지속농업을 위한 생물비료로서의 유용세균관련 식물검정 연구 개관 (Research Trends on Plant Associated Beneficial Bacteria as Biofertilizers for Sustainable Agriculture: An Overview)

  • 생물비료는 아직도 한국에서는 생소한 용어다. 한국에서 생물비료라 함은 식물추출액, 퇴비류-다양한 형태의 미생물 혼합제 등으로 인식되고 있다. 그러나 최근에는 식물영양요소의 흡수나 이용도를 증진시키는 토양미생물 사용으로 언급하기도 한다. 본 개관은 식물성장을 증진시키는 것으로 알려진 PGPR 서로 다른 기작과 실질적 역할에 대하여 검토하였다.

송이 자생군락 토양 내 난배양성 세균군집의 계통학적 특성 (Phylogenetic Characteristics of viable but Nonculturable Bacterial Populations in a Pine Mushroom (Tricholoma matsutake) Forest Soil)

  • 송이 자생군락 토양 내 세균군집의 정량적 평가를 수행한 결과 CFDA 형광염색법을 이용해 직접 계수된 생균수는 $7.4{\pm}1.19{\times}10^8{\sim}1.07{\pm}0.17{\times}10^9cells/g$ soil로 육즙영양배지(nutrient broth, NB)에서 배양된 생균수는 CFDA 계수치의 $5{\sim}8%$로 계수되었으며, $10^{-2}$으로 희석한 NB(DNB)배지에서는 $40{\sim}47%$의 계수치를 나타내었다. 이상의 결과로부터 송이 자생군락 토양내에는 배양이 곤란한 난배양성(viable but non-culturable; VBNC)세균이 다수 존재해 있는 것으로 추정되었다. 송이 자생군락 토양내 세균군집의 계통학적 특성을 검토하기 위해 토양으로부터 직접 DNA를 추출하고 16S rDNA-ARDRA cluster 분석을 통하여 대표 clone의 16S rDNA 염기서열 분석을 수행하였다. 송이 자생군락 토양으로부터 구축된 총 115 clone은 31 ARDRA cluster로 분류되었으며, ${\alpha}-,\;{\beta}-,\;{\gamma}-$ Proteobacteria, Acidobacteria, Actinobacteria 그리고 Firmicutes의 6개 계통군이 확인되었다. 이들 계통군 중 약 85%가 Acidobacteria 계통군에 속하여 압도적인 우점군임이 확인되어 매우 독특한 계통학적 특성을 나타내었다.

  • 논토양(土壤)에 볏짚을 시용(施用)하고 투수조건(透水條件)으로 수도(水稻)를 재배(栽培)하면서 경시적(經時的)으로 질소대식(窒素代識)에 관여(關與)하는 미생물(微生物)의 소장(消長)과 토양성분(土壤成分)의 변화(變化)를 조사(調査)하여 얻어진 결과(結果)는 다음과 같다. 토양중(土壤中) Eh는 경시적(經時的)으로 저하(低下)했으며 볏짚시용구(施用區)의 Eh가 더욱 낮다. pH는 상층(上層)이 하층(下層)보다 약간 높았다. 이가철(二價鐵)은 함량(含量)이 볏짚시용(施用)으로 증가(增加)되었으며 경시적(經時的)으로 증가(增加)하는 경향(傾向)이었다. $NH_4$-N는 생육초기(生育初期)에 볏짚시용구(施用區)가 함량(含量)이 낮고 $NO_3$-N는 거의 검출(檢出)되지 않았다. 탈실균(脫室菌)은 담수초기(湛水初期)에 상층(上層)에서 높았으며 볏짚시용(施用)으로 생육초기(生育初期)에 약간 균수증가(菌數增加)가 있었다. 경시적(經時的)으로 탈질균(脫窒菌)은 감소(減少)하는 경향(傾向)이었으며 질산환원균(窒酸還元菌)은 증가(增加)하였다. 암모니아 산화균(酸化菌)은 상층(上層)의 균수(菌數)가 하층(下層)보다 많았다. 아질산산화균(亞窒酸酸化菌)은 경시적(經時的)으로 감소(減少)하고 볏짚시용(施用)으로 균수(菌數)가 더욱 감소(減少)되었다. 토양중(土壤中) 이가철함량(二價鐵含量)과 탈질균(脫窒菌)과는 부(負)의 상관(相關)이 비교적 높으며 질산환원균(窒酸還元菌)과는 정(正)의 상관(相關)은 있으나 유의성(有意性)은 없었다. 암모니아산화균(酸化菌)과 그 이가철함량(二價鐵含量)과는 일정(一定)한 상관관계(相關關係)를 나타내지 못하고 있으며 $NH_4$-N 함량(含量)과도 뚜렷한 유의상관(有意相關)이 없었다.

Appropriate Soil Heat Treatment Promotes Growth and Disease Suppression of Panax notoginseng by Interfering with the Bacterial Community

  • In our greenhouse experiment, soil heat treatment groups (50, 80, and 121℃) significantly promoted growth and disease suppression of Panax notoginseng in consecutively cultivated soil (CCS) samples (p < 0.01), and 80℃ worked better than 50℃ and 121℃ (p < 0.01). Furthermore, we found that heat treatment at 80℃ changes the microbial diversity in CCS, and the inhibition ratios of culturable microorganisms, such as fungi and actinomycetes, were nearly 100%. However, the heat-tolerant bacterial community was preserved. The 16S rRNA gene and internal transcribed spacer (ITS) sequencing analyses indicated that the soil heat treatment had a greater effect on the Chao1 index and Shannon's diversity index of bacteria than fungi, and the relative abundances of Firmicutes and Proteobacteria were significantly higher than without heating (80 and 121℃, p < 0.05). Soil probiotic bacteria, such as Bacillus (67%), Sporosarcina (9%), Paenibacillus (6%), Paenisporosarcina (6%), and Cohnella (4%), remained in the soil after the 80℃ and 121℃ heat treatments. Although steam increased the relative abundances of most of the heat-tolerant microbes before sowing, richness and diversity gradually recovered to the level of CCS, regardless of fungi or bacteria, after replanting. Thus, we added heat-tolerant microbes (such as Bacillus) after steaming, which reduced the relative abundance of pathogens, recruited antagonistic bacteria, and provided a long-term protective effect compared to the steaming and Bacillus alone (p < 0.05). Taken together, the current study provides novel insight into sustainable agriculture in a consecutively cultivated system.

In vitro 및 토양에서 추출물, 삼출물, 용액에 대한 세균의 화학주성 (Bacterial Chemotaxis to Extracts, Exudates, Solutions in Vitro and Soil)

  • 토양추출물, 인삼삼출물과 용액이 Pseudomonas sp. P. fluorescens 및 Erwinia carotovora에 미치는 화학주성은 토양추출물처리, 삼출물 및 용액은 증류수 보다 이들 세균이동에 영향을 나타냈다. 고무관내 흙속으로 세균의 이동성은 P. fluorescens가 토양추출물처리 삼출물에서 주성을 나타냈으나, P. sp 와 E. caotovora는 주성이 없었다. 세균주성에 미치는 진균의 삼출액은 이동에 영향력을 나타내지 못하였다. 고무관내 흙을 진균으로 접종한 후에 세균의 주성은 Pseudomonas sp. 에서는 주성이 나타나지 않았으나, P. fluorescens와 E. carotovora는 F. solani, F. oxysporum 및 혼합균처리에서는 주성이 나타났다. Alternaria panax는 이들 세균에 대해 주성이 없었으나, E. carotovora는 A. panax에 대해 주성이 있었다. 토양유기물함량은 김포, 강화의 나병지와 건전지에는 낮았고, K함량은 김포의 건전지에 높았다. 세균분포는 괴산의 나병 및 건전지와 강화건전지에 진균은 금산 괴산의 나병 및 건전지에 많이 분포되었다. 방선균은 괴산건전지에 특히 많이 분포하였고 다음이 풍기나병지였다.

유기물원이 다른 퇴비연용이 밭토양의 물리성 및 미생물상 변화에 미치는 영향 (The Effect of Long-term Application of different Organic Material Sources on Soil Physical Property and Microflora of Upland Soil)

  • 본 연구는 밭 토양에서 종류가 상이한 퇴비 연용이 토양 중 물리적 특성과 토양 미생물상 변동에 미치는 영향을 구명하기 위하여 수행하였다. 밭 토양 토성은 양토와 사양토를 공시하여 계분퇴비, 우분퇴비, 분뇨잔사 및 식품오니퇴비등 4종을 사용하였다. 퇴비 시용량은 0, 40, $80Mg\;ha^{-1}$ 수준으로 처리하여 1994년 1997년도까지 4년간 시험한 주요연구결과를 요약하면 다음과 같다. 양토의 용적밀도는 퇴비 무시용구가 $1.49Mg\;m^{-3}$인 반면에 퇴비시용구는 $1.07{\sim}1.32Mg\;m^{-3}$, 사양토는 무시용구 $1.25Mg\;m^{-3}$에서 시용구는 $1.00{\sim}1.20Mg\;m^{-3}$ 수준으로 감소하였다. 재배작물별 용적밀도변화는 옥수수재배는 나지와 비슷하였다. 퇴비시용시 유기물 분해미생물은 $5^{\circ}C$보다 $25^{\circ}C$ 조건에서 배양초기에 균수가 급속히 증가되었고, 토성별로는 양토에서 사양토보다 민감하게 증가되었다. 질소순환에 관련된 미생물의 변화는 암모니아 산화균은 배양초기에 많았고, 탈질균은 1~4주까지 증가되다가 다시 감소되었다. 토성별로는 암모니아 산화균은 양토에서, 탈질균은 사양토에서 $25^{\circ}C$ 조건에서 균수가 증가되었다.

Bacterial Diversity in the Rhizosphere of Halophyte Phragmites communis at the Western Coastal Mudflats of Korea

  • This study investigated the population densities and diversity of heterotrophic bacteria, and the rhizosphere-to-soil ratios (R/S) in the rhizosphere soil of halophyte Phragmites communis at the western coastal mudflats of Korea. The population densities of aerobic heterotrophic bacteria on the rhizosphere soil of P. communis were in the range of $3.3\;{\pm}\;0.9\;{\times}\;10^7\;{\sim}\;1.2\;{\pm}\;0.5\;{\times}\;10^8\;cfu\;g^{-1}$ dry weight (d. wt.). Population densities of amylolytic bacteria ranged from $1.1\;{\pm}\;0.2\;{\times}\;10^6$ to $3.0\;{\pm}\;1.2\;{\times}\;10^6\;cfu\;g^{-1}\;d.\;wt.$, while those of cellulolytic bacteria and proteolytic bacteria ranged from $5.6\;{\pm}\;2.3\;{\times}\;10^6$ to $1.5\;{\pm}\;0.3\;{\times}\;10^7\;cfu\;g^{-1}\;d.\;wt.$ and from $1.4\;{\pm}\;0.3\;{\times}\;10^6$ to $3.5\;{\pm}\;2.3\;{\times}\;10^7 \;cfu\;g^{-1}\;d.\;wt.$, respectively. The R/S ratios ranged from 2.26 to 6.89. Genetic (16S DNA) analysis of fifty-one isolates from the roots of P. communis suggested that the dominant species were closely related to the ${\gamma}$-proteobacteria group (18 clones) and the ${\alpha}$-proteobacteria group (14 clones). We found that halophyte species and mudflat environment both affected the rhizosphere bacterial communities.

Analysis of Bacterial Diversity and Communities Associated with Tricholoma matsutake Fruiting Bodies by Barcoded Pyrosequencing in Sichuan Province, Southwest China

  • Endophytes play an important role in the growth and development of the host. However, the study of endophytes is mostly focused on plants, and reports on bacteria associated with fungi are relatively rare. We studied the bacteria associated with fruiting bodies of Tricholoma matsutake picked from seven main T. matsutake-producing areas in Sichuan, China, by barcoded pyrosequencing. About 8,272 reads were obtained per sample, representing 40 phyla, 103 classes, and 495 genera of bacteria and archaea, and 361-797 operational taxonomic units were observed at a 97% similarity level. The bacterial community was always both more abundant and more diverse than the archaeal community. UniFrac analysis showed there were some difference of bacterial communities among the samples sites. Three bacterial phyla, Proteobacteria, Bacteroidetes, and Firmicutes, were dominant in all samples. Correlation analysis showed there was a significant correlation between some soil properties and bacterial community associated with T. matsutake. This study demonstrated that the bacteria associated with T. matsutake fruiting bodies were diversified. Among these bacteria, we may find some strains that can promote the growth of T. matsutake.

Structure and Diversity of Arsenic-Resistant Bacteria in an Old Tin Mine Area of Thailand

  • The microbial community structure in Thailand soils contaminated with low and high levels of arsenic was determined by denaturing gradient gel electrophoresis. Band pattern analysis indicated that the bacterial community was not significantly different in the two soils. Phylogenetic analysis obtained by excising and sequencing six bands indicated that the soils were dominated by Arthrobacter koreensis and $\beta$-Proteobacteria. Two hundred and sixty-two bacterial isolates were obtained from arsenic-contaminated soils. The majority of the As-resistant isolates were Gramnegative bacteria. MIC studies indicated that all of the tested bacteria had greater resistance to arsenate than arsenite. Some strains were capable of growing in medium containing up to 1,500 mg/l arsenite and arsenate. Correlations analysis of resistance patterns of arsenite resistance indicated that the isolated bacteria could be categorized into 13 groups, with a maximum similarity value of 100%. All strains were also evaluated for resistance to eight antibiotics. The antibiotic resistance patterns divided the strains into 100 unique groups, indicating that the strains were very diverse. Isolates from each antibiotic resistance group were characterized in more detail by using the repetitive extragenic palindromic-PCR (rep-PCR) DNA fingerprinting technique with ERIC primers. The PCR products were analyzed by agarose gel electrophoresis. The genetic relatedness of 100 bacterial fingerprints, determined by using the Pearson product-moment similarity coefficient, showed that the isolates could be divided into four clusters, with similarity values ranging from 5-99%. Although many isolates were genetically diverse, others were clonal in nature. Additionally, the arsenic-resistant isolates were examined for the presence of arsenic resistance (ars) genes by using PCR, and 30% of the isolates were found to carry an arsenate reductase encoded by the arsC gene.

Biological Control of Pythium Damping-off of Bush Okra Using Rhizosphere Strains of Pseudomonas fluorescens

  • A severe damping-off disease of bush okra caused by Pythium aphanidermatum, was diagnosed in plastic houses in Der Attia village, 15 km southwest of El-Minia city, Egypt, during the winter of 2001. Bush okra seedlings showed low emergence with bare patches inside the plastic houses. Seedlings that escaped pre-emergence damping-off showed poor growth, stunting and eventually collapsed. Examination of the infected tissues confirmed only Pythium aphanidermatum, showing its typical intercalary antheridia, and lobulate zoosporangia. P. aphanidermatum was shown to be pathogenic on bush okra under pot and field experiments. Bacteria making inhibition zones against the damping-off fungus P. aphanidermatum were selected. Agar discs from rhizosphere soil of bush okra containing colonies were transferred onto agar plate culture of P. aphanidermatum. After 2 days of incubation, colonies producing clear zones of non-Pythium growth were readily detected. The two bacteria with the largest inhibition zones were identified as Pseudomonas fluorescens. Bush okra emergence(%) in both pot and plastic houses experiments indicated that disease control could be obtained by applying P. fluorescens to the soil or coating the bacteria to the bush okra seeds before sowing. In the plastic houses, application of the bacteria onto Pythium-infested soil and sowing bush okra seeds dressed with bacteria gave 100% emergence. In addition, This was the first reported disease of bush okra by this oomycete in Egypt.
