Characteristics of $G_{418}$-sensitive mitochondrial ATPase/ATP synthase from pleurotus florida (사철느타리버섯 중 $G_{418}$-sensitive 미토콘드리아성 ATPase/ATP synthase의 특성)

  • The mitochondrion was purified at 44% sucrose layer from pleurotus florida by using ultracentrifuge and sucrose density gradient method. Optimum pH and temperature of ATPase and ATP synthase were pH 7.4, $60^{\circ}C$ and pH 7.5, $57^{\circ}C$ respectively, also their Km values were determined as 11.6mM and 8.4mM. ATPase was activated at 5~6mM ATP substrate concentration, then ATP synthase was 5~10mM range ADP. ATPase/ATP synthase were $Mg^{2+}$-dependent enzyme, partially inhibited by their substrate, and then showed an none competitive inhibition pattern by $G_{418}$. Amino acid composition of ATPase/ATP synthase was as follows, hydrophobic amino acid residue was 50.5%, small residue, 56.1%, hydrogen bonding residue, 43.7% and helix breaking residue, 55.2%. Phosphatidyl choline, phosphatidyl ethanolamine and phosphatidyl glycerol were contained but not phosphatidyl inositol and phosphatidyl serine. Palmitate(51.31%), stearate(18.32%) and unsaturated fatty acids($C_{18:1}$, $C_{18:2}$ and $C_{16:1}$) were predominated.

Volatile Flavor Components in the Mashes of Takju Prepared Using Different Yeasts (효모종류를 달리한 탁주 술덧의 휘발성 향기성분)

  • In this study, we examined the volatile flavor components in the mashes of takju prepared using different yeasts such as Saccharomyces coreanus, S. ellipsoideus, S. carlsbergensis, S. cerevisiae (Baker's yeast), and S, rouxii by GC and GC-MS. Fourteen alcohols, 13 esters, 5 acids, 3 aldehydes, 7 amines, and 2 other compounds were identified in the mash after 6 days of fermentation. On day 6, the takju fermented by S. coreanus had the greatest variety of volatile flavor components. Fifteen flavor components, including ethanol, isobutyl alcohol, isoamyl alcohol, methyl pentanol, 1,3-butanediol, 3-methylthio-1-propanol, benzeneethanol, ethyl lactate, acetic acid, acetaldehyde, and 1,3-cyclohexane diamine, were typically detected in all the treatments. The relative peak areas of the volatile components were as follows: alcohols (96.758-99.387%), esters (0.081-0.968%), acids (0.040-0.640%), aldehydes (0.266-0.959%), and amines (0.011-0.047%). In particular, 1-propanol, isobutyl alcohol, 3-methyl-1-butanol, 2,3-butanediol, trimethyl benzylalcohol, heptene-2,4-diol, ethyl lactate, diethyl succinate, ethyl nonanoate, methyl hexadecanoate, linoleic acid, hexadecanoic acid, and acetaldehyde were hish in the takju made by S. coreanus. Also, ethyl stearate was high in the takju made by S. carlsbergensis, and hexanoic acid was high in the takju made by S. cerevisiae. Finally, methyl pentanol, 1,3-butanediol, 3-methylthio-1-propanol, benzene ethanol, ethyl octadecanoate, acetic acid, pentanal, and 1,3-cyclohexane diamine were high in the takju made by S. rouxii.

Mode of Action of Several Surfactants on Paraquat Efficacy (Paraquat 활성에 미치는 계면활성제의 작용기구)

  • The effects of 24 ionic and nonionic surfactants on paraquat (1, 1' -dimethyl-4 4'-bipyridinium) efficacy were investigated with several annual plant species under greenhouse conditions. The paraquat efficacy was decreased or even lost when treated with the anionic surfactants tested. However, the efficacy of paraquat was significantly increased by 7 nonionic surfactants such as sorbitan palmitate, sorbitan stearate, polyoxyethylene sorbitan monopalmitate, polyoxyethylene sorbitan monostearate, polyoxyethylene stearyl ether, polyoxyethylene laurylamine ether, and polyoxyethylene stearylamine ether. Among these tested surfactants, 0.08% of polyoxyethylene laurylamine ether most significantly increased the paraquat activity, and the $GR_{50}$ value of paraquat with polyoxyethylene laurylamine ether was 1.6 times lower than the $GR_{50}$ value without polyoxyethylene laurylamine ether. In in vitro experiments, cellular leakage and chlorophyll contents between the application with and without polyoxyethylene laurylamine ether did not show significant changes. The absorption rate of $^{14}C$ paraquat in the treatment with polyoxyethylene laurylamine ether showed an absorption rate of 1.6 times higher than without surfactant. These results suggest that using compatible surfactants would increase the paraquat efficacy, and this increasing are due to improved absorption rate with the surfactant.

Structures of OH Emulsion Prepared with Saccharide Surfactants (당류계 계면활성제로 제조된 O/W 에멀젼의 구조)

  • The o/w emulsions were prepared with saccharide surfactants which were sucrose monostearate(S160), sucrose distearate(S110), and POE(20) methyl glucose stearate(SSE20). And for emulsion the oils used were n-hydocarbon, squalane(SQ), liquid paraffin(LP), octylpalmitate(OP), octylstearate(OS), alkyl benzoate(AB), isostearyl benzoate(ISB). The structures of o/w emulsion droplet were investigated by laser light scattering and the fractal dimensions were calculated from light intensity curves. Increasing of concentration, chain length, and nonpolarity of oils, fractal dimensions of emulsion droplets were found greater. In general fiactal dimensions were varied from 1.7 to 2.8 and its structures were fractal But the fractal dimensions of octadecane( $C_{18}$), 50, and LP emulsified with S110 and S160 were varied from 3.0 to 3.2 and its structures were more dense. The overall fractal dimensions of S110 and S160 were varied from 2.1 to 2.6, that of SSE20 were varied from 1.5 to 2.1. So it was found that the structures of SSE20 system were less compact than that of S110 and S 160 system, because the hindrance effect of polyoxyehtylene group of SSE20 was stronger than that of sucrose of S160. The strucures of emulsion droplets changed according to the nature of emulsifiers and to compositions of oil substances which they contained, and the structures were found similar when the hydophilic moiety of emulsifiers was same.

Cloning and Characterization of an Esterase from Xanthomonas oryzae pv. oryzae

  • The gene encoding a putative esterase of Xanthomonas oryzae pv. oryzae was cloned using PCR technique. The gene was expressed with His6 tag in E. coli. One-step purification of the recombinant esterase with Ni-NTA resin resulted in one band by SDS-PAGE analysis. The purified enzyme showed a molecular weight of 30 kDa, as expected, therefore the enzyme was a mononer. The enzyme was the most active toward p-nitrophenyl (p-NP) acetate and p-NP-butyrate to a lesser extent. However, the enzyme could not hydrolyze p-NP-myristate, palmitate, and stearate. Therefore, the enzyme is considered as an esterase, very different from lipase. The purified esterase had optimal pH at around 8.0 and was stable in a broad range of pH values. The optimal temperature ranged from 30 to $40^{\circ}C$, and the residual activity observed after heat treatment at $55^{\circ}C$ for 20 min was 72 % of the initial activity. The activity was inhibited by the presence of copper and cobalt ions.

The Mechanical Appearances and Microscopic Tissue Reactions of the Suture Materials in vivo (생체내 이식된 흡수성 봉합사들의 물리적 성질 변화와 조직 소견에 관한 연구)

  • A comparative study of three absorbable suture materials of chromic catgut, Dexon II (a polyglycolic acid suture with a polycaprolate coating system) and Coated Vicryl (a polyglactin 910 suture with a glycolide-lactide-calcium stearate coating system) was undertaken in terms of tensile strength, breaking elongation, appearances of the suture materials in the subcutaneous layer of rats and of tissue reaction in the intramuscular layer of rats. The initial tensile strength of chromic catgut and the tensile strength after 21 days were about 1.55 kg and 0.19 kg, respectively. Those of Dexon II were about 2.01 kg and 0. 20 kg, respectively. Those of Coated Vicryl were about 2.39 kg and 0.48 kg, respectively. Coated Vicryl showed the highest tensile strength among the three materials during the whole period. On the other hand, Dexon II showed the highest breaking elongation among the three materials during the first week. But the breaking elongation of Dexon II rapidly continuously decreased during the whole period. The appearances of the suture materials in the subcutaneous layer of rats showed that chromic catgut was the fastest among the three materials, whereas it formed comparatively much connective tissue. The intramuscular absorption rate and tissue reaction of Dexon II were similar to those of Coated Vicryl. The intramuscular absorption rate of these was fairly late when compared with chromic catgut. And the tissue reaction appearance of these showed the formation of a granuloma with foreign body giant cells, macrothages and epitheloid cells by the 28th day in the experimental period.

Synthesis and Evaluation of Stearic Acid Derivatives as Cetane Number Improvers

  • 1,2,4,5-tetraoxane, mono and dinitrate glycerol carbonate ester derivatives of stearic acid were synthesized along with the known 9(10)-keto methyl sterate, methoxy mono-nitrate and dinitrate of methyl stearate. Their cetane numbers (CNs) were investigated to evaluate their viability for use as CN improvers. The CN performances of tetraoxane and all of the nitrate derivatives were investigated at 500 and 1000 ppm concentrations and compared to that of a traditional CN improver 2-ethylhexyl nitrate (2-EHN). The experimental results suggest that all derivatives evaluated in this study showed better CN improvement than base diesel fuel. Specifically, the 1,2,4,5-tetraoxane derivative of stearic methyl ester was superior to all derivatives studied, also being superior to 2-EHN. We also discussed the correlations between the observed CN trends and thermo-analytical data resulted from thermo gravimetric analysis curves (TGA) and differential scanning calorimetry (DSC).

Design and Optimization of Solid Dispersed Osmotic Pump Tablets of Aceclofenac, A Better Approach to Treat Arthritis

  • The aim of this work was to prepare porous osmotic pump tablets for controlled delivery of Aceclofenac. Aceclofenac solid dispersion was prepared to improve the solubility by using the drug - carrier (Mannitol) ratio of 1:1. The osmotic pump tablets were prepared using the solid dispersed product of Aceclofenac. The formulation contains potassium chloride as osmotic agent, cellulose acetate as semipermeable membrane, poly ethylene glycol (PEG 4000) as pore former and sodium lauryl sulphate (SLS) as solubility enhancer. The formulations were designed by the general factors such as osmotic agent and pore former. All formulations were evaluated for various physical parameters and, the in vitro release studies were conducted as per USP. The drug release kinetic studies such as zero order, first order, and Higuchi and Korsmeyer peppas were determined and compared. All the formulations gave more controlled release compared to the marketed tablet studied. Numerical optimization techniques were applied to found out the best formulation by considering the parameter of in vitro drug release kinetics and dissolution profile standards. It was concluded that the porous osmotic pump tablets (F7) composed of Aceclofenac solid dispersion/Potassium chloride/Lactose/Sodium lauryl sulphate/Magnesium Stearate (400/40/95/10/5, mg/tab) and coating composition with Cellulose acetate/ PEG 4000 (60/40 %w/w) is the most satisfactory formulation. The porous osmotic pump tablets provide prolonged, controlled, and gastrointestinal environment-independent drug release.

Preparation of Stick Type Solid Glue as Paper Adhesive Using Mixed Seaweed Extract

  • Seaweed extracts, namely carrageenan obtained from Grateloupia elliptica and algin obtained from Laminaria, were employed as adhesive agents to synthesize solid adhesives for paper. Carrageenan from Grateloupia elliptica with the highest adhesive strength and lgin from Laminaria with the highest compressive strength was selected. The selected carrageenan and algin were mixed in a ratio of 7:3, and the mixture was employed as an adhesive agent. At a high temperature, sodium stearate(used as a solidifying material) oxidized the seaweed extracts. Consequently, carrageenan and algin were added to the final manufacturing process. The adhesive strength of the final synthesized solid adhesive is found to be 3.02 MPa and the compressive strength is found to be 30.5 N. Compared to the adhesive strength (2.95 MPa) and compressive strength (30.11 N) of commercial solid adhesives, the obtained results indicate superior adhesion characteristics. Furthermore, the proposed adhesive is environment-friendly because the presence of volatile organic compounds, formaldehyde, and heavy metals(such as chromium, lead, and cadmium) were not detected. Moreover, when used, the flatness of paper was twice that of commercial solid paper adhesives. Hence, the proposed adhesive can provide excellent adhesion, stability, and usability.

Polyetherimide/Dicyanate Semi-interpenetrating Polymer Networks Having a Morphology Spectrum

  • The morphology, dynamic mechanical behavior and fracture behavior of polyetherimide (PEI)/dicyanate semi-interpenetrating polymer networks (semi-IPNs) with a morphology spectrum were analyzed. To obtain the morphology spectrum, we disported PEI particles in the procured dicyanate resin containing 300 ppm of zinc stearate catalyst. The semi-IPNs exhibited a morphology spectrum, which consisted of nodular spinodal structure, dual-phase morphology, and sea-island type morphology, in the radial direction of each dispersed PEI particle due to the concentration gradient developed by restricted dissolution and diffusion of the PEI particles during the curing process of the dicyanate resin. Analysis of the dynamic mechanical data obtained by the semi-IPNs demonstrated that the transition of the PEI-rich phase was shifted toward higher temperature as well as becoming broader because of the gradient structure. The semi-IPNs with the morphology spectrum showed improved fracture energy of 0.3 kJ/$m^2$, which was 1.4 times that of the IPNS having sea-island type morphology. It was found that the partially introduced nodular structure played a crucial role in the enhancement of the fracture resistance of the semi-IPNs.
