해바라기 유(油)의 탈납 (Dewaxing of Sunflower Seed Oil)

  • 해바라기유의 재래식 탈납공정 경제성을 높이기 위해서 여러가지 유화제(0.2% sodium bexametaphosphate, 0.05% sodium lauryl sulfate, 0.001% dioctyl sodium sulfosuccinate)를 이용한 새로운 탈납공정을 개발하였다. 해바라기 원유를 회분식 (batch process)과 연속식(continuous process)으로 유화제 수용액과 유화시킨후 원심분리기로 분리시켰을 때 재래식 탈납공정에 비견할 만한 탈납을 할 수있음을 알았다. 탈납 공정에서 위에 설명한 탈납정도(degree of dewaxing)외에 또 하나의 중요인자는 수율(dewaxirg loss)인바 회분식 공정의 경우에는 재래식 탈납공정의 수율과 같았으나 연속식 공정의 경우에는 개량할 여지가 있음을 알았다. 실험결과에 의하면 회분식 공정의 경우 탈납전 원유에 함유된 납(wax)의 다과에 관계없이 수율은 $0.62{\sim}0.82%$로 재래식 공정과 같았으나 연속식 공정인 경우에는 탈납전 원유에 함유된 납의 양이 적은 경우와 많은 경우 각각 2.28%와 5.68%로 현격한 차이를 보였다. 또한 연속식 공정의 경우 원유와 유화제를 유화시키는 방법에 따라 수율에 많은 영향이 미치는 것으로 밝혀졌으며 contactor와 static mixer를 썼을때 각각 5.2%와 2.8%이었다.

항균제 처리에 따른 수삼의 미생물 저감화 효과 (Efficacy of antibacterial treatments of fresh ginseng (Panax ginseng C.A. Meyer))

  • 건강 기능성 소재로 많이 활용되고 있는 수삼은 정제수에서 1시간 세척 후에도 일반세균은 6.5 log CFU/g, 곰팡이는 4.3 log CFU/g 수준으로 높게 나타나 미생물 안전성을 확보하기 위해 상업적 항균제 8종의 적용 실험을 진행하여 다음과 같은 결과를 얻었다. 다양한 박테리아와 곰팡이류에 대한 항균효과가 있는 것으로 알려져 있는 1% (w/w) 구연산나트륨, 이초산나트륨, 초산나트륨, 구연산 및 젖산나트륨 용액에 세척한 수삼을 각각 1시간 동안 침지한 결과 유기산 종류별로 31.0-97.5%의 총세균의 저감화 효과를 보였다. 유기산 계통 항균소재 중 구연산나트륨이 6.5 log CFU/g에서 4.3 log CFU/g으로 2 log CFU/g 내외의 오염 미생물이 저감화 되어 가장 살균력이 좋았다. 수삼에 대한 유기산 항균효과는 물에 해리된 pH 2.4 내외의 산 성분이 미생물의 세포막을 통해 세포질로 확산되어 생육에 필수 인자인 단백질과 핵산의 구조와 기능을 변화시켜 발생하였다. 바실러스 포자에 대해 항균력이 있는 것으로 알려진 계면활성제 계통의 항균제 실험 결과 세척만 적용한 무처리구 6.5 log CFU/g 대비 비타민B1라우릴 황산염 처리시 3.8 log CFU/g으로 2.7 log CFU/g 감균되어 99.8%의 우수한 살균효과를 나타내었다. 계면활성제의 친수성 및 소수성 부위가 세포막의 유사 부위와 각각 결합하여 구조 변형을 가져오게 되어, 미생물의 세포막이 파괴되면 생육에 관련된 효소, 뉴클레오타이드 및 뉴클레오시드 및 당의 누출을 초래하게 되어 효과적으로 세포 사멸을 유도하게 된다. 통마늘과 통생강과 같은 향신채소의 미생물 제어에 효과적인 1.0% (w/w) 산화칼슘 용액에서는 수삼내 총균이 3.5 log CFU/g으로 무처리구 대비 3 log CFU/g 내외 감균되어 99.9%의 우수한 살균효과를 나타내어 산화칼슘이 수삼의 미생물 감균 소재로 활용도가 높은 것으로 나타났다. 소성 산화칼슘은 pH 12.5의 강한 알칼리성과 활성산소로 미생물의 생리 활성에 영향을 주어 효과적인 살균 효과를 나타내는 것으로 밝혀졌다.

카르복실산 합성전구체(合成前驅體)로서의 옥탄니트릴의 생성반응(生成反應)에 관(關한) 연구(硏究) (A Study on the Formation of Octanenitrile as a Precursor for Synthesis of Carboxylic Acid)

  • Using the quarternary ammonium salts as phase transfer catalyst, the nucleophilic substitution reaction of 1-chlorooctane with sodium-cyanide was investigate kinetically with respect to the formation of octanenitrile. The product was analyzed with gas chromatograph, and quantity of octanenitrile was measured. The reaction condition was considered by the effect of the reaction temperature, of the species and the amount of catalyst, of the speed of strirring, and of the concentration of reactants. The reaction was carried out in the first order on the concentration of 1-chlorooctane and sodium cyanide, respectively. The over-all order was 2nd. The activation energies for the nucleophilic substitution reaction of 1-chlorooctane and 1-bromooctane under tetrabutylammonium hydrogen-sulfate were calculated as 2.05 and 10.08kcal/mol, respectively. The effect of various caltalysts was decreased in the order of tetrabutylammonium bromide, terabutylammonium, tetrabutylammonium hydrogensulfate, and tetrabutylammonium iodide. The reaction rate was dependent on the concentration of sodium-cyanide dissolved in the aqueous phase, and the good result was shown when the mol ratio between 1-chlorooctane and sodium cyanide was one per three.

산 및 황산염 저항성에 미치는 시멘트 종류의 영향 (Influence of Cement types on the Resistance to Acid and Sulfate)

  • 본 연구에서는 산 및 황산염 저항성에 미치는 시멘트 종류의 영향을 평가하기 위하여 물-결합재 비 32% 및 43%에 대해서 보통 포틀랜드 시멘트(OPC), 2성분계 시멘트(BBC), 및 3성분계 시멘트(TBC)와 같은 3종류의 시멘트를 사용한 콘크리트 공시체를 제작하였다. 제작된 콘크리트 공시체에 대해서 JSTM C 7401에 따라 5% 황산, 10% 황산나트륨 나트륨 및 10% 황산마그네슘 용액에 의한 침지실험을 통하여 재령에 따른 외관 변화 및 질량 감소율을 평가하였다. 재령 91동안의 침지실험 결과로부터, 콘크리트의 산 및 황산염에 대한 저항성은 물-결합재비가 감소할수록 증가하고, BBC와 TBC 콘크리트가 OPC 콘크리트보다 우수한 것으로 나타났다.

Production of Sulfuric Acid and Ammonia Water from Ammonium Sulfate Using Electrodialysis with Bipolar Membrane and Ammonia Stripping

  • The feasibility of producing sulfuric acid and ammonia water from ammonium sulfate was investigated by an integrated process including ammonia stripping (AS) and electrodialysis with bipolar membrane (EDBM). It was suggested that the production of sulfuric acid using ammonia stripping-electrodialysis with bipolar membrane (ASEDBM) was effective in obtaining high concentration of sulfuric acid compared with EDBM alone. AS was carried out over pH 11 and within the range of temperatures, $20^{\circ}C{\~}60^{\circ}C$. Sodium sulfate obtained using AS was used as the feed solution of EDBM. The recovery of ammonia increased from $40\%$ to $80\%$ at $60^{\circ}C$ due to the increased mobility of ammonium ion. A pilot-scale EDBM system, which is composed of two compartments and 10 cell pairs with an effective membrane area of $200 cm^2$ per cell, was used for the recovery of sulfuric acid. The performance was examined in the range of 0.1 M${\~}$1.0 M concentration of concentrate compartment and of $25 mA/cm^2{\~}62.5 mA/cm^2$ of current density. The maximum current efficiency of $64.9\%$ was obtained at 0.1 M sulfuric acid because the diffusion rate at the anion exchange membrane decreased as the sulfuric acid of the concentrate compartment decreased. It was possible to obtain the 2.5 M of sulfuric acid in the $62.5 mA/cm^2$ with a power consumption of 13.0 kWh/ton, while the concentration of sulfuric acid was proportional to the current density below the limiting current density (LCD). Thus, the integrating process of AS-EDBM enables to recover sulfuric acid from the wastewaters containing ammonium sulfate.

  • 토양으로부터 alkaline protease 생성능이 강한 Streptomyces griseus HC-1141을 분리하였으며, 효소생산의 최적 배양조건은 0.5 casein, 0.05 ammonium chloride, 0.1 ferrous sulfate, 2.0의 lactose, pH 8.0에서 84시간 배양했을 때이다. 효소의 정제는 ammonium sulfate 침전, DEAE-cellulose ion exchange chromatography, Sephadex G-150 gel filtration, crystallization으로 하여 53.23배 정제할 수 있었으며 polyacrylamide gel 전기영동상 단일밴드를 나타내었다.

비타민 B1 유도체(Thiamine Di-lauryl Sulfate:TDS)의 나노입자화를 통한 고추탄저병균의 항진균 활성 증진 (Enhancement of Antifungal Activity of Anthracnose in Pepper by Nanopaticles of Thiamine Di-lauryl Sulfate)

  • This study was performed to enhance antifungal activity of anthracnose in chili pepper by nanopaticles of thiamine di-lauryl sulfate (TDS) through high pressure homogenization process. Yield of TDS was 79.14% by reaction of thiamine hydrochloride and sodium lauryl sulfate. TDS nanopaticle solution was manufactured through high pressure homogenization process. The turbidity of nanoparticles solution was increased with increasing the concentration of TDS, and nanoparticles solution of 100 ppm was showed the highest turbidity with absorbance of 3.212. The size of nanoparticles solution was measured as average 258.6 nm by DLS. Nanoparticles solution of 100 ppm showed growth inhibition activity with higher than about 80% compared to the control group against Colletotrichum gloeosporioides. Finally, nanoparticles solution was increased effectively the penetration of the TDS nanopaticles on attached cell membrane of hyphae and started to destruct the cells under microscope observation. Consequently, we suggested that the TDS nanoparticle solution by high pressure homogenization process might be suitable biochemical pesticides for improving the antifungal activities against anthracnose in pepper.

Spectrophotometer 및 HPLC 에 의한 식용달팽이의 황산콘드로이틴 분석 (Spectrophotometric and High-Performance Liquid Chromatographic Assay of Chondroitin Sulfate in Edible Snail, Achatina Fulica Bowdich)

  • Chondroitin sulfate (Chs) contents in edible snail , Achatina fulica Bowdich , andits processed meat extracts were determined by high-performance liquid chormatogrpahy(HPLC) and spectrophotometric method. Spectrophotometric method was based on the precipitation of acriflavine by ChS, and HPLC method was based on the detection of two unsaturated disaccharides, 2-acetamido-2-deoxy-3-O-($\beta$ -D-gluco-4-enepyranosyluronic acid)-4-O-sulfo-D-galactose($\Delta$Di-4S) and 2-acetamido-2deoxy-3-O-($\beta$-D-gluco-4-eepyranosyluronic acid)-6-O-sulfo-D-galactose ($\Delta$야-6S) librated from ChS byenzymeatic digestion with chondroitinase ABc. the ratio of 125$\mu$mol of sodium hydroxide to mg of ChS and 8$0^{\circ}C$ of reaction temperature were proper for alkaline hydrolysis to remove protein residue form ChS. In assay preparation for HPLC ethod, the iptimum concentration of the enzyme chondroitinase ABc was 0.15 unit per 50 $\mu\textrm{g}$ of ChS at a fixed reaction time (30 min) and pH 8.0 using Tris buffer. ChS content in edible snail was 177.6mg% by spectrophotometric method and 153.5mg% by HPLC method and those in the processed meat extract was 71.3mg% by spectrometric method ad 62.8mg% by HPLC method, respectively.

Pseudomonas sp. Aryl Acylamidase의 정제 및 성질 (Purification and Characterization of Aryl Acylamidase from Pseudomonas sp.)

  • Acetaminophen 자화성 Pseudomonas sp.에 존재하는 aryl acylamidase[EC 3.5.1.13]는 ammonium sulfate fractionation, DEAE-Sephacel anion exchange chromatography, Phenyl-Sepharose CL-4B hydrophobic interaction chroamtography 및 Sephadex G-100 gel-permeation chromatography를 통해 순수 정제되었다. 정제된 효소는 SDS-PAGE상에서 분자량을 측정한 결과 56 kDa, Sephadex G-100 gel-permeation chromatography로 측정한 결과 57 kDa이었으며, 따라서 단일한 subunit로 구성된 효소이었다. 정제된 효소의 최대 활성을 위한 pH와 온도는 각각 pH 10.5와 4$0^{\circ}C$이였다. 5$0^{\circ}C$에서 30분간 처리시 34%의 잔존활성을 나타내었다. Acetaminophen과 4'-nitroacetanilide쉐 대한 Km값은 각각 0.10 mM과 0.11 mM 이었다.

Controlled Release of Gentamicin Sulfate from Poly(3-hydroxybu-tyrate-co-3-hydroxyvalerate) Wafers for the Treatment of Osteomyelitis

  • Biodegradable wafers were prepared with poly (hydroxybutyrate-co-hydroxyvalerate) (PHBV;5, 10, and 15 mole% for 3-hydroxyvalerate) by simple heat pressing method for the sustained release of antibiotic agent, gentamicin sulfate (GS) to investigate the possibility of the treatment for osteomyelitis. The effects of hydroxyvalerate (HV) content, thickness of wafers, various types of additives such as sodium dodecyl sulfate (SDS), microcrystalline cellulose, polyvinylpyrrolidone, and hydroxypropylcellulose (HPC), and different initial drug loading ratio on the release profile have been investigated. In vitro release studies showed that different release patterns and rates could be achieved by simply modifying factors in the preparation conditions. PHBV wafers with 3 mm thickness, 10% of GS initial loading, 15% of HV content and addition of 5% of SDS and HPC were free from initial burst and a near-zero-order sustained release was observed for over 30 days. It might be suggested that the mechanisms of G5 release may be more predominant simple dissolution and diffusion of GS than erosion of PHBV in our system.
