Skin-derived precursor cells (SKPs) are multipotent, sphere-forming and embryonic neural crest-related precursor cells that can be isolated from dermis. It is known that the properties of porcine SKPs can be enhanced by leukemia inhibitory factor (LIF) which is an essential factor for the generation of embryonic stem cells in mice. In our present study, to enhance or maintain the properties of murine SKPs, LIF was added to the culture medium. SKPs were treated with 1,000 IU LIF for 72 hours after passage 3. Quantitative real time RT-PCR was then performed to quantify the expression of the pluripotent stem cell specific genes Oct4, Nanog, Klf4 and c-Myc, and the neural crest specific genes Snai2 and Ngfr. The results show that the expression of Oct4 is increased in murine SKPs by LIF treatment whereas the level of Ngfr is decreased under these conditions. Interestingly, LIF treatment reduced Nanog expression which is also important for cell proliferation in adult stem cells and for osteogenic induction in mesenchymal stem cells. These findings implicate LIF in the maintenance of stemness in SKPs through the suppression of lineage differentiation and in part through the control of cell proliferation.
Background: Vitiligo is a chronic autoimmune disease in which the destruction of melanocytes causes white spots on the affected skin. Janus kinase (JAK) is a family of intracellular, non-receptor tyrosine kinases that transduce cytokine-mediated signals via the JAK-signal transducer and activator of transcription pathway. The aim of the present study is to explore the possible role of JAK1 in the pathogenesis of vitiligo using immunohistochemical methods. Methods: The current study was conducted in a sample of 39 patients who presented with vitiligo and 22 healthy individuals who were age and sex matched as a control group. We used immunohistochemistry to evaluate JAK1 status (intensity and distribution) and assess the percentage of residual melanocytes using human melanoma black 45 (HMB45). Results: Intense and diffuse JAK1 expression was significantly more likely to indicate vitiliginous skin compared to normal skin (p<.001). Strong and diffuse JAK1 expression was associated with short disease duration, female sex, and lower percentage of melanocytes (detected by HMB45) (p<.05). Conclusions: JAK1 may be involved in the pathogenesis of vitiligo, as indicated by intense and diffuse expression compared to control and association with lower percentage of melanocytes detected by HMB45 immunostaining.
This study aims to investigate how the woman's beauty makeup trend had been changed in 1960s by analyzing the characteristics of woman's make-up in that period. For this study, a lot of documents and papers related to the woman's make-up were collected and reviewed. In particular, to analyze the characteristics of woman's make up trend in 1960s, a lot of women's pictures and makeup-related articles from 143 volumes of woman's magazines such as Yeowon and Jubusaenghwal were collected. Among the 624 pictures, fifty two pictures which were good enough for studying the total face and makeup were used for this study. The period of this study ranges from 1960 through 1969 and the analysis is focused on skin expression, eyebrow, eye shadow, eye line, lipstick, and brusher and through the related articles, the intention and contents of make up. were delivered and found out. The results are as follows. In early 1960s, the woman's make up was characterized by the make up ranging from unnatural and gray skin expression to dense pink skin expression, giving natural and glossy tint onto the skin and in particular putting a bright accent on the eyelid, eye or lip. In the mid-1960s, the make up style expressed skin more naturally, giving more shading on the eyelid, nose and lip, thus having cubic make-up. In the late 1960s, the make up became more refined and harmonized by using colors according to the TPO (time, place and objective) and skin color. The study results show that the women in 1960s pursued the "cute and young looking image" and used the make up to express their images young and cute. In the mid 1960s, the minimalism which pursued the pure and simple make up appeared in US. That trend affected the make up style of Korean women and hence Korean women showed more natural make up style in the mid and late 1960s.
본 연구에서 루꼴라(Eruca sativa)에 대한 다양한 생리활성을 조사하여 기능성소재 응용가능성을 검토하였다. 루꼴라 추출물은 B. subtilis, E. coli, C. albicans에 대한 항균활성이 매우 우수하게 나타났으며 특히 그람양성세균 B. subtilis에 대한 항균활성이 높았다. 또한, 인볼루크린 발현에 대한 면역조직화학 분석을 인공피부를 이용하여 분석한 결과 루꼴라 추출물을 처리(50 mg/L)한 경우에 0.1% DMSO로 처리한 대조군에 비하여 인볼루크린 발현이 뚜렷하게 증가하였으며 동일농도로 처리된 합성물질 WY14643과 비교하였을 때에도 비슷한 발현양상을 보여 주었다. 이와 같은 결과로 미루어 볼 때 루꼴라 추출물은 피부염 개선 및 피부장벽 기능을 향상할 수 있는 천연물 기반의 소재로 활용하기 위한 매우 효과적인 재료가 될 수 있다고 사료된다.
Objectives : This study was conducted to investigate the effects of Gwakhyangjeonggi-san(GJS) on atopic dermatitis(AD) induced by 2,4-dinitrochlorobenzene(DNCB) in mice. Methods : The mice(Balb/c mice) were divided into three groups; normal Balb/c mice with oil treatment(Sham group), DNCB-induced AD mice(AD group), and GJS treated AD mice(GJS group). GJS group were orally administered GJS daily for 2 weeks. We observed changes of clinical skin severity score, the expression of thymic stromal lymphopoietin(TSLP), interleukin(IL)-4 and tumor necrosis factor(TNF)-${\alpha}$ in skin and mast cell infiltration. Also, serum immunoglobulinE(IgE), IL-4, $TNF-{\alpha}$ and IL-6 were evaluated. Results : The clinical skin severity score of GJS group was decreased compared to AD group. In hematoxylin and eosin staining results, GJS group showed a significant reduction of epithelial skin thickness. In addition, expression of TSLP and mast cell infiltration in skin were also reduced by GJS treatment compared to those of AD group. Thus, we evaluated expression of IL-4, Th2-dependent cytokine, and $TNF-{\alpha}$, pro-inflammatory cytokine in skin. GJS significantly reduced both IL-4 and $TNF-{\alpha}$ compared to AD mice. Moreover, levels of IgE, IL-4, $TNF-{\alpha}$ and IL-6 in plasma also significantly decreased by oral GJS treatment. Conclusion : The present study suggests that GJS can significantly reduced symptoms of AD, therefore it can be a promising candidate for anti-atopic dermatitis treatment.
얼굴의 표정은 얼굴의 구성요소같은 기하학적 정보와 조명이나 주름 같은 세부적인 정보들로 표현된다. 얼굴 표정은 기하학적 변형만으로는 실감적인 표정을 생성하기 힘들기 때문에 기하학적 변형과 더불어 텍스처 같은 세부적인 정보도 함께 변형해야만 실감적인 표현을 할 수 있다. 표정비율이미지 (Expression Ratio Image)같은 얼굴 텍스처의 세부적인 정보를 변형하기 위한 기존 방법들은 조명에 따른 피부색의 변화를 정확히 표현할 수 없는 단점이 있다. 따라서 본 논문에서는 이러한 문제를 해결하기 위해 서로 다른 조명 조건에서도 실감적인 표정 텍스처 정보를 적용할 수 있는 비선형 피부색 모델 기반의 표정 합성방법을 제안한다. 제안된 방법은 동적 외양 모델을 이용한 자동적인 얼굴 특징 추출과 와핑을 통한 표정 변형 단계, 비선형 피부색 변화 모델을 이용한 표정 생성 단계, 유클리디 거리 변환 (Euclidean Distance Transform)에 의해 계산된 혼합 비율을 사용한 원본 얼굴 영상과 생성된 표정의 합성 등 총 3 단계로 구성된다. 실험결과는 제안된 방법이 다양한 조명조건에서도 자연스럽고 실감적인 표정을 표현한다는 것을 보인다.
High levels of inflammatory cytokines were proposed contributors to the pathogenesis of a various inflammatory skin disorders. Therefore, investigating the immune response of the inflammatory skin disorder allows a better understanding of pathogenesis of a various inflammatory skin disorders and therapeutic approaches. The aim of this study was to analyze of the immune response in dogs with chronic inflammatory skin disease. To this aim, the present study evaluated relative mRNA expression of canine $IFN-{\gamma}$, IL-4, $TGF-{\beta}$ and IL-10 using TaqMan realtime PCR assays and semi-quantitative RT-PCR in freshly isolated peripheral blood mononuclear cells from twenty dogs with chronic inflammatory skin disease and ten normal dogs. The relative mRNA expression levels of IL-4 mRNA were significantly higher in dogs with chronic inflammatory skin disease than those in normal dogs (P < 0.01). The results of present study also showed a tendency towards increased expression of IL-10 transcripts in dogs with chronic inflammatory skin disease. However, there were no significant differences in the levels $IFN-{\gamma},\;TGF-{\beta}$ between normal and chronically inflammed dogs. In addition, the concentration of serum IgE was significantly increased in dogs with chronic inflammatory skin disease compared with those in normal dogs (P < 0.01). In histopathological examination, we found that there were markedly increased mast cell counts in chronically inflammed dogs (P < 0.05). These results suggest that the pathogenesis of chronic inflammatory skin disease might be associated with a T-cell mediated inflammatory responses characterized by a Th2-skewed immune response. Based on these results, the modulation of Th1/Th2 balance may be an effective therapeutic strategy for the treatment of chronic inflammatory skin disease.

Recently, there have been considerable efforts to search for naturally occurring substances for the intervention of carcinogenesis. Curcumin, a yellow coloring ingredient of turmeric (Curcuma longa L., Zingiberaceae), has been shown to inhibit experimental carcinogenesis and mutagenesis, but molecular mechanisms underlying its chemopreventive activities remain unclear. In the present work, we found that curcumin inhibited 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced expression of CPX-2 in female ICR mouse skin when applied topically 30 min prior to TPA as determined by both immunoblot and immunohistochemical analyses. Rab25, a member of the Rab11 small GTPase family, is central to achieving cellular polarity in epithelial tissues. Rab25 is highly expressed in epithelial cells of various tissues including breast, vagina, cervix, the gastrointestinal tract, and skin. Rab25 plays key roles in tumorigenesis, mainly by regulating epithelial differentiation and proliferation. However, its role in skin physiology is relatively unknown. In this study, we demonstrated that Rab25 knock-out (KO) mice show a skin barrier dysfunction with high trans-epidermal water loss and low cutaneous hydration. To examine this observation, we investigated the histology and epidermal differentiation markers of the skin in Rab25 KO mice. Rab25 KO increased cell proliferation at the basal layer of epidermis, whereas the supra-basal layer remained unaffected. Ceramide, which is a critical lipid component for skin barrier function, was not altered by Rab25 KO in its distribution or amount, as determined by immunohistochemistry. Notably, levels of epidermal differentiation markers, including loricrin, involucrin, and keratins (5, 14, 1, and 10) increased prominently in Rab25 KO mice. In line with this, depletion of Rab25 with single hairpin RNA increased the expression of differentiation markers in a human keratinocyte cell line, HaCaT. Transcriptomic analysis of the skin revealed increased expression of genes associated with skin development, epidermal development, and keratinocyte differentiation in Rab25 KO mice. Collectively, these results suggested that Rab25 is involved in the regulation of epidermal differentiation and proliferation.
Objectives : Hedgehog skin is one of the animal medicines in Traditional Korean Medicine for hematochezia and hemorrhoids. In this study, we examined cytotoxicity and anti-inflammatory effects. Methods : Cytotoxicity of hedgehog skin extracts was measured by MTT assay in vitro. We investigated the inhibition of lipopolysaccharide (LPS) stimulated nitric oxide (NO) production in RAW 264.7 cells. The phosphorylation of mitogen-activated protein kinases (MAPKs) was measured by western blot. And we observed the effect of hedgehog skin extracts on the expression of IL-6 genes using real time PCR. Results : As a result of MTT assay for cytotoxicity, there were no significant differences between non-treatment group and hedgehog skin extracts treatment groups. $500{\mu}g/m{\ell}$ of hedgehog skin extracts treatment significantly decreased nitric oxide production in comparison with non-treatment in LPS-induced RAW 264.7 cells. In measurement of the phosphorylation of MAPKs using western blot analysis, LPS stimulation increased the phosphorylation of MAPKs and $500{\mu}g/m{\ell}$ of hedgehog skin extracts treatment decreased the phosphorylation of ERK1, ERK2 and p38 significantly. But there were no significant differences the phosphorylation of JNK1 and JNK2. As a result of confirmation of the IL-6 mRNA gene expression using real time PCR, IL-6 mRNA gene expressions were significantly decreased in $50{\mu}g/m{\ell}$, $100{\mu}g/m{\ell}$ and $500{\mu}g/m{\ell}$ hedgehog skin extracts treated groups by comparison with non-treatment group. Conclusion : These results could provide a mechanistic explanation for the anti-inflammatory effects of the hedgehog skin.
