NMR Spectroscopic Assessment of the Structure and Dynamic Properties of an Amphibian Antimicrobial Peptide (Gaegurin 4) Bound to SDS Micelles

  • The structure and dynamics of a 37-residue antimicrobial peptide gaegurin 4 (GGN4) isolated from the skin of the native Korean frog, Rana rugosa, was determined in SDS micelles by NMR spectroscopy. The solution structure of the peptide in SDS micelles was determined from 352 NOE-derived distance constraints and 22 backbone torsion angle constraints. Dynamic properties for the amide backbone were characterized by $^1H-^{15}N $heteronuclear NOE experiments. The structural study revealed two amphipathic helices spanning residues 2-10 and 16-32 and that the helices were connected by a flexible loop. An intraresidue disulfide bridge was formed between residues Cys31 and Cys37 near the C-terminus. The loop region (11-15) connecting the two helices are were slightly more flexible than these helices themselves. From the fact that since there is no contact NOEs between two helices, it is implied that the GGN4 peptide shows an independent motion of both helices which has an angle of about $ 60^{\circ}-120^{\circ}$ from each other.

Self-Diffusion of Hydrophobically End-Capped Polyethylene Oxide Urethane Resin by Using Pulsed-Gradient Spin Echo NMR Spetroscopy

  • Hydrophobically End-capped polyethylene oxide Urethane Resin(HEUR)-associating polymers, HEUR 35(8), HEUR 35(12), and HEUR 35(18), comprise a polyethylene oxide (PEO) having a molecular weight of 35,000 that is end capped with two C$\_$8/H$\_$17/, C$\_$12/H$\_$25/, and C$\_$18/H$\_$37/ alkyl chains, respectively. These associating polymers were synthesized by condensation reactions with polyethylene oxides and alkyl isocyanates. The self-diffusion coefficients of HEUR-associating polymers were measured in aqueous solution by pulsed-gradient spin-echo (PGSE) nuclear magnetic resonance (NMR) spectroscopy. All polymers underwent a decrease in their mean diffusion coefficients as the concentration was increased. However, the dispersion of the diffusion coefficients, ${\beta}$, about the mean fluctuated with changes in concentration. The large dispersion at low concentrations of HEUR 35(8) and HEUR 35(12) is related to the interaction between hydrophobic end groups, and the large dispersion at high concentrations of HEUR 35(18) is correlated with transient network formation. These results are valuable for predicting the associating mechanism of the large aggregates before and after their critical micelle concentration.

NMR Structural Studies on Novel Disintegrin, Saxatilin from Gloydius saxatilis Venom

  • A new disintegrin protein named saxatilin was purified from Korean snake venom (Gloydius saxatilis). Saxatilin is a 73 residue small ploypeptide, which has a primary recognition motif in extracellular matrix, Arg-Gly-Asp (RGD) sequence. Data from inhibition activity assay for the ${\alpha}_v{\beta}_3$ integrin showed that saxatilin showed about 5000-fold higher activity than those of RGD peptides, suggesting that RGD sequence may not be sufficient to induce full cellular function of this site. The solution structures calculated from NMR data were well converged for backbone atoms except RGD loop. The structure revealed that most of tight turns are stabilized by medium range NOE contacts and the RGD motif is located far from the rigid core of the C-terminal domain. The three-dimensional fold and biological function of saxatilin are discussed with those of salmosin, which is a disintegrin protein derived from Agkistrodon halys brevicaudus.

NMR-based structural characterization of transthyretin in its aggregation-prone state

  • Transthyretin (TTR) is an abundant protein in blood plasma and cerebrospinal fluid (CSF), working as a homo-tetrameric complex to transport thyroxine (T4) and a holo-retinol binding protein. TTR is well-known for its amyloidogenic property; several types of systemic amyloidosis diseases are caused by aggregation of either wild-type TTR or its variants, for which more than 100 mutations were reported to increase the amyloidogenicity of TTR. The rate-limiting step of TTR aggregation is the dissociation of a monomeric subunit from a tetrameric complex. A wide range of biochemical and biophysical techniques have been employed to elucidate the TTR aggregation processes, among which nuclear magnetic resonance (NMR) spectroscopy contributed much to characterize the structural and functional features of TTR during its aggregation processes. The present review focuses on discussing the recent advances of our understanding to the amyloidosis mechanism of TTR and to the structural features of its monomeric aggregation-prone state in solution. We expect that the present review provides novel insights to appreciate the molecular basis of TTR amyloidosis and to develop novel therapeutic strategies to treat diverse TTR-related diseases.

O3에 의한 아조염료 Acid Red 27의 산화분해시 구조 변화에 관한 연구 (Studies on the Oxidative Structural Change of Azo Dye Acid Red 27 by Ozone)

  • The structural change of azo dye (Acid Red 27) by oxidation with ozone has been investigated using FT-IR and $^1H$ NMR. The solution pH was observed to decrease during oxidation reaction, which was considered to be due to the generation of several organic and inorganic acids as the result of the decomposition of azo compound. The FT-IR analysis showed that changes of specific absorption bands of Acid Red 27 were observed after ozonation. When azo dye was oxidized by ozone, several new peaks were shown to appear by $^1H$ NMR analysis and the peaks were generally shifted to the direction of up field. This was presumably due to the breakage of benzene ring contained in the molecular structure of Acid Red 27 by the oxidation and the shape of peaks was shown to change according to the reaction time.

Cacao Bean Husk 추출물로부터 Glucosyltransferase 저해 활성 물질의 분리 및 구조 동정 (Purification and Structure Determination of the GTase Inhibitor from Cacao Bean Husk Extract)

  • 충치 예방 물질의 탐색 연구 일환으로, Cacao bean husk(CBH) 추출물로부터 얻어진, gluco-syltransferase(Gtase) 저해 활성 분획을 분리하였 다. 이를 다시 MCI-gel CHP-20와 Sephadex LH 20의 column chromatography법으로 분리, 정제하였으며, 이로부터 얻어진 2개의 GTase 활성 저해 물질에 대한 구조 해석을 행하였다. 분리.정제된 2개의 GTase 저해 활성 물질은 anisaldehyde-$H_2SO_4$ 및 FeCb에서 각각 갈색 및 청색 반응을 하며, 또 TLC상에서 dimer의 flavan-3-ol $R_r$값을 나타내는 단일 물질이었다. NMR(PMR, C CMR) 빛 mass spectrum으로 두 정제 분획에 대 한 화학 구조를 분석한 결과, G Tase 저해능을 나타 내는 단물질은 (-)-epicatechin-($\beta\rightarrow8$)-catechin 으로 결합된 procyanidin B-1 ($C_{30}H_{26}O_{12}$) 및 cate chin의 2량체인 procyanidin B一3($C_{30}H_{26}O_{12}$)이었다.

An NMR study on the intrinsically disordered core transactivation domain of human glucocorticoid receptor

  • A large number of transcriptional activation domains (TADs) are intrinsically unstructured, meaning they are devoid of a three-dimensional structure. The fact that these TADs are transcriptionally active without forming a 3-D structure raises the question of what features in these domains enable them to function. One of two TADs in human glucocorticoid receptor (hGR) is located at its N-terminus and is responsible for ~70% of the transcriptional activity of hGR. This 58-residue intrinsically-disordered TAD, named tau1c in an earlier study, was shown to form three helices under trifluoroethanol, which might be important for its activity. We carried out heteronuclear multi-dimensional NMR experiments on hGR tau1c in a more physiological aqueous buffer solution and found that it forms three helices that are ~30% pre-populated. Since pre-populated helices in several TADs were shown to be key elements for transcriptional activity, the three pre-formed helices in hGR tau1c delineated in this study should be critical determinants of the transcriptional activity of hGR. The presence of pre-structured helices in hGR tau1c strongly suggests that the existence of pre-structured motifs in target-unbound TADs is a very broad phenomenon.

몇가지 페닐 알카놀의 Sodium Dodeylsulfate 수용액 미셀내에서의 가용화 위치 (The Solubilization Site of Some Phenyl Alkanols in Aqueous Sodium Dodecylsulfate Micelle)

  • Sodium dodecylsulfate(SDS) 0.2M수용액 미셀내에 몇 가지 페닐 알카놀$[C_6H_5(CH_2)_nOH;$ 페놀(n=1), 벤질 알코올(n=1), 펜에틸 알코올(n=2), 3-페닐-1-프로판올(n=3)]이 가용화(solubilization)될 때 이들의 가용화 위치를 이차원 이핵 상관 NMR분광법(Two dimensional heteronuclear correlation spectroscopy (2D C-H COSY)으로 조사하였다. 실험 결과 $^1H$-NMR 신호의 적분에 의하여 조사한 이전의 연구결과보다 훨씬 정량적이며 정확한 가용화 위치를 알 수 있었다. 이들이 SDS 미셀 중심의 중간부 메틸렌기에 침투하는 깊이는 ${\alpha}$메틸렌기로부터 6.5~7.0 단위까지 임을 알았다.

Characterization of Vanadium Oxide Supported on Zirconia and Modified with MoO3

  • Vanadium oxides supported on zirconia and modified with MoO₃were prepared by adding Zr(OH)₄powder into a mixed aqueous solution of ammonium metavanadate and ammonium molybdate followed by drying and calcining at high temperatures. The characterization of prepared catalysts was performed using FTIR, Raman spectroscopy and solid-state $^{51}V$ NMR. In the case of a calcination temperature of 773 K, for samples containing low loading of $V_2O_5$, below 15 wt %, vanadium oxide was in a highly dispersed state, while for samples containing high loading of $V_2O_5$, equal to or above 15 wt %, vanadium oxide was well crystallized because the $V_2O_5$ loading exceeded the formation of a monolayer on the surface of $ZrO_2$. The $ZrV_2O_7$ compound was formed through the reaction of $V_2O_5\;and\;ZrO_2$ at 873 K and the compound decomposed into $V_2O_5\;and\;ZrO_2$ at 1073 K, which were confirmed by FTIR spectroscopy and solid-state $^{51}V$ NMR. IR spectroscopic studies of ammonia adsorbed on $V_2O_5-MoO_3/ZrO_2$ showed the presence of both Lewis and Bronsted acids.

Solid-State $^{51}V$ NMR and Infrared Spectroscopic Study of Vanadium Oxide Supported on $TiO_2-ZrO_2$

  • Vanadium oxide catalyst supported on TiO2-ZrO2 has been prepared by adding Ti(OH)4-Zr(OH)4 powder to an aqueous solution of ammonium metavanadate followed by drying and calcining at high temperatures. The char-acterization ofthe prepared catalysts was performed using solid-state 51V NMR and FTIR.In thecase ofcalci-nation temperature at 773 K, vanadium oxide was in a highly dispersed state for the samples containing low loading V2O5 below 25 wt %, but for samplescontaining high loading V2O5 equal to or above 25 wt %, vana-dium oxidewas well crystallized due to the V2O5 loading exceeding the formation of monolayer on the surface of TiO2-ZrO2.The ZrV2O7 compound was formed through the reactionof V2O5 and ZrO2 at 773-973 K, where-as the V3Ti6O17 compound was formedthrough the reaction of V2O5 and TiO2 at 973-1073 K. The V3Ti6O17 compound decomposed to V2O5 and TiO2 at 1173 K, which were confirmed by FTIR and 51V NMR.
