전통 생약재에서의 항노화와 주름 개선 활성 신소재 탐색 (Screening of Traditional Herbal Medicines to Develop New Materials for Anti-aging and Anti-wrinkle in the Skin)

  • 본 연구는 화장품 소재개발에 있어 보다 안전하고 우수한 활성을 나타내는 천연 소재를 개발하고자 하였다 본초학과 식품공전에 수재되어 있는 전통생약재에 대한 연구는 국내 화장품 원료개발의 중요한 부분으로써 우리나라가 경쟁력을 가질 수 있는 부분으로 사료되며, 본 연구에서는 폐와 피부 질환에 사용되는 전통생약재 121종을 선별하였다. 이들 전통생약재 추출물들을 대상으로 DPPH free radical 소거법과 SOD 활성 측정법에 의한 항산화 활성과 주름 생성의 주 기전에 관련된 elastase 효소 억제 활성을 관찰하였다. 항산화 활성을 보이면서도 주름 형성관련 효소인 엘라스타제에 대한 억제 활성을 지니는 전통생약재 9종을 선별하였다. 항주름/피부탄력 활성을 나타내리라 사료되는 9종의 생약재에 대한 각각의 정상 세포에 대한 세포독성을 측정한 결과, 효과를 나타내리라 여겨지는 0.1 g/mL 이하의 농도에서 세포 독성이 없는 것으로 확인되었다. 또한 인체피부 일차 자극 실험을 통해 피부자극 유무를 측정한 결과 피부자극이 없음을 확인하였다 본 연구를 통해 식품공전과 동의보감 자료에 의거, 폐와 피부질환에 관련된 전통생약재 121종 중 9종에 대해 항주름과 피부탄력에 관련한 향장 소재로써의 개발 가능성을 확인하였다.

화피 추출물이 염증 및 알레르기 반응에 미치는 영향 (Effect of Phellinus igniarius Quel Extract on the Anti-inflammatory, Anti allergy, Anti-oxidant, Anti-wrinkle reaction)

  • Objectives : Betula platyphylla var. japonica extract (BPE) was used to determine the modulation of cytokine secretion, the activation of inflammatory and allergic factor and the inhibition of gene expression. Inflammatory and allergic cytokines as IL-$1{\beta}$, IL-2, IL-4, IL-5, IL-6, IL-8, TNF-${\alpha}$, NO and COX-2 were measured to use effectively on improvement or treatment of atopic dermatitis. Methods : We used NC/Nga mouse induced by atopic dermatitis to observe the effects of BPE on the weight, water and feed, blood test, weight of organs, histological change, total IgE and histological change of main organs. Results : BPE is effective on anti-inflammatory and allergic reaction. However, further study is needed to prove which component of BPE indicates effective pharmacological action. Conclusions : The above results suggest that Phellinus igniarius Quel extract could be applicable for improvement of several skin functions.

Oral Administration of KTNG0345 Prepared from Red Ginseng Extracts Reduces UVB-induced Skin Wrinkle Formation in Hairless Mice

  • Chronic ultraviolet (UV) exposure is the major cause of photoaging that causes skin wrinkling, roughness, dryness, laxity, and pigmentation. Recently, increasing efforts are being made to understand the relationship between foods and skin health. Ginsenosides are present in ginseng (Ginseng Radix Rubra) extract, and are known to have biomedical properties, such as, anti-oxidant and anti-inflammatory effects. In this study, we investigated whether KTNG0345 prepared from red ginseng extracts delivered orally reduces skin wrinkling and ultraviolet B (UVB)-induced wrinkle formation in hairless mouse skin. KTNG0345 was administrated orally to the mice (5 times a week) during the period of UVB-irradiation (3 times a week) for 8 weeks at three different doses of 300 mg/kg, 500 mg/kg and 1000 mg/kg (w/v). UV doses were increased weekly by 1 MED (1MED = 75 $mJ/cm^2)$ up to 4 MED and then maintained at this level. After the 8-week administration period, it was found that orally administered KTNG0345 significantly inhibited UVB-induced wrinkle formation in a dose-dependent manner. Increases in skin thickness caused by UVB were prevented by KTNG0345. Moreover, it also significantly inhibited matrix metalloproteinases (MMP) -13 and MMP-9 expressional inductions by UVB. In addition, KTNG0345 was observed to prevent UVB-induced water loss of epidermis in hairless mouse skin. Our results demonstrate that orally administered KTNG0345 has anti-wrinkling effects in hairless mouse skin, and suggest that dietary red ginseng and herbal mixture may be considered a functional beauty food for preventing UVB-induced skin wrinkles.

대황(Rheum undulatum L.)으로부터 항주름성 Elastase 저해제의 추출 및 특성 (Extraction and Characterization of an Anti-wrinkle Elastase Inhibitor from Rheum undulatum L.)

  • To develop a new anti-wrinkle agent from medicinal plants, this study investigated the optimal conditions for extracting elastase inhibitor from Rheum undulatum L. Maximal extraction occurred by using 70% methanol at $50^{\circ}C$ for 24 hr; the elastase inhibitory activity was 60.4%($IC_{50}:6.7{\times}10^3{\mu}g/m{\ell}$). Systematic solvent extraction, thin layer chromatography, silica gel column chromatography, Sephadex LH-20 column chromatography, and reverse-phase high performance liquid chromatography were used in the partial purification of the elastase inhibitor. The compound was soluble in dimethylsulfoxide, methanol, and ethanol, and had maximum absorption spectra at 231.5 nm and 275.5 nm.

  • Indole-3-acetic acid (IAA) molecule has been successfully encapsulated in biocompatible inorganic matrix in order for the cosmetic application such as superfacial fine line reduction. The encapsulation was realized through chemical reaction involving simultaneous formation of inorganic lattice and 1M giving rise to an 1M-inorganic nanohybrid (IAA-brid) which shows excellent storage stability and sustained releasing property of indole-3-acetic acid. The clinical efficacy of essence cream containing IAA-brid as anti-wrinkle formulation was also carefully evaluated by measuring the roughness of the skin replica before and after treatment. Upon administration of the cream on the eye-area, the fine-line is drastically reduced.

Agastache rugosa Kuntze Attenuates UVB-Induced Photoaging in Hairless Mice through the Regulation of MAPK/AP-1 and TGF-β/Smad Pathways

  • Chronic exposure to ultraviolet (UV) radiation, regarded as a major cause of extrinsic aging or photoaging characterized by wrinkle formation and skin dehydration, exerts adverse effects on skin by causing the overproduction of reactive oxygen species. Agastache rugosa Kuntze, known as Korean mint, possesses a wide spectrum of biological properties including anti-oxidation, anti-inflammation, and anti-atherosclerosis. Previous studies have reported that A. rugosa protected human keratinocytes against UVB irradiation by restoring the anti-oxidant defense system. However, the anti-photoaging effect of A. rugosa extract (ARE) in animal models has not yet been evaluated. ARE was orally administered to hairless mice at doses of 100 or 250 mg/kg/day along with UVB exposure for 12 weeks. ARE histologically improved UVB-induced wrinkle formation, epidermal thickening, erythema, and hyperpigmentation. In addition, ARE recovered skin moisture by improving skin hydration and transepidermal water loss (TEWL). Along with this, ARE increased hyaluronic acid levels by upregulating HA synthase genes. ARE markedly increased the density of collagen and the amounts of hydroxypoline via two pathways. First, ARE significantly downregulated the mRNA expression of matrix metalloproteinases responsible for collagen degradation by inactivating the mitogen-activated protein kinase/activator protein 1 pathway. Second, ARE stimulated the transforming growth factor beta/Smad signaling, consequently raising the mRNA levels of collagen-related genes. In addition, ARE not only increased the mRNA expression of anti-oxidant enzymes but also decreased inflammatory cytokines by blocking the protein expression of nuclear factor kappa B. Collectively, our findings suggest that A. rugosa may be a potential preventive and therapeutic agent for photoaging.

Development of Vaccinium uliginosum L. extracts for whitening & anti-wrinkle functional food

  • This study is peformed to investigate the effect of water extract from Vaccinium uliginosum L., on melanin production in B 16 melanoma cells, procollagen production and matrix metalloproteinase-1(MMP-1) inhibition in human fibroblast cells. One hundred grams of the Vaccinium uliginosum L. was extracted with 2000 mL of water($90^{\circ}C$, 16h, 2times). The water extracts were lyophilized and stored at $4^{\circ}C$ until used. Dry weight yields of extracts of Vaccinium uliginosum L. were $3\%$(w/w). Extracts from Vaccinium uliginosum L. showed scavenger activities on DPPH radical, superoxide anion radical, hydroxyl radical, hydrogen peroxide and singlet oxygen radical. And these substances inhibited release of cyiokines from human keratinocyte after UV B exposure. Therefore we confirmed that extracts from Vaccinium uliginosum L. had antioxidative effect. These substances inhibited purified tyrosinase activity and melanogenesis in B 16 melanoma cells treated/untreated IL-$1{\alpha}$. Moreover this extract stimulated procollagen production and inhibited MMP-1 production in human fibroblast cells treated/untreated IL-$1{\beta}$. Therefore we confirmed that extracts from Vaccinium uliginosum L. had whitening effect. And these substances decreased degree of wrinkle in hairless mouse skin that induced by UV B irradiation. Therefore we confirmed that extracts from Vaccinium uliginosum L. had anti-wrinkle effect. From the above results, it is possible that Vaccinium uliginosum L. may be developed to be an anti-melanogenesis agent and anti-wrinkle agent.

사인추출물의 인슐린 유사 성장인자-3의 합성과 피부 노화 개선에 대한 효과 (Effects of Amomi, Semen Extract on Synthesis of Insulin-like Growth Factor-1 and Anti-wrinkle in Skin)

  • 본 연구에서 프로모터-리포터 분석방법을 통해 인슐린 유사 성장인자-1의 프로모터를 자극하는 천연물을 선별한 결과 사인 추출물이 가장 좋은 프로모터 자극효과를 나타냈으며, 무모생쥐에서 패쇄첩포 후 RT-PCR로 실험한 결과 IGF-1 mRNA를 $35\%$ 증가시키는 것으로 나타났다. 사인 추출물의 피부 주름개선 효과를 알아보기 위하여 인체 섬유아세포에서는 type-I collagen과 MMP-1 합성 변화를 관찰하였으며, 무모생쥐에서는 콜라젠의 증가와 진피 두께를 관찰하였다. 그 결과, 동위원소를 이용한 콜라젠 증가실험에서 type-I collagen은 $38\%$ 증가하였으며 무모생쥐에서 실시한 RT-PCR 결과에서는 mRNA가 $21\%$ 증가하는 것으로 관찰되었다. MMP-1 효소발현의 경우 ELISA 분석을 통해서 $63\%$의 높은 발현저해능을 확인하였고 Western blot에서도 발현이 저해되는 것을 확인하였다 추출물을 무모생쥐에 패쇄첩포 하였을 경우 대조군에 비해 진피 두께가 두꺼워지고 콜라젠 양도 증가되는 것으로 조직염색 관찰을 통해 확인하였다. 이상의 결과를 통해 사인추출물은 피부 주름개선에 좋은 효과를 나타내는 것으로 보이며, 여기에는 인슐린 유사성장 인자-I의 발현증가와 관련된 기전이 관여하는 것으로 판단된다.

신규한 Palmitoyl Tripeptide의 피부 주름개선 효과에 관한 연구 (A Study on the Skin Anti-wrinkle Effect of Novel Palmitoyl Tripeptide)

  • 콜라겐은 포유동물의 가장 풍부한 동물성 단백짙로, 전체 단백질의 약 30 %를 차지하며, 결합 조직에 존재하며 대부분의 장기의 구조적 지지에 기여한다. Tripeptide (glycine-proline-hydroxyproline: INCI name Tripeptide-29) 는 collagen type 1의 주성분이며, palmitoyl tripeptide (palmitoyl-glycine-proline-hydroxyproline: INCI name Palmitoyl Tripeptide-29) 는 콜라겐의 합성을 촉진하는 항노화 불질로서 디자인된 합성소재이다 합성된 웹타이드 유도체는 HPLC를 이용하여 분식하였다. in vitro test를 통하여 콜라겐합성과 섬유아세포 증식 효능을 확인하였고, 비침습적 기기를 사용하여 피검자에 대한 8주간의 적용결과 피부주름과 탄력의 상당한 개선을 확인하였다. Palmitoyl tripeptide는 우수한 항노화 효능을 갖는 화장품 소재로 사료된다.

Effects of the Chestnut Inner Shell Extract on the Expression of Adhesion Molecules, Fibronectin and Vitronectin, of Skin Fibroblasts in Culture

  • The inner shell of the chestnut (Castanea crenata S. et Z., Fagaceae) has been used as an anti-wrinkle/skin firming agent in East Asia, and preliminary experiments have found that a 70% ethanol extract from this plant material can prevent cell detachment of skin fibroblasts from culture plates. In order to examine the molecular mechanisms underlying this phenomenon, its effects on the expression of adhesion molecules, such as fibronectin and vitronectin, were investigated using the mouse skin fibroblast cell line, NIH/3T3. Using fixed-cell ELISA, Western blotting and immunofluorescence cell staining, it was clearly demonstrated that the chestnut inner shell extract enhanced the expression of the cell-associated fibronectin and vitronectin. Scoparone (6,7-dimethoxycoumarin), isolated from the extract, also possessed similar properties. These findings suggest that the enhanced expression of the adhesion molecules may be one of the molecular mechanisms for how the chestnut inner shell extract preventing cell detachment and may be also responsible for its anti-wrinkle/skin firming effect.
