Effect of Isoflavone Supplementation on Bone Metabolism in Ovariectomized Rats at Different Ages (이소플라본 보충이 다른 나이에 난소를 절제한 흰쥐의 골대사에 미치는 영향)

  • Osteoporosis that is associated with estrogen deficiency in menopause is by far the most common cause of age-related bone loss. Since isoflavone had been reported as a natural substance that minimizes bone loss, we have begun this study to examine the effect of the substance on bone metabolism in ovariectomized rats. Five week-old (n=22) and 25 week-old (n=22) Sprage-Dawley female rats were classified into young (Y) and adult (A) groups. Each group consisted of three subgroups : sham operated group (SH), ovariectomized group (OVX), and isoflavone supplemented group (OVX+ISO 80 mg/kg B.W.). They were fed chow for 9 weeks. The result showed that body weight gain was increased in YOVX in comparison to YSH group, (p<0.05) serum osteocalcin concentration and urinal deoxypyridinoline (DPD) excretion had significantly increased in YOVX more than in YSH group, and significantly decrease in OVX+ISO than in YOVX group (p<0.05). We concluded that soy isoflavones may decrease bone turnover in young rats. However, isoflavone supplement didn't show significant influence on bone metabolism of adult rats.

Analysis of the General and Functional Components of Various Soybeans (여러 종류 콩의 일반 및 기능성 성분 분석)

  • The objective of this study was to analyze the general and functional components of various soybeans, used for producing soy sauce, tofu, bean sprouts, and for cooking with rice. The moisture, crude protein, crude fat, and ash content of soybeans were in the ranges of 5.50~6.16%, 38.49~41.08%, 14.89~21.89%, and 4.89~5.86%, respectively. Mineral and functional components varied by sample and showed no relationship with usage type. Somyeong for bean sprouts had the highest level of Ca (406.36 mg/100 g), and Sinpaldal for soy sauce and tofu had the highest level of Mg (247.79 mg/100 g). However, Jinpum No. 2 for soy sauce and tofu had the lowest level of Ca (199.51 mg/100 g), and Seonhuk had the lowest level of Mg (90.03 mg/100 g) among the soybeans tested. The isoflavone content in soybeans was in the range of 97.54~402.00 mg/100 g. Somyeong for bean sprouts had the highest level and Seonheuk for cooking with rice had the lowest level. Total oligosaccharides were in the range of $5,838.52{\sim}9,345.90{\mu}g/100g$. Sucrose content was approximately 50% of total oligosaccharides in all samples. Raffinose content was $516.28{\sim}806.95{\mu}g/100g$, and stachyose content was $2,047.13{\sim}3,454.10{\mu}g/100g$. Phytosterols including bracassisterol, campesterol, stigmasterol, and ${\beta}$-sitosterol ranged from 19.25~35.34 mg/100 g. ${\beta}$-sitosterol represented 50% of total phytosterols in almost all samples, followed by campesterol, stigmasterol and bracassisterol. The phytic acid content in soybeans tested was around 2%, except for Sinpaldal No. 2 and Jinpum No. 2 at 0.86% and 1.65%, respectively. The dietary fiber of soybeans was in the ranges of 24.20%~29.20%.

Effects of Soy Protein Concentrate and Age on Plasma Lipids and Phospholipid Fatty Acid Patterns in Female Rats (콩 단백 성분 및 연령이 암컷 흰쥐의 혈장 지질 농도와 인지질 지방산 패턴에 미치는 영향)

  • Effects of soy protein concentrate (SPC) containing isoflavone and casein diets on plasma phospholipid (PLs)-fatty acid patterns were investigated in 7-& 40- wk old female rats. Diets containing 16% SPC (soy/young:SY, soy/old: SO) and casein (casein/young : CY, casein/ old: CO) supplemented with 0.5% cholesterol were fed for 4 wks. Fatty acid compositions of plasma PLs were determined by TLC and GLC. Compared to the dietary protein effects, age effects on serum lipids were more profound. The levels of total cholesterol (Chol.), triglyceride, HDL-Chol., (LDL+VLDL)-Chol. and atherogenic index (AI) were higher in older groups (OC & OS) than younger groups (YC &. YS). Soy groups had higher Ell)L-Chol. level and lower (LDL+ VLDL)-Chol. and AI, compared with casein groups. The compositions of C22:0, Cl8:1 $\omega$9 and sum of MUFA in plasma PLs were significantly higher in casein group (CY & CO) than soy group (SY & SO), but those of sum of SFA were higher in soy group. The compositions of C22:0, Cl8:1 $\omega$9, C22:1, Cl8:3$\omega$3 and C22:4$\omega$6 were higher and those of C22:6$\omega$3, sum of $\omega$3, Cl8: 2$\omega$6 C20:4$\omega$6, sum of $\omega$6 and sum of PUFA were lower in plasma PLs of younger rats. The average P/S and $\omega$3/$\omega$6 ratio in older group was higher. The $\Delta$-7 desaturation index (16:0⇒16:1$\omega$7) and $\Delta$-9 desaturation index (18:0⇒18:1$\omega$9) were lower in soy group than casein group, while $\Delta$-6 and $\Delta$-5 desaturation index were not affected by dietary protein. The $\Delta$-4 desaturation index (22:4$\omega$6⇒22:5$\omega$6) were higher and elongation index (20:4$\omega$6⇒22:4$\omega$6) were lower in older group. The ratio of the products of $\omega$3 fatty acid series (Cl8:3) was significantly higher in older group, which indicated that age affected the plasma PUFA metabolism. On the other hand, older rats had higher serum cholesterol level compared with younger rats. Taken together, these changes in fatty acid composition might cause minimal changes in tile membrane fluidity induced by the increase serum cholesterol level.

  • Bacillus sp. SP-KSW3 is an auxothroph bacteria that is being used for starter in fermentation. Physico-chemical characteristics, enzyme activities, ACE inhibitor and antimutagenicity in fermented soybean (Kanjang) inoculated with Bacillus sp. SP-KSW3 starter was investigated for the ripening duration of fermentation. Tyrosinase and ACE showed 7% higher activity degree on the Kanjang maturated fermented 2 years with Bacillus sp. SP-KSW3 (Type I) than test field than Kanjang maturated 2 years (control). For antimutagenicity using S. enterica serovar Typhimurium TA100 against MNNG and NPD showed 35.17% and 28.37% (Type I). Similarly, S. enterica serovar Typhimurium TA98 was used against NPD and NQO showed 25.48% and 21.64% (Type I), respectively. Hydrogen donating ability 2 year for maturing (Type I) appeared most highly in the test eulogy 83.1% which it makes. Daidzin of isoflavone in fermented soybean showed similarly. Genistein was not detected The initial test field for daidzin and genistein contained 3.95 mg/kg and 1.25 mg/kg (Type I), respectively.

Influence of Sowing Date on Seed Yield and Quality of Black Soybean (Glycine max (L.) Merrill cv. C heongja-3ho) in the Southern Paddy Field (남부지역 논에서 파종 시기별 검정콩의 종실 생산량 및 품질 특성)

  • Growth and seed productivity of black soybean (cv. Cheongja-3ho) sown on four different dates were investigated in paddy fields in the southern region to cope with climate change, trends in consumption of soy foods, and the spread of double cropping region. Sowing date of black soybean showed a significant correlation with above-ground growth, seed yield, useful components, etc. When sown in May, the above-ground part was plentiful, while seed yield significantly decreased. On the other hand, when sown in June and July, reproductive growth was vigorous resulting in high seed yield, exceeding 200 kg/10a, and pod injury and seed coat cracking were reduced. Furtherm ore, the isoflavone content of seed increased significantly as the sowing date was delayed. These results suggests that sowing from early June to early July is appropriate. Nevertheless, late June sowing oppears the m ost appropriate for black soybeans in the southern paddy fields, in order to avoid a risk of overlapping with sowing dates of winter crops in the double cropping.

Inhibitory Mechanism of Daidzein on Helicobacter pylori Growth (Helicobacter pylori 의 생육에 대한 Daidzein의 저해 특성)

  • This study was conducted to investigate the inhibitory effects of daidzein against H. pylori and its cholesterol $\alpha$-glucosyltransferase ($CHL{\alpha}GcT$). $CHL{\alpha}GcT$ is responsible for the production of $\alpha$-glucosyl cholesterol which constitutes more than 25% of cell wall lipids in H. pylori, and it has been suggested that it is essential for H. pylori viability. $CHL{\alpha}GcT$ was inhibited by daidzein, in a dose-dependant manner, of which $IC_{50}$ value was $128.5\;{\mu}M$. Daidzein also showed the inhibitory effect toward H. pylori growth by paper disc diffusion assay. Therefore, it is thought that the inhibition of daidzein on $CHL{\alpha}GcT$ was related to its anti-Helicobacter activity.

Effects of Genistein Supplementation on Fatty Liver and Lipid Metabolism in Rats Fed High Fat Diet (고지방식이를 섭취하는 흰 쥐에서 제니스테인 보충이 지방간 및 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of genistein, a kind of soy isoflavones, on fatty liver and lipid metabolism in rats fed high fat diet. Twenty four male Sprague-Dawley rats were divided into four groups by dietary fat and genistein contents then raised for six weeks. The rats(n=6/group) were fed normal fat diet(NOR), high fat diet (HF), high fat with 0.1% genistein(HF+0.1%G) or high fat with 0.2% genistein(HF+0.2%G). Hepatic total lipid, triglyceride, total cholesterol and Serum GPT, as a marker for fatty liver, were significantly increased by high fat diet. Also, serum total lipid, triglyceride, total cholesterol, glucose and insulin concentration, hepatic lipogenic enzyme (fatty acid synthase and malic enzyme) activities were significantly increased by high fat diet. However, hepatic total lipid, triglyceride, total cholesterol and Serum GPT were significantly decreased by genistein intake. Also, genistein supplementation decreased serum total lipid, triglyceride, glucose and insulin concentration, hepatic lipogenic enzyme (fatty acid synthase and malic enzyme) activities. There were no differences by genistein level except for serum insulin. These results suggest that fatty liver induced by high fat diet was caused by increased serum lipid profiles and hepatic lipogenesis, whereas, genistein may be useful in inhibiting of fatty liver by reducing serum lipid profiles and hepatic lipogenesis.

Effects of Calcium and Genistein on Body Fat and Lipid Metabolism in High Fat-induced Obese Mice (칼슘과 제니스테인 섭취가 고지방식이로 유도된 비만 모델 마우스의 체지방과 지질대사에 미치는 영향)

  • The study was conducted to investigate the effects of dietary calcium and soy isoflavone on body fat and lipid metabolism in high fat-induced obesity. Four week old female C57/BL6J mice, known as a good model of diet-induced obesity, were fed low Ca and high fat diet for 6 weeks. After induced obesity, mice were divided into six groups according to diets varying calcium contents (0.1 or 1.5%) and genistein contents (0 or 500 or 1,000 ppm). Body weight, fat pad (perirenal fat and parameterial fat), adipocyte size, serum total lipid and total cholesterol were significantly decreased by both high Ca intake and genistein supplementation. However, the effect of genistein supplementation showed in low Ca-fed groups. Serum LDL-cholesterol and TG were significantly decreased by high Ca intake and genistein supplementation, respectively. In liver, lipogenic enzymes (fatty acid synthase and malic enzyme) activity and TG were significantly decreased by both high Ca intake and genistein supplementation. This inhibitory effect of genistein on lipogenic enzymes showed in low Ca-fed groups. But liver total cholesterol and total lipid were significantly decreased by high Ca intake and genistein supplementation, respectively. Fecal excretion of total lipid, total cholesterol and TG were significantly increased by high Ca intake, not by genistein supplementation. In conclusion, high calcium intake and genistein supplement may be beneficial for suppression of obesity through direct anti-adipogenesis by decreasing fat weight and size and indirect anti-lipo-genesis by inhibiting lipogenic enzymes activity and improving lipid profile.

Genistein Reinforces the Inhibitory Effect of Cisplatin on Liver Cancer Recurrence and Metastasis after Curative Hepatectomy

  • Background: The high recurrence rate after hepatic resection in hepatocellular carcinoma (HCC) is a major obstacle to improving prognosis. The objective of the present study was to explore the function of genistein, a soy-derived isoflavone, in enhancing the inhibitory effect of cisplatin on HCC cell proliferation and on tumor recurrence and metastasis in nude mice after curative hepatectomy. Methods: Proliferation of human HCC cells (HCCLM3) was detected by 3-(4, 5-dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide (MTT) assay. Synergistic effects of genistein and cisplatin were evaluated with the median-effect formula. Nude mice bearing human HCC xenografts underwent tumour resection (hepatectomy) 10 days post implantation, then received intraperitoneal administration of genistein or cisplatin alone or the combination of the two drugs. 33 days after surgery, recurrent tumours and pulmonary metastasis were evaluated individually. MMP-2 level in recurrent tumours was detected by immunohistochemistry and real-time PCR; MMP-2 expression in HCCLM3 was detected by immunocytochemistry. Results: Genistein and cisplatin both suppressed the growth and proliferation of HCCLM3 cells. The two drugs exhibited synergistic effects even at relatively low concentrations. In vivo, mice in the combined genistein and cisplatin group had a smaller volume of liver recurrent tumors and fewer pulmonary metastatic foci compared with single drug treated groups. Cisplatin upregulated the expression of MMP-2 in both recurrent tumours and HCCLM3, while genistein abolished cisplatin-induced MMP-2 expression. Conclusions: Genistein reinforced the inhibitory effect of cisplatin on HCC cell proliferation and tumour recurrence and metastasis after curative hepatectomy in nude mice, possibly through mitigation of cisplatin-induced MMP-2 upregulation.

Photoprotective Effects of Soybean Extract against UV-Induced Damage in Human Fibroblast and Hairless Mouse Model

  • Soy isoflavones have been reported to possess many physiological activities such as antioxidant activity and inhibition of cancer cell proliferation. This study investigated the photoprotective effects of soybean extract in human fibroblast cell line and hairless mice model. Human fibroblast was treated with soybean extract before and after ultraviolet B (UVB; 290-302 nm) irradiation. In the soybean extract treated group, the cells showed better resistance to ultraviolet (UV) than control group. The amount of type I collagen recovered from the soybean treated group was higher than the vehicle group exposed to UV-induced damage. Moreover, increased expression of metalloproteinases-1 as a result of UV irradiation was suppressed by the soybean extract. Female mice were orally administered soybean extract and irradiated with UVB light for 8 weeks. The effects of the soybean extract on the skin appearance, collagen deposition and epidermal thickness in the UV-damaged mouse skin were analyzed using histopathological methods. In soybean extract treated group, the skin had a better morphology than that of the control group. Furthermore, the amount of type I collagen was increased and overexpression of MMP-1 was reduced in the soybean extract group compared to vehicle group. Additionally, up-regulation of pro-inflammatory cytokines induced by UV irradiation was suppressed by dietary soybean extract treatment. It appears that soybean extract had a photoprotective effect, including anti-aging and anti-inflammatory effect, from UV-induced damage in not only human fibroblast, but also hairless mice. We confirmed that these effects were possibly due to promotion of collagen synthesis and inhibition of MMP-1 expression.
