High-speed countercurrent chromatography를 이용한 인삼 saponin의 대량 분리 농축 (Preparative Isolation of Ginseng Saponin from Panax ginseng Root Using High-speed Countercurrent Chromatography)

  • 이상계 용매시스템을 이용하여 물질을 고순도로 대량 분리 할 수 있는 기술인 countercurrent chromatography를 이용하여 인삼으로부터 생리황성 성분인 saponin을 대량 분리 농축하였다. 용매 조성별 인삼 saponin의 분배계수에 따른 인삼 saponin 분리에 적합한 용매시스템은 chloroform/methanol/water(40/39/21, v/v/v)으로 결정되었으며 HSCCC의 작동 조건은 chloroform/methanol/water 용매시스템의 하층부를 이동상으로 한 head to tail mode에서 이동상의 유속 5mL/min, 인삼추출물 injection량 $200{\mu}L$, 컬럼회전속도 800 rpm의 조건이 적합한 것으로 판단되었다. 이러한 조건하에서 분리된 인삼saponin의 양은 $550.7{\mu}g$으로 HSCCC에 주입한 인삼 추출물 $200{\mu}L$중에 존재하는 총 saponin의 양 $865.5{\mu}g$에 비교하여 전체 수율은 63.6%로 나타났으며 TLC로 각 분획의 순도를 확인할 수 있었다.

정지 및 회전하는 원주에 의한 난류후류의 응집구조 (An Investigation of the Coherent Structures in Turbulent Wake Past a Stationary and Rotating Cylinder)

  • Turbulent coherent structures in the intermediate wake of a stationary and rotating cylinder, spin rate S=0.7, situated in a uniform were experimentally investigated using a conditionalphase average technique. Measurements were carried out at a section of 8.5 diameters downstream form the center of cylinder and a Reynolds number of $Re=6.5{\times}10^{3}.$/TEX> The phase averaged velocity and velocity vector fields, contours of vorticity, turbulent intermittency function and velocity fluctuation energy are presented and discussed in relation to the large scale coherent structures by Karman vortices that shed periodically from the cylinder. Coherent wake structures of the rotating cylinder is almost identical with stationary cylinder, but the lateral displacement and shrinkage of turbulent wake region is occured by rotation. Rotation of the cylinder result in that the deflection of wake center to deceleration region(Y/D${\simeq}-0.3)$ and the decrease of mean velocity defect(10%), vorticity strength of large scale structures(19%), total velocity fluctuation energy(12%).

A New Control Strategy for a Three-Phase PWM Current-Source Rectifier in the Stationary Frame

  • This paper presents a novel power control strategy for PWM current-source rectifiers (CSRs) in the stationary frame based on the instantaneous power theory. In the proposed control strategy, a virtual resistance based on the capacitor voltage feedback is used to realize the active damping. In addition, the proportional resonant (PR) controller under the two-phase stationary coordinate is designed to track the ac reference current and to avoid the strong coupling brought about by the coordinate transformation. The limitations on improving steady-state performance of the PR controller is investigated and mitigated using a cascaded lead-lag compensator. In the z-domain, a straightforward procedure is developed to analyze and design the control-loop with the help of MATLAB/SISO software tools. In addition, robustness against parameter variations is analyzed. Finally, simulation and experimental results verify the proposed control scheme and design method.

The Diversity of Lysine-Acetylated Proteins in Escherichia coli

  • Acetylation of lysine residues in proteins is a reversible and highly regulated posttranslational modification. However, it has not been systematically studied in prokaryotes. By affinity immunoseparation using an anti-acetyllysine antibody together with nano-HPLC/MS/MS, we identified 125 lysine-acetylated sites in 85 proteins among proteins derived from Escherichia coli. The lysine-acetylated proteins identified are involved in diverse cellular functions including protein synthesis, carbohydrate metabolism, the TCA cycle, nucleotide and amino acid metabolism, chaperones, and transcription. Interestingly, we found a higher level of acetylation during the stationary phase than in the exponential phase; proteins acetylated during the stationary phase were immediately deacetylated when the cells were transferred to fresh LB culture medium. These results demonstrate that lysine acetylation is abundant in E. coli and might be involved in modifying or regulating the activities of various enzymes involved in critical metabolic processes and the synthesis of building blocks in response to environmental changes.

Function of Cell-Bound and Released Exopolysaccharides Produced by Lactobacillus rhamnosus ATCC 9595

  • The physiological characteristics and function of the exopolysaccharide (EPS) produced by Lactobacillus rhamnosus ATCC 9595 were determined. The total quantity of EPS was rapidly increased to 496$\pm$20 mg/l during the exponential phase, and then maintained steadily during the stationary phase. During the exponential phase (18 h), the total EPS consisted of 61% cell-bound EPS (cb-EPS) and 39% released EPS (r-EPS), whereas the relative proportion of EPS during the stationary phase (48 h) was convered to 23% cb-EPS and 77% r-EPS. On gel permeation chromatography, cb-EPS was fractionated as a single peak of 8.6$\times10^6$ Da, whereas r-EPS was fractionated as two peaks with average molecular weights of 4.3$\times$10$^4$ and 8.6$\times10^6$ Da. Interestingly, both EPS species exhibited anticancer properties and cholera toxin-binding activities. Our results suggest that the EPS generated by L. rhamnosus ATCC 9595 might be suitable for use as a functional food or food supplement.

달걀 단백질 분석을 위한 HPLC 연구 (HPLC Study for Egg White Analysis)

  • RP-HPLC에서 주요 변수인 이동상의 조성과 컬럼의 종류를 변화시켜 난백 단백질을 실험하였다. C4, C8 C18 컬럼을 비교 실험하여 C18 컬럼에서 가장 많은 피크를 보여 C18 컬럼을 선택하였다. 등용매 용리에서 ACN : water의 조성이 50 : 50에서 가장 많은 피크를 보여 이 결과를 토대로 기울기 용리를 하여 실험하였다. 기울기 용리에서 얻어진 4가지 피크를 확인하기 위하여 단일성분인 lysozyme와 ovalbumin의 체류시간을 측정하여 확인하였고 논문을 통해 2가지 피크를 예측할 수 있었다.
