The objective of this study is to estimate the inhibition of skin melanin formation by extract of Angelica koreana and Cnidium monnieri and the possibility of functional cosmetic materials through anti-irritation and stability test. The extract used in this experiment is White-$AK^{TM}$ and the INCI name is Osthole. The main component of White-$AK^{TM}$ was identified as coumarin and EC50 value was 2.7ppm by mouse melanoma B 16 cell test. White-$AK^{TM}$ showed inhibitory effects 100 times lower concentration than arbutin. The main mechanism for skin whitening effect thought to be inhibition of tyrosinase-related gene expression. The basic essence formulation of White-$AK^{TM}$ 5% solution applied to the skin showed the effect of relieving skin irritation. White-$AK^{TM}$ in an opaque container, under UV conditions for 4 weeks, and showed close to 100% recovery and 97% recovery under $50^{\circ}C$ for 4 weeks. Therefore, it is thought that White-$AK^{TM}$ which helps skin whitening, relieving skin irritation and stable in UV condition is able to be used as the functional component in the cosmetic formulation.
These days there is a constant possibility of exposure to UV radiation which can cause abnormal production of melanin and result in skin disease such as hyperpigmentation and melanoma. Many materials were investigated for skin whitening and protection against UV radiation. In this study, we assessed the melanogenesis inhibitory activities of Korean Red Ginseng (KRG, Ginseng Radix Rubra) and Ginkgo (EGb 761 Ginkgo Biloba) in an attempt to develop a new skin whitening agent derived from natural products. B16F10 melanoma cells were treated for 48 hr with KRG and EGb 761. The inhibitory effect on melanogenesis was measured and related cytokines and proteins expression were also investigated by RT-PCR and Western blotting. In addition, we also assessed the effects of these substances on the skin of C57BL/6 mice. Cell growth, melanin content and tyrosinase activity were inhibited effectively in B16F10 melanoma cells treated with KRG and EGb 761. Moreover, tyrosinase mRNA expression was inhibited clearly and melanogenesis related proteins (MRPs) containing tyrosinase, TRP1 and TRP2 were also reduced by KRG and EGb761, while cytokines such as IL-$1{\beta}$ and IL-6 were induced. In the case of UV irradiated mice, we observed induction of cytokine mRNA levels and reduction of MRPs mRNA expression. In addition, a decrease in pigmentation from treatment with KRG and EGb 761 on the skin of mice was observed. These results indicate that KRG and EGb 761 inhibit melanogenesis in B16F10 cells and have display protective activities against UVB. Therefore, we suggest that KRG and EGb 761 are good candidates to be used as whitening agents and UVB protectors for the skin.

Isomaltol glycoside is a hydrophilic furanic glycoside in which the amino acids and sugars of ginseng are thermally denatured during red ginseng production. Various skin whitening tests were conducted on isomaltol glycoside containing a lot of red ginseng extract in order to investigate the skin whitening effect as a cosmetic raw material. We have tested melanin content assay in B16-F10 cells, zebrafish embryo pigmentation assay, mushroom tyrosinase inhibitory activity, western blot analysis to determine skin whitening activity of isomaltol glycosides. In the zebrafish melanin content assay, isomaltol glycoside decreased total melanin content by about 20% and zebrafish tyrosinase activity by about 10% after treatment with 50 and $100{\mu}g/mL$ compared to the untreated control group. Isomaltol glycoside also showed a concentration-dependent decrease in melanin content in B16-F10 melanoma. Furthermore, it increased the expression of MITF phosphorylation factors p-AKT and p-ERK in B16-F10 melanoma and decreased the concentration of MITF. It also inhibited tyrosinase, TRP-1 and TRP-2 expression. The content of isomaltol glycoside was about 3% in the ginseng extract and about 1% in the ginseng root. Thus, isomaltol glycoside is considered as one of the main components that exhibit the whitening activity of ginseng when considered quantitatively as whitening activity.

The half-dried leaves of Stewartia. pseudocamellia were extracted with hot water (SPE) and partitioned with n-hexane (SPEH), dichloromethane (SPED), and ethyl acetate (SPEE) successively. SPE and SPEE showed significant inhibitory effects against melanogenesis and tyrosinase activities. By bioassay-guided isolation, ten phenolic compounds were isolated by column chromatography from SPEE. The whitening effect of the isolated compounds from SPEE were tested for the inhibitory activities against melanogenesis using B16 melanoma cells, in vitro inhibition of tyrosinase, and L-3,4-dihydorxy-indole-2-carboxylic acid (L-DOPA) auto-oxidation assay. A cytotoxic activity assay was done to examine the cellular toxicity in Raw 264.7 macrophage cells. Of the compounds isolated, gallic acid and quercetin revealed significant inhibitory activities against melanogenesis compared to arbutin. In particular, quercetin exhibited similar inhibitory activities against tyrosinase and L-DOPA oxidation without cytotoxicity. These results suggested that SPE could be used as a potential source of natural skin-whitening material in cosmetics as well as in food products.
Mulberry extracts can be incorporated into skin-whitening products. The compound attributed to lighten the skin is arbutin, a form of hydroquinone that inhibits melanin release by suppressing the tyrosinase enzyme. For the cosmetic applications, the physiological effects of mulberry (Morus alba) extracts were investigated. The water soluble fraction of mulberry contains higher amount of protein (16.28~4.47%) in contrast to fat (1.55~1.41%). In addition, the fraction abundantly contains succinic acid (972.4-275.8 mg/g) and phosphoric acid (1,628.4-121.9 mg/g) in different parts of mulberry. The free radical scavenging ability in water soluble fraction was found to display remarkable effects in comparison with methanol and ethyl acetate fraction. The ethyl acetate-soluble of root and leaf showed remarkable tyrosinase inhibition activity by IC 50 (${\mu}g/ml$). The anticancer activity of methanol fraction obtained from mulberry using human cancer cell lines showed growth inhibition effect (270.14 mg/ml in Calu-6 cells, 295.29 mg/ml in HCT-116, and 332.29 mg/ml in MCF-7 cells, respectively). Based on the results, Morus alba extracts include cosmetic ingredients with antioxidizing and whitening properties.

The coffee silver skin is a part of coffee beans. We report that the coffee sliver skin extracts exhibited cosmetic properties of antioxidant, anti-winkle and whitening effects. The ethanol extracts of silver skin showed free radical scavenging activity up to 92.26% in $50{\mu}g/mL$, especially against DPPH radical and ABTS radical cation. The silver skin extracts showed inhibitory effects for tyrosinase activity and DOPA oxidation in a dose-dependent manner, suggesting the extracts retain for the whitening property in cosmetics. The coffee silver skin extracts effectively inhibited the elastase and collagenase. Cytotoxicity of the coffee silver skin extracts was measured by the colorimetric MTS assay. The viability of the human keratinocytes (HaCaT) treated with the coffee silver skin extracts was same as that of untreated cells, indicating the extracts are safe to human cells. Here, we suggest that the silver skin extracts of coffee bean could be a potential natural substance for anti-winkle, whitening, antioxidant properties for cosmetics.
To evaluate the whitening effect of black tea water extract (BT), BT was topically applied to artificially hyperpigmented spots on the back skins of brown guinea-pigs (weight: 450~500 g) induced by 1,500 mJ/$cm^2$ of ultraviolet B (UVB) irradiation. The test compounds of 30 ${\mu}l$ were applied twice a day, six days a week, for four weeks. The artificially hyperpigmented spots were divided into 5 groups: control (UVB + saline, C), vehicle control [UVB + propylene glycol: ethanol: water (5 : 3 : 2), VC], positive control (UVB + 2% hydroquinone, PC), experimental 1 (UVB + 1% BT), experimental 2 (UVB + 2% BT). After 4-week application, the spots were removed by biopsy punch under anesthetic condition and used as specimens for the histological examination. The total polyphenol and flavonoid contents of BT were 104 and 91 mg/g, respectively. The electron-donating ability of BT revealed a dose-dependent response, showing the excellent capacities of 86% at 800 ${\mu}g$/ml. The artificially hyperpigmented spots treated with the PC and BT were obviously lightened compared to the C and VC groups. At the fourth week, the melanin indices for the PC and BT groups were significantly lower (p < 0.001) than those of the C and VC groups. In histological examination, PC and BT groups were significantly reduced in the melanin pigmentation, the proliferation of melanocytes and the synthesis of melanosomes compared to the C and VC groups. It is found that BT inhibits the proliferation of melanocytes and synthesis of melanosomes in vivo using brown guinea pigs, thereby showing a definite skin whitening effect.

Dendropanax morbifera Leveille (Araliaceae) is an endemic species growing in the south-western part of South Korea and has been used in folk medicine. However, the effects of Dendropanax morbifera Lev. on skin biology remain to be elucidated. In this study, we isolated 1-tetradecanol and ${\beta}$-sitosterol from the n-hexane fraction of Dendropanax mobifera Lev. and To investigate the whitening effect of the fraction, we tested the inhibition of tyrosinase activity of 1-tetradecanol. The results show that the inhibitory effect of the 1-tetradecanol was higher than water extract and n-hexane fraction. And 1-tetradecanol significantly reduced melanin contents of B16F10 cells compared to more than water extract and n-haxane fraction dose-dependantly without cell cytotoxicitiy (below $100{\mu}g/mL$). We also investigated the skin moisturizing effect using HR-1 hairless mice. The transepidermal water loss (TEWL) in the 1-tetradecanol treated group was significantly smaller than that in the other groups. To investigate the effect of the preventing hair loss by ${\beta}$-sitosterol, we observed HR-1 hairless mice through periodic growth feature. The results suggest that hair loss of mice by ${\beta}$-sitosterol was delayed and it's hair density showed the highest. These data provide evidence that Dendropanax morbifera Lev. may be a potent candidate for the improvement of both skin whitening, moisturizing and alopecia from the point of cosmetic industry view.
The aim of this study was to investigate the effect of deformable liposomes with sodium cholate on the skin permeation and skin deposition of arbutin, a hydrophilic skin-whitening agent. Various compositions of liposomes were prepared by the extrusion method. Particle size distribution and entrapment efficiency were determined by the laser light scattering and the gel permeation chromatography, respectively. The in vitro rat skin permeation and deposition of arbutin in various skin layers were investigated using the Keshary-Chien diffusion cells at $37^{\circ}C$. The average particle size of the deformable liposomes ranged from 217.4 to 117.4 nm, depending on the composition. The entrapment efficiency was dependent on surfactant concentration and loading dose of arbutin. The permeation rate of 5% arbutin in deformable liposomes was $8.91({\pm}1.33){\mu}g/cm^2/h$, and was not significantly different from 5% arbutin aqueous solution $[9.82({\m}0.86){\mu}g/cm^2/h]$. The deposition of arbutin was $43.34({\pm}12.13)$ and $16.99({\pm}7.83){\mu}g/cm^2$ in stratum corneum layer and epidermis/dermis layer, respectively, after 12 h of permeation study. These results are consistent with several earlier studies for the localization effect of liposomal formulations in stratum corneum, and demonstrated the feasibility of the deformable liposomes as a promising carrier for the skin deposition of hydrophilic skin-whitening compounds.

Grape skins are a natural resource rich in antioxidants, but people only eat grape flesh and have discarded the skins. This study investigated the possibility of using grape skin extract as a raw material for functional cosmetics. The dried grape skin powder was put in distilled water and stirred for 1 h, and then the supernatant separated from the solid was used as an extract. The extract yield was 17.8 ~ 31.4%, and the total flavonoid and polyphenol contents in the extract were 1.8 ~ 2.5 mg-QE g-extract-1 and 16.9 ~ 20.3 mg-GAE g-extract-1, respectively. The extract effectively removed radicals of DPPH and ABTS, and the degree of scavenging increased with the concentration of the extract. The extract inhibited the collagen hydrolysis activity of collagenase, and the activity inhibition rate increased to 84.2% as the extract concentration increased. However, notable inhibition of tyrosinase by the extract was not found. As the extract of Chamaecyparis obtusa was added to the grape-skin extract, the tyrosinase inhibition rate increased, but the DPPH radical scavenging activity decreased. This study found that grape skin extract has a high antioxidant capacity and anti-wrinkle effect but a low whitening effect. However, by mixing the grape skin extract with the extract of C. obtusa in an optimal ratio, the whitening effect was improved with excellent antioxidant and anti-wrinkle effects.
