Lab-on-a-chip technology is attracting great interest because the miniaturization of reaction systems offers practical advantages over classical bench-top chemical systems. Rapid mixing of the fluids flowing through a microchannel is very important for various applications of microfluidic systems. In addition, highly sensitive on-chip detection techniques are essential for the in situ monitoring of chemical reactions because the detection volume in a channel is extremely small. Recently, a confocal surface enhanced Raman spectroscopic (SERS) technique, for the highly sensitive biological analysis in a microfluidic sensor, has been developed in our research group. Here, a highly precise quantitative measurement can be obtained if continuous flow and homogeneous mixing condition between analytes and silver nano-colloids are maintained. Recently, we also reported a new analytical method of DNA hybridization involving a PDMS microfluidic sensor using fluorescence energy transfer (FRET). This method overcomes many of the drawbacks of microarray chips, such as long hybridization times and inconvenient immobilization procedures. In this paper, our recent applications of the confocal Raman/fluorescence microscopic technology to a highly sensitive lab-on-a-chip detection will be reviewed.
탄소나노튜브를 이용하여 변형한 콜레스테롤 측정을 위한 바이오 센서를 개발하였다. 개발된 콜레스테롤 바이오 센서는 일회용으로 콜레스테롤의 효소반응에서 전자전달을 증진시키기 위하여 탄소잉크로 스크린 프린트된 전극을 탄소나노튜브를 이용하여 변형하였으며, 변형한 탄소나노튜브 전극위에 콜레스테롤 산화효소와 과산화효소, 전자매개 물질로 페로시안 칼륨을 도포하여 제작하였다. 탄소나노튜브를 이용하여 변형한 콜레스테롤 바이오 센서는 전기화학적으로 콜레스테롤 측정시 매우 신속하고 안정된 신호를 나타내었다. 개발된 콜레스테롤 센서는 소량의 (0.5 μL) 시료로 총 콜레스테롤 100~400 mg/dL 영역에서 5초 이내에 직선적인 감응을 나타내었으며 좋은 재현성을 나타내었다(CV 4.0% 이하).

A development of disposable strip-type galactose sensor for point-of-care testing (POCT) was studied, which was constructed using screen-printed carbon electrodes. Galactose levels were determined by the redox reaction of galactose oxidase in the presence of potassium ferricyanide as an electron transfer mediator in a small sample volume (i.e., less than 1 µL). The optimal performance of biosensor was systematically designated by varying applied potential, operating pH, mediator concentration, and amount of enzyme on the electrode. The sensor system was identified as a highly active for the galactose measurement in terms of the sensitivity (slope = 4.76 ± 0.05 nA/µM) with high sensor-to-sensor reproducibility, the linearity (R2 = 0.9915 in galactose concentration range from 0 to 400 µM), and response time (t95% = <17 s). A lower applied potential (i.e., 0.25 V vs. Ag/AgCl) allowed to minimize interference from readily oxidizable metabolites such as ascorbic acid, acetaminophen, uric acid, and acetoacetic acid. The proposed galactose sensor represents a promising system with advantage for use in POCT.
The periodicity of the physiological flow has been the major interest of analytic research in this field up to now Among the mechanical forces stimulating the biochemical reaction of endothelial cells on the wall, the wall shear stresses show the strongest effect to the biochemical product. The objective of present study is to find the effects of velocity waveform on the wall shear stresses and pressure distribution along the artery and to present some correlation of the velocity waveform with the clinical observations. In order to investigate the complex flow phenomena in the bifurcated tube, constitutive equations, which are suitable to describe the rheological properties of the non-Newtonian fluids, are determined, and pulsatile momemtum equations are solved by the finite volume prediction. The results show that pressure and wall shear stresses are related to the velocity waveform of the physiological flow and the blood viscosity. And the variational tendency of the wall shear stresses along the flow direction is very similar to the applied sinusoidal and physiological velocity waveforms, but the stress values are quite different depending on the local region. Under the sinusoidal velocity waveform, a Newtonian fluid and blood show big differences in velocity. pressure, and wall shear stress as a function of time, but the differences under the physiological velocity waveform are negligibly small.
Some pretreatment methods have been proposed to enhance the biodegradability and to shorten the hydrolysis reaction time. By means of efficient pretreatment the suspended solids (SS) can be made of better accessible for the anaerobic bacteria. There are several ways how this can be accomplished, which include biological, mechanical, thermal, and chemical methods. For the sludge solubilization using the cavitation phenomenon, we have tried to develop a pretreatment process consisted of a reactor and pumps. The objectives of this study were to develop a advanced wastewater treatment consisted of IABR and the cavitation with PGA. The most effective removal for organic matter and nutrients were occured when both cavitation pretreatment and ${\gamma}$-PGA were applied at the IABR process. Only small portion of ${\gamma}$-PGA at a rate of 1.38mg/L, was enough to improve sedimentation ability, SS removal efficiencies, and sludge volume reduction. After the sludge solubilization by the cavitation, SCOD increased to 193% and SS decreased to 36%. The removal ratio of BOD was 94.5%, T-N removal ratio was 85.5% and T-P removal ratio was 84.9%. The combination process of the IABR with the cavitation and PGA addition seems to be very effective alternative wastewater treatment process.
A rapid liquid chromatography/tandem mass spectrometry (LC/MS/MS) assay method was developed for the determination of topotecan levels in rat serum. The assay utilized a single liquid-liquid extraction with a mixture of ethy l acetate and acetonitrile (6:1 v/v) and isocratic elution. The multiple reaction monitoring was based on the transition of m/z 422.0$\rightarrow$376.5 for topotecan and 315.1$\rightarrow$226.6 for clomipramine (internal standard). The developed assay was validated to demonstrate the specificity, recovery, lower limit of quantification (LLOQ), accuracy and precision. The assay was linear over a concentration range from 0.5-100 ng/mL, with LLOQ being 0.5 ng/mL using a small volume of rat serum (0.1 mL). The mean intra- and inter-day assay accuracy was 87.7-111.0% and 97.8-108.3, respectively, and the mean intra- and interday precision was between 1.6-4.3% and 3.8-10.3, respectively. The developed assay was applied to a pharmacokinetic study after a bolus i.v. injection of topotecan in rats.
An optimization procedure to find the inlet concentration profile that yields the most uniform deposition rate in a cylindrical CVD chamber has been developed. Assuming that the chemical reaction time is negligibly small, a SIMPLE based finite-volume method is adopted to solve the fully elliptic equations for momentum, temperature, and concentration. The inlet concentration profile is expressed by a linear combination of Chebyshev polynomials and the coefficients of which are determined by the local random search technique. It is shown that the present method is very effective in improving the uniformity of the deposition rate, especially when Re is high and/or the wafer is placed close to the inlet. The optimal profiles have been obtained for various Re, Gr, and geometry combinations.
착물 $trans^-[Cr(en)_2F(H_2O)]^{2+}$의 hydrolysis에 대한 속도상수를 분광광도법으로 여러가지 온도와 압력에서 측정하였다. 온도 범위는 30${\circ}C$ ~ 50${\circ}C$까지이고 압력은 1bar에서 1500bar까지 변화시켰다. 1bar, 30${\circ}C$에서 측정된 속도상수는 $2.632{\times}10^{-5}sec^{-1}$였다. 속도상수는 일정온도에서 압력이 증가함에 따라 감소하였다. 활성화 부피와 다른 활성화 파라미터들은 이러한 속도상수로 부터 계산되었다. 활성화 부피는 모두 양의 값을 가졌으며 0.447에서 3.152$cm^3$/mol내에 있고, 활성화 엔트로피는 작은 값을 가진다. 이 반응은 흡열반응이고, 실험적인 온도에서 엔탈피 지배 반응이다.
SiC particles, 8.3 ${\mu}m$ in volume average diameter, were chlorinated in an alumina tubular reactor, 2.4 cm in diameter and 32 cm in length, with reactor temperature varied from 100 to $1200^{\circ}C$. The flow rate of the gas admitted to the reactor was held constant at 300 cc/min, the mole fraction of chlorine in the gas at 0.1 and the reaction time at 4 h. The chlorination was negligibly small up to the temperature of $500^{\circ}C$. Thereafter, the degree of chlorination increased remarkably with increasing temperature until $900^{\circ}C$. As the temperature was increased further from 900 to $1200^{\circ}C$, the increments in chlorination degree were rather small. At $1200^{\circ}C$, the chlorination has nearly been completed. The surface area of the residual carbon varied with chlorination temperature in a manner similar to that with the variation of chlorination degree with temperature. The surface area at $1200^{\circ}C$ was 912 $m^{2}/g$. A simple model was developed to predict the conversion of a SiC under various conditions. A Langmuir-Hinshelwood type rate law with two rate constants was employed in the model. Assuming that the two rate constants, $k_{1}$ and $k_{2}$, can be expressed as $A_{1e}^{-E_{1}/RT}$ and $A_{2e}^{-E_{2}/RT}$, the four parameters, $A_{1}$, $E_{1}$, $A_{2}$, and $E_{2}$ were determined to be 32.0 m/min, 103,071 J/mol, 2.24 $m^{3}/mol$ and 39,526 J/mol, respectively, through regression to best fit experimental data.
토마토황화잎말림바이러스(Tomato yellow leaf curl virus; TYLCV)는 온실가루이(Bemisia tabaci)에 의해서 영속전염되는 DNA 바이러스로 토마토에 발생하는 가장 중요한 병 중 하나이다. 우리나라에서 TYLCV는 2008년 최초로 보고된 이래 급속하게 전국적으로 확산되어 토마토 생산에 심각한 경제적 손실을 일으키고 있다. 토마토 생산과정에서 TYLCV의 확산을 최소화하기 위해 바이러스의 조기진단이 매우 중요하다. 본 연구에서는 바이러스의 신속진단을 위해 초고속 정밀 PCR 진단기술을 개발하였으며, 이는 마이크로칩을 기반으로 하여 $5{\mu}l$의 시료만으로 PCR을 수행할 수 있도록 고안된, 휴대가 가능할 정도의 소형 GenSpector$^{TM}$ TMC-1000 PCR 기기를 이용한 새로운 기술이다. 본 연구에서 개발된 초고속 정량 PCR을 이용하였을 때 TYLCV 진단을 위한 30 cycle의 PCR과 용융점분석(melting curve analysis)에 15분 이내의 시간이 소요되었으며, GenSpector$^{TM}$ TMC-1000 PCR을 이용한 초고속 정밀진단 기술은 향후 TYLCV의 대발생을 모니터링하는데 유용하게 사용될 수 있을 것으로 생각한다. 본 연구결과는 GenSpector$^{TM}$ TMC-1000 PCR기반의 초고속정량 PCR 기술을 이용한 식물 바이러스의 진단기술 개발로는 최초의 보고이다.
