The essential oil fractions from six plant parts including leaf of Zanthoxylum piperitum and flower of Lindera obtusiloba have revealed to possess resistance inhibitory activity on antibiotic resistant Staphylococcus aureus SA2 when combined with ohloramphenicol (Cm). The combination of Cm and essential oil mixtures showed potent resistance inhibition in the level of 10∼20 $\mu\textrm{g}$/ml.
One subfraction from the hexane fraction of Acri graminei Rhizoma, the E4 fraction which is mainly consisted of acorenone, showed a potential inhibitory activity against chloramphenicol acetyltransferase (CAT) of S. aureus SA2 that is a multidrug-resistant strain to 10 usual antibiotics. The combination therapy of this fraction with chloramphenicol resulted in reduction of the minimal inhibitory concentration from 128 $\mu\textrm{g}$/ml to 8 $\mu\textrm{g}$/ml. The E4 fraction also revealed to prevent the induction of CAT from this strain.

The pKH2 isolated from the multidrug-resistant Staphylococcus aureus SA2 is a 40.98-kb plasmid and mediates resistance to ampicillin, clindamycin, erythromycin, kanamycin, and streptomycin. The 3.4-kb HindIII fragment conferring kanamycin resistance was cloned from the pKH2 into pBluescriptII $KS^+$ and partial sequence determination of that fragment was carried out. Sequence analysis revealed that the kanamycin resistance gene which encoded aminoglycoside 3'-phosphotransferase was linked to the streptothricin resistance gene. But a nonsense mutation was found in the streptothricin resistance gene and this mutation resulted in a truncated protein of streptothricin acetyltransferase. Homology comparison with nucleotide sequence databases revealed that the 3.4-kb HindIII fragment of pKH2 had been derived not from S. aureus but from Gram-negative Campylobacter coli.
This study was designed to assess the possibility of using medicinal plant extracts as β-lactamase inhibitors to control antibiotic-resistant Staphylococcus aureus. The susceptibilities of S. aureus ATCC 15564 (SAWT), ciprofloxacininduced S. aureus ATCC 15564 (SACIP), oxacillin-induced S. aureus ATCC 15564 (SAOXA), and clinically-isolated S. aureus CCARM 3008 (SACLI) to ampicillin were determined in the absence and presence of medicinal plant extracts, including Cleyera japonica (CJ), Carpinus laxiflora (CL), Euphorbia helioscopia (EH), Euscaphis japonica (EJ), Oenothera erythrosepala (OE), and Rosa multiflora (RM). The phenotypic change in the clear inhibition zones around ampicillin disc was observed for SAWT, SACIP, and SAOXA, indicating the production of ampicillinase. Compared to the controls, the MICs of ampicillin against SAWT, SACIP, and SAOXA were decreased from 4 to 0.5 ㎍/mL in the presence of CL, 16 to 4 ㎍/mL in the presence of RM, and 32 to 2 ㎍/mL in the presence of CL, EH, and RM, respectively. The medicinal plant extracts, OE, EJ, and CL, effectively inhibited the β-lactamase activities of SAWT (78%), SACIP (57%), and SAOXA (76%) when compared to the control. This results suggest that the medicinal plant extracts can be used as BLIs to control the antibiotic-resistant S. aureus.
The 2.4-kb cryptic plasmid (pKH8) of multidrug-resistant Staphylococcus aureus SA2 was characterized by complete nucleotide sequencing and homology comparison. pKH8 was found to contain three open reading frames. Protein analysis of pKH8 showed that pKH8 was a multidrug resistance plasmid and mediated resistance to ethidium bromide and quaternary ammonium compounds.
The complete nucleotide sequence of pKH6, a tetracycline-resistance (Tc$^{r}$) plasmid isolated from multi-drug resistant Staphylococcus aureus SA2, has been determined and compared with that of the staphylococcal Tc$^{r}$ plasmid pTl8l. The nucleotide sequences of the two plasmids are in agreement except for 7 nucleotides. All differences are caused by base pair substitutions. Among 6 substitutions, 3 occurred in coding regions. However, only two base substitutions in coding regions resulted in changes of amino acid sequences in two different ORFs of repC and Pre proteins.
Background: Chronic bovine mastitis is linked to biofilm-producing Staphylococcus aureus (bp-Sa) or Staphylococcus coagulase-negative (bp-Scn). Objectives: Bp-Sa and bp-Scn were treated with intramammary preparations of either enrofloxacin HCl·2H2O-dimethyl-sulfoxide-chitosan (enro-C/DMSO/chitosan) or enro-C alone. Their potential to inhibit and degrade biofilm formation in vitro was also assessed. Methods: Milk samples were obtained from the affected quarters in a herd. Phenotypical and genotypical identifications as biofilm-producing Staphylococcus species were carried out. Enro-C/DMSO/chitosan and enro-C alone were assessed to determine their in vitro efficacy in interfering with biofilm formation and their bactericidal effects. A prolonged eight-day treatment with a twice-daily intramammary insertion of 10 mL of enro-C/DMSO/chitosan or enro-C alone was set to evaluate the clinical and bacteriological cures on day 10 in 15 cows per group and the biofilm-inhibiting ability. Results: Fifty-seven percent of the isolates were identified as Staphylococcus spp., of which 50% were bp-Sa, 46% bp-Scn, and 4% Staphylococcus pseudintermedius. One hundred percent of the S. aureus isolated and 77% of Staphylococcus coagulase-negative were biofilm producers. In both groups, the icaA and icaD biofilm-producing genes were identified. The experimental preparation could inhibit biofilm formation, degrade mature biofilms, and have well-defined microbicidal effects on planktonic and biofilm bacteria. The respective clinical and bacteriological cure rates were 100% and 80% for enro-C/DMSO/chitosan and 41.7% and 25% for enro-C alone. Conclusions: Enro-C/DMSO/chitosan eliminates bp-Sa and bp-Scn from cases of chronic bovine mastitis.

This report is to observe the rate of multiple resistance to various antibiotics among staphylococcus aureus and E. coli which were isolated with high frequency at our department during 1969. 1. One hundred fifty two strains(20.6%) resistant to four antibiotics multiple resistant was the highest incidence among 739 strains of Staphylococcus aureus. 2. Regarding the multiple antibiotic resistant of Staphylococcus aureus, 75strains(10.1%) to SA. PC. TC the three antibiotics resistant, have been observed, with the highest incidence, and next to this, 69 strains(9.3%) to SA. PC. SM. TC the four autibiotics resistant, and 68 strains(9.2%) to SA. PC. SM. CM. TC. SM. the six antibiotics resistant, have been observed in descending order. 3. The case of one hundred twenty seven strains(28.0%) of the six antibiotic multiple resistant among 454 strains of Escherchia coli was the highest incidence. 4. Concerning the multiple antibiotic resistant of Escherichia coli, 70 strains(l5.4%) to SA. SM. CM. TC. EM. FD. the six antibiotics resistant, have been observed, with the highest incidence, and next to this, 59 strains(l3.0%) to SA. SM. CM. TC. EM. FD. AC the seven antibiotics resistant, and 42 strains(9.3%) to SA. SM. CM. TC. EM. FD. KM. AC the eight antibiotics resistant have been observed in descending order.
Once known as a major community-genotype, sequence type (ST) 72 methicillin-resistant Staphylococcus aureus (MRSA) strains have been increasingly identified in hospital-associated infections in Korea. Here, we report the complete genome sequence of SA520 isolated from a patient in Korea.
Plasmids were isolated from 15 tetracycline (Tc) resistant S. aureus. Two small tetracycline resistance plasmids, pKH16 and pKH17, have been isolated from Staphylococcus aureus JY10 and Staphylococcus aureus JY22, respectively and the complete nucleotide sequences of those plasmids have been determined. pKH16 consisted of 4,442 bp and showed high identity to pKH6 (99% matching percentage) isolated in 1989 from S. aureus SA2. pKH17 consisted of 4,441 bp and showed less identity to pKH6 (95% matching percentage) than pKH16. PCR analysis showed that tetK and tetM did not exist in ten large plasmids isolated from ten Tc resistant S. aureus. Twelve Tc resistant S. aureus showed reistance both to Tc and Mn and we might analogize that twelve Tc resistant S. aureus had tetM in their chromosome.
