For identification of four sibling species of the Anopheles hyrcanus complex found in Korea, the 5.8 rDNA-ITS2-28S rDNA region of each species was sequenced and the species-specific primers wee designed The amplified PCR products obtained from each species were analyzed by agarose gel electrophoresis. The result showed a single species- specific band, I.e. 559bp, 432bp, 322bp and 192bp for An. sinensis, An. sp., An. lesteri and An. pullus, respectively. In conclusion, the species-specific PCR primers designed from ITS2 variable regions functioned successfully and specifically, and can be applied as a useful tool for identifying species of the Anopheles hyrcanus complex found in Korea.
In recent years, the knowledge about bifidobacteria has considerably evolved thanks to recent progress in molecular biology. The analysis of the whole genome sequences of 48 taxa of bifidobacteria offers new perspectives for their classification, especially to set up limit between two species. Indeed, several species are presenting a high homology and should be reclassified. On the other hand, some subspecies are presenting a low homology and should therefore be reclassified into different species. In addition, a better knowledge of the genome of bifidobacteria allows a better understanding of the mechanisms involved in complex carbohydrate metabolism. The genome of some species of bifidobacteria from human but also from animal origin demonstrates high presence in genes involved in the metabolism of complex oligosaccharides. Those species should be further tested to confirm their potential to metabolize complex oligosaccharides in vitro and in vivo.

Several species belonging to the Gibberella fujikuroi species (Gf ) complex are commonly associated with rice and corn, not only causing serious diseases, but also producing fumonisins, a group of mycotoxins harmful to animals and humans. To characterize the population structure of the putative fumonisin-producing Gf complex in Korea, we obtained 276 candidate isolates from rice and corn harvested in 2009 and 2010 by diagnostic polymerase chain reaction with several specific primer sets. Phylogenetic trees were constructed using multilocus sequences (combined RPB2 and EF1A, totaling 1.6 kb) from these isolates. Among the 135 isolates from rice, F. fujikuroi (teleomorph: G. fujikuroi; 59.3%) and F. proliferatum (G. intermedia; 13.3%) were predominant, followed by F. concentricum (5.9%). Additionally, twenty-five (18.5%) rice isolates belonged in a distinct subclade of F. commune, a non-member of the Gf complex. In contrast, F. verticillioides was the most predominant species (38.3%) among the 141 corn isolates, and followed by F. fujikuroi (27.7%), F. proliferatum (14.9%), F. subglutinans (7.1%), and F. concentricum (2.8%). A single mating type (MAT1-1) was found predominantly among the Gf complex isolates examined. Possible distinct subclades were detected within the populations of F. fujikuroi and F. proliferatum; however, this needs further confirmation. This is the first reported population-level characterization of putative fumonisin-producing Gf complex associated with rice and corn in Korea.

The formation of brown colonies due to phenol oxidase activity on classic agar media containing natural material extracts of Helianthus annuus or on medium containing L-3,4-dihydroxyphenylalanine has been used to identify Cryptococcus species complex. In this study, various natural materials were used to develop a modified medium and to identify five major serotypes of Cryptococcus species complex. Serotypes A, D, and A/D were pigmented on medium using Perilla frutescens var. japonica Hara (PerJ agar) after a three-day incubation. Serotypes B and C were pigmented on PerJ agar after four- and five-day incubations, respectively. Growth time and pigmentation of the five serotypes occurred more rapidly on PerJ agar than on the other media. In addition, colony morphology, size, and pigmentation were specific by serotype. In conclusion, PerJ agar should be used in clinic settings to identify Cryptococcus species complex and its serotypes rapidly.
Members of the genus Acinetobacter are recognized as newer pathogens of the nosocomial infection with an increasing frequency in recent years. Strains that belonged to A. calcoaceticus A. baumannii complex (genomic species 1, 2, 3, and 13TU) were major groups associated with nosocomial infection. Phenotypic identification was unreliable and laborious method to classify Acinetobacter strains into 19 genomic species. Rapid and reliable identification of clinical isolates is essential to diagnosis and epidemiology of Acinetobacter. We investigated the suitability of amplified ribosomal DNA restriction analysis (ARDRA) to identify genomic species of 131 Acinetobacter isolates. The 16S rRNA genes (ribosomal DNA) were enzymatically amplified and the amplified PCR products were restricted independently with the enzymes, AluI, CfoI, and MboI. Genomic species of Acinetobacter was classified by the combinations of restriction patterns. The analysis was showed that restriction profiles were characteristic for each genomic species. One hundred fourteen isolates were identified as A. baumannii, twelve were identified as genomic species 13TU, and one was identified as genomic species 3. Four isolates were found to be unknown organisms. All of the isolates which were identified to A. baumannii by phenotypic tests were completely discriminated into A. baumannii and genomic species 13TU by ARDRA. This study demonstrates that ARDRA is a rapid and simple techniques for the identification of Acinetobacter species according to the genomic species.
Quantifying the abundance of Chattonella species is necessary to effectively manage the threats from ichthyotoxic raphidophytes, which can cause large-scale mortality of aquacultured fish in temperate waters. The identification and cell counting of Chattonella species have been conducted primarily on living cells without fixation by light microscopy because routine fixatives do not retain their morphological features. Species belonging to the Chattonella marina complex, including C. marina and C. marina var. ovata, had high genetic similarities and the lack of clear morphological delimitations between the species. To estimate the abundance of C. marina complex in marine plankton samples, we developed a protocol based on the droplet digital polymerase chain reaction (ddPCR) assay, with C. marina complex-specific primers targeting the internal transcribed spacer (ITS) region of the rDNA. Cell abundance of the C. marina complex can be determined using the ITS copy number per cell, ranging from 25 ± 1 for C. marina to 112 ± 7 for C. marina var. ovata. There were no significant differences in ITS copies estimated by the ddPCR assay between environmental DNA samples from various localities spiked with the same number of cells of culture strains. This approach can be employed to improve the monitoring efficiency of various marine protists and to support the implementation of management for harmful algal blooms, which are difficult to analyze using microscopy alone.
The purpose of this study was to establish the more rational and practical basic theory for the landscape planning in the aparment complex. In this study, the actual conditions of exterior space, the residens' satisfaction degree and the correlation between exterior space and the residents' satisfaction defree were investigated and analyzed on the 13apartment complexes in Taegu City. Through the statistical analysis, the main results were obtained as fellows : It was found that the size of green area ratio in the apartment complexes were in the order of Jugong Apt. (Korean Housing Corporation), Siyoung Apt. (City Operation), Minyoung high-rise Apt. (Private Business) and Minyoung low-rise Apt. (Private Business) complex. The highest ratio of footway and Parking area were shown in Minyoung high-rise Apt. complex. The important factors at the exterior space of apartment complex were composed by 6 major factors, importance degrees of which are Visual, Practical, Convenient, Sentimental, Recreational and Spatial factors for teenagers in orders. The residents' satisfaction degrees to the exterior space were highly correlated with the spatil organization, size of green area and quantity of trees, view, management situation, defensibility of outside noise, rest place, water landscapings by turns. In order to get more than the average satisfaction degree 18 exterior space, the green area should be occupied by 35% of the apartment complex area and more than 76% of the green area (i. e. 16% of the apartment complex area) should be planted with trees. In the Taegu City regulation, the ratio of tree composition is proposed for only the number of tree. But it was shown that the satifaction degree are more correlated with the species and afforestation of trees than the nuts her of trees in this study. therefore, the species of tree and the afforestation of tree should be considered when the landscape planning of the apartment complex begins. It was found that the ratio of afforestation to make the more desirable exterior space In the apartment complex shoule be 8 to 2 in the ratio of arbor to shrub. It was also required that 30 species of arbor and 15 species of shrub should be planted for the more desirable landscape of the apartment complex.
In order to estimate effects of air pollution for herbs in the plnus thunbergii forests arrounding onsan industrial complex, this study was surveyed. number of species of herb also had a difference over 10 species by distance at the source of air pollution, and species diversity and evennes were also increasing at remote sites in industrial complex as woody plants. At the vicinity of industrial complex, $SDR_4$ of calamagrostis arundinatios arundinacea., miscanthus sinensis and cymbopogon tortilis var. goeringii were very high, and those of lianas as cocculus trilobus, lonicera japonica and paederia scandens, and phytolacca americana were low, but because of being higher than other sites, it was able to inferred that lianas had a tolerance to air pollution. The composition of biological spectra of herbs showed to type of $H-D_1-R_{2-3}-1$ at near site in the source of air pollution and $H-D_1-R_{1-3}-t$ at the other sites, and the propotion of lians forms were relatively high at the vicinity of industril complex.
Drosophila 의 수컷 앞다리 부절에 존재하는 성즐의 유무는 Sophophora subgens와 Drsophila sub-genus의 분류기준이 되며, 전자의 경우는 종 특이적인 성즐을 가지나 후자에는 일반적으로 존재하지 않는다. 수종 Drosophila 의 성즐에 관하여 비교 조사한 결과 Sophophora subgens내의 D. melanoga-ster subgroup의 경우는 모두 제 1부절에만 존재하였다. 그중 D. melanogaster complex의 D. mauti-tiana 가 약 12.75개, D. simulans는 평균 8.35정도였으며, D. yakuba complex는 D. melanogaster complex에 비하여 다소 적었다. D. melanogaster complex의 종간 교배에 의한 분석으로는 성즐유전양시겡 대한 뚜렷한 방향성을 제시할 수 없었다. Sophophora subgemus의 D. melanogaster species group에 속하는 다른 3종의 경우(D. auralia, D. lutecens 및 D. suzuki)는 성즐의 분포 양식에서 D. immigrans 및 D. virilis)에서는 성즐이 존재하지 않았다.
Drosophila melanogaster와 D. Simulan는 전세계적으로 분포하며, D. Sechellia 는 Africa의 Seychelles 제도에만 서식하는 지역종으로, 모두 D. melanogaster complex에 속하는 동포종이다. 이들 D. melanogaster complex 3종을 대상으로 종간 교배를 통한 잡종을 형성하여 성증(sex-bomb)과 생식궁(genital arch)의 표현형적 유연관계를 부모계통과 상호 비교하였다. 상기 3종들 사이의 교배를 통해 4가지 유형의 잡종 수컷을 얻었으며 이들은 모두 불임이었다. 성즐의 평균 치열 수는 D. melanogaster(OR)가 10.73개, D. sechellia(Ja)는 10.69개였으며, D. simulans($O_9$)에서는 8.35개였다. 종간 잡종의 치열수 분석으로 뚜렷한 성즐의 유전양식을 제시할 수가 없었다. 외부 생식기를 구성하는 각 부속기관 중 분류상 중요 기준이 되는 생식궁은 종 특이적 모양을 띠고 있었으며, 종간 접종 생식궁의 일반적인 형태는 부모종간의 중간형인 mosaic 구조였다.
