Identification of Ozone-induced Skin Damage and Screening of Antioxidant for Ozone (오존에 의한 피부손상 확인 및 이를 방어하는 피부 외용제 소재의 탐색)

  • Ozone(O$_3$), one of best-known toxic air pollutant, act as a strong oxidant. It is possible that skins exposed to the air can be easily damaged by such oxidative air pollutants. Therefore, in the present study, anti-oxidative effects of natural product. on $O_2$ㆍ and ㆍOH were investigated by EPR. Ozone caused protein damage and lipid oxidation, in HaCaT and B16F10 leading ultimately to programmed cell death. It also reduced the level of antioxidant molecules including ascorbic acid and tocopherol in stratum comeum. However, antioxidants originated from natural products could protect skin from these products could protect skin from these oxidative damages. We concluded that eight natural extracts including Rosa davurica, Ligularia sibrica, Green tea acted as strong antioxidants against ozone.

  • Polyester, polyester/cotton blend, nylon and cotton fabrics were ultraviolet cutting finished with padding method or exhaustion method using several UV absorbers. The transmittance of ultraviolet ray in the textiles can be greatly depressed by the processing and it is expected that human skin can be kept safe from sun-burn or damages by ultraviolet ray.

Protective effects of red orange (Citrus sinensis [L.] Osbeck [Rutaceae]) extract against UVA-B radiation-induced photoaging in Skh:HR-2 mice

  • BACKGROUND/OBJECTIVES: The skin is the outermost organ of the human body and plays a protective role against external environmental damages, such as sunlight and pollution, which affect anti-oxidant defenses and skin inflammation, resulting in erythema or skin reddening, immunosuppression, and epidermal DNA damage. MATERIALS/METHODS: The present study aimed to investigate the potential protective effects of red orange complex H extract (ROC) against ultraviolet (UV)-induced skin photoaging in Skh:HR-2 mice. ROC was orally administered at doses of 20, 40, and 80 mg/kg/day for 13 weeks, along with UV irradiation of the mice for 10 weeks. RESULTS: ROC improved UV-induced skin barrier parameters, including erythema, melanin production, transepidermal water loss, elasticity, and wrinkle formation. Notably, ROC inhibited the mRNA expression of pro-inflammatory cytokines (interleukin 6 and tumor necrosis factor α) and melanogenesis. In addition, ROC recovered the UV-induced decrease in the hyaluronic acid and collagen levels by enhancing genes expression. Furthermore, ROC significantly downregulated the protein and mRNA expression of matrix metalloproteinases responsible for collagen degradation. These protective effects of ROC against photoaging are associated with the suppression of UV-induced phosphorylation of c-Jun NH2-terminal kinase and activator protein 1 activation. CONCLUSIONS: Altogether, our findings suggest that the oral administration of ROC exerts potential protective activities against photoaging in UV-irradiated hairless mice.

Anti-aging Effect on Skin with the needles of red pine, Pinus densiflora (적송엽(赤松葉)이 피부(皮膚)의 노화(老化)에 미치는 영향(影響))

  • Objectives : We investigated the anti-aging effect on skin with the extract of the needles of red pine, Pinus densiflora. Methods : We measured various effects related to skin such as the anti-oxidant effect, the protection against ultraviolet (UV) irradiation, the inhibition of reactive oxygen species (ROS) generation, the induction of heat shock protein 70 (HSP70), the reduction of matrix metalloproteinase-2 (MMP-2) synthesis and senescent cell. Results : The results were as follows : The extract of the needles of red pine (RP) had the potent anti-oxidant effect and the ROS scavenging effect. Also RP preserved the systemic anti-oxidant enzyme system (superoxide dismutase and catalase) from UVB irradiation. RP protected the cell membrane from the damages induced by UVB irradiation. RP induced HSP70, a mediator of resistance to UVB irradiation. RP reduced the synthesis of MMP-2 induced by UVB irradiation. And RP inhibited the amount of senescent-associated (SA) ${\beta}-galactosidase$ staining, as a marker of replicative senescence. Conclusions : The results of our study indicate that the extract of the needles of red pine, Pinus densiflora, has anti-aging effects on skin.

A Case of Acute Multiple Nummular Eczema with Exudation (급성 다발성 삼출성 화폐상 습진 한방 치험 1례)

  • Objective : The purpose of this report is to know the effect of oriental medical treatments on acute multiple nummular eczema with exudation. Methods : We treated a 41-years-old woman patient with nummular eczema on right shin and left upper arm with Korean medicine. After Korean medical treatment for 31 days, we measured the decrease of exudation and recovery of skin damages. We recorded pictures of changes on symptoms. Results & Conclusion : The symptoms of nummular eczema were significantly improved except scars. Exudation was disappeared and skin damages were almost recovered. Pruritus was decreased by 70%. Thus Korean medical treatments are effective on acute multiple nummular eczema with exudation.

A Case of Acute Allergic Contact Dermatitis with Multiple Nummular Eczema (화폐상 습진에 병발한 급성 알레르기성 피부염 한방 치험 1례)

  • Objective : The purpose of this report is to know the effect of oriental medical treatments on acute allergic contact dermatitis with multiple nummular eczema. Methods : We treated a 34-years-old woman patient with acute allergic contact dermatitis on face and multiple nummular eczema on both arm, right ankle and left inguinal region by Korean medicine. After Korean medical treatment for 42 days in hospital, we measured the decrease of exudation and pruritis as well as recovery of skin damages. We recorded pictures of changes on symptoms. Results & Conclusion : The symptoms of ACD and nummular eczema were significantly improved except scars. Exudation and pruritis were disappeared and skin damages were almost recovered until discharge from hospital. After 20 days we could confirm the symptoms were clearly disappeared. Thus Korean medical treatments are effective on ACD and nummular eczema.

A Clinical Report of Exfloliative Dermatitis (박탈성 피부염의 한방 치험 1례)

  • Objective : The purpose of this report is to know the effect of Korean medical treatments on exfloliative dermatitis. Methods : We treated a 25-years-old man patient with exfloliative dermatitis on the limbs and back with Korean medicine. After Korean medical teatment for 26 days, we measured the decrease of dermatitis and recovery of skin exfloliate. We recorded pictures of changes on symptoms. Result & Conclusion : The symptoms of exfloliative dermatitis were significantly improved. The dry skin and itching were disappeared and skin damages were almost recovered. Thus Korean medical treatments are effective on exfloliative dermatitis.

Protective Effect of Paeoniae Radix Alba against UVB Photodamage ( UVB 광손상에 대한 백작약의 보호 효과)

  • Objective : UVB damages skin health by causing skin redness and intense inflammation, sunburn, and skin cancer. Paeoniae Radix Alba has been used to relieve gynecological symptoms, muscle spasms, and skin ailments. This study was conducted to confirm whether it has a protective effect against UVB photodamage. Methods : Ethanol extract of Paeoniae Radix Alba (PRA) was prepared by extracting 100 g Paeoniae Radix Alba in 1 L of ethanol for 48 h. Apoptosis was monitored by the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay and expression levels of apoptosis indicator proteins, and tyrosinase activity was measured with a colorimetric commercial kit. Results : In human keratinocyte HaCaT cells, PRA reduced UVB-induced cell death through apoptosis by inhibiting PARP cleavage and caspase-3 and -9. UVB-induced increase in cellular reactive oxygen species (ROS) was suppressed by PRA pretreatment. PRA also showed dose-dependent ABTS and DPPH radical scavenging activities. Furthermore, the inhibitory effect of tyrosinase activity by PRA was confirmed. Conclusion : These results demonstrated the protective role of PRA in UVB photodamage of human keratinocytes, mainly due to its antioxidant and antiapoptotic properties. We also suggest that PRA can be considered as an effective natural agent to prevent skin photodamage.

The Effects of Baekho-tang Extracts on Regulating Th2 Differentiation through Improving Skin Fat Barrier Damage (백호탕 추출물의 지방장벽 손상 개선을 통한 상피 내 Th2 분화 조절 효과)

  • Objectives The purpose of this study is to confirm the regulate effect of T helper (Th) 2 differentiation that Baekho-tang extract may produce to improves skin lipid barrier damages. Methods Four-weeks-old NC/Nga mice were divided into four groups: control group (Ctrl, n=10), lipid barrier eliminated group (LBE, n=10), Dexamethasone treatment after lipid barrier elimination group (DxT, n=10), and Baekho-tang extract treatment group after lipid barrier elimination group (BHTT, n=10). Baeko-tang extract was administered for 3 days after removal of the skin fat barrier in BHTT group. Then, we identified changes in external symptoms of the skin, factors affecting skin barrier such as potential of hydrogen (pH), filaggrin (FLG), transepidermal water loss (TEWL) and Th2 differentiation factors like Interleukin (IL)-4, Kallikrein Related Peptidase 7 (KLK7) and protease activated receptor 2 (PAR-2) through our immunohistochemistry. Results After lipid barrier elimination, the reduction of morphological skin inflammations was less in BHTT group than in LBE group and DxT group. Also, pH and TEWL were significantly decreased with BHTT group. However, FLG was significantly increased in BHTT group compared to LBE, DxT, and Ctrl group. All kinds of Th2 differentiation factors (IL-4, KLK7 and PAR-2) were also decreased in BHTT compared to the LBE and DxT. Conclusions As a result of this study, BHT administration decreased pH, TEWL, and increased FLG, thus participating in recovering damaged skin barrier. Since Th2 differentiation factors were decreased as well, BHT's regulatory effect in sequential immune reactions may be a possible explanation of how it enhances recovery of the damaged lipid barrier.

Anti-aging Effects of Marine Natural Extracts against UVB-induced Damages in Human Skin Cells (UVB로 손상이 유도된 피부세포에 해양소재 추출물의 항노화 효능)

  • The skin is continuously exposed to environmental stresses. One of the most important stress factor is UV radiation. UV radiation causes a variety of biological effects on the skin, including inflammation, pigmentation, photoaging and cancer. Therefore in this study, we tried to search for skin-protective antioxidant materials from marine natural products (Porphyra Thalli, Laminariae japonicae thallus, Ostreae Concha, Sargassum Thallus, Undaria thallus, Haliotidis Concha, Agar, Codium thalli, Hizikia fusiforme thalli; HFE, Thalli) which exhibit protective activities against UVB-induced cytotoxicity and oxidative cell death and antiaging effects. As a results, UVB-induced cytotoxicity and cell death were effectively suppressed by treatment of Sargassum Thallus, Agar, Haliotidis Concha, Codium thalli, Thalli ethanol extracts. UVB-induced cell death was mediated by intracellular accumulation or ROS, which was significantly inhibited by treatment with marine natural products extracts. Also, The protective effect of these marine natural products seemed to be mediated by increased expression of type I collagen and Type I procollagen. These results suggest that marine natural products may have anti-aging effects new functional materials against oxidative stress-mediated skin damages.
