The purpose of this study was to investigate the antioxidant and skin whitening effects of Artemisia iwayomogi extract. Artemisia iwayomogi was extracted with 100% ethanol and water. The antioxidative and skin whitening effects of these extracts were determined with in vitro assays by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) method assessing the inhibitory effects on tyrosinase activity and melanogenesis in B16 melanoma cells. Radical scavenging activity of the extracts was tested by DPPH assay which showed a high DPPH radical scavenging activity ($SC_{50}$; 17.1 ppm in EtOH, 198.4 ppm in water). In term of tyrosinase inhibitory activity, Artemisia iwayomogi ethanol extract showed high inhibition activity ($IC_{50}$: 481.8 ppm). In B16 mouse melanoma cells, the ethanol extract significantly inhibited melanin synthesis by 36.8% at a concentration of 50 ppm. These results suggest that Artemisia iwayomogi ethanol extract has significant antioxidant activity and whitening activity.

In this study, we investigated the anti-oxidative, anti-wrinkle and whitening effects of Platycarya strobilacea bark extracts. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$) of extract / fractions of Platycarya strobilacea was in the order: 50% ethanol extract ($6.75{\mu}g/mL$) < deglycosylated aglycone fraction ($6.62{\mu}g/mL$) < ethyl acetate fraction ($4.15{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of some Platycarya strobilacea extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol-dependent chemiluminescence assay. The order of ROS scavenging activity was ethyl acetate fraction (OSC50, $0.56{\mu}g/mL$) < 50% ethanol extract ($0.02{\mu}g/mL$) < deglycosylated aglycone fraction ($0.01{\mu}g/mL$). The deglycosylated aglycone fraction showed the most prominent scavenging activity. The protective effects of extract / fractions of Platycarya strobilacea on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethanol extract (50%) suppressed photohemolysis in a concentration dependent manner, particularly ethyl acetate fraction exhibited the most prominent cellular protective effect (${\tau}_{50}$, 717.27 min at $10{\mu}g/mL$). The inhibitory effect of Platycarya strobilacea extracts on tyrosinase were investigated to assess their whitening efficacy. Finally, their anti-elastase activities were measured to predict the anti-wrinkle efficacy in the human skin. The inhibitory effect ($IC_{50}$) on tyrosinase of some Platycarya strobilacea extracts was 50% ethanol extract ($243.98{\mu}g/mL$) < ethyl acetate fraction ($153.87{\mu}g/mL$) < deglycosylated aglycone fraction ($137.53{\mu}g/mL$). Also, The inhibitory effect of elastase ($IC_{50}$) of some Platycarya strobilacea extracts was 50% ethanol extract ($31.01{\mu}g/mL$) < ethyl acetate fraction ($14.42{\mu}g/mL$) < deglycosylated aglycone fraction ($1.48{\mu}g/mL$). The cream containing the ethyl acetate fraction of Platycarya strobilacea extracts was formulated. The skin hydration, transepidermal water loss, and the whitening effects were investigated after topical application of the cream. The skin hydration of cream containing extract was increased by $2{\sim}8%$ than the placebo cream, transepidermal water loss was decreased. The cream containing extract suppressed the melanogenesis of skin by 9.55% than the placebo cream. These results indicate that extract / fractions of Platycarya strobilacea can function as antioxidants in biological systems, particularly skin exposed to UV radiation by anti-oxidative activity and protect cellular membranes against ROS. The inhibitory effect on elastase and tyrosinase, and the increase of skin hydration and the whitening effect of the cream containing extract could be applicable to new functional cosmetics for antiaging.

Melanin plays a key role in the protection of skin from ultraviolet light that generates reactive oxygen species (ROS), such as superoxide, hydroxyl radical, singlet oxygen and hydrogen peroxide. However, the ROS leading to the oxidation of lipids, proteins and DNA are involved in the overproduction of melanin that is known to cause melasma, age spots and freckles. Among the herb medicines, Ulmus macrocarpa used in this study was reported to contain flavonoids as a main component. The aim of this study is to investigate the whitening and anti-oxidant effects of Ulmus macrocarpa ethanolic extracts (UMEE) in B16F1 cells. UMEE below $3.12{\mu}g/ml$ did not show cytotoxicity. In an anti-oxidant experiment, UMEE showed not only high reducing power and scavenging activity on DPPH, but it was also observed that UMEE exhibit an inhibitory effect on lipid peroxidation. UMEE did not display an inhibitory effect on tyrosinase activity in vitro. However, UMEE inhibited melanin synthesis in B16F1 cells. In addition, UMEE reduced the expression levels of tyrosinase and tyrosinase-related protein-2 (TRP-2), which are key enzymes in melanogenesis. These results indicate that UMEE exert a whitening effect through the inhibition of both tyrosinase and TRP-2 expressions as well as anti-oxidant activity, suggesting that UMEE could have the functional potential for a whitening effect on the skin.

The imports of Centella asiatica L. Urban are increasing year-by-year due to the fact that its extract is a raw material used for skin wounds and in cosmetics. However, studies on the cultivation and identification of native C. asiatica species in Korea have been extremely rare. Therefore, this study was conducted in order to investigate the physiological and functional activity of Korean native C. asiatica plant cultivated in Hapcheon, Gyeongsangnam-do, Korea. As a result, the highest antibacterial and anti-inflammatory activities were examined with methanol extract while skin-whitening and wrinkle improvement were examined with water extract. Seven bacterium and one fungus were treated with 50% methanol extracts of C. asiatica and most of the bacterium showed similar or low levels of antibacterial activity compared to the control group of Omiza (Schisandra chinensis) extract, except for Streptococcus pyogenes, which showed higher antimicrobial activity than that of Omiza extract. However, neither C. asiatica and Omiza extracts showed antimicrobial activity against the fungus, C. albicans. The results of anti-inflammatory effect analyses with Raw 264.7 cells confirmed that the treatment of methanol extract reduced the level of NO by 50% or more compared to the control group. In addition, the water extract showed the highest reduction of melanin content of up to 20% more than the control group when examined with B16F10 cell line, indicating a significant skin-whitening effect. Furthermore, we were able to show the significant skin wrinkle improvement caused by C. asiatica extract with NHDF cell as an indicator, but strong cytotoxicity was also observed, suggesting that further studies are necessary.
There is growing interest in natural cosmetic ingredients as natural cosmetics become popular. As a part of effort to look for natureal cosmetic agent, seaweed Sargassums were tested for functional cosmetic agents. Effective materials were extracted from Sargassum coreanum. Sargassum hemiphyllum, and Sargassum patens by simple hot water extraction. Antioxidation, whitening, anti-wrinkle, UV absorption and anti-inflammation effects were studied for functional cosmetic agents. Sargassum extracts indicated excellent cell viability, strong anti-oxidation effect by DPPH radical scavenging activity and showed significant whitening effect from tyrosinase inhibition. However, effects of antiwrinkle, UV absorption and anti-inflammation were negligible. In conclusion, Sargassum coreanum extracts showed good possibility for anti-oxidation and whitening cosmetic agent.
Hwang Chil (Dendropanax morbifera Lev.) belongs to Aralia elata group. This plant is only found in Korea and is considered to be along with ginseng. The physiological activities of it have been reported as having scavenging effect on oxygen free radicals and anti-carcinogenic effect. As most of the anti-oxidants were effective on protecting hepatotoxicity from alcohol, hard tissues restoring and skin whitening, the effectiveness of Hwang Chil extracts was screened. (omitted)

Silkworm pupae, a by-product of the silk industry are known to be valuable resource of nutrients for humans as well as animals besides encompassing diverse bioactive constituents. However, there is a paucity of knowledge on their role in amelioration of oxidative stress and anti-skin aging properties. In the present study, we evaluated the inhibitory effect of aqueous as well as ethanolic (30% and 70%) extracts from distinct stages of male and female silkworm pupae belonging to two silkworm varieties on skin aging-related enzymes. The activities of collagenase, elastase and tyrosinase were effectively inhibited by 70% ethanolic silkworm pupal extracts (SPE), followed by 30% with aqueous extracts exhibiting meager inhibitory potential. SPE were also investigated for their antioxidant activity in oxidative-stressed murine fibroblasts (L929). The intracellular ROS and lipid peroxidation induced by tert-butyl hydroperoxide (t-BHP) in fibroblasts was better attenuated by pre treatment with ethanolic (30%) and aqueous extracts, respectively. The safety of the extracts was determined by studying their effect on fibroblast cell viability and it was found that none of the extracts were cytotoxic. Our findings indicate the potential utility of SPE as anti-aging components in cosmeceuticals.
Reactive oxygen species (ROS) are known to lead to oxidation of lipids, proteins, and DNA and cause skin damage. Moreover, ROS promote melanogenesis, which causes melasma, age spots, and freckle. The main compounds of the herbal medicines Poria cocas, Glycyrrhiza uralensis, and Ulmus macrocarpa were reported to be parchymic acid, glabridin, and flavonoids, respectively. The aim of this study was to investigate the whitening and antioxidant effects of a mixture of P. cocas, G. uralensis, and U. macrocarpa extracts (PGUE) in B16F1 cells to develop whitening cosmetics. PGUE inhibited DPPH radicals and lipid peroxidation, in addition to high reduction power, compared with Glycyrrhiza uralensis ethylacetate extracts (GUEE). Furthermore, PGUE exhibited a protective effect against DNA oxidation induced by the hydroxyl radicals. In addition to its antioxidant activity, the inhibitory activity of PGUE against tyrosinase, which is associated with melanogenesis, was greater than that of arbutin used as a positive control. Moreover, PGUE exerted an inhibitory effect on melanin synthesis in live melanoma cells and reduced the expression levels of superoxide dismutase-1 (SOD-1) and tyrosinase related protein-1 (TRP-1). These results indicate that PGUE has skin whitening and antioxidant effects, suggesting that this mixture can be used as the main ingredient in the development of effective whitening cosmetics.
We investigated antioxidative activity, tyrosinase inhibitory activity, and melanin production inhibitory activity of the methanol extract of Paeoniae Radix and its fractions. The total polyphenol content of the extract was 73.45 mg/g, and content of the ethyl acetate fraction was 514.50 mg/g as the highest content of fractions. In DPPH radical scavenging ability, $SC_{50}$ values of the ethyl acetate was 3.86 ${\mu}g/ml$ as a result of greater activity in the positive control (ascorbic acid). Moreover, tyrosinase inhibitory activity of the ethyl acetate fraction showed higher activity than arbutin used as a positive control. In the cytotoxicity measurement by MTT assay, the extract and fractions were exhibited cell viabilities of 76.96~157.26% against Raw 264.7 and B16F10 cell in concentration of 10~100 ${\mu}g/ml$. In nontoxic concentration range, the ethyl acetate fraction showed strong melanin production inhibitory effect in ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH)-stimulated B16F10 cell. As a result, the ethyl acetate fraction of the methanol extract from Paeoniae Radix could be applicable to functional materials for skin-whitening agents.
The study was conducted to investigate functional materials as skin whitening and anti-inflammatory agent from Taraxacum officinale and Taraxacum coreanum. The total polyphenol and flavonoid content in the ethanol extract of Taraxacum officinale were found to be 64.07mg/g and 32.46mg/g, respectively. In tyrosinase inhibitory activity, the hot water extract of Taraxacum coreanum was higher than the other extracts. However, in nitric oxide (NO) scavenging ability, the ethanol extract of Taraxacum coreanum was higher than the other extracts. the ethanol extract of Taraxacum coreanum showed strong NO production inhibitory effect in lipopolysaccharide (LPS)-stimulated Raw 264.7 cell. In the cell viability measurement by MTT assay and the lactate dehydrogenase (LDH) assay against L929 cell, the extracts were exhibited fine cell viabilities and normal LDH release levels as nontoxic result in sample concentration of $250{\sim}1000{\mu}g/m{\ell}$. As a result, the ethanol extract and the hot water extract of Taraxacum coreanum could be applicable to functional materials for anti-inflammatory and skin whitening related fields, respectively.
