하천 퇴적토에서 분리한 Citrobacter strain SE4-1에 의한 아셀렌산염의 원소상 셀레늄으로의 환원 (Selenite Reduction to Elemental Selenium by Citrobacter Strain SE4-1 Isolated from a Stream Sediment)

  • 아셀렌산염(selenite, ${SeO_3}^{2-}$)을 환원시키는 세균 SE4-1을 복합 산업단지 내 위치한 오염된 하천 퇴적토로부터 분리하였고, 계통분석결과 이 균주는 Citrobacter freundii와 가장 유사하였다. 이 균주는 혐기적 환경에서 아셀렌산염을 원소상 셀레늄(elemental selenium, $Se^0$)으로 환원시켰고, 그 형상은 구형으로 확인되었다. 이러한 고체상의 침전은 셀레늄 산소음이온의 이동성을 줄이고 생물가용성을 줄여, 생태계 독성을 줄이는 잠재성을 제시해 줄 수 있을 것이다.

  • The remote monitoring system of water salinity was assessed in Wando reclaimed land lake during a farming season in 2009. Increasing of water salinity in this lake used to bring about salt damage on rice plant occasionally. At the early stage of the rice growing period, rice growth was not damaged due to enough rainfall with more than 120 mm from the mid-May to the first ten days of June. Data collection using on-site water salinity measuring sensors every 2 hours and real-time transmission in system were carried out for the experiment. We compared the transmitted values from the sensor system with water sample values collected and analyzed by a local technical office. Salt concentrations measured by sensor in real-time monitoring system were available data. The regression equation between rainfall and water salinity was presented as (water salinity after rainfall) = $0.621{\times}$(water salinity before rainfall)${\times}exp(-0.0139{\times}rainfall)$, ($r^2=0.579$, p<0.01). It is suggested that the system is useful for stable farming in the area where farmer use water in reclaimed lakes as an irrigation source.

Cr(VI) Resistance and Removal by Indigenous Bacteria Isolated from Chromium-Contaminated Soil

  • The removal of toxic Cr(VI) by microorganisms is a promising approach for Cr(VI) pollution remediation. In the present study, four indigenous bacteria, named LY1, LY2, LY6, and LY7, were isolated from Cr(VI)-contaminated soil. Among the four Cr(VI)-resistant isolates, strain LY6 displayed the highest Cr(VI)-removing ability, with 100 mg/l Cr(VI) being completely removed within 144 h. It could effectively remove Cr(VI) over a wide pH range from 5.5 to 9.5, with the optimal pH of 8.5. The amount of Cr(VI) removed increased with initial Cr(VI) concentration. Data from the time-course analysis of Cr(VI) removal by strain LY6 followed first-order kinetics. Based on the 16S rRNA gene sequence, strain LY6 was identified as Pseudochrobactrum asaccharolyticum, a species that had never been reported for Cr(VI) removal before. Transmission electron microscopy and energy dispersive X-ray spectroscopy analysis further confirmed that strain LY6 could accumulate chromium within the cell while conducting Cr(VI) removal. The results suggested that the indigenous bacterial strain LY6 would be a new candidate for potential application in Cr(VI) pollution bioremediation.

  • 다양한 지진 규모 및 주파수 특성을 가지는 지반운동에 대하여 사장교에 장착된 준능동 제어시스템의 제어효과를 분석하고 비용효율성을 평가하였다. Dyke 등에 의하여 제시된 벤치마크 사장교 제어문제에 준능동 제어시스템을 설계하였으며, LQG 최적제어기에 기반한 bi-state 제어방법을 적용하였다. 제어시스템의 비용효율성은 제어시스템을 장착하지 않은 교량의 생애주기 비용에 대한 제어시스템을 장착한 교량의 생애주기비용의 비로서 정의하였으며, 손상비용 규모와 준능동 제어장치의 가격을 매개변수로 하여 그 변화에 따른 비용효율성 평가를 수행하였다. 분석 결과, 제어시스템의 경제적 효율성은 준능동 제어장지의 가격에 크게 민감하지 않은 반면, 손상비용 규모에 따라 민감하게 변화하였다. 또한 중진규모의 연약지반과 강진규모의 견고한 지반에 해당하는 지반운동에 대하여 준능동 제어시스템의 비용효율성이 높은 것으로 평가되었다.

Epidemiology and Control of Strawberry Bacterial Angular Leaf Spot Disease Caused by Xanthomonas fragariae

  • Strawberry bacterial angular leaf spot (ALS) disease, caused by Xanthomonas fragariae has become increasingly problematic in the strawberry agro-industry. ALS causes small angular water-soaked lesions to develop on the abaxial leaf surface. Studies reported optimum temperature conditions for X. fragariae are $20^{\circ}C$ and the pathogen suffers mortality above $32^{\circ}C$. However, at the nursery stage, disease symptoms have been observed under high temperature conditions. In the present study, results showed X. fragariae transmission was via infected maternal plants, precipitation, and sprinkler irrigation systems. Systemic infections were detected using X. fragariae specific primers 245A/B and 295A/B, where 300-bp and 615-bp were respectively amplified. During the nursery stage (from May to August), the pathogen was PCR detected only in maternal plants, but not in soil or irrigation water through the nursery stage. During the cultivation period, from September to March, the pathogen was detected in maternal plants, progeny, and soil, but not in water. Additionally, un-infected plants, when planted with infected plants were positive for X. fragariae via PCR at the late cultivation stage. Chemical control for X. fragariae with oxolinic acid showed 87% control effects against the disease during the nursery period, in contrast to validamycin-A, which exhibited increased efficacy against the disease during the cultivation stage (control effect 95%). To our knowledge, this is the first epidemiological study of X. fragariae in Korean strawberry fields.

재사용이 가능한 나노복합재료 Fe3O4-ACCS-Ag의 제조 및 항균 특성 평가 (Investigation of Synthesis and Antibacterial Properties of a Magnetically Reusable Fe3O4-ACCS-Ag Nanocomposite)

  • In this study, Fe3O4-ACCS-Ag nanoparticles (NPs) were successfully synthesized using silica extracted from corn cob ash. The synthesized Fe3O4-ACCS-Ag NPs were characterized using X-ray diffraction (XRD), scanning electron microscopyenergy dispersive X-ray spectroscopy (SEM-EDX), transmission electron microscopy (TEM) and fourier transform infrared spectroscopy (FTIR). In addition, the potential application of Fe3O4-ACCS-Ag NPs as an antibacterial material in water disinfection was investigated using Escherichia coli ATCC 8739 as model bacteria. The antibacterial activity of synthesized composite material showed 99.9% antibacterial effect within 20 min for the tested bacteria. From this experiment, the synthesized Fe3O4-ACCS-Ag nanocomposites also hold magnetic properties and could be easily recovered from the water solution for its reuse. The reused nanocomposites presented the decreasing antibacterial efficiencies with the reuse cycle but the composite used three times still killed 90% of bacteria in 20 min.

Slope stabilization with high-performance steel wire meshes in combination with nails and anchors

  • Slope draperies in soil and rock are a well known method to avoid rockfalls into the roads or onto housings. Common wire mesh or a combination of wire mesh and wire rope nets are pinned to the slope by the means of fully grouted nails or anchors. Most of these installations have not been designed to stabilize the slope, but simply avoid the rocks from bouncing. The combination of soil- or rocknailing with a designable flexible facing system offers the advantage of a longterm stabilization of slopes and can replace other standard methods for slope stabilization. The capability to transfer axial and shear loads from the flexible facing system to the anchor points is most decisive for the design of the stabilization system. But the transfer of forces by mesh as pure surface protection devices is limited on account of their tensile strength and above all also by the possible force transmission to the anchoring points. Strong wire rope nets increase the performance for slope stabilizations with greater distances between nails and anchors and are widely used in Europe. However, they are comparatively expensive in relation to the protected surface. Today, special processes enable the production of diagonally structured mesh from high-tensile steel wire. These mesh provide tensile strengths comparable to wire rope nets. The interaction of mesh and fastening to nail / anchor has been investigated in comprehensive laboratory tests. This also in an effort to find a suitable fastening plates which allows an optimal utilization of the strength of the mesh in tangential (slope-parallel) as well as in vertical direction (perpendicular to the slope). The trials also confirmed that these new mesh, in combination with suitable plates, enable substantial pretensioning of the system. Such pretensioning increases the efficiency of the protection system. This restricts deformations in the surface section of critical slopes which might otherwise cause slides and movements as a result of dilatation. Suitable dimensioning models permit to correctly dimension such systems. The new mesh with the adapted fastening elements have already been installed in first pilot projects in Switzerland and Germany and provide useful information on handling and effects.

The Effect of Seed-borne Mycoflora from Sorghum and Foxtail Millet Seeds on Germination and Disease Transmission

  • The seed-borne mycoflora of sorghum and foxtail millet collected from different growing areas in South Korea were isolated and taxonomically identified using dry inspection, standard blotter and the agar plate method. We investigated the in vitro and in vitro germination rates of disinfected and non-disinfected seeds of sorghum and foxtail millet using sterilized and unsterilized soil. The percent recovery of seed-borne mycoflora from the seed components of sorghum and foxtail millet seeds was determined and an infection experiment using the dominant species was evaluated for seedling emergence and mortality. A higher number of seed-borne fungi was observed in sorghum compared to that of foxtail millet. Eighteen fungal genera with 34 fungal species were identified from the seeds of sorghum and 13 genera with 22 species were identified from the seeds of foxtail millet. Five dominant species such as Alternaria alternata, Aspergillus flavus, Curvularia lunata, Fusarium moniliforme and Phoma sp. were recorded as seed-borne mycoflora in sorghum and 4 dominant species (Alternaria alternata, Aspergillus flavus, Curvularia lunata, Fusarium moniliforme) were observed in foxtail millet. The in vitro and in vitro germination rates were higher using disinfected seeds and sterilized soil. More seed-borne fungi were recovered from the pericarp compared to the endosperm and seed embryo. The percent recovery of seed-borne fungi ranged from 2.22% to 60.0%, and Alternaria alternata, Curvularia lunata and 4 species of Fusarium were isolated from the endosperm and embryo of sorghum and foxtail millet. Inoculation of the dominant seed-borne fungi showed considerable mortality of seedlings. All the transmitted seed-borne fungi might well be a primary source of infection of sorghum and foxtail millet crops.

전남지방에서 쯔쯔가무시병 숙주와 매개체의 서식환경 조사 (Environmental survey on the vectors and hosts of Tsutsugamushi disease in Jeonnam province, Korea)

  • Tsutsugamushi disease or scrub typhus cause by Orientia tsutsugamushi is an endemic disease in Korea. Chigger mites and field rodents play roles in transmission of the disease by the vector and host of the agent. The purpose of this study is to investigate the density of the chigger mites and field rodents due to environmental factors such as temperature, relative humidity, soil thickness and the various vegetations to the 9 field rodent collection sites. The total 62 field rodents was captured by the Sherman collapsible traps from April to October 2009 at the Jangseong of Jeonnam Province, Korea. The trapping rate of the field rodents by the different collecting sites was dominant at subside storage water (24%), bush near by dam (22%), bank around field (20%), followed by 18% of grassy field and surround cattle shed. The distribution of chigger mites by the different collecting sites was the highest at Bush near by dam (28.7%). And the sites of subside storage water, bank around field and surround cattle shed were 20.4%, 18.8%, 16.4%, respectively. On the other hand the collecting sites of stream bank and ridges between rice paddies were not collected. The temperature to the collecting sites was showed $24.1^{\circ}C$ in June and $24.2^{\circ}C$ in October which was higher than April ($10.6^{\circ}C$), whereas lower than May ($25.3^{\circ}C$) and September ($26.8^{\circ}C$). The highest number of mites was collected at $24.2^{\circ}C$ and 46.6% relative humidity in October. The chigger mites and field rodents were highly collected between 18 and 24% at the sites where are loosely in the superficial layers of the soil from 8.0 cm to 10.2 cm. Total 25 species of vegetation were distributed at the collecting sites. In the present study, strong evidence was found that bank around field and grassy field were provided for the prevalence sites of tsutsugamushi disease.

재순환수 주입에 따른 매립장 함수율 변화특성 분석 (Analysis of the Characteristics of the Change in the Moisture Rate of Landfill with Recirculation Water Injection)

  • Analysis of the change in water content and distribution was conducted according to the supply of recirculation water to a landfill. An excavation sample analysis showed that the recirculation water injection zone had water content 8.8% point higher than that of the non-injection zone, after 8 months of operation. And due to the influence of recirculation water supply by vertical wells in injection zones, the water content increases along with depth more clearly than non-injection zone. According to an electrical specific-resistivity survey after 13 months of operation, the water content got higher towards the bottom of the landfill. The water transmission coefficient is 8.72×10-4 cm/sec for injection zones and 3.36×10-5 cm/sec for the intermediate cover layer; analysis shows that the intermediate cover layer may affect the penetration velocity of water supplied by the horizontal injection tube. For the scientific design and operation of re-injection facilities, it was deemed necessary to follow-up research on the residence time and behavior of re-injection water considering the ratio of recirculation water supply in horizontal and vertical tubes, and pitcher coefficient of intermediate and waste layers.
