Variation of Peroxidase 8 in Maize, Zea mays, L. (옥수수에 있어서 Peroxidase 8의 변이)

  • As one way of evaluating polymorphism in maize, variation of peroxidase 8 (Px.8) in maize plants was investigated by means of horizontal acrylomide electrophoresis. The specific part of maize plants was stele and other parts of plants were also studied for 34 different maize lines and hybrids. The results obtained indicate that Px.8 has three distinct migrating patterns on gel such as fast, slow and fast-slow. The band pattern varied with materials used. Most hybrids were showing fast-slow band, indicating that those hybrids are heterozygous at least for Px.8 allele. Variation of band pattern was obseerved within a single inbred line. The Px.8 in stele tissue and in young leaf and nodal tissue was found to be identical, indicating the possible use of those tissues as an alternative tissue for Px.8 study. It was also found that there might be some definite relationship between Px.8 and Px.3 activities in the same tissue.

Petrological Characteristics and Deterioration phenomena of the rocks consisting the Naju-Dongmunoi-Seogdanggan(Stone Stele) (나주동문외석당간 구성암석의 암석학적 특징과 훼손양상)

  • The rock consisting the Naju-Dongmunoi-Seogdanggan (Stone Stele)(Treasure number 49) is biotite granite of medium to coarse grains which might be taken from nearby area. The rock is mainly composed of quartz, plagioclase, alkali feldspar and biotite. Due to strong weathering grail peel-off and surface exfoliation are well developed. The rock surface is changed into pale brown or pale black colors according to weathering, organism and weathering product of iron band. Major deterioration phenomena are grain peel-off, surface exfoliation, cracks and damage which may be originally classified into weathering, effect of weathering product of iron band, organisms, structural unstabilities and impact.

Isozymic Characteristics of Multiple-Ear and Tiller Maize Lines (다수다얼성 옥수수의 동위효소 특성)

  • This experiment was conducted to determine the isozymic differences between normal maize and maize inbreds of multiple ears and tillers (MET). Two maize inbreds Euisung, Iri and their hybrid having tillers and multiple ears were compared with normal maize. With usual electrophoresis using 6% polyacrylamide gel, peroxidase and esterase enzymes were studied. Matured leaf, culm, leaf sheath, root and young ear tissues showed different isozymic patterns between METs and normal maize in peroxidase. The Euisung inbred grown for 7 days under dark condition showed typical peroxidase. bands compared with checks in the tissues of coleoptile and stele. Better observation of isozymic bands was made during early part of maize growth. Parental inbreds showed more active and apparent band differences than their hybrids in esterase. Bands for esterase were also apparently different in the stele, coleoptile and young ear tissues of the METs and the checks. The maize lines infected with black streaked dwarf virus showed obvious differences in peroxidase and esterase isozymes.

  • The seasonal variation in the activity of sucrose synthase, ADP-glucose pyrophosphorylase and UDP-glucose pyrophosphorylase in roots of Panax ginseng C.A.Meyer have been studied. It was revealed that sucrose synthase and ADP-glucose pyrophosphorylase are adaptive enzymes and can serve as markers of sink strength, while UDP-glucose pyrophosphorylase is the maintenance enzyme. The average day temperature exceeded 24。C appeared to cause the disturbance in refilling process, affecting the starch synthesis. Study on the dependence of oxygen consumption in stele tissue with temperature revealed the sharp accelerating of this process after 24。C.

Rusty-Root Tolerance and Chemical Components in 4-year old Ginseng Superior Lines (4년생 인삼계통의 적변내성 및 화학성분 특성)

  • Experiments were carried out to examine the rusty tolerance in 61 inbred lines of ginseng cultivated in field, and chemical components were analyzed to clarify the difference between healthy and rusty ginseng roots. Among them, 10 lines showed rusty tolerance (RT) while 10 lines showed rusty sensitivity (RS). The content of phenolic compound in RT was lower than that in RS in cortex, epidermis and branch & fine roots, but it was not difference between RT and RS in stele. The contents of K, Ca, Na in RT were lower than RS in cortex, and the content of Mg, Fe, Na, Mn, AI, Si in RT were lower than RS in epidermis, and the content of Fe in RT were lower than RS in branch & fine roots, but mineral contents were not difference between RT and RS in stele. The content of phenolic compound in healthy cortex was lower than that in rusty cortex in same 6-year roots, but the mineral contents were not difference between healthy and rusty cortex in same 6-year roots. In root of seedlings, the contents of phenolic compound, K and Na in RT were lower than RS. It was suggested that the contents of phenolic compound, K and Na might be marker to select rusty tolerance ginseng lines.

Genetic Analysis of Esterase Isozymes in Tillering Maize (분벽성 옥수수에 대한 Esterase 동위효소의 유전분석)

  • This study was conducted to investigate the genetic analysis of esterase isozymes in maize with tillers. The materials used for the study were stele tissue for five day old seedlings of IK inbred line with tiller and A-type inbred line with no tiller, their Fl and F,. The methods employed for the study were same as previous report by Lee and Choe. A total of thirteen isoesterase enzyme bands were identified and five zones were distinguised according to both migration distance and genetic segregation patterns. The E$\sub$0.3/, E$\sub$0.4/ and E$\sub$0.5/ loci appeared from orgin to 0.5cm migration distance were controlled by the two alleles in (IK/A-type)F$_2$ and the E$\sub$0.3/+E$\sub$0.4/ of variants was controlled by codominant alleles. The E$\sub$1.0/, E$\sub$1.2/, E$\sub$1.5/ and E$\sub$1.8/ loci appeared from 1.0cm to 1.8cm were also controlled by the two alleles. However, the null band was functioned alleles. The E$\sub$2.8/, E$\sub$3.0/ and E$\sub$3.5/ loci appeared from 2.8cm to 3.5cm migration distance were very active and near location. A total of individuals with two paried bands of these loci were more than those of three paired bands(x$^2$=0.327$\^$**/). The activity of bands appeared over 3.8cm were very low and these were controlled by the two alleles. In above results, genetic segregations of stele tissue of maize with tillers were suggested to be controlled by Mendelian genetic laws.

Petrological Characteristics and Provenance Interpretation of Daejiguksatapbi (State Preceptor Daeji's Memorial Stele) in Samcheonsaji Temple Site, Seoul (서울 삼천사지 대지국사탑비의 암석학적 특성과 산지해석)

  • Daejiguksaptapbi (Tapbi) is a memorial stele which is inscribed with personal history of a state preceptor, Daejiguksa Beopkyeong. The Tapbi comprises tortoise-shaped pedestal, dragon top and body stone, and the body stone is remained in 255 pieces by excavation in Samcheonsaji temple site of Seoul. The Tapbi is composed of tuffaceous breccia for the body, coarse-grained pink feldspar granite for the pedestal and fine-grained granite for the top. As a result of geological survey to find a rock provenance of the Tapbi rock, the tuffaceous breccia is found in the banks of Gangwha straits, and the pink feldspar granite and fine-grained granite are found around the temple site in Mt. Bukhansan. Each rock shows similarities to the Tapbi in lithology and mineralogy. Also, geochemical data of major, some trace and rare earth elements verify their cognation in each rock type. It can be interpreted that the provenance of the body stone is distant as Gangwha area, and the provenance of the pedestal and top stones are near the temple site. The results can be utilized to restore the Tapbi by using the same rocks.

Pharmacognostical Studies on a Folk Medicine "SinKyungCho" (민간약 신경초의 생약학적 연구)

  • A Korean folk medicine, "SinKyungCho" has been used to cure common cold and rheumatis. While this crude drug has similar morphological feature to that of Rubia species, the botanical origin has never been studied pharmacognostically. Therefore, in order to clarify the botanical origin of "SinKyungCho", the morphological and anatomical characteristics of Rubia species growing in Korea, i.e. R. akane, R. chinensis var. glabrescens, R. cordifolia var. pratensis, R. cordifolia var. sylvatica were studied. Especially, differing from the other three species, R. cordifolia var. pratensis has 2-4 cork cell layers under the endodermal cell layer. R. cordifolia var. sylvatica showed the largest number of parenchyma cell layers in cortex by 8-17. While the average ratio of stele to root transverse section is over 45% for R. akane and R. cordifolia var. pratensis, the ratio is under 45% for R. chinensis var. glabrescens and R. cordifolia var. sylvatica. According to these morphological criteria, the folk medicine "SinKyungCho" from Korean traditional market was proved to be the underground part of R. akane.

Observation and Distribution of Ectomycorrhizal Fungi in Pinus Roots

  • Detailed structures of ectomycorrhizae formed in Pinus roots were observed with various microscopes: light, fluorescence, and scanning electron microscopes. The mantles and Hartig nets commonly found in the structure of ectomycorrhiza were newly observed according to developmental stage by various staining. The mycelia were observed to be composed of coiled types on the surface of epidermal root during early stage and fused to form mantles of smooth fungal layers, loosing mycelia with some viscous liquid secreted. The ectomycorrhizal hyphae in anatomical roots penetrated the cortical layer and formed obviously mantle and Hartig net. The round spots of ectomycorrhizal mycelia were observed morphological distribution from the cortical layer to vascular bundle of stele in the ectomycorrhizal roots of Pinus species and especially scattered at the area of meristem at the root tip as longitudinal sections. Those mycelia penetrated seemed to move into other roots by means of vascular bundle of ectomycorrhizal roots and newly form ectomycorrhizal roots of dichotomous branches.

Origin of the Korean Screen Planisphere with both Old and New Star-charts

  • The origin of the Korean Screen Planisphere with both Traditional and New Star-Charts, made by Korean Astronomers in the Royal Astronomical Bureau of the Joseon Dynasty by adopting the knowledge of the European astronomy, is investigated by analyzing its inscriptions and star charts. The considerations on both the changes in notations or representations of names of asterisms and the naming taboos used in the Old-style planisphere imply that the star-chart is originated from either the Sukjong-Stele-Replica of Cheonsang-Yeolcha-Punyajido(天象列次分野之圖). The New style planisphere is just the reproduction of Huangdao-congxingtu (黃道總星圖), with the exception of the non-Chinese-traditional stars. The Huangdao-congxingtu was made in 1723 CE by Ignatius K$\ddot{o}$gler who was a Jesuit missionary and worked for the Bureau of Astronomy (欽天監) in the Qing Dyansty. I find that the star chart was imported in 1742 CE from the Qing by An Gukrin (安國麟) who was an astronomer in the Royal Astronomical Bureau of Joseon. The chart became model for the screen star-chart made in 1743 CE and now housed in Bopju temple. I found that the inscriptions are extracted from the sentences in both Xinzhi Lingtai Yixiangzhi (新製靈臺儀象志) and Qinding Yixiangkaocheng (欽定儀象考成). Korean historical records in either Daily Records of the Royal Secretariat of the Joseon Dynasty (承政院日記) or Annals of the Joseonn Dynasty (朝鮮王朝實錄) show that Xinzhi Lingtai-Yixiangzhi was imported from the Qing Dynasty in 1708 CE, and the Qinding Yixiangkaocheng was imported in 1766 CE. Thus, the Korean Screen Planisphere with both Old and New Star-charts was certainly made after 1766 CE.
