The effect of five element constitutional acupuncture(Sa-am acupuncture) therapy on the six yin meridians on the high fat diet-induced obese rats (염전보사(捻轉補瀉)를 시행(施行)한 육음경(六陰經) 승격(勝格) 혈위(穴位) 침자(鍼刺)가 고지방식이(高脂肪食餌) 비만(肥滿) 백서(白鼠)에 미치는 영향(影響))

  • Objectives : This research was performed to investigate the effect of five element constitutional acupuncture(Sa-am acupuncture) therapy at acupoints on the Yin meridians on obese rats(Sprague Dawley strain, male) induced by high fat diet. Methods : The control and acupuncture groups were provided with high fat diet, and the acupuncture groups were treated with five element constitutional acupuncture(Sa-am acupuncture) every other day for five weeks. We observed the body weight gain, food intake, riglyceride, total cholesterol, HDL-cholesterol, AST, and ALT. Result : The acupuncture group treated with five element acupuncture on the meridians of spleen, heart, pericardium showed significant decrease of body weight, the groups treated on the meridians of lung, heart, pericardium showed signigicant decrease of food intake. Concentration of HDL-cholesterol was significantly increased in the groups treated on the meridians of spleen, heart, pericardium, liver. Conclusion : Reviewing these experimental results, it appears that the five element constitutional acupuncture(Sa-am acupuncture) therapy at acupoints on the meridians of spleen, heart and pericardium have an efficacy on obesity induced by high fat diet.

Effect of Prostaglandin $E_1$ and Acetazolamide upon Carbonic Anhydrase Activity of Whole Blood in Rat (Prostaglandin $E_1$과 Acetazolamide가 흰쥐 전혈(全血)의 Carbonic Anhydrase 활성에 미치는 영향)

  • This study was undertaken to investigate the influence of prostaglandin $E_1(PGE_1)$ upon the activity of carbonic anhydrase and upon the inhibitory action of acetazolamide on carbonic anhydrase. The heparinized blood was sampled by cardiac puncture from Sprague-Dawley strain rats under ether anesthesia and was hemolysed by adding distilled water 1,000 times the amount of the blood. The activity of carbonic anhydrase of 0.1 ml of the hemolysate was measured by Maren's simplified micro-method. In the first experiment, the 7 rats were used, and the activity was measured by adding 0.1 ml of various concentrations of $PGE_1$(0.5, 1.25, 2.5, 5.0, 10 and $20\;{\mu}g/ml$). In the second experiment, the 6 rats were used and the activity was measured by adding 0.1 ml of $PGE_1(5\;{\mu}g/ml)$ and 0.1 ml of acetazolamide$(6{\times}10^{-7}M/l)$ respectively or simultaneously. Obtained results were as follows: 1) The activity of carbonic anhydrase was significantly inhibited by $PGE_1$ at doses of $0.5{\sim}10\;{\mu}g/ml$, maximally at a dose of $2.5\;{\mu}g/ml$, but inhibition was no more observed at a dose of $20\;{\mu}g/ml$. 2) The activity of the acetazolamide group was significantly less than that of the control group. 3) The activity of the $PGE_1+acetazolamide$ group was significantly less than those of the $PGE_1$ group and the acetazolamide group. It is inferred from the above results that the $PGE_1$ inhibits the activity of carbonic anhydrase dose-dependently and strengthens the inhibitory effect of acetazolamide on carbonic anhydrase.

Effect of Acidosis and Prostaglandin $E_1$ upon Acid Secretion in Isolated Whole Stomach in Rat (흰쥐의 적출한 위(胃)에서 산증(酸症)과 Prostaglandin $E_1$이 위산(胃酸) 분비에 미치는 영향)

  • This study was undertaken to investigate the effect of the acidosis on the gastric acid secretion in the isolated whole stomach of the rat and the effect of prostaglandin $E_1$ on the gastric acid secretion influenced by the acidosis. Twenty-two male albino rats(Sprague-Dawley strain) were used. The isolated whole stomach from each rat was introduced into the Kreb's solution which was continuously gassed with $95%O_2-5%CO_2$ for 1 hour, after irrigation of the lumen with cold physiological saline$(4^{\circ}C)$. Thereafter, each stomach was irrigated again with 5% dextrose solution (pH 7.4, $37^{\circ}C$), and filled with the dextrose solution. All the stomachs with the dextrose solution were divided into 4 groups according to the Kreb's solutions in which each stomach was incubated for 30 min: 1) control group, in the pH 7.4 solution, 2) $PGE_1$ group, in the pH 7.4 solution containing $5\;{\mu}g/ml$ of $PGE_1$, 3) acid group, in the pH 7.0 solution, and 4) $acid+PGE_1$ group, in the pH 7.0 solution containing $5\;{\mu}g/ml$ of $PGE_1$. After incubatory period, the contents of each stomach were collected and centrifuged(1,500 rpm, room temperature) for 15 min. The acid output in the supernatant was determined with 0.012 N NaOH by means of autotitrator(Dosimat, Metrohm Herisau Co.) at pH 7.4. Results obtained were as follows: 1) The acid output of the acid group increased significantly in comparison with the control value. 2) The acid output of the $acid+PGE_1$ group decreased significantly in comparison with the acid group. It is inferred from the above results that the acidosis facilitates the gastric acid secretion and $PGE_1$ inhibits the gastric acid secretion induced by the acidosis.

Effect of Old Antler Extracts on the Galactosamine-induced Hepatotoxicity in Rats (녹각이 Galactosamine에 의해 유도된 간장해에 미치는 영향)

  • This study was conducted to investigate the biochemical efforts of old antler extracts on the enzyme activities in serum of galactosamine (GaIN)-induced rats. Male rats of Sprague-Dawley strain weighing the average of 110$\pm$10g were divided into four groups and each group was fed either AIN-76 diet or modified AIN-76 diet with old antler extracts (water extract, neutral extract, ether extract) for four weeks. CaIN (400mg/ kg of body weight) was injected to CaIN treatment groups intraperitoneally on the same day every week. CaIN treatment decreased growth performance of rats. But dietary supplementation of old antler extracts prevented this decrement. Serum aminotransferase activities were increased by CaIN treatment, but this increment was reduced by feeding the old antler extracts, and the effect of water extract was the most prominent of the old antler extracts. Activities of lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) in serum of CaIN treatment groups were higher than those of control groups and were decreased by old antler extracts supplementation.

Efficiency of ATP Synthesis and Impairment of Glucose Tolerance in the NIDDM-Prone Rat

  • This study was designed to determine whether genetic defects in the efficiency of ATP synthesis existed in the NIDDM-prone BHE/cdb rat and to determine whether these defects caused the development of glucose intolerance. Thyroxine treatment provided an excellent clue as to the nature of the genetic defects in this rat. The characteristics of hyperhyroid and control Sprague-Dawley(SD) and BHE/cdb rats were studied. Hyperthyroidism was induced through the addition of thyroxine($T_4$) to the diet(2mg/kg of diet). Active proton conductances and passive proton conductances were tested. Mitochondria from hyperhyroid BHE/cdb rats were less efficient iii active proton conductances than mitochondria from hyperhyroid SD rats. It showed that decreased efficiency of ATP synthesis in the BHE/cdb rat was probably related to defects in active proton conductance, Indicating aberrant FoATPase. The levels of $F_1F_0$ATPaseATPase activity were tested. Mitochondria from hyperthyroid BHE/cdb rats were less active than mitochondria from hyperthyroid SD rats. This may be an attribute of aberrant F$_1$ATPase and may contribute to the BHE/cdb strain s characteristic of reduced ATP synthesis efficiency. Glucose tolerances were tested. BHE/cdb rats were profoundly affected by thyroxine, whereas SD rats were less so. It showed that the diabetes phenotype in BHE/cdb rats was related to defects in thyroxine-induced uncoupling. These results showed the decreased efficiency of ATP synthesis due to genetic defects in $F_1F_0$ATPase had relevance to the characteristic of impaired glucose tolerance in the NIDDM-prone BHE/cdb rat.

Effects of Fly Maggot Extracts on the Liver and Plasma Lipid in Rat Fed High-Fat Diets (고지방식이 유도된 흰쥐의 혈액지질 및 간에 관한 파리유충 추출물의 효과)

  • The bioactive effects of ethanol extracts from fly maggot (ME) on reduction of plasma lipids levels in rats fed high-fat diets (Expt. Ⅰ), and on liver function recovery of hepatotoxicity rats by intraperitoneal injection of carbon tetrachloride ($CCl_4$) or by orally administration of alcohol (Expt. II) were investigated. In expt. I, twenty seven, male rat SDS(sprague dawley strain) were randomly assigned to three treated groups, including normal control group, HF (group with high fat diets which have no extracts) and HFE (HF plus orally administered doses of ME extract at 5.0 mg/100g of body weight). In expt. II, forty five, male rats (SDS) were randomly assigned to each of the five groups: T1 (control), T2 (intraperitoneal injection of $CCl_4$), T3 (intraperitoneal injection of $CCl_4$ after orally administered with ME), T4 (orally administered with combination of ME and alcohol), T5 (orally administration of ME after orally administered with alcohol). There were significant decreases in plasma (TAG), (TC), (LDL-C) in the HFE group with orally administered doses of ME at 5.0 mg/100g of body weight, respectively, however, the (HDL-C) were significantly increased in HFE group as compared to HF group with high fat diets which have no extracts (p<0.05). The levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), ${\gamma}$-glutamyl transferse(${\gamma}$-GTP) and bilirubin were highest in T2 or T3, and high in order T4 or T5, and lowest in T1 except for bilirubin which has same with T4, T5 (p<0.05). The high recovery of liver damage by $CCl_4$ from the light microscopic appearance was observed in rats (T3) with extracts, and also high in T4 than T5 by orally administrated with alcohol. In conclusion, the ethanol extracts from fly maggot may have a bioactive effects to prevent for human lipids disorder and alcoholic disease.

Anti-Menopausal Effect of Heat-Killed Bifidobacterium breve HDB7040 via Estrogen Receptor-Selective Modulation in MCF-7 Cells and Ovariectomized Rats

  • Menopause is induced by spontaneous ovarian failure and leads to life quality deterioration with various irritating symptoms. Hormonal treatment can alleviate these symptoms, but long-term treatment is closely associated with breast and uterine cancer, and stroke. Therefore, developing alternative therapies with novel anti-menopausal substances and improved safety is needed. In our study, heat-killed Bifidobacterium breve HDB7040 significantly promoted MCF-7 cell proliferation in a dose-dependent manner under estrogen-free conditions, similar to 17β-estradiol. This strain also triggered ESR2 expression, but not ESR1, in MCF-7 cells. Moreover, administrating HDB7040 to ovariectomized (OVX) Sprague-Dawley (SD) female rats reduced estrogen deficiency-induced weight gain, fat mass, blood triglyceride, and total cholesterol levels. It also recovered collapsed trabecular microstructure by improving trabecular morphometric parameters (bone mineral density, bone volume per tissue volume, trabecular number, and trabecular separation) and decreasing blood alkaline phosphatase levels with no significant changes in uterine size and blood estradiol. HDB7040 also significantly regulated the expression of Tff1, Pgr, and Esr2, but not Esr1 in uteri of OVX rats. Heat-killed B. breve HDB7040 exerts an anti-menopausal effect via the specific regulation of ERβ in vitro and in vivo, suggesting its potential as a novel substance for improving and treating menopausal syndrome.

Effects of Feral Haw (Crataegus pinnatifida Bunge) on Hepatic Functional Enzyme Activity in Dyslipidemic Rats (야생 산사(Crataegus pinnatifida Bunge)가 이상지질혈증 흰쥐의 간 기능 대사효소의 활성 변화에 미치는 영향)

  • Haw (Crataegus pinnatifida Bunge) has been used in traditional medicine for treating various ailments such as obesity and digestive trouble in Far East countries, including Korea. The present study was carried out to investigate the effects of feral haw on hepatic functional enzymes in dyslipidemic rats. Four groups of male rats (Sprague Dawley strain) were fed different diets for 5 weeks: NND (normal-nondyslipidemic diet) group, NNDH (normal-nondyslipidemic diet + haw extract) group, CDD (control-dyslipidemic diet) group and DDH (dyslipidemic diet + haw extract) group. ALP (alkaline phosphatase), LDH (lactate dehydrogenase), AST (aspartate aminotransferase) and ALT (alanine aminotransferase) activities were significantly higher in the CDD group than the NND group. However, haw extract supplement significantly reduced hepatic functional enzyme activities compared to the CDD group. Lipid deposition of the DDH group decreased compared to the CDD group. The size of adipose tissue decreased significantly in the DDH group compared to the CDD group. These results suggest that feral haw could be used as a food resource and functional food material.

Effect of Dietary Fibers on Serum and Liver Lipids of Cholesterol-fed Rats (식이성(食餌性) 섬유소(纖維素)가 콜레스테롤 식이(食餌) 흰쥐의 혈청(血淸) 및 간장지질(肝臟脂質)에 미치는 영향(影響))

  • This experiment was designed to investigate the effect of dietary fibers on serum and liver lipids of cholesterol-fed rats. Forty-two male rats of Sprague-Dawley strain weighed $145{\pm}10\;g$ were divided into 6 groups, each group receiving a different diet for 6 weeks, i.e., basal diet, basal diet plus 0.5% cholesterol without fiber, basal diet plus 0.5% cholesterol and 5% pectin, basal diet plus 0.5% cholesterol and 5% agar, basal diet plus 0.5% cholesterol and 5% pectin plus tannic acid mixture and basal diet plus 0.5% cholesterol and 5% tannic acid. The lowest net weight gain and digestibility were found in 5% tannic acid-containing group. The weight of kidney, heart and lung was significant by different, however, those of liver and spleen was not significantly different among the groups tested. GOT and GPT of serum were significantly higher in 0.5% cholesterol-containing group without fiber, whereas those of 5% pectin-containing group were significantly lower. Highest total serum protein content was found in 0.5% cholesterol-containing group without fiber. However, albumin and A/G ratio were not significant. The content of total lipid and cholesterol in serum were not significant by different among the groups studied, whereas crude lipid contents of liver in 5% tannic acid and pectin plus tannic acid-containing groups were significantly lower. Cholesterol content in the liver was significantly lower in 5% tannic acid-containing group. Crude lipid and sterol content of feces were significantly higher in 5% pectin-containing group.

Accumulation of Metallothionein in Rat Liver and Kidney by Cadmium Administration (카드뮴으로 중독된 흰쥐의 간장 및 신장에서의 Metallothionein 합성에 관한 연구)

  • This study was carried out to investigate accumulation of metallothionein(MT) in rat liver and kidney by cadmium administration. After male rats of Sprague-Dawley strain weighing 60$\pm$5g were fed basal diet ad libitum for 4 weeks, two types of experiments were performed. In the first set of experiment, rats were divided into five groups. Control groups was fed basal diet without injection of cadmium. Dose groups of A, B, C and D were i.p. injected 0.625, 1.25, 2.5, 5mg Cd/Kg of body wt, once a day for two days. In the second set of experiment, rats were also divided into five groups. Control group was fed basal diet without injection of cadmium. Number groups of I, II, III and IV were i.p. injected 1, 2, 3, and 4 times every 24hrs, respectively and injection doses were 2.5mg Cd/Kg of body wt. in a day. In the first of experiment, hemoglobin contents in C, D groups were lower than control group. MT concentrations in liver and kidney were increased with increasing Cd injection doses to 2.5mg Cd/Kg of body wt. Liver - SH group values in C, D groups were higher than control group. Hematocrit values did not differ among groups. In the second of experiment, hemoglobin contents and hematocrit values were decreased. MT concentration in liver and kidney were progressively increased with increasing number of Cd injection. In both sets of experiments, liver MT concentrations were higher than kidney.
