The focus of this study was to investigate that cellular parameters and glucose uptake might be altered by extracellular calcium and starvation. Addition of 1 mM $Ca^{++}$ to hepatocytes (equalling to the free calcium concentration of blood) significantly increased intracellular $Na^+$ and decreased $Na^+$ & LDH leakage. This pertains to the hepatocytes of control rats as well as those of rats fasted for 24 and 48. hr. These effects might be come from the membrane-stabilizing effects of calcium. But calcium had no effects on cell volumes, superoxide-formation and glucose uptake. Actually hepatocytes of starved rats showed changes in several cellular parameters. Starvation increased LDH leakage, glucose uptake and the total concentration of $Na^+$ and $Na^+$ whereas it markedly decreased cell volumes. Since total tonicity remained unchanged, intracellular $Na^+$ and $Na^+$ could contribute to a higher share of total osmolarity in starvation. Starvation increased the cytoplasmic pH because $R-NH^{3+}$ions and their corresponding counterions disappeared. This increase may be related to suppress the protonization of amino groups in proteins. Starvation decreased hepatic glycogen, a major compound that affects cytosolic volume of hepatocytes. The data indicate that starvation increases the glucose transport activity. The possible molecular basis will be discussed.
This study was designed to compare by the body composition in 4 groups of Sprague-Dawley rats during starvation previously fed with standard, high Carbohydrate, high protein or high fat diet. A) In different internal organs and skeletal muscles, the weights were differently reduced during starvation. Especially the weight reduction of liver was the highest level at $50{\sim}70%$, and adrenal glands did not change significantly. After death due to starvation, body weight decreased morethan 40% comparing to the initial body weight and epididmal fat pads disap peared completely. B) In rats starved to death, the changes of body composition (%) were following; lipid content was markedly reduced, while protein was similar to that of zero day starvation, ash increased slightly and water content increased. C) Five days starvation did not change body composition and body weight significantly. D) Effects of succeeding starvation after feeding 4 different diets were not significant, and it seemed that 4 different diets did not affect life span due to starvation.

Capsaicin has been shown to affect lipid metabolism. However, it is currently not known whether capsaicin would lower the intestinal absorption of cholesterol. Thus, this study was conducted to investigate the effect of intraduodenally infused capsaicin on the lymphatic absorption of cholesterol and lipids in rats. Male Sprague-Dawley rats weighing 300-350 g were starved for 16 hr and the mesenteric lymph duct was cannulated. Each rat was infused at 3.0 mL/hr for 8 hr via the duodenal catheter with a lipid emulsion, which contained 33.3 kBq [$^{14}C$]-cholesterol, 20.7 μmol cholesterol, 452 μmol triolein, 3.1 μmol $\alpha$-tocopherol, and 396.0 μmol Na-taurocholate without (control) or with 5.0 mg capsaicin in 24 mL PBS buffer (pH 6.4). Simultaneously, lymph was collected hourly for 8 hr. There was no significant difference in lymph flow between the groups. However, the lymphatic absorption of 14C-cholesterol for 8 hr was significantly lower in rats infused with capsaicin than in those infused with no capsaicin. Also, the output of oleic acid for 8 hr was significantly decreased by capsaicin. However, the intestinal absorption of $\alpha$-tocopherol did not differ between the groups. The results indicate that the luminal infusion of capsaicin inhibits the intestinal absorption of cholesterol and lipids in rats.
Objectives : The purpose of this study is to examine what are the effect of Carthami Flos on Blood according to the administered dosage. Methods : thrity Sprague-Dawleys rats of starved during 3 days were used and divided 3 groups ; Normal group ; Experimental group that were administered Carthmi Flos 117mg/200g(Sample1) ; Experimental group that were administered Carthmi Flos 936mg/200g(Sample2). and the observerd blood(RBC, Hct, Hb, MCV, MCH, MCHC, PLT, WBC NEUT, LYM, MONO, EOSIN). Result : the result were obtained as follows ; 1. RBC, Hb, MCHC were significantly increased, and MCV were decreased in Sample1.(p<0.05) 2. PLT, MCHC, LYM were significantly increased, and Hct, MCV were decreased in Sample2.(p<0.05) Conclusion : According in the above result, it was consided that a small quantity dosage of Carthami Flos was nourished the blood, and a large quantity of that was curative for thrombosis and elevated blood viscosity and it is suggested that more interest and study in the mechanism and clinical use were needed.

Purpose : Pulmonary vascular hypertension is a common problem in congenital heart disease, the most common cardiac condition in childhood. However, the mechanisms responsible for this pathologic change, treatment, and prevention are poorly understood. Therefore, we studied the gene expression of vascular endothelial growth factor(VEGF) by using a hypoxic model of the pulmonary artery smooth muscle cells. Methods : The main pulmonary artery and its proximal branches of a 6 wk old Fischer rat were excised. They were cut into multiple small pieces and suspended in DMEM medium supplemented with 20% fetal bovine serum and incubated in 5% $CO_2$-95% air atmosphere. The smooth muscle cells were confirmed by immunostaining with smooth muscle myosin and ${\alpha}$-smooth muscle actin antibodies. The VEGF gene expression in the hypoxic group was compared with the one in control the group as well as the one in the starved group by RT-PCR and Northern blot hybridization. Results : There was no statistically significant difference among the control, hypoxic and starved groups. Conclusion : There are few studies of pulmonary vascular hypertension at the molecular level in Korea. Therefore, we studied the expression of VEGF gene in hypoxic pulmonary vascular smooth muscle cells. Further studies will be needed to find the difference between newly born and adult rats, or human and rat pulmonary vascular smooth muscle cells in gene expression. We hope that the study will lead to a better understanding of pulmonary vascular hypertension.
The present study was aimed at establishing what changes occur in the dopamine levels and pattern of TH-immunoreactive neurons of certain areas of rat brain during food intake suppression produced by intraperitoneally administration of CCK-8. CCK-8 in dose of $10\;{\mu}g/kg$ was injected intraperitoneally to 48 h food-deprived rats. In the fasted group, the contents of dopamine were decreased in the frontal, striatum, hypothalamus and amygdala as compared to those of the fed control group. The administration of CCK-8 showed significant decrease on the dopamine levels of the hypothalamus, in comparison to those of the sated and starved group. During deprived condition, the density and number of TH-immunoreactive neurons in the paraventricular nucleus, arcuate nucleus, median eminence and substantia nigra were lower than those of the fed control group. After administration of CCK-8, the pattern and distribution of TH-positive neouons in the hypothalamic areas and substantia nigra were increased when compared to those of the starved group. It is concluded that the results demonstrate the partial involvement of hypothalamic dopamine-containing neurons in the feeding inhibition of CCK-8. Furthermore, the results indicate that TH-immunoreactive neurons play on important role in the hypothalamus and substantia nigra for eating behavior

Restricted supply of nutrients may affect genes at the molecular level as well as physiological functions. Understanding the cellular responses during starvation is necessary for developing strategies to reduce damage caused by starvation stress. After 1 h of starvation, Got1 gene expression was increased but its expression returned to the normal state after 24 h. Mat1 gene expression continuously increased with starvation from 1 h until 24 hr. Rats starved for 1-3 days showed significant changes in expression of the Got1 and Mat1 genes, which were significantly reduced in the cerebral cortex and cerebellum. In the lung, gene expression was increased by starvation for 1-2 days but decreased on the third day. No differences were observed in gene expression in the heart. Strong Got1 lung gene expression was seen in the starvation group one day after restoration of the food supply. Muscle mass was significantly reduced at the start of starvation and remained the same after two days of starvation and one day after the food supply was restored. The Mat1 gene expression did not change. The Got1 was induced by NaCl and showed strong expression in the lung and the thymus, but the apparent decrease of the remaining changes were not observed in male rats. The Mat1 gene was not as sensitive as the Got1 gene to induction by NaCl. However, differences in gene induction by NaCl were evident between males and females, indicating that diet control of gene expression is associated with hormones.
