Effect of Bradykinin on Oxygen Consumption in the Distal Tubule and Cortical Collecting Tubule of Rat (흰쥐 원위세뇨관과 피질집합관의 산소소비량에 대한 Bradykinin의 영향)

  • Infusion of bradykinin (BK) into the renal arteries increases sodium excretion. However, it is not clear whether natriuresis results from the renal hemodynamic effects or from the direct effect on renal tubular sodium transport. Therefore, we examined the effects of BK on the transport-dependent oxygen consumption in the distal tubule (DT) and cortical collecting tubule (CCT) of deoxycorticosterone-treated rats. BK inhibited oxygen consumption in a dose-dependent way with a maximal reduction at $0.1\;{\mu}M$ BK. The inhibitory effect of BK was not present in the absence of sodium or in the presence of ouabain (1 mM). These data imply that the inhibitory effect of BK is restricted to the sodium transport-dependent oxygen consumption. We also investigated the relationship between the effect of BK on oxygen consumption and arachidonic acid metabolism. Mepacrine $(10\;{\mu}M)$, an inhibitor of membrane phospholipases, prevented the inhibitory effect of BK, but indomethacin (0.5 mM) didn't. These results suggest that BK decreases the sodium transport-related oxygen consumption in the rat DT and/or CCT, and that it may be mediated by products of enzymes other than cyclooxygenase.

  • The effects of ethanol on the toxicity of 2, 4-Dichlorophenoxyacetic acid in ICR mice were examined. The results were summerized as follows; 1. The LD$\sub$50/ of 2.4-dichlorphenoxyacetic acid sodium injected intraperitoneally in mice was approximately 367 mg/kg. When the animals were administered with ethanol, 2 ml/kg or 4 ml/kg the LD$\sub$50/ of 2.4-D sodium were decreased to 338 mg/kg or 32 mg/kg, respectively. 2. In the acute experimental group, spontaneous motor activity in the ANIMEX system exhibited dose-dependent decrease in mice administered with 2.4-D sodium and ethanol. 3. However, in subacute experimental group, spontaneous motor activity in the ANIMEX system exhibited dose-dependent increase in mice treated with ethanol.

Mechanisms of Contraction Induced by Sodium Depletion in the Rabbit Renal Artery

  • In the rabbit renal artery, mechanisms of contraction by sodium depletion were investigated. The helical strips of isolated renal artery were immersed in the Tris-buffered salt solution. The contractions were recorded isometrically using a strain-gauge transducer. Na-free solution (Na was substituted by Li, choline or sucrose) produced contractions which were dependent on the nature of the Na substitutes. Na-free solution (choline) produced the contraction in ouabain-pretreated artery (Na loaded artery) even in the presence of verapamil. The amplitude of the contraction was dependent on the duration of the pretreatment with ouabain $(10\;^5M)$. Monensin potentiated the effect of ouabain on the contraction. Removal of Ca from bathing solution abolished the contraction and the substitution of Sr for Ca produced the contraction. Divalent cations such as Mg, Mn blocked the depolarization-induced contraction, while they had little effect on the Na-free contraction in Na loaded artery. These results suggest that the contraction induced by Na removal is dependent on the cellular Na content and may be caused by Ca influx via the Na-Ca exchange carrier.

Sodium Salicylate Induces the Cyclin-dependent Kinase Inhibitor p21 (Waf1/Cip1) through PI3K-related Protein Kinase-dependent p53 Activation in A549 Cells

  • Sodium salicylate (NaSal), a chemopreventive drug, has been shown to induce apoptosis and cell circle arrest depending on its concentrations in a variety of cancer cells. In A549 cells, low concentration of NaSal (5$\sim$10 mM) induces cell cycle arrest, whereas it induces apoptosis at higher concentration of 20 mM. In the present study, we examined the molecular mechanism for NaSal-induced cell cycle arrest. NaSal induced expression of p53, p21 (Wafl/Cipl), and p27 (Kipl) that play important roles in cell cycle arrest. p53 induction was mediated by its phosphorylation at Ser-15 that could be prevented by the PI3K-related kinase (ATM, ATR and DNA-PK) inhibitors including wortmannin, caffeine and LY294002. In addition, NaSal-induction of p2l (Wafl/Cipl) was detected in P53 (+/+) wild type A549 cells but not in p53 (-/-) mutant H1299 cells, indicating p53-dependent p21 (Wafl/Cipl) induction. In contrast, p27 (Kipl) that is a negative regulate. of cell cycle with p21 (Wafl/Cipl) was observed both in A549 cells and H1299 cells. Thus, 5 mM NaSal appeared to cause cell cycle arrest through inducing the cyclin-dependent kinase inhibitor p21 (Wafl/Cipl) via PI3K-related protein kinase-dependent p53 activation as well as by up-regulating p27 (Kipl) independently of p53 in A549 cells.

  • Electroless plated Ni-Co-P films have been used to suppress the electromagnetic waves from magnetic recording media, and the suppression is known to be achieved with films made with optimized plating composition and plating condition. Effects of complexing agents on the deposition rate and bath stability of Ni-Co-P film were studied using sodium citrate, sodium tartrate and multi-complex agents containing both of them. Deposition of electroless Ni-Co-P platings was dependent upon the complexing agents. Deposition rate was twice when using sodium tartrate compared to that using sodium citrate. And it was slightly slower with multi-complex agents than with sodium tartrate, bath stability being declined in the former. Deposition rate increased with increasing pH until pH 11. Excellent bath stability and good deposition rate were obtained using multi-complex agent as sodium citrate 0.10 mol/L and sodium tartrate 0.15 mol/L in the electroless Ni-Co-P plating films.

Gold Sodium Thiomalate Inhibits iNOS Gene Expression in RAW 264.7 Macrophage: Differential Regulation by Gold Sodium Thiomalate and Sodium Salicylate (Gold Sodium Thiomalate에 의한 유도성 Nitric Oxide Synthase Gene의 발현억제: Gold 제제와 Sodium Salicylate의 차이점)

  • Gold sodium thiomalate (GST, gold compound) is a widely used anti-arthritic, anti-rheumatic and anti-inflammatory drug that is considered a good alternative to sodium salicylate (NaSA) for individuals who cannot tolerate salicylates. Nitric oxide (NO) synthesized by inducible nitric oxide synthase (iNOS) has been implicated as a mediator of inflammation. Recent evidence suggests that anti-inflammatory effect of NaSA lies in the inhibition of iNOS, but nothing has been reported about the direct effect of iNOS expression by GST. The present study was designed to elucidate sequentially the action mechanisms of GST and NaSA on lipopolysaccharide (LPS) plus interferon-gamma (IFN-$\gamma$) induced iNOS expression in RAW 264.7 macrophages. Both GST and NaSA inhibited NO production and iNOS protein expression in a dose dependent manner. GST inhibited iNOS mRNA expression induced by LPS plus IFN-$\gamma$, whereas NaSA did not. These findings suggest that GST may exert anti-arthritic, anti-rheumatic and anti-inflammatory effect by inhibiting iNOS expression induced by LPS plus IFN-$\gamma$ at transcriptional level, whereas NaSA exert its effect by inhibiting iNOS expression at the translational or posttranslational level.

Proton Effect on the Degradation of Phenolic Compound by Activated Sludge and Nocardia asteroides (활성슬러지 혼합미생물과 Nocardia asteroides에 의한 페놀화합물 분해시 양성자이온의 영향)

  • This study was investigated to evaluate the effect of the sodium ion and pH on toxicity of dinitrophenol at high concentrations (0.41 to 0.54 mM), over a sodium concentration range of 0.1 mM to 107 mM and over a pH range of 5 to 9. The concentration of sodium ions in the activated sludge mixed liquor seemed to have very little effect on dinitrophenol toxicity. However, lack of sodium in the growth media resulted in a reduction of the dinitrophenol degradation rate by bacterial isolate from the activated sludge culture, which has been identified as Nocardia asteroides. Dinitrophenol inhibition was found to be strongly dependent on mixed liquor pH. The dinitrophenol degradation rate was highest in the pH range of 6.95 to 7.84; at pH 5.94 degradation of 75 mg/L dinitrophenol was significantly inhibited; at pH < 5.77, dinitrophenol degradation was completely inhibited after approximately 30% of the dinitrophenol was degraded.

Effect of Sodium Butyrate on the Production of IDS by CHO-DG44 Cell

  • Mammalian cell culture in the presence of sodium butyrate has been shown to enhance protein biosynthesis. In the present study, the effect of sodium butyrate on growth of recombinant Chinese Hamster Ovary cells and on the production of Iduronate 2-sulphatase were investigated in serum-containing and serum-free media. The culture with addition of 0-5mM sodium butyrate showed enhancement of both intracellular and extracellular IDS production. But, Cell death was observed in a dose-dependent manner. The optimal sodium butyrate concentration was observed to be 5mM. Also, The relative productivity of IDS was significantly increased when sodium butyrate was added to medium at 48 hour, the rapid growth phase. These results suggest that sodium butyrate are efficient agent for increasing the productivity of IDS with recombinant CHO cells.

Effects of Hyunsamkabhangbang on the Hyperthyroidism Induced by Sodium Levothyroxine in Rats (현삼갑항방(玄參甲亢方)이 Sodium Levothyroxine 투여로 유발된 백서의 갑상선중독증에 미치는 영향)

  • Objective : This study was to elucidate the effects of Hyunsamkabhangbang (HSK) on the hyperthyroidism induced by sodium levothyroxine. Method : Sprague-Dawley rats were separated into five groups. Except the normal group, the other four groups were treated with sodium levothyroxine 160${\mu}g/kg/day$ for 5 days by oral administration. Among the four groups, all except one (as control) were treated with various concentrations of HSK (0.5, 1.0, 2.0 g/kg/days) for 3 days separately. T3-uptake, T3, T4, TSH, total cholesterol, free fatty acid, ${\beta}$-lipoprotein, glucose, ALP, AST, ALT change in the serum and body weight of rats were measured after medication of solid extract of HSK. Results : HSK decreased significantly serum T3, T4, T3-uptake, ALP, free fatty acid and glucose level in a dose-dependent manner compared with control. HSK increased significantly serum TSH and total cholesterol level and body weight in a dose-dependent manner compared with control. HSK also increased serum ${\beta}$-lipoprotein level compared with control, but not statistically meaningfully. HSK increased significantly serum AST and ALT level compared with control, but relationship to hyperthyroidism is not found. Conclusions : From these results, it could be concluded that HSK lowers excess thyroid hormone and increased metabolism, resulting in improvement of hyperthyroid state.

Dose-dependent Ultrastructural Changes of the Odontoblasts in the Neonatal Rat after Ingestion of the Fluoride to the Pregnant Rat (음용수 불소농도에 따른 흰쥐태아 상아모세포의 미세구조적 변화)

  • The pregnant rats were given a drinking water administration of the sodium fluoride and normal saline for control animals. The sodium fluoride produced cellular changes of odontoblast with consistent response. Compare to control group, the odontoblasts that were administrated by sodium fluoride, showed significantly ultrastructural differences including large number of free ribosomes and swelled mitochondria in dose-dependent manner (300 ppm). From fine structural and morphological investigations of the changes in odontoblast, there were three distinctive structural changes: (1) destruction of the endoplasmic reticulum, (2) swelling of the mitochondria, and (3) severe cellular derangement of the endoplasmic reticulum and mitochondria. From this consecutive structural change, we observed that sodium fluoride temporarily affects the cell organelles in odontoblasts (100, 200 ppm), suggesting it is important that optimal concentration of the sodium fluoride in developing fetus of the rat.
