A report of 10 unrecorded bacterial species of Korea, belonging to the phylum Firmicutes

  • To investigate the indigenous prokaryotic species diversity in Korea, various environmental samples from diverse ecosystems were examined taxonomically. The isolated bacterial strains were identified based on 16S rRNA gene sequences, and those exhibiting at least 98.7% sequence similarity with known bacterial species but never reported in Korea were selected as unrecorded species. As an outcome of this study, 10 unrecorded bacterial species belonging to the phylum Firmicutes were discovered from various sources such as soil, tidal flat, fresh water, sea water, kimchi and gut of Fulvia mutica. The unrecorded species were assigned to 7 different genera of 5 families, namely Bacillus and Ornithinibacillus of Bacillaceae, Exiguobacterium of Exiguobacteriaceae, Brevibacillus and Paenibacillus of Paenibacillaceae, Staphylococcus of Staphylococcaceae, and Lactococcus of Streptococcaceae. The selected isolates were subjected to further taxonomic characterization including the analysis of Gram reaction, cellular and colonial morphology, biochemical activities, and phylogenetic trees. The descriptive information on the 10 unrecorded species are provided.

Dithiocarbohydrazone류의 합성과 항균작용에 관한 연구 (I) (Synthesis and Antimicrobial Activity of Dithiocarbohydrazones (I))

  • In order to study 2-formylpyridine dithiocarbohydrazones (DTCH's) and their Cu(II) chelates as potential effective antimicrobial agents, twelve new compounds of six DTCH's and their Cu(II) chelates were synthesized. The compositions of Cu(II) chelates were determined on the basis of the data obtained from elemental analysis, electronic and IR spectrophotometry, and other method. They were tested for antimicrobial activity in vitro against a gram-positive, seven gram-negative bacterial species and three fungal species. DTCH's exhibited high antibacterial activity against the gram-positive Staphylococcus aureus, but low activity against the various gram negative bacterial species. In contrast to DTCH'S, their Cu(II) chelates exhibited higher antibacterial activity against St. aureus, but weaker, if any, activity against the gram-negative bacterial species. Both DTCH's and their Cu(II) chelates showed relatively potent antifungal activity against Cryptococcus neoformans, but weak activity against Candida albicans and Candida pseudotropicalis.

나린게닌의 항균력에 미치는 관련 후라보노이드의 병용효과 (Antimicrobial Effects of Naringenin alone and in Combination with Related Flavonoids)

  • As part of our search for less toxic antimicrobial substances from natural resources, naringin was isolated from feels of Citri fructus and then hydrolyzed to naringenin. The antimicrobial activity of naringenin was examined by measuring the minimal inhibitory concentration(MIC) against fourteen species of bacteria. The antimicrobial activity of narngenin in combination with rutin or hesperetin was evaluated by checkerboard method. Among fourteen species tested, the antimicrobial activity of naringenin was the most prominant against Staphylococcus aureus and Shigella boydii showing MIC of $100\;{\mu}g/ml$ for both species. Combinations of naringenin with rutin or hesperetin showed synergism against several species of bacteria, but no antagonism was observed.

  • This report was concerned with the isolation and identification of bacterial flora in the human dental plaque and the dextransucrase activity of isolated species. The results were obtained as follows: 1. The bacterial flora, isolated from the human dental plaque, was identified as 3 species of resembling streptococci, Streptococcus salivarius strain SD-1, Streptococcus bacilli, Lactobacillus brevis strain SD-3, Lactobacillus acidophilus strain SD-2 and SD-7, resembling Staphylococcus sp, and one species of resembling Leuconostoc mesenteroides strain SD-6. 2. The dextransucrase activites of resembling Streptococcus mitis strain SD-9 and Streptococcus salivarius strain SD-1 were exhibited the highest among the isolated species of human dental plaque.

서울시내 중학생의 손오염에 관한 세균학적 연구 (Bacterial Contamination on the hands of the Middle School Students in Seoul City)

  • 활동성이 강한 중학생을 대상으로 남$\cdot$여, 학생에 따른 세균 오염현황을 파악함으로써 중학생의 보건 교육에 관심을 높이고자 하여 1985년 2월 4일부터 2월 15일까지 서울 시내 중학교에 재학중인 학생 29명을 대상으로 양손으로 부터의 세균을 분리하여 다음과 같은 결과를 얻었다. 1. 손에 오염된 평균 세균수는 1학년 남학생 $3.08\times10^4$개 여학생$2.48\times10^4$개, 2학년 남학생 $3.32\times10^4$, 여학생 $1.64\times10^4$개로 2학년 여학생에 있어 특히 적은 수가 나타남을 알 수 있었다. 2. 중학생 손에서 검출된 세균의 속명은 Staphylococcus, Streptococcus, Micrococcus, Bacillus, Neisseria spp., Branhamella, Acinetobacter, Moraxella, Chromobacterim, Alcaligenes, Flavobacterium, Pseudomonas, Actinobacillus, Pasteurella, Vibrio, Enterobacteria와 Plesiomonas로 17속으로 나타났다. 3. 속명이 밝혀진 균주 중 종명까지 밝혀진 것은 Staphylococcus aureus, Staphylococcus epidemidis, Micrococcus varians, Branhamella catarrhalis, Acinetobacter anitratus, Moraxella bovis, Moraxella urethralis, Pseudomonas aeruginosa와 plesiomonas shigelloid로 9종이 밝혀졌다. 4. Genus 중 전체 학생중 나타난 출현 빈도가 가장 높은 것은 Staphylococcus $(96.6\%)$이었고 그 다음 Enterobacterial $(14.4\%)$ Bacillus$(20.7\%)$, Branhamella$(17.2\%)$순으로 나타났다.

부산지역 치과환경에서 분리된 coagulase-negative staphylococci의 특성 (Characteristics of Coagulase-negative Staphylococci Isolates from Dental Clinic Environments in Busan, Korea)

  • 본 연구는 치과 병의원 진료실 주변환경과 치과종사자의 휴대전화에서 분리된 coagulase-negative staphylococci (CNS)의 분리율과 항균제 내성양상 및 분자 역학적 특성을 분석하고자 하였다. 2014년 12월부터 2015년 1월까지 부산지역 10개의 치과 병의원에서 총 154개의 샘플을 수집하여 MALDI-TOF분석법을 통해 동정하였다. 항균제 감수성검사는 디스크 확산법을 시행하였고 mupA, mecA 유전자 보유현황 및 SCCmec type은 PCR과 염기 서열분석에 의해 결정하였다. 154개의 샘플 중 10개(6.5%)에서 CNS 균주(Staphylococcus epidermidis 5주, Staphylococcus capitis 2주, Staphylococcus warneri 2주, Staphylococcus haemolyticus 1주) 가 분리되었다. 항균제 감수성검사에서 penicillin 10주, mupirocin 6주, gentamicin 5주, tetracycline 3주 및 cefoxitin과 erythromycin 2주가 내성이었고 clindamycin, ciprofloxacin, teicoplanin, trimethoprim-sulfamethoxazole에 내성인 세균은 없었다. 2개의 CNS균주(S. warneri, S. haemlyticus)에서 mecA 유전자가 검출되었고 1개의 CNS균주(S. epidermidis)에서 mupA가 확인되었다. Methicillin 내성 CNS균주 가운데 1주는 SCCmec I형이었고 1균주는 SCCmec의 유전형이 구분되지 않았다. 본 연구를 통하여 다약제 내성을 보이는 CNS균주가 더 이상 우리나라 치과병의원 환경에서 드물지 않음을 알 수 있었다.

Detection of Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus and Listeria monocytogenes in Kimchi by Multiplex Polymerase Chain Reaction (mPCR)

  • We developed an mPCR assay for the simultaneous detection, in one tube, of Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus and Listeria monocytogenes using species-specific primers. The mPCR employed the E. coli O157:H7 specific primer Stx2A, Salmonella spp. specific primer Its, S. aureus specific primer Cap8A-B and L. monocytogenes specific primer Hly. Amplification with these primers produced products of 553, 312, 405 and 210 bp, respectively. All PCR products were easily detected by agarose gel electrophoresis, and the sequences of the specific amplicons assessed. Potential pathogenic bacteria, in laboratory-prepared and four commercially available kimchi products, were using this mPCR assay, and the amplicons cloned and sequenced. The results correlated exactly with sequences derived for amplicons obtained during preliminry tests with known organisms. The sensitivity of the assay was determined for the purified pathogen DNAs from four strains. The mPCR detected pathogen DNA at concentrations ranging from approximately 0.45 to $0.05\;pM/{\mu}l$. Thus, this mPCR assay may allow for the rapid, reliable and cost-effective identification of four potentially pathogens present in the mixed bacterial communities of commercially available kimchi.

냉동보존한 자가지방의 오염률에 대한 임상적 고찰 (Clinical Considerations on Contamination Rates of Cryopreserved Autologous Fat)

  • Purpose: Autologous fat graft is a widely accepted technique used for soft tissue augmentation. Nonetheless, the use of fat graft is limited by unpredictable survival rates and repeated grafting. To avoid repeated grafting, cryopreserved fat graft technique has recently been widely used. On the other hand, the number of patients with chronic infection(who received cryopreserved fat injection) has currently been increasing. Therefore, this study was focused on the safety of cryopreserved fat injection from the infection. Methods: We collected 150 samples from local aesthetic clinics to examine the safety of cryopreserved autologous fat. To test for microbacterial contaminations of the cryopreserved fat specimens, microbacterial cultures & antibiotics sensitivity tests were performed. Then, we examined possible correlation between the preservation period and donor sites, focused on the results of microbacterial culture. Results: Cultures were positive for Staphylococcus epidermidis in 5 samples(methicillin - resistant Staphylococcus epidermidis in 4 samples), Micrococcus species in 3 samples. An average duration of preservation was 191 days and there was no significant correlation between the duration of preservation and microbacterial growth. Conclusion: Staphylococcus epidermidis was the leading cause of cryopreserved fat contamination, and the resistance to methicillin is common. Based on the above results, aseptic handling of fat during harvesting and preservation appeared to be most important.

In Vitro: Antimicrobial Effect of Lactobacillus salivarius on Staphylococcus pseudintermedius

  • Lactobacillus spp. are the bacteria most commonly used as probiotics and it has been proven that they inhibit pathogenic bacterial growth and improve skin repair in humans. This study was conducted to investigate the growth inhibitory effect of Lactobacillus on Staphylococcus pseudintermedius, the most commonly isolated pathogen in canine pyoderma, and whether Lactobacillus could inhibit the adhesion capability of S. pseudintermedius to canine corneocytes. For this study, L. salivarius and S. pseudintermedius were isolated from healthy beagle fecal samples and the skin surface of dogs with skin infection, respectively. S. pseudintermedius was co-cultured with L. salivarius to assess the inhibitory effect. For the adhesion assay, corneocytes were collected from healthy beagle ventral abdominal skin. Both bacterial species attached to corneocytes and were assessed in number. As a result, L. salivarius significantly inhibited the growth of S. pseudintermedius in the culture medium. Moreover, L. salivarius reduced attachment of S. pseudintermedius in the adhesion assay. These results suggest that L. salivarius has an inhibitory effect on S. pseudintermedius and may be effectively used in the topical therapy of canine skin infections.

Uncovering the Antibacterial Potential of a Peptide-Rich Extract of Edible Bird's Nest against Staphylococcus aureus

  • The diverse pharmacological properties of edible bird's nest (EBN) have been elucidated in recent years; however, investigations into its antibacterial effects are still limited. In the present study, we explored the antibacterial activity of a peptide-rich extract of EBN against Staphylococcus aureus, a notorious pathogen. The EBN extract (EEE) was prepared by soaking EBN in 80% ethanol for 2 days at 60℃. Biochemical analyses showed that peptides at the molecular weight range of 1.7-10 kDa were the major biochemical compounds in the EEE. The extract exhibited strong inhibition against S. aureus at a minimum inhibitory concentration (MIC) of 125 ㎍/ml and a minimum bactericidal concentration (MBC) of 250 ㎍/ml. This activity could be attributed to the impact of the extract on cell membrane integrity and potential, biofilm formation, and reactive oxidative species (ROS) production. Notably, the expression of biofilm- and ROS-associated genes, including intercellular adhesion A (icaA), icaB, icaC, icaD, and superoxide dismutase A (sodA), were deregulated in S. aureus upon the extract treatment. Our findings indicate a noteworthy pharmacological activity of EBN that could have potential application in the control of S. aureus.
