랫트를 이용한 정자독성평가 연구 (A Study on the Spermatotoxicity Evaluation in Rats)

  • The present study was carried out to establish several spermatotoxicity test methods. For this purpose we investigated following parameters in the fertility study of DA-125, a new anticancer agent, in rats: testicular spermatid counts, epididymal sperm counts, daily sperm production rate, sperm morphology, and serum testosterone concentration. Motility and velocity of sperms were also measured using non-treated rats. At 0.3 mg DA-125/kg, spermatids per 1g testis and daily sperm production rate per 1g testis were significantly decreased, when compared with those of control group. Several types of abnormal sperms, such as no head, pin head, double head, hook at wrong angle, no tail, and small sperm, were found in both treated and control groups at a low frequency. Serum testosterone concentration at 0.3 mg DA-125/kg was close to the control value. Sperm motility and velocity measured with non-treated rats were in a good agreement with the results of other investigators. In our study established spermatotoxicity test methods can be used as a tool not only for the close examination of the cause of drug- or chemical-induced infertility, but also for the effective evaluation of reproductive toxicity.

Monthly variations in semen parameters in a Sri Lankan population undergoing fertility evaluation

  • Objective: Seasonal variations in semen quality are known to occur in temperate regions, but results regarding tropical areas remain inconclusive. The aim of this study was to determine whether monthly variations in semen parameters are present among men in a tropical region. Methods: Data were retrospectively collected from semen analyses of 3,000 men over a 10-year period, from 2012 to 2022. Analysis of variance and the independent-samples t-test were employed to observe variations in semen parameters throughout the entire period and between months, respectively. Results: The mean±standard deviation sperm concentration was significantly lower in June, at 42.5±31.4 million/mL, compared to other months. The highest sperm concentration was found in March, at 57.8±42.6 million/mL, constituting a mean difference of 15.3 million/mL between the lowest and highest concentrations. The total sperm count displayed a similar pattern of monthly variation, with a difference of 47.2 million between the highest and lowest months. No significant monthly differences were observed in other parameters, such as sperm motility, morphology, and semen volume. Conclusion: Significant monthly variations in sperm concentration and total sperm count were evident in this Sri Lankan population. March, which displayed the highest sperm counts, is in the spring in temperate regions, while the month with the lowest counts, July, is part of the summer. Fluctuations in photoperiod appear to most strongly influence these variations.

정계정맥류로 인한 불임증에 대한 췌장성 Kallikrein의 효능 (Efficacy of Pancreatic Kallikrein on Semen Parameters of Men with Varicocele)

  • It has been reported that oral kallikrein therapy exerts a favourable effect on sperm motility in asthenozoospermic patients. In order to evaluate the efficacy of kallikrein on asthenozoospermia, a total of 20 subfertile male patients with varicocele, whose sperm counts were less than $40{\times}10^6/ml$ and sperm motility was less than 30%, was subjected to this clinical study (Table 1). They were divided into 2 study groups: 1) Varicocelectomy group consisted of 10 patients with varicocele (grade II-III) who underwent varicocelectomy. 2) Kallikrein group was composed of 10 patients with varicocele (grade I) who were given kallikrein orally 600 KU (kallikrein unit) daily divided 3 times after meal for 3 to 9 months. Semen analyses were repeated twice before the study, once a month during the study and twice after the study. Effective results designate that sperm parameters improved more than 30% from the basical levels after varicocelectomy or kallikrein exposure. Sperm counts increased from $32.5{\times}10^5/ml$ to $45.5{\times}10^6/ml$ after varicocelectomy in 3 patients and sperm motility increased from 25% to 38.5% after varicocelectomy in 3 patients. Pregnancy occurred in 2 patients of 3 responders and 1 patient of 7 non-responders 3 to 6 months after varicocelectomy in Varicocelectomy group. Sperm motility increased from 28% to 40.2% after kallikrein treatment in 3 patients. Pregnancy occurred in 2 patients of the 3 responders in Kallikrein group (Tables 2-3). There were no significant changes in volume and morphology in Varicocelectomy group before after varicocelectomy and no significant changes in volume, counts, and morphology before and after kallikrein exposure. No remarkable side effects were noted with kallikrein treatment.

Gelatinized and Fermented Powders of Lepidium meyenii (Maca) Improve Physical Stamina and Epididymal Sperm Counts in Male Mice

  • Lepidium meyenii, known as Maca, is traditionally employed in the Andean region for its supposed properties to improve energy and fertility. In the present study, we investigated the effects of gelatinized and fermented Maca on improvement of physical stamina and epididymal sperm counts, and on blood biochemical parameters related to fatigue and tissue injury: creatine phosphokinase, aspartate transaminase, lactate dehydrogenase, blood urea nitrogen, glucose, total cholesterol and total proteins. Adult male mice was divided at random into two main groups (resting and excercise groups). The excercise group was separated into three subgroups (exercise only, exercise with gelatinized Maca and fermented Maca-treatment groups). Gelatinized or fermented Maca (800 mg/kg) were orally administered for 30 days. All animals in exercise groups were subjected to daily 30-min swimming for 28 days 30 min after Maca treatment. Daily exercise decreased the body weight gain, and fermented Maca further attenuated the body weight increase. Gelatinized and fermented Maca significantly increased the maximum swimming time on 14 and 28 days of treatment (p<0.05), respectively, suggestive of a long-term stamina-enhancing effect of fermented Maca. Both Maca fully or significantly recovered blood parameters of energy as well as muscular and hepatocytic injuries changed by repeated exercise and maximum swimming performance (p<0.01). Moreover, gelatinized and fermented Maca increased epididymal sperm counts 22.0% and 32.0%, respectively. In conclusion, the results indicate potential benefits of Maca for improving both physical stamina by minimizing muscular and hepatic damage and preserving energy during swimming exercise and male reproductive function by increasing epididymal sperm counts.

Styrene-Mediated Oxidative Stresses in Rat Sperm Cells

  • Styrene is a commercially important chemical used mainly in the production of plastics. A toxic effect exerted by styrene exposure may cause infertility, congenital anomalies or death in offspring. Treatment with styrene for 0, 50, 100, and 500 mg/kg for 5 days in Sprague-Dawley rats significantly decreased sperm motilities and sperm counts while sperm abnormalities were significantly increased. To determine the relationship between changes in sperm motilities and roles of reactive oxygen species (ROS), we determined the effect of styrene on ROS production and mRNA expression of antioxidant enzymes in rats. ROS production was enhanced by styrene treatment in a dose-dependent manner. The mRNA expression of catalase and superoxide dismutase (SOD) 2 was strongly suppressed by styrene treatment although SOD1 or glutathione peroxidase (GPX) 4 expressions were not significantly changed. Taken together, these results indicate that styrene may cause toxic effect in rat sperm cells by enhancing oxidative stresses.

  • The objectives of present study is to investigate genetic damage of radiation in mammalian male germ cell and. to establish available screening method for determining genetic hazard by radiation. Several methods were employed to measure the genetic damage of radiation as follows: Sperm head counts, frequency occurrence of sperm with abnormal head shape, fertility, activity of LDH-X, and the induction of unscheduled DNA synthesis (U.D.S.) in male mouse were performed with the passing of time after irradiation by making use of the sequence of event that occurs during spermatogenesis. Sperm head counts and activity of LDH-X in testes were gradually reduced by increased radiation dose and with the passing of the time after irradiation. Frequency occurrence of sperm with abnormal head shape, sterile period, and the induction of unscheduled DNA synthesis were increased by increased radiation dose. It is suggested that since germ cell is a direct reflection of genetic complement, the use of male germ cell is rapid and convenient method for measuring genetic damage by radiation.

  • The aims of the study were to establish a short-term screening test for detecting testicular toxicity of chemicals in rats and to determine whether a 2-week administration period is sufficient to detect testicular toxicity of 2-bromopropane (2-BP) as an example. Male Sprague-Dawley rats were subcutaneously administered with 1000 mg/kg/day of 2-BP or its vehicle for 2 weeks. Ten male rats each were sacrificed on days 3, 7 and 14 after the initiation of treatment. Parameters of testicular toxicity included genital organ weights, testicular sperm head counts, epididymal sperm counts, motility and morphology, and qualitative and quantitative histopathologic examinations. The early histopathological changes observed on day 3 of treatment included degeneration of spermatogonia and spermatocytes, multinuclear giant cells, mature spermatid retention, vacuolization of Sertoli cells, and decreased number of spermatogonia in stages II and V. On day 7 of treatment, atrophy of seminiferous tubules, exfoliation of germ cells, degeneration of spermatogonia and spermatocytes, multinuclear giant cells, mature spermatid retention, vacuolization of Sertoli cells, decreased number of spermatogonia in stages II and V, and decreased number of spermatocytes in stages VII and XII. On day 14 after treatment, a significant decrease in the weights of testes and seminal vesicles was found. Atrophy of seminiferous tubules, exfoliation of germ cells, degeneration of spermatogonia and spermatocytes, mature spermatid retention, vacuolization of Sertoli cells, decreased number of spermatogonia in stages II and V, and decreased number of spermatocytes in all spermatogenic stages were also observed. In addition, a slight non-significant decrease in testicular sperm head counts, daily sperm production rate and epididymal sperm counts was found. The results showed that 2 weeks of treatment is sufficient to detect the adverse effects of 2-BP on male reproductive organs. It is considered that the short-term testicular toxicity study established in this study can be a useful tool for screening the testicular toxic potential of new drug candidates in rats.

토끼 음경해면체평활근 이완 및 10주간 경구투여한 흰쥐의 생식기능에 미치는 KH204의 효과 (Effects of KH204 on the Relaxation Response of Rabbit Corpus Cavernosum and Reproductive Function in Male Rats)

  • KH204가 음경해면체 이완과 생식기능에 미치는 영향을 규명하기 위해 효능을 실험한 결과 토끼의 음경해면체를 $1{\times}10^{-6}M$ pheylephrine으로 수축시킨 후 KH204를 61, 183 및 549 mg/L의 축적용량으로 처리하여 각각 6.8, 10.1 및 16%의 용량의존적인 이완효과를 관찰하였다. 흰쥐를 이용한 생식기능 실험은 KH204를 61, 183 및 549 mg/kg으로 10주간 장기투여에서 생식장기인 고환조직의 중량 및 정자수 증가에 유의한 효과를 나타내었다. 정자 형태검사는 대조군과 비슷한 수치로 나타내어 고농도로 장기투여에도 정자군과 비슷한 수치로 나타내어 고농도로 장기투여에도 정자 기형에 문제가 없음을 확인하였다. 이상의 결과로 KH204가 특별한 부작용이 없이 음경해면체를 농도의존적으로 이완시키는 효과가 있어, 음경발기를 개선시키는 효과뿐만 아니라 고환성 원인 또는 정자생산 기능부전으로 보이는 남성불임에서 또한 효능이 있음을 시사하였다.

Nitric oxide, 8-hydroxydeoxyguanosine, and total antioxidant capacity in human seminal plasma of infertile men and their relationship with sperm parameters

  • Objective: Oxidative stress plays a key role in the pathogenesis of male infertility. But, the adverse effects of oxidative biomarkers on sperm quality remain unclear. This study aimed to investigate the levels of nitric oxide (NO), 8-hydroxydesoxyguanosine (8-OHdG), and total antioxidant capacity (TAC) oxidative biomarkers in seminal plasma and their relationship with sperm parameters. Methods: A total of 77 volunteers participated in the study, including fertile (n = 40) and infertile men (n = 37). NO, 8-OHdG, and TAC levels were measured using the ferric reducing ability of plasma, Griess reagent method and an enzyme-linked immunosorbent assay kit, respectively. Results: The mean values of sperm parameters in the infertile group were significantly lower than those in the fertile group (p< 0.001). The mean 8-OHdG in the seminal plasma of infertile men was significantly higher (p= 0.013) than those of controls, while the mean TAC was significantly lower (p= 0.046). There was no significant difference in NO level between the two groups. The elevated seminal 8-OHdG levels were negatively correlated with semen volume, total sperm counts and morphology (p< 0.001, p= 0.001 and p= 0.052, respectively). NO levels were negatively correlated with semen volume, total sperm counts and morphology (p= 0.014, p= 0.020 and p= 0.060, respectively). Positive correlations between TAC and both sperm count and morphology (p= 0.043 and p= 0.025, respectively) were also found. Conclusion: These results suggested that increased levels of NO and 8-OHdG in seminal plasma could have a negative effect on sperm function by inducing damage to the sperm DNA hence their fertility potentials. Therefore, these biomarkers can be useful in the diagnosis and treatment of male infertility.

야콘 50% 에탄올 추출물의 성인 남성의 정자 수 증가효과 (The Spermatogenic Effect of 50% Ethanol Extracts of Yacon in Healthy Male Volunteers)

  • Male reproductive function seems to have deteriorated considerably in the past 4 to 5 decades. It was observed a significant decline in mean sperm counts from $113{\times}10^6/ml$ in 1940 to $66{\times}10^6/ml$ in 1990; a fall of $0.94{\times}10^6/ml/year$. We reported that Yacon tuber extracts have spermatogenic effects in rat. In the present study, we tested the spermatogenic effect of Yacon tuber extracts in healthy male volunteers. Subjects were assigned randomly to the control group and the Yacon ethanol extracts administered group (each 12 subjects). And, placebo or Yacon tuber extracts (100 ml) were administered two times daily, by oral for 3 months. Sperm numbers, biochemical parameters and hormone levels were recorded before starting administration, then every month. The Yacon tuber extracts administered group showed significant time dependant increases in according to administration period. Especially, the numbers of sperm increased by 54% after 3 months of administration. And, in Yacon tuber extracts administered group, testosterone and estradiol level were significantly higher than placebo group. On the other hands, Yacon tuber extracts didn't show any toxicity in glucose and lipid metabolism and liver and kidney function. The results of the present study suggest that Yacon tuber extract is a possible therapeutic for the treatment of sperm deficiency.
