Antioxidant Activities of Leaf, Stem and Root of Panax ginseng C. A. Meyer (인삼의 부위별 항산화 활성)

  • To develop a new functional material, leaf, stem and root of Panax gjnseng were analyzed in their antioxidant activities. Root and leaf of ginseng collected from 3 regions, exhibited inhibition activity as $45.2{\sim}54.3%\;and\;90.1{\sim}96.5%$ on peroxidation of low density lipoprotein and linoleic acid, respectively. Scavenging activities of stem, leaf and root of ginseng on superoxide anion radical were $35.6{\sim}76.1%,\;60.1{\sim}69.3%\;and\;-5.6{\sim}20.1%$, respectively. Total phenol contents of leaf, stem and root of ginseng were $147{\sim}200\;mg%,\;110{\sim}153\;mg%\;and\;61{\sim}86\;mg%$ respectively as tannic acid equivalent.

EFFECTS OF bFGF AND PDGF-BB ON OSTEOBLAST DIFFERENTIATION OF BONE MARROW-DERIVED MESENCHYMAL STEM CELL IN RAT (bFGF, PDGF-BB가 백서 골수기원 간엽 줄기세포의 조직골세포 분화에 미치는 영향에 관한 연구)

  • In this study we evaluate the effects of bFGF-BB and PDGF on in vitro proliferation, differentiation and mineralization of mesenchymal stem cells (MSCs) from rat. MSCs were prepared from the bone marrow of 6 or 7-week-old male rats with a technique previously described by Maniatopoulos et al. in 1988. Lineage differentiation to osteogenesis, chondrogenesis and adipogenesis were performed. At first, we characterized the cultured cell on passage 1, 3, 5, 7 with immunocytochemical staining using CD29, 44, 34, 45, ${\alpha}$-SMA and type I collagen. And to study the effects of bFGF and PDGF-BB on proliferation, differentiation and mineralization, we seeded the expanded cell at a density of 6 $6{\times}10^3\;cells/cm^2$ to 100-mm dish for evaluation of cell proliferation and MTT assay was carried out on day 2, 4, 7, 9. We also resuspended the cells with same density $(6{\times}10^3\;cells/cm^2)$ to 24 well plates for subculture. On the following day, the attached cells were exposed to 2.5ng/ml bFGF and/or 25ng/ml PDGF-BB daily during 5 days. The osteocalcin (OC) level was assessed and mineral contents were evaluated with alizarin red S staining on subculture day 2, 7, 14, 21. We identified the mesenchymal stem cell from the bone marrow derived cells of rat through their successful multi-differentiation and stable display of its phenotype. And bFGF and PDGF-BB showed the effect that inhibited osteoblastic differentiation and mineralization mildly in above concentration at in vitro culture. Aggregation of Human Eyelid Adipose-derived Stem Cells by Human Body Fluids

  • Fetal bovine serum (FBS) is the most frequently used serum for the cultivation of mammalian cells. However, since animal-derived materials might not be appropriate due to safety issues, allogeneic human serum (HS) has been used to replace FBS, particularly for the culture of human cells. While there has been a debate about the advantages of HS, its precise effect on human adult stem cells have not been clarified. The present study aimed to investigate the effect of HS on the human eyelid adipose stem cells (HEACs) in vitro. When HEACs were cultivated in a medium containing 10% HS, many cells moved into several spots and aggregated there. The phenomenon was observed as early as 9 days following 10% HS treatment, and 12 days following 5% HS plus 5% FBS treatment. However, the aggregation was never observed when the same cells were cultivated with 10% FBS or bovine serum albumin. To examine whether cell density might affect the aggregation, cells were seeded with different densities on 12-well dish. Until the beginning of aggregation, cells seeded at low densities exhibited the longest culture period of 16 days whereas cells seeded at high densities showed the shortest period of 9 days to form aggregation. The number of cells was $15.1{\pm}0.2{\times}10^4$ as the least for the low density group, and $29.3{\pm}2.8{\times}10^4$ as the greatest for the high density group. When human cord blood serum or normal bovine serum was examined for the same effect on HEACs, interestingly, cord blood serum induced the aggregation of cells whereas bovine serum treatment has never induced. When cells were cultivated with 10% HS for 9 days, they were obtained and analyzed by RT-PCR. Compared to FBS-cultivated HEACs, HS-cultivated HEACs did not express VIM, and less expressed GATA4, PALLD. On the other hand, HS-cultivated HEACs expressed MAP2 more than FBS-cultivated HEACs. In conclusion, human adult stem cells could move and form aggregates by the treatment with human body fluids.

Biomass and Net Primary Production of Pinus densiflora Stands in Gochang regions (고창지역 소나무림의 바이오매스 및 순생산량에 관한 연구)

  • This study was conducted to examine the biomass and net primary production, stem density and biomass expansion factors of Pinus densiflora in Gochang regions. The mean age of Pinus densiflora in both stands was 10 and 48 years. The dry weights (kg/tree) and aboveground biomass (Mg/ha) were 8.59 and 17.55 for 10 years young stand, 166.66 and 122.05 for 48 years old stand. The total biomass (Mg/ha) including the above and belowground were 21.48 and 154.16 in both age stands. The proportion of stem biomass, stem bark biomass and root biomass increased from the young stand to the old stand while on the leaf biomass and branch biomass, tend to decreased. The net primary production of aboveground biomass (Mg/ha) and belowground biomass were 6.30~6.95 for the young stand and 11.61~13.19 for old stand. The stem density ($g/cm^3$) was 0.338 for young stand while on the other hand, 0.448 for old stand was observed. The above and total biomass expansion factors were 2.304~2.508 and 1.318~1.644 in each age stands, respectively.

Effect of Paclobutrazol on Anti-lodging Characteristics and Yields of Rice (Paclobutrazol이 수도(水稻)의 도복형질(倒伏形質)과 수량(收量)에 미치는 영향(影響))

  • To determine anti-lodging effect of paclobutrazol and its influence on yield of rice, paclobutrazol was applied at 12 and 18g ai/10a to cv. Nakdongbyeo and cv. Seomjinbyeo 20, 15, and 10 days before heading under 12 and 20kg/10a nitrogen conditions. 1. Paclobutrazol reduced internode length of rice, especially internode N3. Higher suppression of stem of both cultivars and reduction of ear length in cv. Nakdongbyeo were observed when paclohutrazol was applied at 18g ai/10a 15 and 20 days before heading. 2. Paclobutrazol increased culm cross-section area, culm diameter, linear density and stem flexure resistance which affected anti-lodging efficacy. Although actual lodging was not occurred in field, less stem inclination was observed at paclobutrazol treated plots. 3. There were significant correlation between stem flexure resistance and culm cross-section area, culm diameter and linear density, and negative correlation between straw inclination angle and culm cross-section area and culm diameter. 4. Slight yield reduction was observed when paclobutrazol was applied at 18g ai/10a 20 days before heading. However, when paclobutrazol was applied at 12g ai/10a 10 and 15 days before heading, rice yield was not decreased compared to untreated control.

The Growth Performances of Fraxinus rhynchophylla According to Planting Density over Seven Years after Planting (식재밀도에 따른 물푸레나무 조림목 식재 후 7년간의 생장 특성)

  • This study aimed to determine the optimal planting density of Fraxinus rhynchophylla assessed from the early growth performance at various planting densities over the 7-year period after planting. The study site was in Pyeongchang County, South Korea, and seedlings of 2-year-old (bare-root seedlings) F. rhynchophylla were planted at four densities (3,000, 5,000, 7,000, and 10,000 trees ha-1) in March 2015. The survival rate, root-collar diameter (RCD), and height (H) were measured from 2015 to 2021, and the H/D (H/RCD) ratio and stem volume were calculated. The survival rate (84-97%) and H/D ratio (54.5-59.2%) were not affected by the planting density during the study period, but the RCD, H, and stem volume were significantly higher for 7,000 trees ha-1 than for other planting densities. Especially, the stem volume (cm3 tree-1) at 7 years after planting was highest for 7,000 trees ha-1 (1,356.1), followed by 10,000 trees ha-1 (958.6), 5,000 trees ha-1 (773.0), and 3,000 trees ha-1 (579.5). As the planting density increased, F. rhynchophylla seedlings showed initial rapid growth due to light competition, but relatively low growth at excessive planting densities. In the future, use of a suitable planting density considering planting costs should provide outstanding growth performance of F. rhynchophylla on plantations.

An integrated bioinformatics analysis of mouse testis protein profiles with new understanding

  • The testis is major male gonad responsible for spermatogenesis and steroidogenesis. Much knowledge is still remained to be learned about the control of these events. In this study, we performed a comprehensive bioinformatics analysis on 1,196 mouse testis proteins screened from public protein database. Integrated function and pathway analysis were performed through Database for Annotation, Visualization and Integrated Discovery (DAVID) and ingenuity Pathway Analysis (IPA), and significant features were clustered. Protein membrane organization and gene density on chromosomes were analyzed and discussed. The enriched bioinformatics analysis could provide clues and basis to the development of diagnostic markers and therapeutic targets for infertility and male contraception.

A Simple Method for Cat Bone Marrow-derived Mesenchymal Stem Cell Harvesting

  • Bone marrow (BM) cell harvesting is a crucial element in the isolation of mesenchymal stem cells (MSCs). A simple method for harvesting cat BM cells is described. The results show that a large number of BM cells can rapidly be harvested from the cat by this simple procedure. MSCs prepared by density-gradient method were spindle-shaped morphology with bipolar or polygonal cell bodies and strongly positive for CD9 and CD44 and negative for CD18 and CD45-like. They were capable of differentiation to adipocytic and osteocytic phenotypes when exposed to appropriate induction media. The advantages of this method are its rapidity, simplicity, low invasiveness, and low donor attrition and good outcome.

Effect of Physical Properties of Soil on Ginseng Seedling Growth in Nursery Bed (양식묘단 토양의 물리성이 묘삼생육 및 수량에 미치는 영향)

  • This study was conducted to elucidate the effect of physical properties of soil in nursery bed with different densities on growth of ginseng seedling. Stem length, leaf length and leaf width of ginseng seedling showed the decreasing tendency with increasing the hardness of the nursery soil. Fresh root weight per seedling and number of available seedlings were increased significantly with decrease of the soil hardness. For solid, liquid phases, bulk density and hardness of soil, negative correlations were shown in stem length, leaf length, leaf width, root weight per seeding, and number t of available seedlings. On the other hand, gas phase, air permeability and porosity of soil had positive correlations with stem length, leaf length, leaf width, root weight per seedling and number of available seedlings. Effect of Plant Density on Growth Responses and Yield in Yacon

  • This study was conducted to determine the optimal planting distance in cultivation of yacon (Polymnia sonchifolia Poeppig & Endlicher). Plug seedlings were planted with 6 different plant densities of 80 $\times$ 60, 80 $\times$ 50, 70 $\times$ 50, 70 $\times$ 45, 70 $\times$ 40 and 60 $\times$ 45 cm. The plant height and the petiole length were increased with increasing the planting distance. The tallest plant height of yacon was 165.4cm with the plant density of 80 $\times$ 60 cm. However, branch number per plant, leaf number on main stem and stem diameter were not significantly difference among planting densities. Tuberous root was harvested 31.42 tons/ha in 70 $\times$ 50 cm spacing. The ratio of heavier tuberous roots than 200 g to total tuberous roots decreased significantly according to increase of planting density. Fresh weights of shoot and root, contain the crown bud, were decreased, as planting distance was shorter. Tuberous root number was fewer but its weight was heavier in wide planting than in dense planting. We think that optimal planting density is about 30,000 plants/ha, if it were to be 70 cm row spacing, intrarow spacing should calculate about 47cm.
