 저속 2행정 디젤엔진의 축계 피로강도 평가)

  • Two stroke low speed diesel engines are widely used for marine propulsion or as power plant prime mover. These engines have many merits which includes higher thermal efficiency, mobility and durability. Yet various annoying vibrations occur sometimes in ships or at the plant itself. Of these vibrations, torsional vibration is very important and dictates a careful investigation during the engme's initial design stage for safe operation. With the rule and limit on torsional vibration in place, shaft strength fatigue due to torsional vibration however demands further analysis which possibly can be incorporated in the classification societies' rule and limit. In addition, the shaft's torsional vibration stresses can be calculated equivalently from accumulated fatigue cycles number due to transient torsional vibration in time domain. In this paper, authors suggest a new estimation method combined with Palmgren-Miner equation. A 6S70MC-C ($25,320ps{\times}91rpm$) engine for ship propulsion was selected as a case study. Angular velocity was measured, instead of shaft's strain, for simplified measurement and it was converted to torsional vibration stress for accumulated fatigue cycle numbers in shafting life time. Likewise, the accumulated fatigue calculation was compared with shaft fatigue strength limit. This new method can be further realized and confirmed in ship with two stroke low speed diesel engine.

Finite element model updating of long-span cable-stayed bridge by Kriging surrogate model

  • In the finite element modelling of long-span cable-stayed bridges, there are a lot of uncertainties brought about by the complex structural configuration, material behaviour, boundary conditions, structural connections, etc. In order to reduce the discrepancies between the theoretical finite element model and the actual static and dynamic behaviour, updating is indispensable after establishment of the finite element model to provide a reliable baseline version for further analysis. Traditional sensitivity-based updating methods cannot support updating based on static and dynamic measurement data at the same time. The finite element model is required in every optimization iteration which limits the efficiency greatly. A convenient but accurate Kriging surrogate model for updating of the finite element model of cable-stayed bridge is proposed. First, a simple cable-stayed bridge is used to verify the method and the updating results of Kriging model are compared with those using the response surface model. Results show that Kriging model has higher accuracy than the response surface model. Then the method is utilized to update the model of a long-span cable-stayed bridge in Hong Kong. The natural frequencies are extracted using various methods from the ambient data collected by the Wind and Structural Health Monitoring System installed on the bridge. The maximum deflection records at two specific locations in the load test form the updating objective function. Finally, the fatigue lives of the structure at two cross sections are calculated with the finite element models before and after updating considering the mean stress effect. Results are compared with those calculated from the strain gauge data for verification.

Lichen-Associated Bacterium, a Novel Bioresource of Polyhydroxyalkanoate (PHA) Production and Simultaneous Degradation of Naphthalene and Anthracene

  • Lichens are generally known as self-sufficient, symbiotic life-forms between fungi and algae/cyanobacteria, and they also provide shelter for a wide range of beneficial bacteria. Currently, bacterial-derived biodegradable polyhydroxyalkanoate (PHA) is grabbing the attention of many researchers as a promising alternative to non-degradable plastics. This study was conducted to develop a new method of PHA production using unexplored lichen-associated bacteria, which can simultaneously degrade two ubiquitous industrial toxins, anthracene and naphthalene. Here, 49 lichen-associated bacteria were isolated and tested for PHA synthesis. During the GC-MS analysis, a potential strain of EL19 was found to be a 3-hydroxyhexanoate (3-HHx) accumulator and identified as Pseudomonas sp. based on the 16S rRNA sequencing. GC analysis revealed that EL19 was capable of accumulating 30.62% and 19.63% of 3-HHx from naphthalene and anthracene, respectively, resulting in significant degradation of 98% and 96% of naphthalene and anthracene, respectively, within seven days. Moreover, the highly expressed phaC gene verified the genetic basis of $PHA_{mcl}$ production under nitrogen starvation conditions. Thus, this study strongly supports the hypothesis that lichen-associated bacteria can detoxify naphthalene and anthracene, store energy for extreme conditions, and probably help the associated lichen to live in extreme conditions. So far, this is the first investigation of lichen-associated bacteria that might utilize harmful toxins as feasible supplements and convert anthracene and naphthalene into eco-friendly 3-HHx. Implementation of the developed method would reduce the production cost of $PHA_{mcl}$ while removing harmful waste products from the environment.

Automated Finite Element Mesh Generation for Integrated Structural Systems (통합 구조 시스템의 유한요소망 형성의 자동화)

  • The structural analysis module is an essential part of any integrated structural system. Diverse integrated systems today require, from the analysis module, efficient real-time responses to real-time input such as earthquake signals, extreme weather-related forces, and man-made accidents. An integrated system may also be for the entire life span of a civil structure conceived during the initial conception, developed throughout various design stages, effectively used in construction, and utilized during usage and maintenance. All these integrated systems' essential part is the structural analysis module, which must be automated and computationally efficient so that responses may be almost immediate. The finite element method is often used for structural analysis, and for automation, many effective finite element meshes must be automatically generated for a given analysis. A computationally efficient finite element mesh generation scheme based on the r-h method of mesh refinement using strain deviations from the values at the Gauss points as error estimates from the previous mesh is described. Shape factors are used to sort out overly distorted elements. A standard cantilever beam analyzed by four-node plane stress elements is used as an example to show the effectiveness of the automated algorithm for a time-domain dynamic analysis. Although recent developments in computer hardware and software have made many new applications in integrated structural systems possible, structural analysis still needs to be executed efficiently in real-time. The algorithm applies to diverse integrated systems, including nonlinear analyses and general dynamic problems in earthquake engineering.

Evaluation of Oxalic Acid Pretreatment Condition Using Response Surface Method for Producing Bio-ethanol from Yellow Poplar (Liriodendron tulipifera) by Simultaneous Saccharification and Fermentation (바이오에탄올 생산을 위한 백합나무(Liriodendron tulipifera)칩의 동시당화발효 및 Response Surface Method를 이용한 옥살산 전처리 조건 탐색)

  • The main purpose of this study is to evaluate the potential of producing bioethanol from yellow poplar ($Liriodendron$ $tulipifera$) wood chips by oxalic acid pretreatment and to examine the pretreatment conditions by response surface methodology (RSM). Based on $2^3$ factorial design, adjusted variables were reaction temperature ($^{\circ}C$), residence time (min), and acid loading (g/g), and a series of distinct 15 experimental conditions was organized with duplication at central point (total 16 performances). After pretreatment, simultaneous saccharification and fermentation (SSF) was subjected on solid fraction with yeast strain $Pichia$ $stipitis$. Maximum ethanol yields of the most samples were measured at 72 hours and applied to RSM as a dependent variable. 9.7 g/${\ell}$ of ethanol was produced from the solid pretreated at $180^{\circ}C$ for 40 min with 0.013 g/g of oxalic acid loading. According to the response surface methodology, it was determined that the temperature is the most governing factor via statistic analysis.

Antibiotic sensitivity of the bacterial strains isolated from operating wounds (수술창에서 분리된 균주의 항생제 감수성)

  • Staphylococuus aureus and Escherichia coli is increasingly responsible for outbreaks of nosocomial infection around the world. Because serious infections due to these organisms currently necessiate use of non-$\beta$-lactam antimicrobial therapy and because strains is ofen resistant to many antimicrobial agents, infections with this organism are difficult to treat. Isolated strains from post operaton wounds of PNU hospital patient were tested for the antimicrobial susceptibility, resistant pattern and combined action to the 6 antibiotics. The minimal inhibitory concentraction of each antibiotic anc antibiotics combining in various ratios were measured by checkerboard dilution method. the synergism was determined through calculating the fractional inhibitory concentraction index (FICI). In case of S. aureus, 15 strains was shown to be highly sensitive to streptomycin and 13 strains to cephalothin. In case of E. coli, it is excellent senstitive 16 strains, sensitive 4 strains on cefoperazone, as like S. aureus, and thus the sensitive is most to be 66%. As the result of gaining MIC from S. aureus upon agar dilution method, MIC$_{50}$ was 8$\mu$g/ml, MIC$_{90}$ was 16$\mu$g/ml and thus the streptomycine is shown to be lowest. In case of E, coli, S. MIC$_{50}$ was 4$\mu$g/ml, MIC$_{90}$ was 16$\mu$g/ml, in streptomycin and thus is shown to be lower than S. aureus. As the result of comparing the resistance aspect of combining the antibiotics on S. aureus and E. coli, the resistant strain can be known to be reduced to the large range more than each 40% than combining with only aminoglycoside-series or cephalosporine-series. As the result of combining aminoglycoside-series, streptomycin and cephalothin or cefuroxime sensitive to S. aureus and E. coli in the above mentioned results, the increase or imporovement of effect is over 73% and 80%, respectively, thus the case od combining 2 antibiotics is shown to be better in the effect. Isolated strains from operating wounds were for the antimicrobial susceptibility. In case of S. aureus 15 strains was shown to be sensitive very much on streptomycin. In case of E. coli it is excellent sensitive 16 strains. As the results of combining aminoglycosides-series, streptomycin and cephalosporine series, cephalothin and cefuroxime, the increase or improvement of effect is over 73%, thus case of combining 2 antibiotics is shown to be better in the effect.
