Zymomonas mobilis에 의해 생성된 Fructan (Levan)의 특성 및 화장품 원료로의 개발

  • Fructan(Levan)은 식물체 및 미생물에서 발견되는 탄수화물로 이는 과당(fructose)이 $\beta$-2, 6 결합으로 연결되어 있는 polysaccaride 이다. 본 연구에서는 Fructan을 생성하는 미생물(Zymomonas mobilis)과 10% sucrose(기질), 1-2% 효모 추출물을 주성분으로 하는 배지를 사용하여 30-37$^{\circ}C$, pH 5.0-7.0에서 20-24시간 동안 배양한후 원심분리하여 균체를 제거하고 3배량의 알코올을 가하여 침전, 건조하여 얻은 Fructan의 화장품 원료로서의 가능성을 조사하였다. 보습효과에 있어서는 Hyaluronic acid와 유사하였으며, keratinocyte에 대한 세포증식 효과를 나타내었다. 또한 3-D culture에 의해 구축된 생인공 피부내에 0.05%의 sodium lauryl sulfate (SLS)를 사용하여 피부자극에 의한 초기 염증 반응을 유도한후 0.01mg/m1, 0.05mg/m1의 Fructan을 각각 처리하였을 때, SLS만을 처리한 생인공피부와 비교하여 세포증식효능을 보였고, SLS 자극물질로 유도된 전염증성 조절인자인 interleukin-l$\alpha$(IL-l$\alpha$)의 분비량을 조사 하였을때 0.01mg/ml, 0.05mg/ml의 Fructan을 처리한 생인공피부의 IL-l$\alpha$ 양이 Fructan을 처리하지 않은 것보다 상대적으로 감소하였다. 이러한 결과로 Fructan이 생인공 피부내 피부 세포의 증식효과를 나타낼 뿐만 아니라, 또한 피부자극물질에 의한 염증반응에 대해 자극완화효능이 있음을 알 수 있었다. 섬유아세포 및 동물을 이용한 안전성 시험에서도 독성이 없는 안전한 원료로 평가되었다.

Reproductive Biology of the Washington Clam Saxidomus purpuratus in the Coastal Waters of Yeosu, Korea (한국 여수에 서식하는 개조개(Saxidomus purpuratus)의 산란생태)

  • The reproductive biology of the Washington clam Saxidomus purpuratus, from Yeosu, Korea was investigated based on 699 individuals collected from January to December 2022. The sex ratio of this species was not significantly different from a 1:1 (male:female) sex ratio (P>0.05). Monthly changes in gonadosomatic index were analyzed to estimate the spawning season. The reproductive cycle of this species can be divided into the following stages: active (Jan.-Mar.), ripe (Mar.-Nov.), spent (Apr.-Dec.), and degenerative (Jun.-Dec.) stages. The spawning period ranged from March to December with the main periods being from April to June and from September to November. The shell length at 50% female group maturity (SL50) was estimated to be 60.2 mm.

TENSILE BOND STRENGTH OF ALUNMINA CORE TREATED BY ION ASSISTED REACTION (이온보조반응법으로 처리한 알루미나 코아의 인장결합강도에 관한 연구)

  • This study was undertaken to evaluate the tensile bond strength of In-Ceram alumina core treat-ed by ion assisted reaction(IAR). Ion assisted reaction is a prospective surface modification technique without damage by a keV low energy ion beam irradiation in reactive gas environments or reactive ion itself. 120 In-Ceram specimens were fabricated according to manufacturer's directions and divided into six groups by surface treatment methods of In-Ceram alumina core. SD group(control group): sandblasting SL group: sandblasting + silane treatment SC group: sandblasting + Siloc treatment IAR I group: sandblasting + Ion assisted reaction with argon ion and oxygen gas IAR II group: sandblasting + Ion assisted reaction with oxygen ion and oxygen gas IAR III group: sandblasting + Ion assisted reaction with oxygen ion only For measuring of tensile bond strength, pairs of specimens within a group were bonded with Panavia 21 resin cement using special device secured that the film thickness was $80{\mu}m$. The results of tensile strength were statistically analyzed with the SPSS release version 8.0 programs. Physical change like surface roughness of In-Ceram alumina core treated by ion assistad reaction was evaluated by Contact Angle Measurement, Scanning Electron Microscopy, Atomic Force Microscopy; chemical surface change was evaluated by X-ray Photoelectron Spectroscopy. The results as follows: 1. In tensile bond strength, there were no statistically significant differences with SC group, IAR groups and SL group except control group(P<0.05). 2. Contact angle measurement showed that wettability of In-Ceram alumina core was enhanced after IAR treatment. 3. SEM and AFM showed that surface roughness of In-Ceram alumina core was not changed after IAR treatment. 4. XPS showed that IAR treatment of In-Ceram alumina core was enabled to create a new functional layer. A keV IAR treatment of In-Ceram alumina core could enhanced tensile bond strength with resin cement. In the future, this ion assisted reaction may be used effectively in various dental materials as well as in In-Ceram to promote the bond strength to natural tooth structure.

Bacterial Blight of Sesame Caused by Xanthomonas campestris pv. sesami (Xanthomonas campestris pv. sesami에 의한 참깨의 세균성잎마름병)

  • A new bacterial disease of sesame(Sesamum indicum) was observed on field-grown plants in Suwon, Hongchun and Yeonchun in 2000. Leaf symptoms initially appeared as water-soaked spots that gradually enlarged, became necrotic and were often bordered by a small zone of lemon yellow tissue. In the case of severe infection, dead leaves were defoliated. Isolations made from diseased leaves on yeast extract dextrose calcium carbonate agar yielded nearly pure cultures of a yellow-pigmented bacterium typical of a xanthomonad. Two bacterial strains were purified and used for farther tests. Pathogenicity of strains was confirmed on 3-week-old sesame plants sprayed with bacterial suspensions containing $10^{8}cfu/ml$ of phosphate buffered saline. The Biolog and fatty acid analyses of the two strains(SL3451 and SL3476) 1mm sesame leaf blight showed that they could be identified as ft campestris pv. sesami because of their high similarity to the tester strain(X. campestris pv. sesami LMG865) with a match probability of $100\%$. The bacterium grew well between 18 and 36$^{\circ}C$, but optimum temperature was $27^{\circ}C$ on LB broth. This is the first report of bacterial blight of sesame in Korea. Symptoms of bacterial blight of sesame are difficult to differentiated with those of bacterial leaf spot caused by Pseudomonas syringae pv. sesami.

Analysis of Phytosterols and Tocopherols, and Production of Structured Lipids from the Extracted Plant Oils (선택된 식물자원에서 추출조건에 따른 Phytosterol과 Tocopherol의 함량분석 및 추출유로부터 재구성지질의 합성)

  • To obtain the oil sesame, walnut, whole wheat, and rice bran were extracted for 1, 3, or 6 hr by a shanking water bath(35$^{\circ}C$ and 100 rpm), and by soxhlet extractor(80$^{\circ}C$) for 1, 3 or 6 hr, respectively. The highest yield of extracted oil was obtained from the walnut(63.07% weight) and the whole wheat showed the lowest extraction yield of oil(1.13% weight). Major fatty acids from the extracted oils were linoleic, oleic, and palmitic acid. The maximum contents of total phytosterol in sesame, walnut, whole wheat, and rice bran were 0.44, 1.57, 2.25, and 2.03(% weight), respectively. Besides, total tocopherol contents in sesame, walnut, whole wheat, and rice bran were maxima 3.42, 0.16, 2.92, and 0.07(% weight), respectively. From the extracted oils, structured lipids(SL) were synthesized by the interesterification reaction with conjugated linoleic acid(CLA) in a shanking water bath at 55$^{\circ}C$. When the reactions(1:3 substrate molar ratio, extracted oil:CLA) were conducted for 24 hr, maxima 23.75 mol% of CLA incorporation was obtained from walnut oil and, in other cases, 16.28 - 19.15 mol% of CLA was found in the produced SL triacylglycerol molecules.

First Record of Polymetme elongata (Stomiiformes: Phosichthyidae) from the Southern Yellow Sea and Jejudo Island, Korea (황해 남부 및 제주도 남부 해역에서 채집된 Polymetme elongata (앨퉁이목: 긴앨퉁이과) 한국 첫기록)

  • Two specimens of Polymetme elongata (Phosichthyidae; Polymetme) were collected by a bottom trawl from both the southern coastal waters of Jejudo Island in October 2016 (70.65 mm SL) and Yellow Sea, Korea in August 2022 (54.04 mm SL). They are an elongated body with large mouth, 11 dorsal fin rays, 10 pectoral fin rays, 7 pelvic fin rays, 30~32 anal fin rays, 1 adipose fin, 18 gill rakers, 44 vertebrae, 1 preorbital photophore, 1~2 ventrolateral rows of photophores on body, 1 paired photophores in symphysis of lower jaw, 9 branchiostegal rays, 2 rows of 8 phorophores between pelvic fin origin and anal fin origin, and 1 row of 23 photophores thereafter. At the origin of anal fin, the second photophore is more higher than third photophore and the first photophore is most lowest. Our specimens differ from the other species of the same genus by the ratio of the head length to the standard length (1/5) and the gill rakers (18), absolutely. As this species is the first record of the family Phosichthyidae, genus Polymetme, and P. elongata in Korean waters, we suggest their new Korean names, "Gin-ael-tung-i-gwa", "Gin-ael-tung-i-sog", and "Gin-ael-tung-i", respectively.

Effect of intermittent low-intensity, short duration exercise on Type IImuscle of suspended rats (간헐적인 낮은강도, 짧은기간의 운동부하가 뒷다리 부유쥐의 Type II근육에 미치는 영향)

  • The purpose of this study was to determine the effect of intermittent low - intensity, short duration exercise during hindlimb suspension on the mass, relative weight, myofibrillar protein content, cross-sectional area of Type I and Type II fibers and SDH activity in Type II(plantaris) muscle. To examine the effectiveness of intermittent low-intensity, short duration exercise on mass, myofibrillar protein content and fiber size, the hindlimbs of adult female Wistar rats were suspended(HS) and half of these rats walked on a treadmill for 45 min/day(9 min every 2h) at 5m /min and a 15$^{\circ}$grade (HS-EX). Plantaris wet weight was 19.67% significantly smaller(p<0.005) and relative plantaris weight was 6.25% smaller compared with those of control rats following seven days of hindlimb suspension. Plantaris wet weight and relative plantaris weight increased by 27.66%, 12.22% each through intermit-tent exercise during hindlimb suspension(p<0.005, p<0.05), moreover, plantaris wet weight and relative plantaris weight of the HS-EX rats were similar to those of control rats. Soleus wet weight and relative soleus weight decreased significantly by 31% and 22.0% in the HS rats(p<0.05). Soleus wet weight and relative soleus weight increased by 10.41%, 25.64% respectively through intermittent ex-ercise during hindlimb suspension, furthermore, soleus wet weight and relative weight of the HS-EX rats were closer to those of control rats. Myofibrillar protein content of plantaris and soleus decreased significantly by 51.49%, 59.65% each, following seven days of hindlimb suspension (p<0.005) Myofibrillar protein content of plantaris and soleus increased by 51.79%, 75.47% each with significance through intermittent exercise during hindlimb suspension(p<0.005). Myofibrillar protein content of plantaris and soleus in HS-EX rats was smaller than that of control rats. No change was observed in fiber type percentage following 1 week of hindlimb suspension or exercise during hindlimb suspension. The type I fiber cross-sectional area of both soleus and plantaris muscle was 18.72% and 41.07% lower in the HS than that of the controls (p<0.05, p<.001), that of both muscles was 6.60% and 29. 3% greater in the HS-EX than that of the HS rats. HS plus intermittent low- intensity short duration exercise resulted in Type I fiber cross-sectional area closer to the controls. Type II fiber cross-sectional area of both plantaris and soleus muscle was 22.45% and 22.58% sl nailer in the HS than in the controls, that of both muschles in the HS-EX was 14.10%, 5.78% greater than HS. Intermittent exercise during hindlimb suspension resulted in Type I, II fiber cross-sectional area closer to the control value. There was no change in SDH activity following 1week of hindlimb suspension or exercise during hindlimb suspension in the plantaris muscle. The results suggest that intermittent low intensity short duration exercise can ameliorate Type II muscular atrophy Induced by hindlimb suspension.

  • In this paper, we propose the progressed interworking method between H.323 and SlP, then explain the improved property. The VolP(Voice over Internet Protocol) technology which is able to use a voice service through internet is more cheaper then existing telephone charges, and is easil)· accept the various of multimedia services from internet. Previous connectionmethod of VoIP used H.323 protocol, but it is very complex to connection establishment. so, the SIP(Session Initiation Protocol) protocol that propose in SIP-Working Group is in use recently. Therefore, we need new interworking methodology between H.323 and SIP Products. In this thesis, the progress interworking method between H.323 and SIP are Propose, then interpret unnecessary packet delay for call setup and improved feature of message exchange.

Transcriptional Analysis of Genes Involved in Ectopic Sporulation in Streptomyces griseus (Streptomyces griseus의 특이적 포자형성에 관여하는 유전자의 전사량 분석)

  • Two Streptomyces griseus strains, a wild-type strain and an A-factor-dependent transcriptional activator mutant strain harboring multiple copies of a gene, dasA, that encodes a substrate-binding protein of the ATP-binding cassette transporter, showed severe ectopic sporulation of young substrate hyphae in response to glucose. The effect of dasA overexpression on the ectopic sporulation of Streptomyces strains was evaluated by comparing the transcriptomes of the strain harboring multiple copies of dasA and a strain harboring empty vector. By DNA microarray, 4 genes (SGR794, SGR2469, SGR3656, and SGR3657) and 3 clusters (SGR795-797, SGR2377-2378, and SGR6997-6998) were differentially expressed by more than 2-fold in S. griseus strains harboring dasA. The DNA microarray result was validated by low-resolution S1 nuclease mapping.

Determination of Flavonoids from Allium victorialis var. platyphyllum and Their Effect on Gap Junctional Intercellular Communication

  • This study was carried out to identify and quantify the flavonoids from 6 different plant parts of Allium victorialis var. platyphyllum (AVP), including the flower, leaf, root, stem, flower stalk, and flower seed, using liquid chromatography/ mass spectrometry. Two major flavonoids were structurally identified as quercetin (3,5,7,3'4,'-pentahydroxyflavone) and kaempferol (3,5,7,4'-tetrahydroxyflavone) at contents of 11.8-25.8 and $6.0-64.4\;{\mu}g/mL$, respectively. In particular, the flower and root plant parts contained the highest amounts of quercetin and kaempferol compared to the other parts. We also assessed the recovery effects of each plant-part extract of AVP on gap junctional intercellular communication (GJIC) in WB-F344 cells by the scrape-loading and dye transfer (SL/DT) method. According to the results, GJIC was reduced by approximately 70.2% ($62.3{\pm}12.5$ cells) compared to the control ($209{\pm}9.5$ cells, 100%) when 12-O-tetradecanoylphorbol-13-acetate (TPA) was treated alone in the WB-F344 rat liver epithelial cells. However, the stem extract (0.2 mg/mL) restored GJIC to basal levels (92%, $204{\pm}2.3$ cells, p<0.01) and the flower extract (0.2 mg/mL) stimulated GJIC to 82.5% ($172.6{\pm}8.3$ cells, p<0.05), when applied together with the TPA.
