Development of Probiotic Microcapsules for the Preservation of Cell Viability (생균활성 보존을 위한 유산균 미세캡슐 개발)

  • Lactobacillus fermentum YL-3 was encapsulated to increase acid tolerance and its total viability. After micro-encapsulation of L. fermentum YL-3 cells with sodium alginate and soybean oil, the morphology of the microcapsule was observed using confocal laser scanning microscopy (CLSM) after staining with pyronin Y and fluorescein isothiocyanate. The sizes of the microcapsules were 120-126 ${\mu}m$, 444-486 ${\mu}m$ and 401-463 ${\mu}m$ when manufactured at pH 2, 3 and 4, respectively. The microcapsule could hold live cells of L. fermentum YL-3 up to $1.2{\times}10^{7}$, $8.1{\times}10^{7}$ and $1.1{\times}10^{8}$ CFU/mL at pH 2, 3 and 4, respectively. The acid tolerance and preservative ability of L. fermentum YL-3 in microcapsule and macrocapsule at $4^{\circ}C$ and $25^{\circ}C$ were tested. L. fermentum YL-3 cells were evenly located in the alginate capsule matrix structure and the firmness of microcapsule was highest at pH 2. Micro-encapsulation showed the most effective acid tolerance at pH 2.0 and preservation of viability at $4^{\circ}C$. However, at $25^{\circ}C$, the macrocapsules showed more effective cell protection than the microcapsules. The application range for microcapsules could be wider than for macrocapsules in the food industry.

The Processing Optimization of Caviar Analogs Encapsulated by Calcium-Alginate Gel Membranes

  • We prepared caviar analogs encapsulated by calcium-alginate gel membranes as a means to replace higher priced natural caviars. Processing the caviar analogs (beluga type) was optimized by response surface methodology with central composite design. Concentrations of sodium alginate ($X_1$) and $CaCl_2\;(X_2)$ were chosen as the independent variables. In order to compare characteristics of the caviar analogs with the natural caviar, sphericity ($Y_1$), diameter ($Y_2$), membrane thickness ($Y_3$), rupture strength ($Y_4$), rupturing deformation ($Y_5$), and sensory score ($Y_6$) were used as the dependent variables. The sphericity of the caviar analogs showed a similar value to that of natural caviar (over 94%) in the range of independent variables. Generally, the $CaCl_2$ concentration ($X_2$) affected all dependent variables to a greater extent than the sodium alginate concentration ($X_l$), For the multiple response optimization of the 5 dependent variables ($Y_1,\;Y_2,\;Y_4,\;Y_5$, and $Y_6$), the desirability function was defined as the following conditions: target values ($Y_1\;=\;100%,\;Y_2\;=\;3.0\;mm,\;Y_4\;=\;1,470\;g,\;Y_5\;=\;1.1\;mm,\;and\;Y_6\;=\;10\;points$). Membrane thickness ($Y_3$) was eliminated from the dependent variables for multiple response optimization because it could not be measured with an image analyzer. The values of the independent variables as evaluated by multiple response optimization were $X_1\;=\;-0.093$ (78%) and $X_2\;=\;-0.322$ (1.07%), respectively.

Partially Hydrolyzed Crosslinked Alginate-graft-Polymethacrylamide as a Novel Biopolymer-Based Superabsorbent Hydrogel Having pH - Responsive Properties

  • In this study, a series of highly swelling hydrogels based on sodium alginate (NaAlg) and polymethacryl­amide (PMAM) was prepared through free radical polymerization. The graft copolymerization reaction was performed in a homogeneous medium and in the presence of ammonium persulfate (APS) as an initiator and N,N'-methylenebis­acrylamide (MBA) as a crosslinker. The crosslinked graft copolymer, alginate-graft-polymethacrylamide (Alg-g­PMAM), was then partially hydrolyzed by NaOH solution to yield a hydrogel, hydrolyzed alginate-graft-poly­methacrylamide (H-Alg-g-PMAM). During alkaline hydrolysis, the carboxamide groups of Alg-g-PMAM were converted into hydrophilic carboxylate anions. Either the Alg-g-PMAM or the H-Alg-g-PMAM was characterized by FTIR spectroscopy. The effects of the grafting variables (i.e., concentration of MBA, MAM, and APS) and the alkaline hydrolysis conditions (i.e., NaOH concentration, hydrolysis time, and temperature) were optimized systematically to achieve a hydrogel having the maximum swelling capacity. Measurements of the absorbency in various aqueous salt solutions indicated that the swelling capacity decreased upon increasing the ionic strength of the swelling medium. This behavior could be attributed to a charge screening effect for monovalent cations, as well as ionic cross-linking for multivalent cations. Because of the high swelling capacity in salt solutions, however, the hydrogels might be considered as anti-salt superabsorbents. The swelling behavior of the superabsorbing hydrogels was also measured in solutions having values of pH ranging from 1 to 13. Furthermore, the pH reversibility and on/off switching behavior, measured at pH 2.0 and 8.0, suggested that the synthesized hydrogels were excellent candidates for the controlled delivery of bioactive agents. Finally, we performed preliminary investigations of the swelling kinetics of the synthesized hydrogels at various particle sizes.

  • The alginate degrading bacteria have been screened from the marine environment. Sodium alginate and NaCl were required for cell growth and enzyme production of 145-C strain and the adequate concentrations were 0.7 and 2.5%, respectively. The effective nitrogen source was peptone and adequate temperature was 28$\pm$2$^{\circ}C$. The 145-C strain was identified as Vibrio sp. from biochemical and biological experiment. The extracellular enzyme produced by Vibrio sp. was purified and the molecular weight was estimated to be 27, 000.

Biocompatibility of oxidized alginate/gelatin/BCP -based hydrogel composites

  • In this study, oxidized alginate/gelatin/biphase calcium phosphate (BCP)- based hydrogel composites were fabricated. Alginate sodium was oxidized by periodate. The oxidized product was confirmed by using $^1H$ and $^{13}C$ NMR spectra. The number average molecular weight ($M_n$), the average molecular weight ($M_w$) of the oxidized alginate were determined by Gel Permeation Chromatography (GPC). The hydrogel was formed from the oxidized alginate and gelatin solution via Schift-base reaction. The hydrogel showed a highly porosity by a Scanning Electron Microscope (SEM) and Mercury Intrusion Porosimetry (MIP). Crosslinked density of the gel matrix were assessd by trinitrobenzene sulfonic acid (TNBS) assay that shows a high effect on swelling ratio. Increment of the crosslinked desity resulted in enhancing compressive strength of the hydrogel composite. The cytotoxity of hydrogel was assessed with osteoblast MG-63. The hydrogel composites show a high compatibility. The obtained results showed a potential application for bone regeneration in future.

Controlled Release and Bioavailability of Piracetam (피라세탐의 방출조절 및 생체이용률)

  • This study is purposed to develop the sustained release and bioavailability of piracetam (PA). The use of alginate beads as a means to achieve sustained release of piracetam was evaluated in comparison with that of piracetam alone. In the PA-sodium alginate(SA) beads was confirmed by differential scanning calorimetry thermogram(DSC), indicating a relative shift of an endometric peak of PA to higher temperature. The changes in dissolution rates from PA-SA beads and PASA beads coated by chitosan(CHO) were significantly slower than that of intact PA. The release rate of PA-SA in the gastric fluid was markedly decreased compared with that in the intestinal fluid, suggesting that PA is mostly released in the intestinal fluid. However, the PA/SA ratio scarcely affected the release profile. The blood concentration- time curves of PA, PA-SA and PA-SA-CHO were obtained by oral administration to rats. $T_{max}$ of PA, PA-SA and PA-SA-CHO were 1, 10 and 6 hours, respectively. It was confirmed that the release of PA was prolonged by the formulation of PA-SA beads and PA-SA-CHO beads.

Antibacterial Properties of Silver-alginate/PVP Nanofiber (은-알지네이트/PVP 나노섬유의 항균 특성)

  • In order to incorporate silver ions into the alginate, silver-alginate was prepared with aqueous solutions of silver nitrate. In the study, the silver-alginate was prepared by blending with poly vinylpyrrolidone solutions and the electrospinning was performed by using this blend solution. Antibacterial properties of silver-alginate/PVP solutions were estimated for Escherichia coli and Staphylococcus aureus by the colony counting test. Electrospinning conditions of silver-alginate/PVP solution were the tip-to-collector distance of 22 cm, the flow rate of the solution at 0.01 mL/min, and the voltage at 26 kV. The form and size of silver-alginate/PVP nanofibers were estimated by SEM and Image J. The average diameter of the electrospun SA5P15 fibers was 124 nm and showed a narrow diameter distribution. The reduction of bacteria for SA5P15 exhibited 99.9% after 24 h.

Evaluation of glycerol encapsulated with alginate and alginate-chitosan polymers in gut environment and its resistance to rumen microbial degradation

  • Objective: To determine the effect of gut pH and rumen microbial fermentation on glycerol encapsulated in alginate and alginate-chitosan polymers. Methods: Glycerol was encapsulated at 2.5%, 5%, 7.5%, or 10% (w/w) with sodium alginate (A) and alginate-chitosan (AC) polymers. Surface morphology and chemical modifications of the beads were evaluated using scanning electron microscopy and Fourier transform infrared (FTIR) spectra. Encapsulation efficiency was determined at the 5% glycerol inclusion level in two experiments. In experiment 1, 0.5 g of alginate-glycerol (AG) and alginate-chitosan glycerol (ACG) beads were incubated for 2 h at $39^{\circ}C$ in pH 2 buffer followed by 24 h in pH 8 buffer to simulate gastric and intestinal conditions, respectively. In experiment 2, 0.5 g of AG and ACG beads were incubated in pH 6 buffer at $39^{\circ}C$ for 8 h to simulate rumen conditions. All incubations were replicated four times. Free glycerol content was determined using a spectrophotometer and used to assess loading capacity and encapsulation efficiency. An in vitro experiment with mixed cultures of rumen microbes was conducted to determine effect of encapsulation on microbial fermentation. Data were analyzed according to a complete block design using the MIXED procedure of SAS (SAS Institute, Cary, NC, USA). Results: For AG and ACG, loading capacity and efficiency were 64.7%, 74.7%, 70.3%, and 78.1%, respectively. Based on the FTIR spectra and scanning electron microscopy, ACG treatment demonstrated more intense and stronger ionic bonds. At pH 6, 36.1% and 29.7% of glycerol was released from AG and ACG, respectively. At pH 2 minimal glycerol was released but pH 8 resulted in 95.7% and 93.9% of glycerol released from AG and ACG, respectively. In vitro microbial data show reduced (p<0.05) fermentation of encapsulated glycerol after 24 h of incubation. Conclusion: The AC polymer provided greater protection in acidic pH with a gradual release of intact glycerol when exposed to an alkaline pH.

  • At present, quarternary ammonium salts(cationic surfactant) and Tego compounds (amphoteric surfactant) are used as germicidal agent. In this paper, it was investigated whether their germicidal activities are influenced or not by some suspending agents which are added to them 0.005% benzalkonium chloride aq. solution and 0.05% dodecyl diaminoethyl glycine aq. solution sterilized respectively against Staphylococcus aureus and Escherichia coli within a minute. The solutions aded acacia, carboxymethyl cellulose, sodium alginate solutions to make to 0.005% to above surfactants solution decreased the germicidal activity, not being sterilized bacteria for more than 2 hours however, the solutions decreased the germicidal activity by addition of suspending agents such as acacia, carboxymethyl cellose, sodium alginate and bacteria were survival for more than 2 hrs, 0.05% of benzalkonium chloride and 0.1% dodecyl diaminoethyl glycine respectively would be sterilized within 10 minutes, when added to 5% suspending agents. This result show that bactericidal action of dodecyl diaminoethyl glycine is affected by suspending agents less than that of benzalkonium chloride.

A Suppressive Effect of Dietary Fiber on in Vitro Absorption of Lead (납의 In Vitro 흡수에 미치는 식이 섬유의 억제효과)

  • In order to examine the suppressive effect of dietary fiber toward the intestinal absorption of lead, an in vitro absorption test using a semipermeable membrane was undertaken. Among dietary fiber components, cellulose showed no suppressive effect, guar gum and carboxymethyl-cellulose, a slight effect whereas citrus pectin and sodium alginate exhibited a remarkable effect. Among fibrous foods tested, rice bran, wheat bran, Chinese cabbage, radish and tangle had a higher suppressive effect while mandarin orange, apple and laver showed a lower effect.
