A Study of the Effect of Monosodium Glutamate on the Development of Drosophila melanogaster (Monosodium Glutamate(MSG)가 초파리(Drosophila melanogaster))

  • Monosodium glutamate(MSG) is a widely used food additive. Some reports descried its positive effect and the others, negative effect on mouse, monkey, human or drosophilid flies. Because of the conflicting reports the present investigation was undertaken to study the effects of MSG on the development of Drosophila melanogaster. The two strains of D. melanogaster, Oregon-R and Sinchon-I were used and MSG as well as cane sugar (as the second control) media were prepared by adding MSG or cane sugar at various concentrations to the standard food media for the present study. Ten flies (Male 5, Female 5) were placed in each vial and the numbers of $F_1$ flies emerged from it were counted. The results are presented below: 1. The numbers of $F_1$ flies decrease as the concentrations of MSG increase, implying that MSG has an inhibitory effect in the development of D. melanogaster. 2. The effects of cane sugar show an enhancing effect rather than an inhibitory one. 3. The numbers of $F_1$ fies produced in the Sinchon-I strain are greater than in the Oregon-R. This may be due to the difference in the length of inbred period. 4. The Muller-5 test shows a negative result, suggesting that MSG may be not mutagenic.

Effect of Cordycepin-increased Cordyceps militaris Powder on Tissues Lipid Peroxidation and Antioxidative Activity in Carbon Tetrachloride-induced Hepatic Damage in Rats (Cordycepin이 사염화탄소 유발 간손상 흰쥐의 조직 과산화 지질 농도 및 항산화 활성에 미치는 영향)

  • This study aimed to evaluate the protective effect of cordycepin-increased Cordyceps militaris strain on carbon tetrachloride ($CCl_4$)-induced hepatotoxicity and oxidative stress in rats. Male Sprague-Dawley rats were randomly divided into five groups (n=6) based on six dietary categories: normal (N), $CCl_4$ control (C), $CCl_4$ plus Paecilomyces japonica (CPJ) (3%, w/w), $CCl_4$ plus C. militaris (CCM) (3%, w/w), and $CCl_4$ plus cordycepin-increased C. militaris ($CCM{\alpha}$) (3%, w/w). The activities of the liver marker enzymes ALT, AST, and LDH and the levels of lipid peroxidation were increased in the $CCl_4$-treated groups, but these parameters were significantly decreased in the $CCM{\alpha}$ group. The TBARS content in the liver homogenate, microsome, and mitochondrial fractions of the C group was significantly elevated compared with the N group. However, in the $CCl_4$-treated groups, $CCM{\alpha}$ group was significantly lowered in the TBARS levels of hepatic homogenate and microsomal fractions. The C group showed a significant decrease in the levels of plasma and hepatic glutathione, whereas they were significantly increased in the $CCM{\alpha}$ group. Accordingly, cordycepin-increased C. militaris may be an ideal animal model for studying hepatoprotective effects.

Isolation of a Phenol-degrading Bacterial Strain and Biological Treatment of Wastewater Containing Phenols (Phenol 분해균주의 분리 및 페놀함유 폐수의 생물학적 처리)

  • Aromatic hydrocarbons, such as phenol, have been detected frequently in wastewater, soil, and groundwater because of the extensive use of oil products. Bacterial strains (56 isolates) that degraded phenol were isolated from soil and industrial wastewater contaminated with hydrocarbons. GN13, which showed the best cell growth and phenol degradation, was selected for further analysis. The GN13 isolate was identified as Neisseria sp. based on the results of morphological, physiological, and biochemical taxonomic analyses and designated as Neisseria sp. GN13. The optimum temperature and pH for phenol removal of Neisseria sp. GN13 was $32^{\circ}C$ and 7.0, respectively. The highest cell growth occurred after cultivation for 30 hours in a jar fermentor using optimized medium containing 1,000 mg/l of phenol as the sole carbon source. Phenol was not detected after 27 hours of cultivation. Based on the analysis of catechol dioxygenase, it seemed that catechol was degraded through the meta- and ortho-cleavage pathway. Analysis of the biodegradation of phenol by Neisseria sp. GN13 in artificial wastewater containing phenol showed that the removal rate of phenol was 97% during incubation of 30 hours. The removal rate of total organic carbon (TOC) by Neisseria sp. GN13 and activated sludge was 83% and 78%, respectively. The COD removal rate by Neisseria sp. GN13 from petrochemical wastewater was about 1.3 times higher than that of a control containing only activated sludge.

Antifungal Activity of Bacillus Subtilis HK2 against Trichothecium Roseum Causing Pink Rot of Melon and White Stain Symptom on Grape (멜론 분홍빛썩음병과 포도 흰얼룩병의 원인균인 Trichothecium Roseum에 대한 Bacillus Subtilis HK2의 항균활성)

  • Pink Rot on melon and White Stain Symptom on grape are caused by Trichothecium roseum, one of the most important diseases of grape and melon. These diseases have been occurred in national-wide in Korea and causes irreversible damage on the grape and the melon at harvest season. This research presents the evaluation of the capacity of Bacillus subtillis HK2 to protect both melon and grape against T. reseum and establishes its role as a biocontrol agent. In this study, we isolated a Bacillus strain HK2 from rhizosphere soil, identified it as Bacillus subtillis by 16S rRNA analysis and demonstrated its antifungal activity against T. roseum. Under I-plate assay it was observed that the effect of hyphal growth inhibition was not due to production of volatile compounds. The optimum culture condition of HK2 was found at 30℃ and initial pH of 7.0. Application of HK2 culture suspension reduced 90.2% of white stain symptom on grape as compared to control, resulting in greater protection to grape against T. roseum infestation. Butanol extract of HK2 culture purified using flash column chromatography. The antifungal material was a polar substance as it showed antifungal activity in polar elute. Therefore, our results indicated a clear potential of B. subtilis HK2 to be used for biocontrol of Pink rot in melon and white stain symptom on grape caused by T. roseum.

Rapid in vivo Colonization Screening of Probiotic Bacteria Isolated from Human Infants using Caenorhabditis elegans Surrogate Host (Caenorhabditis elegans 생체대체모델을 이용한 한국 영유아분변 유래 프로바이오틱스 균주의 in vivo 장 우점능 검토)

  • The ability of probiotics to adhere to the intestinal epithelium likely plays an important role in their colonization of the gastrointestinal tract. Here, we performed high-throughput screening (HTS) for suitable characteristics of potential probiotic bacteria using attachment and colonization ability through a C. elegans surrogate in vivo model. A total of 100 strains of lactic acid bacteria (LAB) isolated from infant feces were subjected to the colonization assay using C. elegans intestine. Based on colonization ability, we showed that nine isolates have a high attachment ability during whole experimental periods (up to 168 h), compared to Lactobacillus rhamnosus strain GG as a control. Also, through the use of an in vitro cell attachment model, nine isolates revealed highly binding activity to the mucus layer. Next, the selected 9 isolates were assayed for their survival ability when exposed to acidic and bile conditions as well as cholesterol reduction and the utilization of prebiotic substrates. As a result, the isolated nine strains were determined to be highly resistant to acid and bile conditions. In addition, they have significant activity for the reduction of cholesterol and utilization of several prebiotic substrates as a carbon source. Finally, the selected nine strains were identified by either L. rhamnosus or L. plantarum (4 strains for L. rhamnosus and 5 strains for L. plantarum, respectively). Taken together, we propose that the direct colonization of probiotics using C. elegans may be applicable to the rapid screening of valuable probiotic strains in vivo.

Structural Performance of RC Slab-Wall Joints Reinforced by Welded Deformed Steel Bar Mats (철근격자망을 사용한 슬래브-벽체 접합부의 구조성능)

  • In order to clarify the structural performances of Welded Deformed Steel Bar Mats (WDSBM), the research stated includes the tests for standard hook of top bars of slab in concrete slab-wall joints, the tests for embedment length of top bar of slab, and the development strength tests for standard hook. The test results are as follows; (1) For slab-wall joints using WDSBM as reinforcement in slab, if the top bars of WDSBM are spliced by ordinary bars with sufficient development length and size, it is enough for the strength and crack control. (2) When WDSBM of slab is spliced in joint, the strength is increased with the embedment of bars of this WDSBM into wall. Beyond peak strength, however, ductility is diminished to that as no splice due to pull-out failure. (3) For slab-wall system, ultimate strain of concrete for flexural compression zone in lower surface of slab seems much greater than that of normal concrete beam. The reason is that normal concrete beam has the joint with $180^{\circ}$, however slab-wall joint has the $90^{\circ}$ of which concrete can be confined.

An Effect of Visual Acuity Protection by Natural Pigment (Anthocyanine) Extracted from Fermented Purple Sweet Potato (발효 자색고구마 추출 천연색소(anthocyanine)에 의한 시력보호 효과)

  • Purpose: The purpose of this study was to research any effect on vision protecting or decreasing VDT syndrome of extracted anthocyanine from fermented purple sweet potato and blueberry. Methods: Subjects were aged 19-20 years old who do not have ophthalmic and systemic diseases and over -N4.00 D of refraction error. 40 mg of extracted anthocyanine from fermented purple sweet potato, from blueberry, and control group, placebo were dosed at separate try. After 2 hours later, subjects were directed perform visual display terminal (VDT) work for 2 hours. Objective refractive error was measured before dosing anthocyanine and after VDT work for 2 hours. Degree of head ache, eye pain and strain and subjective symptoms of neck, shoulder and waist was also examined through interviews by dividing its degree into severe, moderate, slight or none. Results: After 2 hours VDT work, vision protection effect in terms of refractive error for dominant eye was decreased by $0.031{\pm}0.21$ D in the group of extracted anthocyanine from fermented purple sweet potato, $0.006{\pm}0.32$ D in the group of extracted anthocyanine from blueberry. However, there was significantly myopic progression in the placebo group by $0.144{\pm}0.28$ D (t=2.27, p=0.03). Conclusions: It is considered that extracted anthocyanine from fermented purple sweet potato inhibits increase of refraction anomalies of dominant eye rather than non-dominant eye after VDT work.

Antimutagenic and Anticancer Effects of Ethanol Extract from Korean Traditional Doenjang Added Sea Tangle (다시마 분말을 첨가한 전통된장 에탄을 추출물의 항돌연변이성 및 항암효과)

  • This study was carried out to investigate antimutagenic and anticancer effects of ethanolic extract of Korean traditional doenjang added sea tangle. Most of the mineral content of doeniang was increased by addition of sea tangle. In the Ames test, the antimutagenic effect of ethanol extract of Korean traditional doenjang added 5% sea tangle was higher than that of control (no additive), 10%, and 15% sea tangle additions. The inhibition rate of ethanol extract (200$\mu\textrm{g}$/plate) of doenjang added 5% sea tangle in the S. typhimurium TA100 strain showed 97.0% inhibition against the mutagenesis induced by MNNG. In addition, the suppression of ethanol extract (200$\mu\textrm{g}$/plate) of doenjang added 5% sea tangle in the S. typhimurium TA98 and TA100 strains showed 60.2% and 69.1% inhibition respectively, against the mutagenesis induced by 4NQO. The suppressions under the same condition against B($\alpha$ )P and Trp-P-1 in the TA98 and TA100 strains were 71.7% and 87.3%, and 66.6% and 80.8%, respectively. In the anticancer effects, the cytotoxicity of doenjang added 5% sea tangle on the cell lines with human lung carcinoma (A549), human hepatocellular carcinoma (HepG2), and human gastric carcinoma (KATOIII) were inhibited with increasing the extract concentration. The treatment of 1.0 mg/mL Doenjang added 5% sea tangle showed strong cytotoxicity of 56.4%, 87.67%, and 89.5% against A549, HepG2, and KATOIII, respectively.

Quality Characteristics of the Chungkookjang Fermented by the Mixed Culture of Bacillus natto and B. licheniformis (Bacillus natto와 B. licheniformis 혼합 Starter로 제초된 청국장의 품질특성)

  • The quality characteristics and sensory evaluation of chungkookjang were investigated. The samples were prepared and fermented by the inoculation of Bacillus strains; B. subtilis, B. natto and B. licheniformis as a single starter, and mixed culture of B. natto and B. licheniformis on the industrialized model system. It was shown that microbial growth, protease activity, contents of amino-and ammonia-nitrogen and contents of organic acid were higher in B. subtilis inoculated sample, and were lower in B. licheniformis inoculated one. General quality characteristics of sample inoculated by mixed culture of B. natto and B. licheniformis took a middle position between each B. natto and B. licheniformis inoculated one. Fifty eight species of odor components were identified. Ethanol, 3-methyl-1-butanol, acetic acid, benzaldehyde and alkyl pyrazines were identified in all samples and most of other flavor components were strain specific. The contents of unpleasant smell components, alkyl pyrazines and benzaldehyde, were lower in B. licheniformis inoculated sample. The sensory evaluations showed that chungkookjang manufactured from mixed culture of B. natto and B.licheniformis was most acceptable. Therefore, results indicated that chungkookjang manufactured from mixed culture of B. natto and B. licheniformis induced better sensory quality than that of the control.

The optimum media size and composition for sawdust culture of Lentinula edodes (Strain: JNM3022) (표고버섯(JNM3022) 톱밥재배에 적합한 배지조성과 배지크기)

  • These experiments were conducted to search for the optimum media size and composition for culture of Lentinula edodes (JNM3022). Considering the incubation period and the successful culturing rate, the optimum media size was length 25 $cm{\times}diameter$ 14 cm (weight 2.5 kg). The saturation moisture amount of Oak sawdust was 251 ml/100 g. It is a half of pine sawdust 495 ml/100 g. Considering the hypha growing speed and hypha density, the optimum media composition was Oak sawdust 80%+ wheat bran 20% and Oak sawdust 80%+ rice bran 20% In 10 treatments of media composition. The optimum moisture amount of media for growing Lentinula edodes (JNM3022) was 55%. Injection pipe with 20 holes in media was reduced the incubation periods to 11 days when 20cm length media and 32 days when 30cm length media compare to control. The best method for browning media surface was change temperature condition above $10^{\circ}C$ and punching the vinyl bag. This treat could shortened the browning periods to 54 days.
