Optimization of a Virus-Induced Gene Silencing System with Soybean yellow common mosaic virus for Gene Function Studies in Soybeans

  • Virus-induced gene silencing (VIGS) is an effective tool for the study of soybean gene function. Successful VIGS depends on the interaction between virus spread and plant growth, which can be influenced by environmental conditions. Recently, we developed a new VIGS system derived from the Soybean yellow common mosaic virus (SYCMV). Here, we investigated several environmental and developmental factors to improve the efficiency of a SYCMV-based VIGS system to optimize the functional analysis of the soybean. Following SYCMV: Glycine max-phytoene desaturase (GmPDS) infiltration, we investigated the effect of photoperiod, inoculation time, concentration of Agrobacterium inoculm, and growth temperature on VIGS efficiency. In addition, the relative expression of GmPDS between non-silenced and silenced plants was measured by qRT-PCR. We found that gene silencing efficiency was highest at a photoperiod of 16/8 h (light/dark) at a growth temperature of approximately $27^{\circ}C$ following syringe infiltration to unrolled unifoliolate leaves in cotyledon stage with a final SYCMV:GmPDS optimal density $(OD)_{600}$ of 2.0. Using this optimized protocol, we achieved high efficiency of GmPDS-silencing in various soybean germplasms including cultivated and wild soybeans. We also confirmed that VIGS occurred in the entire plant, including the root, stem, leaves, and flowers, and could transmit GmPDS to other soybean germplasms via mechanical inoculation. This optimized protocol using a SYCMV-based VIGS system in the soybean should provide a fast and effective method to elucidate gene functions and for use in large-scale screening experiments.

Ideal Freezing Curve Can Avoid the Damage by Latent Heat of Fusion During Freezing (냉동 시 잠재용융열에 의한 피해를 최소화할 수 있는 이상냉동 곡선)

  • Background:Liquid nitrogen freezing techniques have already met with widespread success in biology and medicine as a means of long-term storage for cells and tissues. The use of cryoprotectants such as glycerol and dimethylsulphoxide to prevent ice crystal formation, with carefully controlled rates of freezing and thawing, allows both structure and viability to be retained almost indefinitely. Cryopreservation of various tissues has various con-trolled rates of freezing. Material and Method: To find the optimal freezing curve and the chamber temperature, we approached the thermodynamic calculation of tissues in two ways. One is the direct calculation method. We should know the thermophysical characteristics of all components, latent heat of fusion, area, density and volume, etc. This kind of calculation is so sophisticated and some variables may not be determined. The other is the indirect calculation method. We performed the tissue freezing with already used freezing curve and we observed the actual freezing curve of that tissue. And we modified the freezing curve with several steps of calculation, polynomial regression analysis, time constant calculation, thermal response calculation and inverse calculation of chamber temperature. Result: We applied that freezing program on mesenchymal stem cell, chondrocyte, and osteoblast. The tissue temperature decreased according to the ideal freezing curve without temperature rising. We did not find any differences in survival. The reason is postulated to be that freezing material is too small and contains cellular components. We expect the significant difference in cellular viability if the freezing curve is applied on a large scale of tissues. Conclusion: This program would be helpful in finding the chamber temperature for the ideal freezing curie easily.

A Study on Identifying Problems by the Comparative Analysis of High-Rise Apartment Evacuation Design Codes (고층 아파트 피난설계기준 비교분석을 통한 문제점 도출 연구)

  • The recent tragic outbreak of fire at the Grenfell Tower in the United Kingdom compels all branches of government to reexamine and reassess the safety measures against fires in high-rise buildings from a broad perspective. Accordingly, this study examined what problems stem from the current evacuation codes by comparative analysis of the high-rise apartment evacuation codes that range over various individual laws before introducing a new evacuation code. The outcome of this study can be utilized as basic data for amending the existing evacuation codes for high-rise buildings. The codes related to high-rise building evacuation include the Building Act, the Housing Act, the Skyscraper Disaster Management Act, and the Fire-Fighting System Act. Some local governments have also drawn up their own codes and put them into practice. Comparative analysis of the major evacuation codes has helped identify a variety of problems, such as different codes being applied to each individual law and ordinance, the use of additional codes other than the provisions of laws and ordinances, and the in effectiveness of evacuation codes. Most of these problems occur when diverse codes are applied to a single building, which means that cooperation among the relevant ministries is urgently needed to improve the related codes in the future.

Effect of Phosphorus Stress on Photosynthesis and Nitrogen Fixation of Soybean Plant under $CO_2$ Enrichment (대기 $CO_2$ 상승시 인산공급이 식물체의 광합성 및 질소고정에 미치는 영향)

  • The objective of this study was to examine the effect of phosphorus deficiency on nitrogen fixation and photosynthesis of nitrogen fixing soybean plant under $CO_2$ enrichment condition. The soybean plants(Glycine max [L.] Merr.) inoculated with Bradyrhizobium japonicum MN 110 were grown with P-stressed(0.05 mM-P) and control(1 mM-P) treatment under control$(400\;{\mu}l/L\;CO_2)$ and enrichment$(800\;{\mu}l/L\;CO_2)$ enviromental condition in the phytotron equipped with high density lamp$(1000\;{\mu}Em^{-2}S^{-1})$ and $28/22^{\circ}C$ temperature cycle for 35 days after transplanting(DAT). At 35 DAT, phosphorus deficiency decreased total dry mass by 64% in $CO_2$ enrichment condition, and 51% in control $CO_2$ condition. Total leaf area was reduced significantly by phosphorus deficiency in control and enriched $CO_2$ condition but specific leaf weight was increased by P deficiency. Phosphorus deficiency significantly reduced photosynthetic rate(carbon exchange rate) and internal $CO_2$ concentration in leaf in both $CO_2$ treatments, but the degree of stress was more severe under $CO_2$ enrichment condition than under control $CO_2$ environmental condition. In phosphorus sufficient plants, $CO_2$ enrichment increased nodule fresh weight and total nitrogenase activity(acetylene reduction) of nodule by 30% and 41% respectively, but specific nitrogenase activity of nodule and nodule fresh weight was not affected by $CO_2$ enrichment in phosphorus deficient plant at 35 DAT. Total nitrogen concentrations in stem, root and nodule tissue were significantly higher in phosphorus sufficient plant grown under $CO_2$ enrichment, but nitrogen concentration in leaf was reduced by 30% under $CO_2$ enrichment. These results indicate that increasing $CO_2$ concentration does not affect plant growth under phosphorus deficient condition and phosphorus stress might inhibit carbohydrate utilization in whole plant and that $CO_2$ enrichment could not increase nodule formation and functioning under phosphorus deficient conditions and phosphorus has more important roles in nodule growth and functioning under $CO_2$ enrichment environments than under ambient condition.

Suppression of Rhizome Rot in Organically Cultivated Ginger Using Integrated Pest Management (종합적 방제기술을 이용한 유기재배 생강의 근경썩음병 억제)

  • This study was conducted to control ginger rhizome rot treated with the combined treatment, the hairy vetch, carbonized rice husk and eggshell calcium in organic ginger farm. Early symptoms of leaf yellowing and plant wilt began in the chemical fertilizer treatment on July 1. Ginger rhizome rot was more progressed on October 2, and stem browning and dead plant showed a high disease incidence with from 36.7% to 43.0%. On the other hand, the combined treatment did not occur at all until July 1 and delayed the disease incidence to October 2. It showed a low disease incidence of 1.3% to 1.7%. In the combined treatment, the content of soil Na, Fe, Cu was decreased and organic matter was increased twice with 31.6% than previous. Population density of Pythium sp. is lower in the combined treatment ($0.3-2.0{\times}10^3cfu/g$ than the chemical fertilizer treatments ($12.0-12.3{\times}10^3cfu/g$). The combined treatment, hairy vetch, carbonized rice husk and the eggshell calcium is able to control the ginger rhizome rot in organically cultivated ginger field.

Radial Variation of Anatomical Characteristics of Chestnut Wood (Castanea crenata) Grown in Korea - Vessel Element and Ray - (국내산 밤나무 목재의 해부학적 특성의 방사방향 변이성 - 도관요소 및 방사조직 -)

  • Some wood anatomical characteristics of seven chestnut cultivars (Tanzawa, Ibuki, Arima, Ginyose, Tsukuba, Riheiguri, and Mansung) grown in Korea were examined in this study. We investigated the radial variation of wood components such as diameter and number of earlywood vessel element and ray density in cross section and number and height of ray in tangential section. Discs were taken from the stem of seven chestnut cultivars at breath height and cut into the radial strip. Cross, radial, and tangential sections, 10 to $20{\mu}m$ thick, were sliced with a sliding microtome. These sections were stained, dehydrated, and mounted with Canada balsam according to common procedure. Measurement and observation were performed with a light microscopy. In all chestnut cultivars, radial and tangential diameter of vessel element increased but number of vessel element per $mm^2$ decreased with increasing tree age. Ray spacing (per mm) in cross section from pith to bark showed no significant variation, while number of ray per $mm^2$ increased but ray height decreased with the increase of tree age. Conclusively, the results obtained in present study were thought to be some valuable information for separating juvenile and adult wood as well as identifying chestnut cultivars.

  • Essential oils from Fennel fruit(Foeniculum vulgare Mill), Olibanum resin(Boswellia carteii Birew) and Needl Juniperus stem(Juniperus rigida Sieb.) were extracted by a supercritical fluid extraction system(SFE) and biological activity of each essential oils were observed. SFE technique was applied for the isolation and purification of nonpolar biologically active essential oils from each samples. The quantitative analysis of essential oils was carried out by gas chromatography-mass spectrometer(GC/MS). About 60% of the growth of AGS and A549 cells were inhibited by adding 1.0g/l of the crude essential oils and below 40% was observed by the control. Cytotoxicity on human normal lung cell(HEL299) was scored as $15{\sim}18%$ for the crude essential oils and 12% for control, respectively. It meant that the essential oils were more effective than the control in anti-mutagenecity tested by CHO V79 cells. The effect of the essential oils on the growth of nerve cells, PC12 was observed as follows: The viable cell density was about two times higher than control.

Transformation of Populus tremuloides Using Agrobacterium rhizogenes (Agrobacterium rhizogenes를 이용한 Populus tremuloides의 형질전환)

  • Several factors affecting on transformation efficiency were studied to establish a Agrobacterium rhizogenes mediated system for the transformation of Populus species, and We could obtaine tansgenic plantlets expressing the introduced gene. Leaf sections were more sensitive than stem sections to kanamycin and thought to be better material for transformant screening. The bacterial density did not affect on the efficiency of transformation over the range of $4{\times}10^5{\sim}7{\times}10^9\;cfu$. The optimum period for co-cultivation was one day or shorter. Both of the optimum concentrations of cefotaxime and ampicillin in the medium were $250\;{\mu}g/ml$ for elimination of bacteria from the inoculated leaf sections. The addition of acetosyringone in the bacterial culture medium increased transformation rate, and the highest rate was obtained at $50\;{\mu}M$ of acetosyringone. The transformed galls could be selectively induced and gown on the growth regulator-free medium or on the medium containing $100\;{\mu}g/ml$ or higher contrition of kanamycin. The roots were induced from the galls incited by A. rhizogenes within 3 weeks on the growth regulator-free medium as well as on the medium containing growth regulators. The plantlets were regenerated from the galls cultured for 6 weeks on the medium containing 0.05 mg/ml of NAA and 0.5 mg/ml of BA. The expressions of the introduced opine gene in the transformed galls and plantlets were confirmed by the analysis of agropine and mannopine.

Changes in Water Potential of Pine Seedlings Inoculated with Bursaphelenchus xylophilus (소나무재선충 접종목의 수분포텐셜 변화)

  • The purpose of this study was to understand water stress development in pine seedlings inoculated with Bursaphelenchus xylophilus. Leaf water potentials of four years old Pinus densiflora seedlings inoculated with the nematode were measured with a plant moisture system at predawn and midday. The midday leaf water potentials on the 35th day after the inoculation were -1.04 MPa in the infected seedlings, while -0.94 MPa in the non-infected seedlings. The predawn water potentials on the 56th day were -0.71 MPa in the infected, while -0.26 MPa in the non-infected. At this time the leaves of the infected seedlings became yellow and resin did not exude on the cut basal stem. Density of the nematode was maximum on the 28th day, but dropped sharply on the 68th day. The phloem and pith tissues became brown and the resin exudation reduced in the infected seedlings. In conclusion, pinewood nematode inoculation developed water stress in pine seedlings in a month.

Changes in Soil Chemical Properties after Thinning in Quercus acuta stand (간벌 후 붉가시나무 임분의 토양 특성 변화)

  • This study was carried out to investigate the changes in soil chemical properties after thinning for a Quercus acuta stand in Wan-do Arboretum, Jeollanam-do. The stem density after thinning was determined referring to that of Quercus acutissima with 10 cm diameter at breast height and soil samples were collected 7 years after thinning. There was no considerable difference in soil pH values after thinning. However, the concentrations of soil organic matter, total nitrogen and available phosphorus significantly increased after thinning. In addition, thinning significantly increased cation exchange capacity (C.E.C) and the concentrations of exchangeable cations ($K^+$, $Na^+$, $Ca^{2+}$, and $Mg^{2+}$). From this study, it was revealed that the nutrient concentrations of this study site were generally lower than those of other forest soils. Therefore, it is need to improve the warm temperate forests through thinning and to develop forest treatment for regeneration.
