Quality Characteristics of Seasoned Pork with Fermented Oenothera biennis Juice (달맞이꽃 발효액 첨가 양념 돈육의 품질 특성)

  • The quality characteristics of seasoned pork with sauces containing different concentrations of fermented Oenothera biennis juice such as 5% (5-SP), 10% (10-SP) and 15% (15-SP) during storage for 15 days at $10^{\circ}C$ were investigated. The pH level decreased after storage for 6 days and pH 4.83~4.90 after storage for 15 days in all tested seasoned pork. The titratable acidity increased significantly after 6 days but did not show significant differences between seasoned pork. The total bacteria of seasoned pork were $10^7CFU/g$ (control) and $10^6CFU/g$ (5-SP and 10-SP) after storage for 9 days. The total bacteria showed a lower 1~2 log cycle in seasoned pork with fermented Oenothera biennis juice compared to control. The volatile basic nitrogen contents of seasoned pork were 18.02 mg% (control), 15.41 mg% (5-SP), 14.47 mg% (10-SP), and 17.51 mg% (15-SP) after storage for 15 days at $10^{\circ}C$, respectively. The a value (red) of pork seasoned with fermented Oenothera biennis juice was higher than that of control and decreased during storage. Cooking loss and water holding capacity of 5-SP and 10-SP were better than those of control and 15-SP during storage. The 5-SP showed the best sensory score in taste (4.00), color (4.00), flavor (3.91), and overall acceptability (4.09) among various seasoned pork groups.

  • We constructed the deletion mutants of fission yeast Schizosaccharomyces pombe spNab2 gene that is homologous to poly(A)-binding protein NAB2 in budding yeast Saccharomyces cerevisiae, which plays crucial roles in mRNA 3' end formation and mRNA export from nucleus into the cytoplasm. A null mutant in an $h^+$/ $h^+$ diploid strain was constructed by replacing the spNab2-coding region with an $ura4^+$ gene using one-step gene disruption method. Tetrad analysis showed that the spNab2 is not essential for vegetative growth and mRNA export. However, over-expression of spNab2 cause the severe growth defects and intensive accumulation of poly(A) RNA in the nucleus. Also, the spNab2-GFP fusions were localized mainly in the nucleus. These results suggest that spNab2 is also involved in mRNA export out of the nucleus.

  • The present study was conducted to research some Actinosporeans species in Oligochaetes (Branchiura sowerbyi). From April to May, 2004, Oligochaetes were sampled from the Daewi Reservoir located in Kunsan, Jeonbuk and a drainageway of the fish farm located in Wanju, Jeonbuk. Tow species of Actinosporeans not reported yet in Korea, Raabeia sp. and Triactinomyxon sp., were detected from Oligochaetes. Actinosporeans detected are recorded to Raabeia sp. E and Triactinomyxon sp. F. A total of 200 Oligochaetes were investigated from two sites, and 28 individuals (14%) were found infected: 27 individuals (13.5%) with Raabeia sp. E and one individual (0.5%) with Triactinomyxon sp. F.

Effect of pH on the Production and Characteristics of Protease by Bacillus sp. SH-8 and Bacillus sp. SH-8M (Bacillus sp. SH-8과 Bacillus SP. SH-8M의 Protease 생산 및 특성에 미치는 pH의 영향)

  • The production and the characteristics of protease produced by Bacillus sp. SH-8 and Bacillus sp. SH-8M were investigated under the different pH conditions. Bacillus sp. SH-8 and Bacillus sp. SH-8M showed the maximum activity of protease at 60 hours(70 units/ml) AND 96 houre(50 units/ml) cultivation, respectively, under the alkaline condition(pH 10.2). However, Bacillus sp. SH-8M exhibited the maximum activities in 8 days cultivation at pH 6.9 and in 6 days cultivation at pH 7.7 Bacillus sp. SH-8M showed the protease activity at the pH change from alkaline to neutral condition, whereas Bacillus sp. SH-8 did not. In addition. all the enzymatic characteristics of protease produced by Bacillus sp. SH-8 and Bacillus sp. SH-8M were similar with the regardless of different pH conditions.

Copepods of the genus Asterocheres Boeck, 1859 (Siphonostomatoida: Asterocheridae) from Korean waters

  • Sixteen species of Asterocheres collected from the southern and eastern coasts of Korea are recorded. Two species are redescriptions-Asterocheres aesthetes Ho, 1984 and A. siphonatus Giesbrecht, 1897. The remaining 14 new species described are A. geminus n. sp., A. processus n. sp., A. culicis n. sp., A. eurychelatus n. sp., A. scutellatus n. sp., A. spiniventer n. sp., A. fici n. sp., A. serratus n. sp., A. tridentatus n. sp., A. nodulosus n. sp., A. tetraodontis n. sp., A. dokdoicus n. sp., A. exilis n. sp., and A. zodius n. sp. These new species are compared with other known species of the genus mainly based on the character states of 17 selected characters. These characters include the length/width ratios of the caudal ramus and genital double-somite, the number of postgenital lateral setules or spinules on the genital double-somite of the female, the number of segments of the antennule of both sexes, the position of aesthetasc on antennular segments in the female, the relative length of terminal claw of the antenna, the relative length of the oral siphon, the number of segments and the relative length of the mandibular palp, the presence or absence of inner coxal seta on legs 1 and 4, the relative lengths of setae on the exopod of female leg 5, the presence or absence of sexual dimorphisms of swimming legs, and the presence or absence of a secondary aesthetasc on the male antennule. A key to 22 species of Asterocheres known in Korea waters is provided.

Isolation and characterization of cellulolytic bacteria, Bacillus sp. EFL1, EFL2, and EFP3 from the mixed forest (혼효림으로부터 셀룰로오스분해 박테리아 분리 및 효소학적 특성규명)

  • This study was conducted to isolate the cellulolytic bacteria able to grow on LB- Carboxymethyl cellulose (CMC) agar trypan blue medium from the mixed forest and Larix leptolepis stands. Three bacterial strains with high activity against both CMC and xylan were isolated. Both API kit test and 16S rRNA gene sequence analysis revealed that the three different isolates belong to the gene Bacillus. Therefore, the isolates named as Bacillus sp. EFL1, Bacillus sp. EFL2, and Bacillus sp. EFP3. The optimum growth temperature of Bacillus sp. EFL1, EFL2, and EFP3 were $37^{\circ}C$. The optimum temperature for CMCase and xylanase from Bacillus sp. EFL1 were $50^{\circ}C$. The optimum pH of Bacillus sp. EFL1 xylanase was pH 5.0 but the optimum pH of CMCase from Bacillus sp. EFL1 was pH 6.0. The optimum temperature of CMCase and xylanase from Bacillus sp. EFL2 was $60^{\circ}C$, respectively. The optimum pH of CMCase of Bacillus sp. EFL2 was 5.0, whereas xylanase showed high activity at pH 3.0-9.0. The optimum temperature for CMCase and xylanase of Bacillus sp. EFP3 was $50^{\circ}C$. The optimum pH for CMCase and xylanse was 5.0 and 4.0, respectively. CMCases from Bacillus sp. EFL1, EFL2, and EFP3 were thermally unstable. Although xylanase from Bacillus sp. EFL1 and EFP3 showed to be thermally unstable, xylanase from Bacillus sp. EFL2 showed to be thermally stable. Therefore, Bacillus sp. EFL2 has great potential for animal feed, biofuels, and food industry applications.

  • Relationship between structure and biological activity was performed using the five substance P-related peptides (SPRPs) [mammalian-SP (M-SP), cod-SP (C-SP), trout-SP (T-SP), lungfish-SP (L-SP) and Ranakinin] and four SP analogs [[$His^5$]-SP (5H-SP), [$Gly^5$]-SP (5G-SP), ($Ile^8$)-SP (8I-SP) and ($Trp^8$)-Ranakinin (8W-Ranakinin)]. The circular dichroism (CD) spectra showed that all of the peptides took an unordered structure in buffer solution and artificial liposomes. However, 8W-Ranakinin undergoes conformational changes by being transferred into neutral and acidic liposomes from an unordered structure to more ordered structure. The arterial relaxing effect of the peptides was also studied with guinea-pig aorta (GPA), As a result of the studies, L-SP was about 14-fold more potant than M-SP. The order of potency compared to $EC_{50}$ value was $L-SP{\gg}M-SP>5G-SP{\ge}8I-SP>5H-SP>T-SP$, C-SP, Ranakinin, 8W-Ranakinin.

Isolation of Microorganisms and Development of Microbial Augmentation for Treatment of Paper Mill Wastewater (제지폐수 처리용 미생물의 분리 및 복합 미생물제제의 개발)

  • This study was performed to investigate the effects of microbial augmentation on the biological treatment of paper mill wastewater. Three bacteria (KN11, KN13, KN27) capable of degrading aromatic compounds and a bacterial strain (GT21) producing an extracellular cellulase were isolated from soil and wastewater by selective enrichment culture. Through morphological, physiological, and biochemical taxonomies, isolated strains of KN11, KN13, KN27, and GT21 were identified as Acinetobacter sp., Neisseria sp., Bacillus sp., and Pseudomonas sp. and named Acinetobacter sp. KN11, Neisseria sp. KN13, Bacillus sp. KN27, and Pseudomonas sp. GT21, respectively. For analysis of non-biodegradable and chemical oxygen demand (COD)-increasing matter in a paper mill wastewater, we utilized GC/MS to detect aromatic compounds and their derivatives containing several substituted functional groups. The microbial augmentation, J30 formulated with the mixture of bacteria including Acinetobacter sp. KN11, Neisseria sp. KN13, Bacillus sp. KN27, and Pseudomonas sp. GT21, was used for the treatment of paper mill wastewater. The optimum temperature and pH for COD removal of the microbial augmentation, J30, were $30^{\circ}C$ and 7.5, respectively. For evaluation of the industrial applicability of the microbial augmentation, J30 in the pilot test, treatment efficiency was examined using paper mill wastewater. The microbial augmentation, J30, showed a COD removal rate of 87%. On the basis of the above results, we designed the wastewater treatment process of the activated sludge system.

  • A series of experiments have been made on the mass culture of Phaeodactylum tricornutum, Platymonas sp. and Chlorella sp. in the laboratory. The shortest lag phase was found In the culture of Ph. tricornutum followed by Platymonas sp. and Chlorella sp. As compared to the aeration culture, the stagnant culture, in general, showed long duration of the lag phase, short period of the exponential phase and extremely small daily increment. The relative growth constants of Ph. tricornutum, Chlorella sp. and Platymonas sp. were $0.302{\pm}0.028$, $0.226{\pm}0.013$, and $0.151{\pm}0.008$, respectively The maximum daily increment of the three species and then daily ages are as follows: Ph. tricornutum Maximum daily increment : 47.5, Daily age : 10, Platymonas sp. Maximum daily increment : 5.6, Daily age : 14, Chlorella sp. Maximum daily increment : 21.1, Daily age : 14 Comparing the packed cell volume with a certain number of cells, the largest value was found in the population of Ph. tricornutum followed by Platymenas sp. and Chlorella sp. A straight line relationship exists between the two values, and the magnitude of the relationship coincides well with the size of the cells. The culture of Ph. tricornutum was proved satisfactory for feeding the larvae of bivalves at about 12 days after innoculation and both of Platymenas sp. and Chlorella sp. were about 16 days respectively.

Subacute Toxicity of SP-102 (Sulbactam. Piperacilline) in Rats Administered Intraperitoneally (복합항생제 SP-102(설박탐.픽페라실린)의 랫드 복강내 투여에 의한 아급성 독성)

  • The subacute toxicity of combined antibiotics, SP-102 (Sulbactam.Piperacilline), was examined in S.D.rats. Four groups of rats were administered intraperitoneally with 0, 512, 1280 and 3200 mg/kg/day of SP-102 for 30 days. Hain clinical sign related to the compound was soft stool. The body weight gain was slightly decreased in male rats treated with 1280, 3200 mg/kg and in female rats treated with 1280 mg/kg of SP-102. Water consumption was significantly increased in rats administered with SP-102. There were no dose-related changes of urinalysis, biochemical examination and hematological findings in all the groups treated with SP-102. Gross necropsy and histopathology revealed no evidence of specific toxicity related to SP-102. Our data indicate that no-observed effect level of SP-102 is below 512 mg/kg in male and female rats. Maximum tolerated dose of SP-102 was estimated to be above 3200 ma/kg in this study.
