A Clinical Research about Herbal Cosmetics Containing Caffeic acid phenethyl ester Isolated from Vespae Nidus on the Whitening Effects (노봉방에서 추출된 카페인산 페네틸 에스테르가 함유된 한방화장품의 미백 개선에 관한 임상적 연구)

  • Objectives: The purpose of this clinical research was to investigate the effects of herbal cosmetics containing Caffeic acid phenethyl ester (CAPE), an ingredient compound of various herbs including Vespae Nidus, on the Whitening Effects. Methods: A total of 20 subjects who visited Pusan National University Hospital from March 2nd, 2017 to April 11th, 2017 were included. In this study, we evaluated the visual evaluation, the melanin index (M) and erythema index (E) through skin analysis equipment, the subjective whitening improvement analysis, and the adverse reaction according to product use. Statistical analysis was performed with independent t-test and Mann-Whitney's U test. Statistical significance was achieved if the probability was less than 5% (p<0.05) Results: As a result of the visual evaluation, it was effective in improving skin whitening. The subjective whitening improvement analysis also showed positive results. However, no statistically significant differences were found between the test and control cosmetics in the melanin index (M) and erythema index (E) through skin analysis equipment. In the safety evaluation, some adverse reactions were reported, but no significant were observed directly from cosmetics. Conclusions: Considering the above results, we have confirmed the possibility of herbal cosmetics containing containing CAPE of Vespae Nidus Extracts.

A Study on Antioxidant, Skin-Whitening and UV Absorption Effect of Beta vulagaris Using Lactiplantibacillus plantarum SM4 (Lactiplantibacillus plantarum SM4 를 이용한 레드비트 생물전환물의 항산화, 피부 미백과 자외선 흡수 효능 연구)

  • The purpose of this study was to confirm the skin whitening and UV absorption effects of bioconverted Beta vulagaris (BBE) using Lactiplantibacillus plantarum SM4, and the effects were evaluated by measuring total polyphenol content (TPC), total flavonoid content (TFC), radical elimination activity (RSA), and tyrosinase activity inhibition (TAI). TPC and TFC of BBE were 25.0 mg GAE/g DM and 8.05 mg QE/g DM, which were 1.3 and 1.1 times higher than hot-water extract (HWE) respectively. RSA, an indicator of antioxidant activity, and TAI, an indicator of skin-whitening effect, were 36.8%, and 68.6%, respectively, 1.1 and 1.2 times higher than that of HWE. UVA and UVB absorption of BBE were 21.4 and 87.6%, which was 1.4 and 1.7 times higher than that of HWE respectively. When main substances of BBE were analyzed using LC-MS/MS, kaempferol and isorhamnetin, a type of polyphenols known to have high antioxidant and skin-whitening effects, were identified. As a results, BBE is expected to be used as a functional cosmetic material as it has excellent antioxidant, skin whitening, and UV absorption properties.

The Antioxidant and Skin-whitening Effects of Saccharomyces cerevisiae FT4-4 Isolated from Berries Grown in Sunchang (화장품 소재로서 순창 베리류 유래 Sacchromyces cerevisiae FT4-4의 항산화 활성 및 미백 효과)

  • Saccharomyces lysate has the well-known function of soothing the skin in various ways: it is an anti-irritant and can treat skin care conditions, such as skin whitening and antioxidative activity. However, data on the safety for use of Saccharomyces lysate in cosmetics and skin care products are still limited. To design a new cosmetic material with antioxidant and skin-whitening effects, 80 yeast strains were isolated from berries grown in Sunchang. Among the isolates, the FT4-4 strain, which exhibited superior biological activities, was selected for further experiments. The FT4-4 strain was identified as Saccharomyces cerevisiae by 18S rRNA gene sequencing analysis. S. cerevisiae FT4-4 showed higher DPPH radical-scavenging (51.41%), superoxide dismutase (62.23%), and tyrosinase inhibition (64.75%) activities. The highest yield of biomass (3.16 g/l) and maximum growth rate of S. cerevisiae FT4-4 were observed within 16 h. Furthermore, the cytotoxicity potential of S. cerevisiae FT4-4 on B16F10 melanoma cells was measured by an MTT assay, and the results indicated that S. cerevisiae FT4-4 had a capacity to inhibit melanin up to 72.02% at an initial 10 mg/ml concentration. These results suggest that S. cerevisiae FT4-4 could be a promising candidate as a multi-functional material for application in the cosmetic industry, especially because of its antioxidant and skin-whitening effects.

Skin-whitening effects of hot water extract from domestic edible mushrooms (국내 시판 식용버섯 열수 추출물의 피부미백 기능 연구)

  • Hot water extracts from 16 domestic edible mushrooms including Pleurotus ferulae (Lanzi) X.L. Mao, Lentinula edodes (Berk.) Pegler, and Hypsizygus marmoreus (Peck) H.E. Bigelow, which are commercially available, were used for determining the cosmetic potential of these mushrooms. In this study, we carried out in vitro functional experiments to determine the inhibitory effects of these extracts on L-DOPA oxidation of tyrosinase and melanin synthesis. Based on the results of these experiments, H. marmoreus (Peck) H.E. Bigelow No. 10 and No. 15 were selected for further analysis. We analyzed the melanin synthesis inhibitory activity, TRP1 and MITF expression via real-time PCR, and Fontana Masson staining in artificial skin $Neoderm^{(R)}-ME$. Taken together, we observed that the hot water extract from H. marmoreus (Peck) H.E. Bigelow (No. 15) had better whitening effect than the extracts of other mushrooms. Thus, it can be a potential source of skin-whitening agent.

Enhancement of Skin-Whitening and UV-Protective Effects of Centella asiatica L. Urban by Utrasonification Process (초음파 병행을 통한 병풀의 미백 및 자외선 차단 활성 증진 효과)

  • Enhancement effect of ultrasonification process on UV-protection and skin-whitening activities using Centella asiatica L. Urban extract was investigated. Cytotoxicity of the extracts measured on human skin fibroblast cells, CCD-986sk, and then, ultrasonification associated extracts showed 5~9% lower cytotoxicity then normal crude extracts on 1.0 mg/$m{\ell}$ of highest sample concentration. The associated extrats showed highest inhibition activity of hyaluronidase on 1.0 mg/$m{\ell}$ of concentration as 54.2%. Also, the associated extract reduced expression of MMP-1 on UV-irradiated CCD-986sk cells down to 100.2% from 136.1%, and revealed high inhibitory potency on tyrosinase as 74.6% by adding 1.0 mg/$m{\ell}$ of concentration. Ultrasonification associated extract showed strong inhibition effect of melanin production on Clone M-3 cells as 84.2% by adding 1.0 mg/$m{\ell}$ of concentration. From the preliminary observations, we considered that the extracts from C. asiatica could be potent natural materials for skin-whitening and anti-aging agent, and could enhance the activities by ultrasonification process.

Effects of Lycii Fructus Extracts(LFE) on Skin whitening and Elasticity using Melanoma cells (구기자 추출물이 피부 미백 및 주름에 미치는 영향)

  • Objective : Lycii Fructus Extracts(LFE) can do Anti-hypertension activity, Antidepressant, Anti-diabetic activity. This study was designed to investigate effects of LFE on skin whitening and elasticity using melanoma cells. Methods : In this experiment, effect of LFE on cell viability, inhibition of melanin synthesis and inhibitory effect on tyrosinase and elastase. Results : More than $250{\mu}g/ml$ of LFE treated group showed lowered proliferation rates significantly compared to non-treated group. More than $125{\mu}g/ml$ of LFE treated groups were lower levels of melanin synthesis respectively. LFE showed inhibitory effect on tyrosinase activities in vitro. And, LFE suppressed tyrosinase activities in B16F10 cells significantly. Finally, LFE suppressed elastase type I and IV activities in dose-dependent manner in vitro. And LFE also slightly suppressed elastase activities in vivo. Conclusion : These results suggest that LFE can inhibit melanin synthesis through ihhibitory action on tyrosinase activity and inhibt elastase activity, and also suggest that these results can be used for the study on maintaining skin elasticity or whitening.

Potential Skin Whitening Agents of Natural Origin from South Asian Region (남아시아지역의 천연 미백제의 연구 현황)

  • South Asian's growing obsession with fair skin has made the cosmetic industry into a multibillion-dollar trade over the last decade alone. With reports of toxicity and potential mutagenicity of conventional skin lightening agents, cosmetic industries are looking for plant-based skin whitening formulations. In this review some potential depigmentation agents from South Asian region are discussed, including their historical background, biochemical characteristics and recent findings on their depigmenting activity.

Idescarpin Isolated from the Fruits of Idesia polycarpa Inhibits Melanin Biosynthesis

  • Tyrosinase is an enzyme that catalyzes the biosynthetic pathway of melanin pigments participating in the coloring of skin, hair, and eyes, and is widely distributed in nature. The inhibitory compounds of tyrosinase have been extensively used as a cosmetic agent with a skin-whitening effect. In this paper, several plant extracts were screened using Melan-a cells for the melanin biosynthesis inhibition activity, and Idesia polycarpa was selected. A melanin biosynthesis inhibitor was isolated from I. polycarpa fruits by activity-guided fractionation, and the inhibitor was identified as 6-hydroxy-2-[[[(1-hydroxy-6-oxo-2-cyclohexenl-yl)carbonyl]oxy]methyl]phenyl$\beta$-D-glucopyranoside (idescrapin) by comparing it with reported spectral data. Idescarpin $(IC_{50}=8{\mu}g/ml)$ reduced melanin content compared with the vehicle. In addition, the inhibitory activity of idescarpin for melanin synthesis is mediated by decreasing tyrosinase protein rather than directly inhibiting the tyrosinase activity. These results suggest that idescarpin isolated from I. polycarpa fruits may be used as a skin-whitening agent.

The Anti-wrinkle Effects and Whitening Effects of Galla Rhois (오배자의 항 주름 효과 및 미백 효과에 미치는 실험적 연구)

  • Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search Galla Rhois (GR) inhibitory activity against aging process of skin in vitro. GR was screened for their inhibitory activity against elastase. GR (final concentration 1 mg/ml) appeared over 40% of inhibition of elastase activity. So we investigated anti wrinkle effects of GR look through MMP-1 inhibition activity. Also Extracts of GR showed higher anti-tyrosinase activity than arbutin (final concentration 1 mg/ml). These results suggest that GR could be potential sources of anti-aging and whitening effects for the skin. Objective : Galla Rhois extracts, Elastase (pancreatic from porcine pancreas), N-Succinyl-(Ala)3-p-nitroanilide, Ursolic acid, mushroom tyrosinase, L-tyrosine, arbutin, Amicon Ultra, Hand Hold UV lamp, MMP-1 primary antibody, MMP-1 secondary antibody Result : 1. GR appeared over 40% of elastase inhibitory activity. 2. GR had MMP-1 inhibition activity. 3. GR showed higher anti-tyrosinase activity than arbutin. Conclusion : According to above results, it is supposed that Galla Rhois could be strong potential sources of anti-aging and whitening effects for the skin.

  • Gentisic acid, a skin-whitening agent, is known to possess tyrosinase inhibition activity. In order to develop an effective skin-whitening agent, hydroquinone derivatives in which the carboxylic acid moiety of gentisic acid was replaced with various functional groups, were selected and evaluated for their ability to inhibit tyrosinase activity as well as to inhibit melanin release.
