To examine the effects of amendments on heavy mineral oil degradation, a pilot scale experiment was conducted for over 105days. During the experiment, soil samples were collected and analyzed periodically for the determination of residual hydrocarbon and microbial activities. At the end of the experiment, the initial level of contamination ($6,205{\pm}173mgkg^{-1}$) was reduced by $33{\sim}45%$ in the amendment amended soil; whereas only 8% of the hydrocarbon was eliminated in the non-amended soil. Heavy mineral oil degradation was much faster and more complete in compost amended soils. Enhanced dissipation of heavy mineral oil in compost amended soil might be derived from increased microbial activities (respiration, microbial biomass-C) and soil enzyme activity(lipase, dehydrogenase, and FDA hydrolase) were strongly correlated with heavy mineral oil biodegradaton (P < 0.01).
This study was conducted to figure out the relationships among soil chemical properties and bacterial biomass related to denitrification and sulfur-reducing and the activity of dehydrogenase, and ultimately to consider the usefulness of dehydrogenase activity as a tool for evaluating the dynamics of forest soil ecosystem. Four sites were selected for the collection of soil samples within two regions(Onsan industrial estate as a polluted region and Mt. Mani at Kanghwa island as a clean area) with two forest types (coniferous and deciduous stands). The soils of Mt. Mani showed higher amount of organic matter, total nitrogen and available phosphorus than those collected from Onsan industrial estate, which indicated that the soils were more beneficial for microbial growth than those of Onsan. The dehydrogenase activity was more sensitive than the denitrifying bacteria or sulfur-reducing bacteria since the activity was significantly different between the regions and season while the two bacterial biomass were not significantly different between the two regions. In addition, the dehydrogenase activity showed relatively high correlation coefficients with organic matter(r=0.53, p=0.004), total nitrogen(r=0.41, p=0.008) and C/Ava. P-ratio(r=-0.52, p=0.001), which was thought to be closely related with microbial activity. Thus, the dehydrogenase activity was thought to be a useful index of soil ecosystem dynamics with considering that the technique need to be applied with the same soil texture for the comparison of the activity as other researchers indicated.
Fire can affect microbial community structure of soil through altered environmental conditions, nutrient availability, and biotic source for microbial re-colonization. We examined the influence of fire on chemical properties and soil enzyme activities of soil for 10 months. We also characterized the soil microbial community structure through ester-linked fatty acid analysis(EL-FAME). For this study, we established five burned plots(1*1 m) and 5 unburned plots outside the margin of fire. Soil was sampled three soil cores in a each plots and composited for analysis at 1, 3, 5, 8, and 10 month after fire. The fire caused an increase in soil pH, exchangeable Ca, and Mg, organic matter, available $P_2O_5$ compared to unburned sites. The content of $NH_4-N$ in burned site was significantly higher than that of unburned site and this effect continued for 8 months after fire. There was no difference of $NO_3-N$ content in soil between burned and unburned site. Fire caused no change in acid phosphatase and arylsulfatase activities but $\beta$-glucosidase and alkaline phosphatase activities in burned site were increased compared to unburned site. Microbial biomass as estimated by total concentration of EL-FAMEs in burned sites was significantly higher than that of unburned sites at one month after fire. Burned site decreased the EL-FAMEs indicative of gram-positive bacteria and tended to increase the fatty acid associated with gram-negative bacteria at one and three months after fire. The sum of EL-FAME compound $18:2{\omega}6,9c$ and $18:1{\omega}9c$ as served fungal biomarkers was decreased in burned site compared to unburned site.
The carbon cycle came into the spotlight due to the climate change and forests are well-known for their capacity to store carbon amongst other terrestrial ecosystems. The annual organic carbon of litter production, forest floor litter layer, soil, aboveground and belowground part of plant, standing biomass, net primary production, uptake of organic carbon, soil respiration, etc. were measured in Mt. Worak in order to understand the production and carbon budget of Quercus serrata forest that are widely spread in the central and southern part of the Korean Peninsula. The total amount of organic carbon of Q. serrata forest during the study period (2010-2013) was 130.745 ton C ha-1. The aboveground part of plant, belowground part of plant, forest floor litter layer, and organic carbon in soil was 50.041, 12.510, 4.075, and 64.119 ton C ha-1, respectively. The total average of carbon fixation in plants from photosynthesis was 4.935 ton C ha-1 yr-1 and organic carbon released from soil respiration to microbial respiration was 3.972 ton C ha-1 yr-1. As a result, the net ecosystem production of Q. serrata forest estimated from carbon fixation and soil respiration was 0.963 ton C ha-1 yr-1. Therefore, it seems that Q. serrata forest can act as a sink that absorbs carbon from the atmosphere. The carbon uptake of Q. serrata forest was highest in stem of the plant and the research site had young forest which had many trees with small diameter at breast height (DBH). Consequentially, it seems that active matter production and vigorous carbon dioxide assimilation occurred in Q. serrata forest and these results have proven to be effective for Q. serrata forest to play a role as carbon storage and NEP.
Soil autotrophic bacterial communities play a significant role in the soil carbon (C) cycle in paddy fields, but little is known about how rhizosphere soil microorganisms respond to different long-term (35 years) fertilization practices under double rice cropping ecosystems in southern China. Here, we investigated the variation characteristics of rhizosphere soil RubisCO gene cbbL in the double rice ecosystems of in southern China where such fertilization practices are used. For this experiment we set up the following fertilizer regime: without any fertilizer input as a control (CK), inorganic fertilizer (MF), straw returning (RF), and organic and inorganic fertilizer (OM). We found that abundances of cbbL, 16S rRNA genes and RubisCO activity in rhizosphere soil with OM, RF and MF treatments were significantly higher than that of CK treatment. The abundances of cbbL and 16S rRNA genes in rhizosphere soil with OM treatment were 5.46 and 3.64 times higher than that of CK treatment, respectively. Rhizosphere soil RubisCO activity with OM and RF treatments increased by 50.56 and 45.22%, compared to CK treatment. Shannon and Chao1 indices for rhizosphere soil cbbL libraries with RF and OM treatments increased by 44.28, 28.56, 29.60, and 23.13% compared to CK treatment. Rhizosphere soil cbbL sequences with MF, RF and OM treatments mainly belonged to Variovorax paradoxus, uncultured proteobacterium, Ralstonia pickettii, Thermononospora curvata, and Azoarcus sp.KH33C. Meanwhile, cbbL-carrying bacterial composition was obviously influenced by soil bulk density, rhizosphere soil dissolved organic C, soil organic C, and microbial biomass C contents. Fertilizer practices were the principal factor influencing rhizosphere soil cbbL-carrying bacterial communities. These results showed that rhizosphere soil autotrophic bacterial communities were significantly changed under conditions of different long-term fertilization practices Therefore, increasing rhizosphere soil autotrophic bacteria community with crop residue and organic manure practices was found to be beneficial for management of double rice ecosystems in southern China.
Understanding N mineralization dynamics in soil is essential for efficient nutrient management. An anaerobic incubation experiment was conducted to examine N mineralization potential and N mineralization rate of the organic amendments with different C:N ratio in paddy soil. Inorganic N in the soil sample was measured periodically under three temperature conditions ($20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$) for 90 days. N mineralization was accelerated as the temperature rises by approximately $10%^{\circ}C^{-1}$ in average. Negative correlation ($R^2=0.707$) was observed between soil inorganic N and C:N ratio, while total organic carbon extract ($R^2=0.947$) and microbial biomass C ($R^2=0.824$) in the soil were positively related to C:N ratio. Single exponential model was applied for quantitative evaluation of N mineralization process. Model parameter for N mineralization rate, k, increased in proportion to temperature. N mineralization potential, $N_p$, was very different depending on C:N ratio of organic input. $N_p$ value decreased as C:N ratio increased, ranged from $74.3mg\;kg^{-1}$ in a low C:N ratio (12.0 in hairy vetch) to $15.1mg\;kg^{-1}$ in a high C:N ratio (78.2 in rice straw). This result indicated that the amount of inorganic N available for crop uptake can be predicted by temperature and C:N ratio of organic amendment. Consequently, it is suggested that the amount of organic fertilizer application in paddy soil would be determined based on temperature observations and C:N ratio, which represent the decomposition characteristics of organic amendments.
Organic farming is rapidly expanding worldwide. Crop growth in organic systems greatly depends on the functions performed by soil microbes, and nutrient supply weed suppression by green manure crops input. Four red-pepper production systems were compared: 1) bare ground (conventional system); 2) hairy vetch monoculture; 3) rye monoculture; and 4) hairy vetch-rye mixture. Soil inorganic N reached the peak at 30 DAI and hairy vetch monoculture was the highest ($192mg\;kg^{-1}$) and soil total carbon was fluctuated sporadically during the experiment. Carbohydrate and phenolic compounds in soil kept significantly higher in green manure crops systems from 10 DBI to 30 DAI, however the level was the maximum at 10 DBI (carbohydrate) and 30 DAI (phenolic comounds). Incorporation of green manure crops residue enhanced soil microbial biomass C and N throughout the growing season except that MBN in rye was reduced after incorporation. Green manure crops systems suppressed weed occurrence and, in particular, it was prominent in rye monoculture. Mineral elements composition and production in red-pepper fruits were markedly decreased in green manure crops systems although hairy vetch monoculture has come close to bare ground (NPK-applied). Therefore, it was suggested that higher biomass production should be performed not only to improve soil quality and suppress weeds but to yield suitable red-pepper fruits in green manure crops-based organic farming.
Biomass is originally photosynthesized from inorgainic compounds such as $CO_2$, minerals, water and solar energy. Recent studies have shown that anaerobic bacteria have the ability to convert recalcitrant biomass such as cellullosic or chitinoic materials to useful compounds. The biomass containing agricultural waste, unutilized wood and other garbage is expected to utilize as feed, food and fuel by microbial degradation and other metabolic functions. In this study we isolated several anaerobic, cellulolytic and chitinolytic bacteria from rumen fluid, compost and soil to study their related enzymes and genes. The anaerobic and cellulolytic bacteria, Clostridium thermocellum, Clostridium stercorarium, and Clostridium josui, were isolated from compost and the chitinolytic Clostridium paraputrificum from beach soil and Ruminococcus albus was isolated from cow rumen. After isolation, novel cellulase and xylanase genes from these anaerobes were cloned and expressed in Escherichia coli. The properties of the cloned enzymes showed that some of them were the components of the enzyme (cellulase) complex, i.e., cellulosome, which is known to form complexes by binding cohesin domains on the cellulase integrating protein (Cip: or core protein) and dockerin domains on the enzymes. Several dockerin and cohesin polypeptides were independently produced by E. coli and their binding properties were specified with BIAcore by measuring surface plasmon resonance. Three pairs of cohesin-dockerin with differing binding specificities were selected. Two of their genes encoding their respective cohesin polypeptides were combined to one gene and expressed in E. coli as a chimeric core protein, on which two dockerin-dehydrogenase chimeras, the dockerin-formaldehyde dehydrogenase and the dockerin-NADH dehydrogenase are planning to bind for catalyzing $CO_2$ reduction to formic acid by feeding NADH. This reaction may represent a novel strategy for the reduction of the green house gases. Enzymes from the anaerobes were also expressed in tobacco and rice plants. The activity of a xylanase from C. stercorarium was detected in leaves, stems, and rice grain under the control of CaMV35S promoter. The digestibility of transgenic rice leaves in goat rumen was slightly accelerated. C. paraputrificum was found to solubilize shrimp shells and chitin to generate hydrogen gas. Hydrogen productivity (1.7 mol $H_2/mol$ glucos) of the organism was improved up to 1.8 times by additional expression of the own hydrogenase gene in C. paraputrficum using a modified vector of Clostridiu, perfringens. The hydrygen producing microflora from soil, garbage and dried pelletted garbage, known as refuse derived fuel(RDF), were also found to be effective in converting biomass waste to hydrogen gas.
In Korea, salt stress is one of the major problems limiting crop production and eco-environmental quality in greenhouse soil. The objective of this study was to evaluate the effectiveness of organic residues (Chinese milk vetch, maize stalk, rice straw, and rye straw) for reducing salt activity in greenhouse soil. Organic residues was incorporated with salt-accumulated soil (EC, 3.0 dS $m^{-1}$) at the rate of 5% (wt $wt^{-1}$) and the changes of electrical conductivity (EC) was determined weekly for 8 weeks under incubation condition at $30^{\circ}C$. The EC, microbial biomass carbon (MBC), and water soluble ions in soil was strongly affected by C/N ratio of organic residues. After 8 weeks incubation, the concentration of water soluble $NO_3{^-},\;Ca^{2+}$, and $Mg^{2+}$ was significantly decreased in organic residues having high C/N ratio (maize stalk, rice straw, and rye straw) incorporated soil compared to organic residues having lower C/N ratio (Chinese milk vetch) incorporated soil. The EC value in Chinese milk vetch incorporated soil was higher than control treatment. In contrast, maize stalk, rice straw, and rye straw amended soil was highly decreased the EC value compared to control and Chinese milk vetch applied soil after 4 weeks incubation. Our results indicated that incorporation of organic residues having high C/N ratio (>30) could reduce salt activity resulting from reducing concentration of water soluble ions.
Glomalin has important roles in soil aggregation in agricultural lands including controlled horticultural lands. The objective of this study was to measure total glomalin content of soils treated conventional farming system (CFS), conventional farming system without pesticides (CFSWP), and organic farming system (OFS) for strawberry cultivation under greenhouse in Goseong-gun, Korea. The average concentration of total glomalin in the soils was significantly higher in the OFS ($2.00mg\;g^{-1}$) compared to the CFS ($1.68mg\;g^{-1}$). In addition, soil microbial biomass C content was 4.9 times higher in the OFS ($821mg\;kg^{-1}$) compared to the CFS ($169mg\;kg^{-1}$). Nitrogen uptake rate of strawberry was higher in the OFS (52.4%) than that in the CFS (13.0%). Furthermore, yield of strawberry in OFS ($51Mg\;ha^{-1}$) was significantly higher compared to CFS ($35Mg\;ha^{-1}$).
