  • Ten strains of Salmonella were isolated from feces and lymph nodes of swine slaughtered at Daegu slaughter-house and the rate of isolation was 6.0 percent. Nine strains of Salmonella were isolated by enrichment in selenite F broth and one strain by direct culture on SS agar, but none of Salmonella were isolated from MacConkey ager and in SS broth. Among Salmonella isolated, Salmonella typhimurium occupied over half (6 strains) and the importance of Salmonella in swine for the incidence of food poisoning in man was discussed.

닭고기 부분육 품질 등급 설정을 위한 도계 및 추가가공장 관리자 의식 조사 (Survey of Manager's Perception of Slaughter and Further Processed House for the Determination of Grading of Poultry Meat Cuts)

  • 본 실험은 닭고기 부분육의 품질 등급을 설정하기 위한 기초 자료를 얻고자 시중 도계장을 대, 중, 소로 구분하여 생산 관리 책임자들을 대상으로 설문지에 의한 방문 조사를 실시하였다. 부분육의 주요 거래처는 대규모 및 중규모 도계장은 중간 상인, 프랜차이즈, 백화점 순이었으나 추가가공장은 단체 급식에 공급하는 비율이 높았다. 도계 과정에서 닭고기 부분육 등급 판정을 수행하기 적당한 곳은 대규모 도계장의 경우 마지막 포장 단계를 선호하였다. 닭고기 부분육 등급은 몇 단계를 선호하는지에 대한 응답은 대규모 도계장 및 추가 가공장은 2단계를 선호하였고 중규모 도계장은 $2\~3$단계를 선호하였다 추가 가공육에 대해서는 도계장 규모와 관계없이 1단계를 선호하였다. 등급 판정 요인으로는 신선도 멍, 외상, 트리밍(손질) 이물질 순으로 중요도를 표현하였다. 절단육에 대한 등급 판정은 중규모 도계장 및 추가 가공장에서는 통닭 상태에서 판정하는 것을 선호하였고 대규모 도계장에서는 절단육 상태에서 판정하자는 의견이 많았다. 닭고기 부분육의 등급 판정 물량에 대해서는 도계장 규모와 관계없이 신청한 물량에 대해서만 판정하자는 의견이 많았다. 부분육등급 판정 도입 시기는 도계장규모와 관계없이 지금 당장은 아니어도 도입되는 것이 좋다는 의견이 다수를 차지하여 닭고기 부분육에 대한 품질 등급제를 선호하였다.

도축처리 단계별 도체 및 환경재료에서 Listeria monocytogenes의 분리 (Isolation of Listeria monocytogenes from animal carcasses and environmental specimens in slaughter house level)

  • To invastigate the epidemiological trait of listeriosis, Listeria monocytogenes were isolated from the carcasses of pigs and cattle, and environmental specimens in slaughter house. Also serotype of isolates were classified by rapid slide agglutination test. In the carcasses of pigs, Listeria sp were isolated from the carcasses after bleeding(62%), after dismemberment(60.0%) and before shipping(76.0%), and L monocytogenes were present in 8% of the carcasses after dismemberment and in 14% of the carcasses before shipping. However, few Listeria sp were isolated from the living body skin and the carcasses after scalding. In the carcasses of cattle, Listeria sp were isolated from the carcasses after bleeding(10%), after dismemberment(36.7%) and before shipping(63.3%), L monocytogenes were present in 3.3% of the carcasses after dismemberment and in 10% of the carcasses before shipping. Overall, L monocytogenes, L innocua, L welshimen, L grayi, and L murrayi were present In 4.8, 40, 2.3, 2.6 and 0.3% of all the carcasses, respectively. Prevalence of Listeria sp in environmental specimens were found to be 80% in slaughter house floors and 100% in sewage, and L monocytogenes were present in 15% of sewage. However, few Listeria sp were isolated from chilled water and from scalding water. Overall, L monocytogenes, L innocua, and L welshimeri were present in 3.8, 45 and 6.3% of all the environmental specimens, respectively. A total 27 strains of L monocytogenes were isolated from samples tested and all of the strains were classified into serotype 1.

ELISA를 이용한 돼지 톡소플라스마병의 조기 진단에 관한 연구 (Use of the enzyme-linked immunosorbent assay for the detection of toxoplasmosis in swine)

  • This study was conducted to evaluate the possibility of application of a microenzyme-linked immunosorbent assay(micro-ELISA) for the serodiagnosis of specific toxoplasma antibodies in swine sera and this test was performed as a microplate system by coating the polystyrene plates with toxoplasma soluble antigen, incubated serially diluted sera, then added horse radish peroxidase labelled goat anti-swine IgG(r) conjugate followed by o-phenylenediamine as substrate. The color development by enzyme-substrate reaction was determined by the photometric reading [ELISA reader at 490nm (OD)] and visual reading. The soluble antigen was prepared from the tachyzoites in mouse peritoneal cavity. A total of 1,200 swine sera from pig slaughter-house and a total of 116 swine sera from pig breeding station (S-C farm) were tested for the detection of antibodies to Toxoplasma gondii. The results obtained were summarized as follows: 1. The optimal reactions of indirect ELISA for the test sera were determined by the dilution of antigen 1:256 and 1:3,200 of horse radish peroxidase conjugate [anti-swine IgG(r)]. 2. The specific togoplasma antibody(IgG) in pigs infected with Tp artificially were detected as the serum titers of 1:64 or 1:128 at one week postinfection. 3. Of a total of 1,200 swine sera from pig slaughter-house 505 samples of sera were detected as positive (42.1%) and of a total of 116 swine sera from S-C pig breeding station 68 samples of sera as positive (58.6%). 4. The specific antibody(IgG) detection rates against a total of 1,200 test sera from pig slaughter-house were not significant between male (43.1%) and female (40.7%). 5. The indirect ELISA was proved to be a sensitive and specific procedure for the serodiagnosis of swine toxoplasmosis and also evaluated as an effective screening test for the large scale of test samples in laboratory.

충남 서부지역 돈군에서 분리된 Bordetella bronchiseptica의 성상에 대한 연구 (Studies on the properties of Bordetella bronchiseptica isolated from the pig herds in Western Chungnam)

  • During 2 years from Octorber 1992 to April 1994, prevalence of general respiratory diseases and atrophic rhinitis in the pig herds located in the Western Chungnam was investigated, and isolation of B. bronchiseptica was attempted for the pigs manifested with the clinical signs of atrophic rhinitis(AR). The isolates were characterized and identified in aspects of biochemical properties, antigenicity, drug sensitivity and pathogenicity. The results obtained through the experiments are summarized as follows; 1. During 2 years of investigation, the overall prevalence of the general respiratory diseases in the pi8 herds in Western Chungnam was 35.3%, consisting of 35.1% in the pig farms and 38.8% in a slaughter house. The prevalence by age groups accounts for 9.2% in adults, 44.7% in rearings and 25.3% in sucklings. By farm size, The highest prevalence of 56.5% was observed in the smallest farm with 1 to 200 heads. 2. The prevalence of clinical cases of artrophic rhinitis was recorded by 12.7% in the group that is the sows and piglets vaccinated, 28.9% in the group that is the sows only vaccinated and 39.8% in the group of the non-vaccinated groups. In the slaughter house, 53(24.8%) of 214 pigs examined exhibit the AR lesions. 3. A total of 189 strains of B. bronchiseptica were isolated from the pig herds. Isolation rates were 12.6% in the group that is the sows and piglets vaccinated, 34.1% in the group that is the sows only vaccinated and 45.7% in the group of the non-vaccinated groups. Isolation rate in the specimen from the slaughter house was 93( 43.5% ) of 214 pigs examined. Of the AR-non-vaccinated group, the piglets aged bet- ween 61 to 90 days revealed the highest isolation rate of 58.5%. 4. The titers of antibody against B. bronchiseptica were measured by tube agglutination test. The group that is the sow and piglet-vaccinated showed the highest titer of 640-2, 560 in sow and 640longrightarrow5, 120 in piglet. The group that is the sows only-vaccinated revealed 640-2, 560 in sows and 640-1, 280 in piglets. Both of the vaccinated groups showed 100% positive reaction. The group of the non-vaccinated sho-wed relatively lower titer of 0-1, 280 in both of sows and piglets. The positive rate of the sera obtained from the slaughter house was 53.3% with the antibody titer of 0-1, 280. 5. Biochemical and serological properities of 189 isolates were very similar to those of the reference B. bronchiseptical phase I type, indicating that most of isolates are B. bronchiseptica phase I type. 6. In antimicrobial drug susceptibility, 87.3% of 189 isolates was susceptible to chloramphenicol, 79.9%, to amikacin, 64.6%, to cephalothin and less than 35.4% to others. 7. In agar-gel immunodiffusion and SDS-PAGE analysis, the isolates presented the identical antigenicity and protein profiles to the reference standard strains. 8. The whole cells and bacterial filtrates of the isolates were inoculated to guinea pigs and mice. The isolates showed the hish pathogenicity and dermonecrotoxiciy.

도축장의 생.해체검사를 통한 경산우 및 미경산우 감별법(I) (Discrimination of cows from heifers by antemortem and postmortem inspection in slaughter house)

  • This study was performed to make helpful standard for discrimination of cows from heifers by uterine and ovarian section inspection. A total of 3,736 heifers and 249 cows were inspected from January to November 2006 in Karak Market Slaughter house. Firstly, it was obvious that non-corpus albicans were observed in almost heifers(3,597/3,736, 96.3%), but there were distinctive corpus albicans in all cows (249/249 100%). Secondly, another evident was that the length and diameter of the horn in uterus was below 30mm in 94.5% (3,267/3,736) heifers, but above 30mm in 90.4% (225/245) cows, and it was easy to distinguish between right and left horn by difference of size in 87.6% (218/249) cows. Thirdly the diameters of pelvic cavity was below 220mm in 96.8% (3,615/3,736) heifers, but was over 200mm in 85.1% (212/249) cows. Fourthly another difference was found in second sacrum union condition and 13th thoracic vertebral process's states. In case of heifers, it was distinctive five lines in each second sacrum, but it was non-distinctive or united in the same each lines of the sacrum in cows. Fifthly in the results of 13th thoracic vertebral colors, the ratio between red color and white color was 7 : 3 in heifers (3,385/3,736, 90.6%), but that was 2 : 8 in cows (199/249, 79.9%). Consequently, it was possible to use the method which discriminated cows from heifers by uterine and ovarian section inspection in large scaled field slaughter house.

Farm to abattoir conditions, animal factors and their subsequent effects on cattle behavioural responses and beef quality - A review

  • The current review seeks to highlight the concerns that have been raised on pre-slaughter stress, contributing factors and its consequent effects on cattle behavioural responses and the quality of beef; inter-linking the activities involved from birth to slaughter. Such information is crucial in light of the consumer concerns on overall animal welfare, quality of meat and food security. Slaughter animals are exposed to different conditions during production and transportation to abattoirs on a daily basis. However; the majority of studies that have been done previously singled out different environments in the meat production chain, while conclusions have been made that the welfare of slaughter animals and the quality of meat harvested from them is dependent on the whole chain. Behaviour is a critical component used to evaluate the animals' wellbeing and it has been reported to have an effect on product quality. Apart from the influence of on-farm, transportation and abattoir conditions, the genetic background of the animal also affects how it perceives and responds to certain encounters. Stress activates the animals' hypothalamic-pituitary-adrenal activity, triggering release of various stress hormones such as catecholamines and cortisol, thus glycogen depletion prior slaughter, elevated ultimate pH and poor muscle-meat conversion. Pre-slaughter stress sometimes results to cattle attaining bruises, resulting to the affected parts of the carcass being trimmed and condemned for human consumption, downgrading of the carcass and thus profit losses.

Influence of Municipal Abattoir Conditions and Animal-related Factors on Avoidance-related Behaviour, Bleeding Times at Slaughter and the Quality of Lamb Meat

  • The objective of this study was to determine the effect of municipal abattoir conditions and animal factors on avoidance-related behaviour (AB) of sheep at slaughter, bleeding times (BT) and mutton quality. The behaviour of 66 castrates and 19 ewes of different age categories was observed at three stages of slaughter. Higher behaviour scores indicated higher levels of AB. Time intervals between the start of blood flow and the time the flow changed from a constant stream into drips were recorded as BT. Thirty two meat samples were obtained to measure quality variables. These were colour ($L^*$, $a^*$ and $b^*$), $pH_{24}$, temperature, cooking loss (CL) and tenderness. Correlations were determined between BT and meat quality variables. Animal behaviour at slaughter differed with breed, gender and age group. Avoidance behaviour was higher in the Dorper breed than in both the Merino breed and their crosses. It was also higher in younger (<10 months) lambs than in older sheep. Castrates were more aggressive or in panic than ewes. Castrates had longer ($72.6{\pm}0.53s$) BT than the ewes ($63.6{\pm}2.82s$). Ewes had higher CL ($39.8{\pm}1.04%$) values than castrates ($35.1{\pm}0.95%$). Meat from castrates was tougher ($32.6{\pm}1.95N$) than the meat from ewes ($24.3{\pm}1.16N$). There were no significant correlations obtained between BT and meat quality variables. It can therefore be concluded that abattoir conditions, breed, age and gender had an effect on AB at slaughter. Gender had an effect on BT and mutton quality.

  • A large plerocercoid was found from abdominal cavity of hen slaughtered in a slaughter-chicken house at Chyung Yang market in Seoul. The worm was identified as Diphyllobothrium mansoni of Genus Spirometra and the length of whole body was approximately 50 cm.
