Bio-Conjugated Polycaprolactone Membranes: A Novel Wound Dressing

  • Background The combination of polycaprolactone and hyaluronic acid creates an ideal environment for wound healing. Hyaluronic acid maintains a moist wound environment and accelerates the in-growth of granulation tissue. Polycaprolactone has excellent mechanical strength, limits inflammation and is biocompatible. This study evaluates the safety and efficacy of bio-conjugated polycaprolactone membranes (BPM) as a wound dressing. Methods 16 New Zealand white rabbits were sedated and local anaesthesia was administered. Two $3.0{\times}3.0cm$ full-thickness wounds were created on the dorsum of each rabbit, between the lowest rib and the pelvic bone. The wounds were dressed with either BPM (n=12) or Mepitel (n=12) (control), a polyamide-silicon wound dressing. These were evaluated macroscopically on the 7th, 14th, 21st, and 28th postoperative days for granulation, re-epithelialization, infection, and wound size, and histologically for epidermal and dermal regeneration. Results Both groups showed a comparable extent of granulation and re-epithelialization. No signs of infection were observed. There was no significant difference (P>0.05) in wound size between the two groups. BPM (n=6): $8.33cm^2$, $4.90cm^2$, $3.12cm^2$, $1.84cm^2$; Mepitel (n=6): $10.29cm^2$, $5.53cm^2$, $3.63cm^2$, $2.02cm^2$; at the 7th, 14th, 21st, and 28th postoperative days. The extents of epidermal and dermal regeneration were comparable between the two groups. Conclusions BPM is comparable to Mepitel as a safe and efficacious wound dressing.

Anti-Inflammatory Effect of Ligularia fischeri, Solidago virga-aurea and Aruncus dioicus Complex Extracts in Raw 264.7 Cells (곰취(Ligularia fischeri), 미역취(Solidago virga-aurea), 삼나물(Aruncus dioicus) 복합 추출물의 항염증 효과)

  • The objective of this study was to evaluate the skin inflammation effects of three herb mixture extracts, Ligularia fischeri, Solidago virga-aurea and Aruncus dioicus, which are from Ullung island in Korea. Regulatory mechanisms of cytokines and nitric oxide (NO) are involved in the immunological activity of Raw 264.7 cells. Tested cells were pretreated with 70% acetone extracts of Ligularia fischeri, Solidago virga-aurea and Aruncus dioicus (LSA-A) and further cultured for an appropriated time after lipopolyssacharide (LPS) addition. During the entire experimental period, 1, 10, and 100 ${\mu}g/ml$ of LSA-A had no cytotoxicity. In these concentrations, LSA-A inhibited the production of NO and prostaglandin $E_2$ ($PGE_2$), tumor necorsis factor-a (TNF-a), interleukin-1${\beta}$ (IL-1${\beta}$), interleukin-6 (IL-6) expression of inducible NO synthase (iNOS), and cyclooxygenase-2 (COX-2). LSA-A showed a 60% $PGE_2$ inhibition rate at 100 ${\mu}g/ml$. iNOS and COX-2 inhibition activities were 54%, and 65% at 100 ${\mu}g/ml$, respectively. In addition, LSA-A extract reduced the release of inflammatory cytokines including TNF-a, IL-1${\beta}$ and IL-6. These results suggest that LSA-A may have significant effects on inflammatory factors, and may be a potential anti-inflammatory therapeutic agent.

Association Analysis of Monocyte Chemotactic Protein-3 (MCP3) Polymorphisms with Asthmatic Phenotypes

  • The monocyte chemotactic protein-3 (MCP3), on chromosome 17q11.2-q12, is a secreted chemokine, which attracts macrophages during inflammation and metastasis. In an effort to discover additional polymorphism(s) in genes whose variant(s) have been implicated in asthma, we scrutinized the genetic polymorphisms in MCP3 to evaluate it as a potential candidate gene for asthma host genetic study. By direct DNA sequencing in twenty-four individuals, we identified four sequence variants within the 3 kb full genome including 1,000bp promoter region of MCP3; one in promoter region (-420T>C), three in intron (+136C>G, +563C>T, +984G>A) respectively. The frequencies of those four SNPs were 0.020 (-420T>C), 0.038 (+136C>G), 0.080 (+563C>T), 0.035 (+984G>A), respectively, in Korean population (n = 598). Haplotypes, their frequencies and linkage disequilibrium coefficients (|D'|) between SNP pairs were estimated. The associations with the risk of asthma, skin-test reactivity and total serum IgE levels were analyzed. Using statistical analyses for association of MCP3 polymorphisms with asthma development and asthma-related phenotypes, no significant signals were detected. In conclusion, we identified four genetic polymorphisms in the important MCP3 gene, but no significant associations of MCP3 variants with asthma phenotypes were detected. MCP3 variation/haplotype information identified in this study will provide valuable information for future association studies of other allergic diseases.

Tuberculous Empyema Necessitatis with Osteomyelitis, a Rare Case in the 21st Century (늑골 골수염에 동반된 흉벽 천공성 농흉 1례)

  • Empyema necessitatis refers to empyema that extends into the extrapleural space through a defect in the pleural surface. Tuberculous empyema necessitatis is a rare complication of tuberculosis. We experienced a 21-month-old boy with tuberculous empyema necessitatis with osteomyelitis in the right $7^{th}$ rib. He presented with a mass on the right lateral chest wall, which was soft and nontender, enlarging for one month. He also had mild fever. The plain radiograph of his chest revealed soft tissue swelling and calcified lymph node on the left axilla, and his PPD skin test was positive. CT scan of the chest showed empyema necessitatis at the right lower chest and upper abdominal walls with osteomyelitis of the right $7^{th}$ rib. He did not have concurrent pulmonary tuberculosis. Surgery was performed for diagnosis and treatment. In histopathologic findings, chronic granulomatous inflammation with caseation necrosis was shown and was positive for acid fast bacilli stain. In addition, M. tuberculosis complex was found as etiology by polymerase chain reaction. The patient has been treated with anti-tuberculous medication without any specific complication.

Beauty food activities of extracts from Pinus densiflora root (동송근(Pinus densiflora root) 추출물의 미용식품활성)

  • The extracted phenolic compounds from Pinus densiflora root were examined biological activities for beauty food. The tyrosinase inhibitory activity which was related to skin-whitening was observed. The tyrosinase inhibitory activity was confirmed to be 92% in ethanol extract at $50{\mu}g/mL$ phenolic. The elastase and collagenase inhibitory activity as anti-wrinkle effect were showed 61 and 78% in ethanol extract at $200{\mu}g/mL$ phenolic, respectively. Astringent effect of ethanol extract was showed to be 82% at $50{\mu}g/mL$ phenolic. Hyaluronidase inhibitory activity of ethanol extract as anti-inflammation effect was confirmed to be 94% of inhibition at $200{\mu}g/mL$ phenolic. These results demonstrated that isolated phenolic compounds from P. densiflora root could be expected to use as a functional cosmetic materials.

Effects of Oral Intake of Gromwell Water Fraction on Ceramides Content and the Development of Atopic Dermatitis in NC/Nga Mice (자초 추출물 극성 성분의 피부 보습 증진 및 아토피 피부염 호전 효과)

  • Gromwell (LE, Lithospermum erythrorhizon), a perennial herbal plant, has been used for the treatment of various problems associated with atopic dermatitis of the skin, such as water loss, epidermal hyperproliferation, and severe inflammation. Previously, it was shown that oral supplementation with a 70% ethanol extract of gromwell prevented the development of atopic dermatitis in NC/Nga mice. In this study, in order to identify the fraction that mediates gromwell's efficacy, the dietary effects of water and ethyl acetate fractions from the ethanol extract of gromwell were assessed in the development of dermatitis using NC/Nga mice. Dietary supplementation of the hot water fraction significantly reduced scores for epidermal hyperproliferation in parallel with a marked increase of ceramides. Supplementation of the gromwell hot water fraction also decreased scratching behavior, which was accompanied by a decrease in plasma levels of IgE. These results showed that the hot water fraction of the gromwell ethanolic extract prevented the development of atopic dermatitis by increasing ceramides in NC/Nga mice.

Improving Effect for Acne with SeleMix AN Composed of Germinating Soy Bean and Magnolia Bark Extract (발아콩 및 목련박피 혼합추출물(SeleMix AN)에 의한 여드름 개선 효과)

  • We investigated new ingredients with real efficacy in both in vitro and in vivo all together. Especially we focused on the real improving effort in the clinical experiments. Because most products containing effective materials evaluated in vitro failed to show a real improving effect in the human with acne. We evaluated the well-known ingredients in a small scale clinical experiment with half-finished goods containing each ingredient. Among these products, product formulating SeleMix AN composed with germinating soy bean and magnolias bark extract remarkably improved acne and acne scar. Moreover skin redness caused by severe acne was improved. There was statistical significance between placebo and sample. Two hundred volunteers participated in our pilot study with written informed consent. After then we performed in vitro efficacy test this ingredient. SeleMix AN inhibited the growth of propionibacterium acnes at a concentration of 0.0125% and suppressed histamine release by 16.9%. Moreover human fibroblast cell activity was increased by 57% compared to control. Lastly, we performed a clinical study. Consisting of groups of 23 volunteers. Although the period of the test was in summer accelerating sebum secretion and recurring a high tate of acne, inflammation lesions were especially improved after applying product containing SeleMix AN for 4 weeks.

Study on Anti-oxidant and Anti-inflammatory Activity of the Tissue-cultured Shoot Clumps from Raoulia australis (라울리아 신초 추출물의 항산화 및 항염 효능에 관한 연구)

  • In this study, the shoot clumps extract of tissue-cultured Raoulia australis using the bioreactor culture system was tested for use a natural cosmetic ingredient. Tissue-cultured R. australis shoot clumps extract was tested anti-oxidant and anti-inflammatory activity for a cosmetic application. R. australis is a wild herbaceous plant of the asteraceae growing in New Zealand and Australia. Previous studies have reported anti-viral activity of the inhibitory effects for the growth of viruses induced meningitis, bronchitis and respiratory diseases but other biological effects are unknown. The shoot clumps extract of tissue-cultured R. australis showed higher anti-oxidant effect and anti-inflammatory effect than the natural R. australis extract. In DPPH, NBT and ABTS assay, the shoot clumps extract of tissue-cultured R. australis enhanced radical scavenging activity (up to 10~25% at $50{\mu}L/mL$) more than the natural R. australis extract. Also, the shoot clumps extract of tissue-cultured R. australis inhibited expression of iNOS and COX-2 protein in LPS-stimulated Raw 264.7 macrophages more than the natural R. australis extract. From this study, the shoot clumps extract of tissue-cultured R. australis displayed strong possibility as a new natural cosmetic ingredient for skin-care products.

Anti-aging and Anti-melanogenesis Effect of Cimicifuge dahurica, Coptis chinensis, Phellodendri amurense and Magnol obovata Extracts Mixture (승마, 황련, 황백, 후박 추출 혼합물의 항노화 및 멜라닌 생성 저해효과)

  • Cimicifuge dahurica (C. dahurica), Coptis chinensis (C. chinensis) and Phellodendri amurense (P. amurense) extracts has an detoxification effect and Magnol obovata bark (M. obovata) extracts has an antibacterial effect in oriental medicine. This study investigated the possibility of the extract mixture as a functional cosmetic ingredient by mixing C. dahurica, C. chinensis, P. amurense and M. obovata extracts. MTT assay was carried out for toxicity test and DPPH/ABTS experiments for antioxidant effects of the extract mixture. Results show that the extract mixture was safer and antioxidant effects in a dose-dependent manner than single extract of the mixture. The mixture effectively inhibited NO (nitric oxide) production, which indicate good efficacy for anti-inflammation. The mixture also protected UVB-induced cell damage and increased type 1 pro-collagen synthesis in fibroblast. In addition, it's treatment inhibited the melanin synthesis and tyrosinase activity by lowering expression of MITF, tyrosinase and TRPs in B16F10 melanoma cell. These results suggest that medicinal herbal extract mixture may be useful as a functional ingredient for anti-aging and skin whitening cosmetic formula.

Anti-inflammaory effects of the Gamroeum in vivo and in vitro (감로음(甘露飮)의 항염증 효과에 대한 실험적 연구)

  • Objectives : The present study was conducted to evaluate the anti-inflammatory effects of the Gamroeum water extracts (GRE) in vivo and in vitro. Methods : The effects of GRE on anti-inflammation were measured by production of NO, $PGE_2$ (Prostaglandin $E_2$), iNOS (inducible Nitric Oxide Synthase), COX-2, $NF{\kappa}B$ (Nuclear Factor kappa B), TNF-$\alpha$ (Tumor Necrosis Factor-alpha) and IL-$1{\beta}$ (Interleukin-$1{\beta}$), IL-6 in Raw 264.7 macrophage cells stimulated with LPS. Results : 1. In machrophage cells, LPS displayed significant stimulatory effects on the production of NO and $PGE_2$. However, GRE showed significant inhibitory effects on NO and $PGE_2$ release. The level of NO and $PGE_2$ was decreased by GRE in a concentration dependent manner as compared with LPS only group. 2. Immunoblot analysis verified that LPS stimulation significantly increased the iNOS and COX-2 protein level, but GRE suppressed the induction of iNOS and COX-2 protein at a concentration dependent manner. 3. GRE reduced the elevated production of TNF-$\alpha$, IL-$1{\beta}$ and IL-6 by LPS. Moreover, the inhibitory effects of GRE was occurred in a dose-dependent manner. 4. GRE significantly reduced the expression of NF-${\kappa}B$ protein in nuclear fraction. 5. GRE effectively inhibited the increases of hind paw skin thicknesses and inflammatory cell infiltrations induced by carrageenan treatment. It, therefore, considered that GRE will be favorably inhibited the acute edematous inflammations. Conclusions : These results indicated that GRE could have anti-inflammatory capacity by inhibiting the production of NO, $PGE_2$ and cytokines in vitro and by reducing the formation of carrageenan-induced paw edema in vivo. Moreover, inhibitory effects of GRE on the macrophage activation were attributable to the reduction of some of inflammatory factors by inhibiting iNOS and COX-2 through the suppression of NF-${\kappa}B$.
