Rhizopus oryzae의 인지질과 그 분해효소에 미치는 계면활성제의 영향 (Effect of Sodium deoxycholate and Sodium dodecy sulfate on Phospholipid Composition and Phospholiases of Rhizopus oryzae)

  • Rhizopus oryzae 균사체 내의 인지질 구성 성분은 6종으로 나타났고 sodium deoxycholate와 sodium dodecyl sulfate를 각각 0.48mM, O.18mM의 낮은 농도로 영양배지에 첨가했을때 성장군략의 형태변화와 아울러 미량 존재하던 cerdiolipin과 phospha tidylinositol 그러고 Iy sophospholipid의 함량이 증가하였다. 또한 phospholipase는 A와 C, 그리고 D가 존재하였으며 A와 D는 sodium deoxycholate에 의하여. C는 sodium dodecyl sulfate에 의하여 활성도 증가를 나타냈다.

계면 활성제 처리에 의한 세균 세포막의 변화에 관한 연구 (The effect of some detergents on the changes of bacterial membrane)

  • The results that the effect of 6 detergents on the structural changes and biochemical composition of bacterial membrane of Escherichia coli and Bacillus cereus are as follows ; 1. Population growth of the bacteria was increased in case of the treatment with palmitoyl carnitine and sodium deoxy cholate but was increased in case of the treatment with palmitoyl carnitine and sodium deoxy cholate but was decreased by sodium dodecyl sulfate and palmitoyl choline, in E.coli and was decreased by palmitoyl carnitine and palmitoyl choline at the low concentration, in B. cereus. 2. The electron micrograph showed that cell wall lysis or cell collapse were observed in the treatment of sodium dodecyl sulfate and palmitoyl choline, and also cell wall was condensed by triton X-100 and sodium deoxy cholate, in E.coli. And in B. cereus, endospore formation of the bacteria was stimulated by palmitoyl choline, and cell lysis or structural changes of the membrane were observed in the treatment of sodium dodecyl sulfate, sodium cholate, and triton X-100, respectively. 3. As to the effect of detergent on the biochemical composition of biomembrane, the content of carnitine, in E.coli, and B.cereus, the content of structural protein and phospholipid were decreased by treatment of sodium dodecyl sulfate and structural protein was denatured by palmitoyl choline. 4. The profile of membrane protein revealed that the bacterial membrane were composed of various proteins. By dint of this result, some of membrane proteins were solubilized or changed to small molecules by the treatment of sodium dodecyl sulfate and palmitoyl choline, in E.coli and membrane protein of the biomembrane by treatment of sodium dodecyl sulfate, sodium deoxy cholate, palmitoyl choline, and palmitoyl carnitine were confirmed to be different profile as compared with those of the control, in B. cereus. Therefore, it is suggested that sodium dfodecyl sulfate and palmitoyl choline soulbilized biomembranes or inhibited membrane transport and that palmitoyl carnitine and sodium deoxy cholate were used as an energy source or stimulating the membrane transport, in E.coli. And, it is suggested that all of detergents were inhibited biomembrane synthesis, expet saponin, in B.cereus.

전해질 첨가에 따른 키토산/Sodium Dodecyl Sulfate 상호작용의 변화 (The Change of Interactions of Chitosan/Sodium Dodecyl Sulfate in the Presence of Electrolytes)

  • The change of interactions of anionic surfactants, sodium dodecyl sulfate(SDS) and sodium tetradecyl sulfate(575) in the presence of electrolytes, to the chitosan-based polyelectrolyte(sol'n and gel phase) were studied. The chitosan gel used in this study were crosslinked with epichlorohydrin(ECH). Binding isotherms were determined by potentiometric technique using a surfactant ion selective solid-state electrode and the results were represented by using the sequence generating function(SGF) method. The results of binding isotherm were shown comparatively high cooperativity. The addition of electrolytes in the chitosan/SDS system resulted in a shift of the binding to higher free surfactant concentration because of screen effect by the electrolytes. Degree of binding of chitosan gel was higher than that of chitosan sol'n. And also a conformational phase transition of the chitosan gel in the presence of electrolytes has been investigated.

  • We propose a method for rapid evaluation and permanent record of sodium dodecyl sulfate polyacrylamide gel electrophoretic runs. This method is based on the photocopy process, rather than on photography and requires no extensive or expensive investment. Comparison of a print obtained through this method and a 35mm photography indicated, on a balanced gel, equal sensitivity.

  • Sodium dodecyl sulfate (SDS) 존재하에서 유기색소인 thionine (TH)의 흡수 및 형광분광학적 성질을 조사 연구하였다. SDS 농도 증가에 따라 측정한 흡수 및 형광스펙트럼의 변화들로부터 i) TH-SDS 착화합물의 회합체 형성, ii)TH-SDS 회합체내의 색소분자의 stacking 과정, iii)dye-rich induced micelles의 형성에 의한 staking의 파괴 및 iv) SDS 미셀의 표면에 TH의 재분포과정 등으로 설명하였다.

  • 순수 물 및 NaCl의 수용액에서 Sodium Dodecyl Sulfate(SDS)와 Tetraethylene glycol monodode-cyl ether(TGME)의 혼합계면활성제가 $25^{\circ}C$에서 나타내는 임계미셀농도값(CMC$^*$)들을 표면장력계로 측정하였으며, 이들 CMC$^*$값들이 음이온 계면활성제인 SDS의 몰분율 조성(${\alpha}_1$)과 첨과제인 NaCl의 농도에 따라 어떻게 변화하는지를 조사하였다. 유사상태분리모델을 기초로하는 비이상적 혼합비셀모델을 적용함으로써 측정된 CMC$^*$값들로부터 SDS/TGME 혼합계면활성제의 미셀화에 대한 여러가지 열역학적 함수값들을 계산하여 분석하였다.

  • $10^{-6}{\sim}10^{-3}mol/{\ell}$ 농도범위에서 PO가 먼저 부가된 sodium poly(oxyethylene(EO), oxyisopropylene(PO)) dodecyl ether sulfates의 표면장력은 EO가 부가된 화합물보다 낮게 나왔다. 그리고 대체적으로 일반적인 음이온성계면활성제보다도 낮게 나왔다. PO가 먼저 부가된 화합물에 대하여 Gibbs' 흡착등온선으로 계산한 흡착능(${\Gamma}=2.2{\times}10^{-10}mol/cm^2$)은 EO가 부가된 화합물보다는 높지만 sodium dodecyl ether sulfate(${\Gamma}=3.2{\times}10^{-10}mol/cm^2$)보다는 낮다. 이러한 현상은 PO가 부가된 화합물의 분자량이 크기 때문으로 사료된다. 기포력, n-hexane과 benzene에 대한 유화력, 돼지기름과 소기름에 대한 세정력 그리고 산화철에 대한 분산성은 PO 화합물이 EO 화합물보다 우수하게 나타났다. 이러한 현상들은 친수성과 소수성 부분이 각각 따로 따로 존재할 때 계면활성이 높음을 의미한다. 수계에서 n-hexane과 benzene에 대한 유화력 측정결과는 n-hexane보다 benzene에 대하여 높게 나왔으며, 불규칙적인 분산특성을 나타내었다.

  • B. subtilis의 항생물질 내성균주에 대하여 여러 plasmid curing 시약인 sodium dodecyl sulfate, acriflavine, ethidium bromide, mitomycin-C등을 처리하여 streptomycin 감수성 변이주를 만들었다. Sodium dodecyl sulfate 처리의 경우 0.1% 농도에서 균체성장 극대기에 도달한 후부터 항생물질 감수성 비율이 증가되었으며, acriflavine 처리의 경우는 4$\mu\textrm{g}$/$m\ell$ 농도에서, ethidium bromide 처리의 경우에는 10$\mu\textrm{g}$/$m\ell$ 농도에서. 그리고 mitomycin-C 처리의 경우에는 200$\mu\textrm{g}$/$m\ell$ 농도에서 가장 curing빈도가 높았다. 또 이들 curing 작용은 B. subtilis의 streptomycin과 terramycin 내성균주에서 보는 바와 같이 경쟁적 작용으로 일어났다.
