  • The chemical analysis and surface chemical properties of biosurfactant formed by Rhodococcus sp. strain IGTS8, which is widely used in biodesulfurization process, in hexadecane/water mixture have been studied. For the chemical analysis, TLC technique was employed. The surface tension, CMC, and emulsion stability of biosurfactant solution were also investigated. The major components of biosurfactant formed by Rhodococcus sp. strain IGTS8 were glucose mycolate and trehalose monomycolate. The CMC of aqueous biosurfactant solution was 0.1 ~0.15 g/100 mL of Water at pH 6.0-6.5 and pH 10~10.5. But the demulsification was faster at pH 10 than at pH 6.3.

  • The antimicrobial activity of Artemisia princeps var. orientalis essential oil against a partial fish pathogenic bacteria was examined. The growth of Aeromonas hydrophila, Aeromonas salomonicida, Aeromonas sorbia, Edwardsiella tarda and Streptococcus sp. (yellowtail) were inhibited at concentrations of 1,000 to 2,000 ppm. The A. salmonicida was inhibited at 1,000 ppm, A. hydrophila, A. sorbia, E, tarda and Streptococcus sp. (yellowtail) at 1,500 ppm, but Vibrio anguillarum, Vibrio ordalii, Edwardsiella ictaluri and Streptococcus sp. (SF 1) were grown on 100-2,000 ppm.

  • Quad band p-HEMT SP6T switch for handset applications was developed. To achieve the low insertion loss and high isolation, trade-off between "On" state and "Off" state was considered by optimization of unit cell. Especially, in case isolation between transmit port and receive port, it was achieved by large capacitors and miniaturization of chip size was achieved by common voltage control and ground using back via process. Designed SP6T switch has size of $950um{\times}100um$ and take into consideration the gate recess error, excellent loss and isolation was confirmed in operating frequency.

  • This study aims to know the effect of water-extracted horseradish Moringa oleifera leaf and KW21 fertilizer combination application on the density of Nannochloropsis sp. It was conducted in the Natural Food Laboratory of State Polytechnique, Tual. The experiment used complete randomized design with 5 treatments and 3 replications: A (25% horseradish leaf extract + 75% KW21), B (50% horseradish leaf extract + 50% KW21), C (75% horseradish leaf extract + 25% KW21), D (positive control of 100% KW21), and E (negative control of 100% horseradish leaf extract). Results showed that Treatment C yielded the best result, both the highest density of Nannochloropsis sp. and suitable harvest time.

  • The examination with 96 bacteria isolates for antibiotics susceptibility was resulted in exploring many antibiotics resistance isolates from the diseased fishes. Vibrio sp. isolates were demonstrated over 90% resistances for Penicillin (P), Amoxicillin (Amc), Erythromycin (I), Colistin (CL). Streptomycin (S). Novobiocin (NV) and Neomycin(N), these isolates were, however, appeared over 80% susceptibilities for Norfloxacin (Nor), CE and UB. In Edwardsiella tarda case, 90 to 100% high resistance was observed for P, Doxycycline (Do), Tetracyclin (Te), Oxytetracycline (T), E, CL, Sulfamethoxasoletrimetoprim (SXT), S, Oxolinic acid (OA), W, Neomycin(N), on the contrary, 90 to 100% high susceptibilities was found for Amc, Nor, Ciprofloxacin (Cip), Orbifloxacin (ORB), Enrofloxacin (ENR), Flumeguine (UB) and NA. CL, Pefloxacin (PEF),S, Flumeguine (UB), OA, NA, NV N was exhibited 90 to 100% resistances for Streptococcus sp., on the other hand, 100% susceptible to AMC and 80% susceptible to Do, Te, ENR and UB was recognised. Lastly, Photobacterium damsela subsp. piscicida was showed 100fe susceptible to Amc and 86% susceptible to NOR, CIP ENR and UB. As a consequence, fish bacterial pathogens isolated from Kyeongnam area, especially Tongyeong-si, Geoje-si, and Goseong-gun, were showed highly resistant to a variety of antibiotics available in the field.

Occurrence of Plant-parasitic Nematodes in Major Potato Production Areas and PCR Identification of Root-knot Nematodes (감자 주산지 식물기생선충 분포와 뿌리혹선층의 PCR 동정)

  • This study was conducted to get basic information on the occurrence of plant-parasitic nematodes for the establishment of nematode management strategy in major potato production areas in Korea. Nationwide soil collection was done in 11 areas of Cheju, Yesan, Gimchun, Goryoung, Hong chun, Pyungchang, Gimjae, Milyang, Namwon, Gangnung, and Inje in 2004-2005. Root-hot nematode juveniles(J2) were detected in 30 samples among the 50 samples. The average density was 12-69 J2/100cc soil. Pratylenchus sp., Helicotylenchus sp., Ditylenchus sp., Tylenchus sp., and Tylenchorhynchus sp. were also detected in various locations, however, their densities were very low. Root-knot nematode females were collected from tomato roots inoculated with the potato field soils for PCR-RFLP identification. The females from Cheju, Milyang, and Goryung showed PCR products of 500 bp. And the Dra I restriction enzyme digestions showing 290 bp and 230 bp fragments confirmed their identity as Meloidogyne hapla.

Report on the Occurrence of Perkinsus sp. in the Manila Clams, Ruditapes philippinarum in Korean

  • Five species of intertidal clams including Ruditapes philippinarum, Tegillarca granosa, Solen strictus, Heteromacoma irus, and Coecella chinensis were tested for the presence of the protozoan parasite, Perkinsus sp. using fluid thioglycollate medium (FTM) fortified with antibiotics and histological techniques. Each individual clam was placed in a test tube filled with 10ml FTM, placed in totally dark place, and incubated over a week. After incubation the clam tissues were stained with Lugol's iodine solution and examined under a light microscope to find out any hypnospores of Perkensus sp. in the tissues. Cross-sections of the clams were also embedded in paraffin, sliced to 3um, and stained with Harry's hematoxylene and Picro eosine to observe the presence of tomont or trophozoites. Perkinsus sp. were found in the presence of tomont or trophozoites. Perkinsus sp. were found in the tissues of R. philippinarum collected from Kangjin and Wando, along the south coast of Korea. However, Perkinsus sp. was not found in four other species of clams nor R. philippinaurm collected from Kimnyong and Waido in Cheju. A size-dependent Perkinsus sp. infection was found in R. philippinarum collected rom Kangjin and Wando the clams smaller than 15mm in shell width do not exhibit and Perkinsus sp. while other clams greater than 20mm in shell width exhibit almost 100% infection. To determine the number of Perkinsus sp. in the clams, FTM cultured clam tissues were digested with 2M NaOH solution and the number of hypnospores in the tube were counted. The number of hypnospores counted from the tissues indicated that each Manila clam contains 100,000 to 3,500,000 Perkinsus cells or 20,000 to 1,000,000 cells per gram tissue wet weight. The results of cell counts also suggests that such a high occurrence of Perkinsus sp. in the clam may cause mortality, as already reported from other studies of Perkinsus spp.

Phylogenetic Diversity of Bacteria Associated with the Marine Sponges, Spirastrella abata and Cinachyrella sp. (해면 Spirastrella abata와 Cinachyrella sp.의 공생 세균의 계통학적 다양성)

  • The bacterial community structure of two marine sponges, Spirastrella abata and Cinachyrella sp. collected from Jeju Island, in April 2009, was analyzed by 16S rDNA-denaturing gradient gel electrophoresis (DGGE). DGGE banding patterns indicated 8 and 7 bands for Spirastrella abata and Cinachyrella sp., respectively. Comparative sequence analysis of variable DGGE bands revealed from 92% to 100% similarity to the known published sequences. The bacterial groups associated with Spirastrella abata were Alphaproteobacteria and Deltaproteobacteria. The bacterial community of Cinachyrella sp. consisted of Alphaproteobacteria, Gammaproteobacteria, and Actinobacteria. Alphaproteobacteria was common and predominant in both the sponge species. Deltaproteobacteria was found only in Spirastrella abata while Actinobacteria and Gammaproteobacteria were found only in Cinachyrella sp. The results revealed that though the common bacterial group was found in both the sponges, the bacterial community profiles differed between the two sponge species obtained from the same geographical location.

  • The vegetative nuclear divisions in hyphae and chromosome numbers were studied with the aid of Giemsa-HCl techniques from 10 strains of Fusarium oxysporum. The entire nuclear division process occurred within an intact nuclear envelope like other fungus. The results confirmed that 2 strains(F. oxysporum S Hongchun D2, F. oxysporum S Jinyang 4) were n=4; 3 strains(F. oxysporum f. sp. lini KFCC 32585, F. oxysporum f. sp. melongenae KFCC 34743 and F. oxysporum f. sp. raphani) n=5; 2 strains(F. oxysporum f. sp. vasinfectum, and F. oxysporum f. sp. mori KFCC 34742) n=6; 3 strains(F. oxysporum f. sp. cucumerium, F. oxysporum f. sp.niveum, and F. oxysporum f. sp. pisi) n=7.

Isolation and Characterization of the Enterococcus sp. RKY1 for Biosynthesis of Succinic Acid. (숙신산 생합성을 위한 Enterococcus sp. RKY1의 분리와 특성)

  • Succinic acid, valuable $C_4$-dicarboxylic acid as a renewable alternative feedstock, is currently produced commercially by the petrochemical process, but extensive efforts have been devoted to establish the biological process for mass production of succinic acid. In this study, the bioconversion of fumaric acid to succinic acid was investigated. We isolated an Enterococcus sp. RKY1 KCTC 8890P, facultative bacterium, capable of the bioconversion of fumaric acid to soccinic acid very rapidly and efficiently. At batch fermentation, the amount of succinic acid production increased with increase in initial fumaric acid from 40 to 100 g/L. With fumaric acid of 70 g/L, the average specific and volumetric production rate, molar yield were reached up to 0.64 g/g.h, 4.87 g/g.h, and 96.5%, respectively. Maximum concentration of succinic acid of 88.9 g/L was achieved with molar yield of 89% with fumaric acid of 100 g/L in less than 20 hours.
