Decrease of Nematode Population by Introduction of Nematophagous Fungi into The Soil as Affected by Inoculum Concentration and Temperature in Vitro (선충 기생 전적 진균의 접종원 농도와 온도조건에 따른 성충감염 및 집단 감소효과)

  • Five nematophagous fungi were evaluated for their nematicidal effect in vitro on Rhabditis sp. and Meloidogyne hapla in soil. Inocula of Arthrobotrys arthrobotryoides, A. conoides, A. oligospora, Dactylella lobata, and Fusarium oxyaporum were grown in moistened corn-sandy soil and chopped potato-sandy soil media, and incubated at 26$^{\circ}C$ for one week. The prepared inocula were incorporated in autoclaved sandy soil, mixing thoroughly at rates equ-invalent to 1:50, 1:100, 1:200, and 1:400, repectively, before 80g of the mixture carrying 100 Rhabditis sp. was put into petri plates. Nematophagous fungi effectively teduced the popuation of Rhabditis sp. in soil in a week or two following treatment of the incula at concentration of 1:50 and 1:100. The optimum was at $25^{\circ}C$ for nematicidial effect as high as 80-100%. The at the rate of 1:100 prepared incula were incorporated in auto-claved soil, where 100 Juveniles M. hapla were introduced per 80% soil. All fungi infected the M. hapla effectively in soil, caysing more than 90% mortality within one week. This result indicated the potential value of these fungi as promising biocontrol agents.

Antimicrobial Activities of Nano Metal Hybrid Materials against the Microorganisms Isolated from Cucurbit Seeds (나노 금속복합체의 박과 작물 종자 분리균에 대한 항균효과)

  • This study was carried out to test the antimicrobial activities of nano metal hybrid materials produced by plasma technologies (radio frequency-thermal plasma system and direct current sputtering system) against microbes isolated from cucurbit (watermelon, pumpkin, and gourd) seeds. Eight different nano metal hybrid materials and four carriers were tested against five different fungal and ten different bacterial isolates in vitro. Among the tested nano metal hybrid material, Brass/CaCO3 (1,000 ppm) exhibited 100% antimicrobial effect against all the five tested fungi. However, nano metal hybrid material Brass/CaCO3 (1,000 ppm) inhibited only four bacterial isolates, Weissella sp., Rhodotorula mucilaginosa, Burkholderia sp., and Enterococcus sp. at 100% level, and did not inhibited other six bacterial isolates. Nano metal hybrid material graphite-nickel (G-Ni) showed 100% inhibition rate against Rhizopus stolonifer and 52.94-71.76% inhibition rate against four different fungal isolates. Nano metal hybrid material G-Ni did not show any inhibition effects against tested ten bacterial isolates. In summary, among the tested eight different nano metal hybrid materials and four carriers, Brass/CaCO3 showed inhibition effects against five fungal isolates and four bacterial isolates, and G-Ni showed variable inhibition effects (52.94-100%) against five fungal isolates and did not show any inhibition effects against all the bacterial isolates.

Physicochemical and Microbiological Properties of Ginseng-Whey Beverages (인삼 유청음료의 이화학적 및 미생물학적 특성)

  • Ginseng-whey beverages were prepared with rennet whey, ginseng, sweetener, honey and Japanese apricot, inoculated with different strains of lactic acid bacteria or unfermented partly. The samples were stored at 4$^{\circ}C$ or 30$\pm$1$0^{\circ}C$ and then physicochemical and microbiological properties were investigated. The yield of whey was 78.8%. The pH-values reduced and acidities increased during the storage period. The contents of solid-substances, ash and lipid in ginseng-whey beverages were 7.90~8.20%, 0.62~0.66% and 0.16%, respectively. The protein contents of ginseng-whey beverages were 0.42~0.56% and the contents were not changed during the storage period. The lactose contents of fermented beverages were higher than those of unfermented beverages. During the storage period (1~5 weeks), the ranges of D(-) - and L(+)- lactic acid contents in fermented ginseng-whey beverages (17.3~156.1 mg/100g, 347.3~1894.2mg/100g) were higher than those of unfermented ginseng-whey beverages (6.2~82.8mg/100g, 7.1~885.5mg/100g). The contents of total saponin in unfermented sample and fermented sample (Lac. casei sub-sp. casei+Str. salivarius sub-sp. thermophilus) were increased during the storage period. But, those of the fermented sample(Lac. acidophilus+Lac. delbrueckii sub-sp. bulgaricus) were reduced. In the electrophoretic results of ginseng-whey beverages, an $\alpha$-lactalbumin and a $\beta$-lactoglobulin bands were shown apparently and there were no changes observed during the storage period. During the storage period (1~3 week) the coliform was not detected and total plate counts and psychrotrophs were increased according to the storage period.

Effects of Yeast Addition as Starter on Fermentation of Kimchi (효모 Starter 의 첨가가 김치발효에 미치는 영향)

  • The purpose of this study is to investigate the effects of yeast addition as starter on kimchi fermentation. The strains used as starter were Saccharomyces sp. YK-17 and Saccharomyces fermentati YK-19 isolated from kimchi, grew under anaerobic condition and low temperature, which showed the acid and base resistances. Chemical and microfloral changes, as well as the sensory properties of starter added kimchi during fermentation were compared with the control fermented without starter. The acidity of kimchi juice was lower and pH was higher in starter added kimchi than the control. Particularly addition of S. fermentati YK-19 prolonged the optimally fermented period (pH 4.0, acidity $0.6{\sim}0.8%$) up to more than 63%. The content of lactic acid, the major non-volatile organic acid in kimchi, was increased rapidly followed by S. sp. YK-17 and S. fermentati YK-19 group. The microfloral changes were found a little different among the samples. Among the microorganisms, Leuconostoc sp. and Lactobacillus sp. showed highest change, and Streptococcus sp. and Pediococcus sp. showed ralatively low change. The growth of Lactobacillus sp. which was the main acidifing microorganism was inhibited by starter addition, particularly by S. fermentati YK-19. The sensory characteristics of acidic and moldy flavor were significantly reduced by the addition, while fresh flavor was increased in starter added group.

Effect of Additives on the Conidial Viability of Aspergillus sp. PS-104 (Aspergillus sp. PS-104의 분생포자 생활력에 미치는 첨가제 효과)

  • A fungus, Aspergillus sp. PS-104, with the high phosphate-solubilizing activities was isolated from Korean upland soil and formulated into a solid powder type with various additives. For the long-time preservation of conidia, some additives (Tween 80, SDS, Triton X-100, glucose, glycerol, corn oil, bio-ceramic, PEG 200, $Cu^{++}$, $Mo^{+++}$, $Fe^{++}$, $Ca^{++}$ and $Zn^{++}$) were supplemented in the rice-cooked hard medium with various concentrations (0, 0.001, 0.01, 0.1, 1.0 and 5.0%). In case of surfactants. the highest relative viability of the Aspergillus sp. PS-104 conidia was recorded nearly to 80% by the addition of 0.01 to 0.1% Tween 80, while 50% in control. The number of conidia were found to be about 100 times higher when treated at 0.01 to 0.1% Tween 80 as compared to control. Relative viability of the conidia was decreased in order of Tween 80 $\geq$ SDS > Triton X-100 during the storage at $25^{\circ}C$. As regards the organic additives, the relative viability of Aspergillus sp. PS-104 conidia was also recorded nearly to 80% by the addition of 1.0% bio-ceramic, and 5.0% glucose and sucrose during the storage at $25^{\circ}C$. In case of metal ions, the relative viability of Aspergillus sp. PS-104 conidia was decreased in order of $Cu^{2+}>Ca^{2+}>Mo^{3+}>Zn^{2+}>Fe^{2+}$ during the storage at $25^{\circ}C$.

Pulp Vitality Evaluation and Comparison with Old Methods Using Pulse Oximetry (맥박산소측정기(pulse oximetry)를 이용한 치수 생활력 측정과 기존 방법과의 비교)

  • Background: This study evaluated pulp vitality of anterior permanent teeth using pulse oximetry (PO), which is already used for monitoring of patient's $SpO_2$ and pulse rates (PR). Also we compared with ice tests and electric pulp test (EPT). Methods: 9 teeth, endodontic treated, were selected as non-vital teeth group. 17 vital teeth were selected as control group. Our aim is to compare sensitivity, specificity, positive predictive value (PPV) and negative predictive value (NPV) of ice test, electric pulp test and pulse oximetry, respectively. Pulse oximetry has two test results, $SpO_2$ and pulse rates. Also we calculated correlation and statistical significances by Pearson's test between EPT and pulse oximetry. Results: Sensitivity, specificity, PPV, NPV were calculated on each tests. Ice test has results of 1.00, 0.89, 0.94 and 1.00, respectively. EPT has results of 0.94, 0.78, 0.89 and 0.88 respectively. $SpO_2$ has results of 0.94, 1.00, 1.00 and 0.90, respectively. PR has results of all 1.00. Conclusions: PO showed relatively accurate, stable and objective results on both $SpO_2$ and PR. Percentage of ability of accurate diagnosis for vital teeth is 94% for ice test, 89% for EPT, 100% for $SpO_2$ and PR. Percentage of ability of accurate diagnosis for non-vital teeth is 100% for ice test, 88% for EPT, 90% for $SpO_2$ and 100% for PR. In additions, PR could be more accurate and significant tests than $SpO_2$.

Production of Algal Biomass and High-Value Compounds Mediated by Interaction of Microalgal Oocystis sp. KNUA044 and Bacterium Sphingomonas KNU100

  • There is growing interest in the production of microalgae-based, high-value by-products as an emerging green biotechnology. However, a cultivation platform for Oocystis sp. has yet to be established. We therefore examined the effects of bacterial culture additions on the growth and production of valuable compounds of the microalgal strain Oocystis sp. KNUA044, isolated from a locally adapted region in Korea. The strain grew only in the presence of a clear supernatant of Sphingomonas sp. KNU100 culture solution and generated 28.57 mg/l/d of biomass productivity. Protein content (43.9 wt%) was approximately two-fold higher than carbohydrate content (29.4 wt%) and lipid content (13.9 wt%). Oocystis sp. KNUA044 produced the monosaccharide fucose (33 ㎍/mg and 0.94 mg/l/d), reported here for the first time. Fatty acid profiling showed high accumulation (over 60%) of polyunsaturated fatty acids (PUFAs) compared to saturated (29.4%) and monounsaturated fatty acids (9.9%) under the same culture conditions. Of these PUFAs, the algal strain produced the highest concentration of linolenic acid (C18:3 ω3; 40.2%) in the omega-3 family and generated eicosapentaenoic acid (C20:5 ω3; 6.0%), also known as EPA. Based on these results, we suggest that the application of Sphingomonas sp. KNU100 for strain-dependent cultivation of Oocystis sp. KNUA044 holds future promise as a bioprocess capable of increasing algal biomass and high-value bioactive by-products, including fucose and PUFAs such as linolenic acid and EPA.

Production of Enantioselective Lipase from Acinetobacter sp. SY-01 (Acinetobacter sp. SY-01로부터 Enantioselective Lipase의 생산)

  • Lipase from Acinetobacter sp. SY-01 plays an important role enzyme that products chiral drug. We investigated optimum condition for mass production of Acinetobacter sp. SY-01 lipase. Addition of among the different oils to medium. olive oil was optimal for enzyme production. When 0.2% olive oil was added as a carbon source, the production of lipase was increased to a maximum. The optimum pH and temperature were pH 7 and $30^{\circ}C$. In the presence of $Fe^{2+}$ and $Ca^{2+}$, the lipase activity was dramatically enhanced by 280% and 160%, respectively. SY-01 lipase was stable in the most of the DMSO among organic solvents. The addition of triton-X 100 increased the SY-01 lipase by 100-fold. The optimum composition of medium for production of the enzyme was 0.8% yeast extract, 0.2% olive oil, 0.4% triton X-100+40% DMSO. 0.1% $NH_4Cl$, 0.4% $K_2HPO_4$ 3.9% $NaH_2PO_4$, 0.03% $CaCl_22H_2O$, 0.01% $FeSO_4$$7H_2O$(pH 7.0).

Effects of Dietary Supplementation of Copper-Sulfate and Copper-Soy Proteinate on the Performance and Small Intestinal Microflora in Laying Hens (Copper-Sulfate와 Copper-Soy Proteinate 첨가가 산란계의 생산성과 장내 미생물 균총에 미치는 영향)

  • The objective of this experimental was to investigate the effect of dietary Cu-sulfate and Cu-soy proteinate on productive performance and small intestinal microflora. A total 1,000 Hy-Line Brown laying hens (35 weeks old) were randomly allotted to 1 of 5 dietary treatments: Control, Cu-sulfate 50, 100 (50, 100 ppm Cu supplementation as Cu-sulfate) and Cu-SP 50, 100 (50, 100 ppm Cu supplementation as Cu-soy proteinate). Each treatment was replicated 4 times with fifty birds per replication, housed in 2 birds cages. Fifty birds units were arranged according to randomized block design. Feeding trial lasted 5 weeks under 16L : 8D lighting regimen. Hen day egg production was significantly (P<0.05) higher in Cu treated groups than control. Feed intake, broken and shell-less egg production was not significantly influenced by treatment. Eggshell color, eggyolk color, haugh unit, and eggshell thickness were not significantly influenced by treatment. However, eggshell strength was significantly (P<0.05) greater in Cu treated groups than control. Concentration of copper of liver was significantly (P<0.05) greater in Cu treated groups than control. Concentration of zinc and iron of liver were not influenced by treatments. Population of Cl. perfrigens and Lactobacilli in the small intestinal content were significantly (P<0.05) influenced by treatments. Population of Cl. perfrigens decreased and that of Lactobacilli increased in the Cu supplement groups. In conclusion, dietary Cu sulfate and Cu-soy proteinate similarly improves egg production, eggshell strength, and favors intestinal microbial population of laying hens.

Cercarial shedding of Echinostoma cinetoychis and experimental infection of the cercariae to several kinds of snails (이전고환극구흡충(Echinostoma cinitorchis)의 cercaria 유출 및 수종 어류에의 cercaria 감염 실험)

  • The development of Echinestcma cinetorchis in several snail species reared in laboratory aquaria was observed. The eggs from adult cukes collected from the intestine of rats were cultivated to miracidia, and exposed to Hippeutis sp. snails. Observations were made for cercarial shedding from the exposed snails. The cercariae shed from the snails were again exposed to several species of fresh water snails in order to observe metacercarial formation in the snails and their infectivity to final hosts. The results obtained in this study were as follows: 1. Twenty miracidia were exposed to each snail of Hippeutis sp. About 58.3% of the above snails (7 out of 12) were dead before shedding the cercariae, anti the remainder shed the cercariae for a period of 7 to 9 days before death. 2. Cercarial shedding from the infected snails started from the 25th day after the exposure to mi.acidia, and the total number of cercariae shed per snail was 684 in average (range; 482-904). 3. The sixte of refine developed in the infected Hippeutis sp. snails was 1$, 242{\times}214{\;}{\mu}m$ in average, and the number of rediae per snail was 350 in average (range; 120-510). 4. About 40 to 50 cercariae shed from the Hippeutis sp. snails were each exposed to several species of snails reared in the laboratory. The metacercarial formation was confirmed by dissecting the infected snails, 12 to 16 days after the infection. The infectivity to each snail species was 100% in Hippeutis sp. (recovery rate; 56.7%) and Radix auricuzaria coreana (recovery rate; 66.4%), 66.7% in Physa acute (recovery rate; 37.5%), and 50% in Cipangopaludina sp. (recovery rate; 8.0%), respectively. 5. The swimming cercariae attached first at the cephalo-podial part of the snails and then migrated to the mantle, internal organ s and hemocele areas to form the metacercariae. 6. Adult worms of E. cinetorchis were obtained from the rats infected with the metacercariae encysted in the experimental snails. Summarising the above results, it is suggested that the mud-snail (Cipangopaludina sp.) may play an important role as a source of human infection with E. cinetorchis in Korea, and that several species of fresh water snails are involved in the life cycle as a second intermediate host.
