A Study on the Establishing a Mid-to Long-term Comprehensive Development Plan Based on Jeongseon-gun's Library Policy Innovation Strategy (정선군의 도서관 정책혁신전략을 기반으로 한 도서관 중장기 종합발전계획 수립연구)

  • Jeongseon-gun has the SaBuk Public Library, 17 small libraries, and the Jeongseon Educational Library. In 2023, with the completion of the Jeongseon County Public Library, there is a need to establish innovative library policy strategies that consider balanced development by region. The primary goal of this study was to set the direction for the future of Jeongseon County's public libraries and provide appropriate strategies. To achieve this, an examination of the local situation and characteristics of Jeongseon County was conducted, followed by an analysis of the operational status of the public libraries. Additionally, a satisfaction survey and demand analysis were conducted among the residents of Jeongseon County regarding the libraries, to analyze directions for library improvements. Ultimately, taking into account the local characteristics, library status, and residents' needs, an optimized strategy for the region was presented. As a result of the study, the operational goal to achieve the vision was set as "a life-friendly library that shares the healthy lives of residents of Jeongseon-gun," and emphasized its role as a companion close to daily life for the healthy lives of Jeongseon-gun residents. The four strategic tasks to promote the operational goals were expressed as "a library that grows with local residents," "a library that provides multicultural exchange," "a sustainable future-oriented library," and "a library that cooperates with the local community."

Identification of piRNAs in Hela cells by massive parallel sequencing

  • Piwi proteins and Piwi-interacting RNAs (piRNAs) have been implicated in transposon control in germline from Drosophila to mammals. To examine the profile of small RNA expression in human cancer cells and explore difference in small RNA transcriptome, small RNA libraries prepared from wildtype, HILI overexpressed and HILI knockdowned Hela cells were sequenced using Solexa technology. piRNAs and other repeat-associated small RNAs were observed in Hela cells. By using in situ hybridization, piR-49322 was localized in the nucleolus and around the periphery of nuclear membrane in Hela cells. Following the overexpression of HILI, the retrotransposon elements LINE1 was significantly repressed, while LINE1-associated small RNAs decreased in abundance. The present study demonstrated that HILI along with piRNAs plays a role in LINE1 suppression in Hela cancer cell line.

  • As small-scale libraries such as Children Library, Small Library, Miracle Library, have recently increased in number, it is required to establish the library consulting system in order to provide them with special knowledges and experiences necessary for library construction and operation. The result of the survey about public librarian's opinions confirms that the latent demand for library consulting is very high. In case of our country where the social experience in library consulting service is insufficient, it is desirable that the library consulting service be executed by the government offices as public service.

Identification and Function Prediction of Novel MicroRNAs in Laoshan Dairy Goats

  • MicroRNAs are a class of endogenous small RNAs that play important roles in post-transcriptional gene regulation by directing degradation of mRNAs or facilitating repression of target gene translation. In this study, three small RNA cDNA libraries from the mammary gland tissues of Laoshan dairy goats (Capra hircus) were constructed and sequenced, individually. Through Solexa high-throughput sequencing and bioinformatics analysis, we obtained 50 presumptive novel miRNAs candidates, and 55,448 putative target genes were predicted. GO annotations and KEGG pathway analyses showed the majority of target genes were involved in various biological processes and metabolic pathways. Our results discovered more information about the regulation network between miRNAs and mRNAs and paved a foundation for the molecular genetics of mammary gland development in goats.

  • In order to balance with supply policy, public housing management and operation policies have been implemented in terms of housing welfare, but citizens have not yet achieved the results that the citizens are experiencing. The purpose of this study is to analysis the residential satisfaction of the including the housing performance through the characteristics of the public housing residents in Seoul. The data used in this study is based on the survey data of public housing panel survey in Seoul (2016). The study method used ordered logistic regression analysis based on the fact that dependent variables appeared as ordered responses. Major research results are as follows. Firstly, housing performance and residential satisfaction may not match. Even though the satisfaction of housing area, type, and management fee is high, satisfaction with residential environment is low if commuting distance, the number of small libraries, and hospitals are small. Secondly, it showed different characteristics of residential environment factors among types of public housing. Rather than focusing on supply, customized supply is needed considering characteristics of public housing types. Thirdly, the policy for public housing needs to be realized by a fair policy on the residential environment. It is necessary to contribute to better housing stability as a customized policy considering the local residential environment.

Neutronic simulation of the CEFR experiments with the nodal diffusion code system RAST-F

  • CEFR is a small core-size sodium-cooled fast reactor (SFR) using high enrichment fuel with stainless-steel reflectors, which brings a significant challenge to the deterministic methodologies due to the strong spectral effect. The neutronic simulation of the start-up experiments conducted at the CEFR have been performed with a deterministic code system RAST-F, which is based on the two-step approach that couples a multi-group cross-section generation Monte-Carlo (MC) code and a multi-group nodal diffusion solver. The RAST-F results were compared against the measurement data. Moreover, the characteristic of neutron spectrum in the fuel rings, and adjacent reflectors was evaluated using different models for generation of accurate nuclear libraries. The numerical solution of RAST-F system was verified against the full core MC solution MCS at all control rods fully inserted and withdrawn states. A good agreement between RAST-F and MCS solutions was observed with less than 120 pcm discrepancies and 1.2% root-mean-square error in terms of keff and power distribution, respectively. Meanwhile, the RAST-F result agreed well with the experimental values within two-sigma of experimental uncertainty. The good agreement of these results indicating that RAST-F can be used to neutronic steady-state simulations for small core-size SFR, which was challenged to deterministic code system.

Election Promises on Library Fields of the Candidates and Implementation of Their Promises of the Elected Heads of Daegu Metropolitan City in the 5th Local Election (대구지역 자치단체장 후보자와 당선자의 도서관 공약 제시 및 이행 분석 - 2010년 제5회 전국동시지방선거를 중심으로 -)

  • This study explored election promises on library fields by the candidates for the mayor, the educational superintendent and the chiefs of districts of Daegu Metropolitan City and implementation of their election promises by the elected heads of Daegu Metropolitan City in the 5th local election of Korea. To do this, election promises of 36 candidates were analyzed by investigating the official gazettes for elections deposited in National Election Commission of Korea. Also, the implementation of their promises of 11 elected heads was analyzed by searching through the official websites, news databases and internet. As a result, election promises and implementation by the candidates for the mayor and the educational superintendent were mostly insufficient both in quality and in quantity. 54% of all candidates for the chiefs of districts presented election promises that were mostly related with construction of public libraries and installation of small libraries. Some elected chiefs actively implemented their promises, while the other elected chiefs did not fulfill their promises.

Bibliometric Analysis to Analyze Topic Areas of Faculty for Academic Library Service (대학도서관 서비스를 위한 서지분석기반 학과의 주제적 특성 분석 연구)

  • As topics of researchers become diverse horizontally or vertically, academic libraries have difficulties to identify the dynamic change of researchers' needs for academic publications. This research aims to illustrate the topic areas of researchers in a department of university by analyzing bibliographies of their publications. First, researchers' publications were used to discover the topic areas where the researchers had published. Second, the cited publications in those papers were analysed to identify the expanded topic areas of these researchers. Finally, highly cited journals were analyzed by network analysis method. The major finding is that the importance of topic areas by the number of journals was not necessarily proportional to that by the number of papers. Researchers have a tendency to use many papers in a small number of journals in a certain topic area. Furthermore, the importance of topic areas discovered by researchers' publications was not the same as that discovered by researchers' citations.

  • The docking and in silico ligand screening procedures can select small sets of lead -like candidates from large libraries of either commercially or synthetically available compounds; however, the vast number of such molecules make the potential size of this task enormous. To accelerate the discovery of drugs to inhibit several targets, we have exploited massively distributed computing to screen compound libraries virtually. The Korea@HOME project was launched in Feb. 2002, and one year later, more than 1200 PC's have been recruited. This has created a 31 -gigaflop machine that has already provided more than 1400 hours of CPU time. It has all owed databases of millions of compounds to be screened against protein targets in a matter of days. Now, the virtual screening software suitable for distributed environments is developed by BMD. It has been evaluated in terms of the accuracy of the scoring function and the search algorithm for the correct binding mode.

Successful Application of the Dual-Vector System II in Creating a Reliable Phage-Displayed Combinatorial Fab Library

  • The dual-vector system-II (DVS-II), which allows efficient display of Fab antibodies on phage, has been reported previously, but its practical applicability in a phage-displayed antibody library has not been verified. To resolve this issue, we created two small combinatorial human Fab antibody libraries using the DVS-II, and isolation of target-specific antibodies was attempted. Biopanning of one antibody library, termed DVFAB-1L library, which has a $1.3{\times}10^7$ combinatorial antibody complexity, against fluorescein-BSA resulted in successful isolation of human Fab clones specific for the antigen despite the presence of only a single light chain in the library. By using the unique feature of the DVS-II, an antibody library of a larger size, named DVFAB-131L, which has a $1.5{\times}10^9$ combinatorial antibody complexity, was also generated in a rapid manner by combining $1.3{\times}10^7$ heavy chains and 131 light chains and more diverse anti-fluorescein-BSA Fab antibody clones were successfully obtained. Our results demonstrate that the DVS-II can be applied readily in creating phage-displayed antibody libraries with much less effort, and target-specific antibody clones can be isolated reliably via light chain promiscuity of antibody molecules.
