  • A recent development in cosmetics has been the pursuit of wrinkling in the skin. The cosmetics composed of anti-wrinkle agent stand out from the point of view of environmental contamination and pollution. Among them, Indole-3-acetic acid (IAA), studied with wrinkling pigmentation and swelling conditions in the area of the eye, showed clinically significant reduction in depth of lines during one month trial using skin treatment. But, IAA has shown some problems when used in cosmetic formulations, such as stability, permeability and toxicity. The results of the clinical examination were shown that its permeability and toxicity didn't matter. To increase the stability of IAA, antioxidants such as Licorice, ubiquinone, tocopherol, Baicalin, ferulic acid, BHT, ascorbic acid, sodium metabisulfite, and so on were employed in cosmetic formulations. Our main purpose is the study for the stability efficiency and effect of each other of cream formulations containing optimal dosage antioxidants (o/w type emulsion), This study evaluated wrinkle reduction effect of IAA, which is used in cosmetics.

Effect of Phellinus igniarius Quel Extract on the Anti-inflammatory, Anti allergy, Anti-oxidant, Anti-wrinkle (상황버섯 추출물이 항염증 및 항알레르기, 항산화, 주름제거에 미치는 실험적 연구)

  • Objectives : This study was carried out to investigate the effects of Phellinus igniarius Quel extract on the Anti-inflammatory, Anti allergy, Anti-oxidant and Anti-wrinkle. Methods : To investigate in vitro anti-oxidant activity assay, ethanol extracts of medicinal plants tested by DPPH method. In the next experiment, to investigate anti-inflammatory test, the RAW 264.7 macrophage cells was cultured using DMEM including the 10% FBS. To study anti-allergic effect, we blended cultured Human Mast Cells(HMC-1), and then observe TNF-$\alpha$, IL-8 by ELISA Results : Phellinus igniarius Quel extract has the effects of anti-inflammation and anti-allergy, which may be due to its inhibitory potential on the macrophage activation. Furthermore, Phellinus igniarius Quel extract has the anti-wrinkle effects through the inhibitory potential on the collagnease, elastase and gelatinase activities. Conclusions : The above results suggest that Phellinus igniarius Quel extract could be applicable for improvement of several skin functions.

Skin Anti-aging and Anti-wrinkle Effects of Pinus koreaiensis Seed Oil (해송자 오일의 피부 항노화 및 주름 개선 효과)

  • Objectives : The aim of this study was to investigated the anti-aging and anti-wrinkles effects of the pinus koreaiensis seed(PKS) oil. Methods : The anti-oxidant effect was performed by beta-carotene bleaching assay and the intracellular proteome was analyzed expression of each 15 proteins by 2-D electrophoresis. And fatty acid was analysed by gas chromatography. Anti-wrinkle effect was analyzing human skin by the PRIMOS system. Results : Fatty acid analysis of PKS oil has shown oleic acid was 49.7% and linoleic acid was 34.1%. And the antioxidant effect was about 125% compared with alpha-tocoperol(0.1%) by beta carotene bleaching assay. In 2D PAGE analysis, fifteen protein changes in five mechanisms which was collagen synthesis pathway, MMPs, ECM-cell interaction, cytokine, antioxidant enzymes were analyzed. In case of anti-wrinkle effect was proved in vivo by analyzing human skin by the PRIMOS system. The analysis results of eye wrinkles for 4 weeks showed an improvement effect of over 6%. Conclusions : In this study, the amount of protein change in the five mechanism through the cell experiment and the skin anti wrinkle efficacy by the human in vivo test were investigated. As a result pinus koreaiensis seed oil by supercritical extraction could be used as a anti-aging and anti-wrinkle substance for the skin.

Anti-skin-aging Effect of Mori Folium through decreased Advanced glycation end product (AGEs) (최종당화산물 억제를 통한 상엽(桑葉)의 항피부노화 효과)

  • Objectives : Recently, numerous studies reported that excessive generation of advanced glycation end products (AGEs) stimulated expression of skin wrinkle related proteins. This study aimed to evaluate inhibits skin wrinkle formation effect of Mori Folium (MF) through decreased AGEs. Methods : To evaluate the skin wrinkle inhibition effect of MF, SD-rats were divided into three groups; normal rats (Nor), AGEs-induced rats (Con), AGEs-induce rats treated with MF at dose of 100mg/kg body weight (MF). To induced AGEs, streptozotocin (50mg/kg) was injected intraperitoneally, and after 3 days, 100mM methyl glyoxal was administered orally for 3 weeks. After the experiment, the animal's dorsal skin tissues and serum were separated and tested. Results : The oral administration of MF suppressed the AGEs level in serum. Also, the AGEs in skin tissues was significantly reduced through treatment of MF compared with control group. Moreover, the expressions of AGEs related proteins such as polyclonal anti-$N^e$-(carboxymethyl) lysine (CML), anti-$N^e$-(carboxyethyl)lysine (CEL), AGE receptors (RAGE) were reduced in MF group compared with the control group in kidney and skin tissues. The matrix metallo proteinase-1 (MMP-1) reduced by MF treatment with the result that collagen type 1 alpha 2 (COL1A2) was improved that reduced by accumulation of AGEs. Conclusion : The evidence of this study indicate that oral administration of MF reduces the levels of AGEs in serum, skin, and kidney tissues. In conclusion, MF inhibit skin wrinkle formation, suggesting the potential of anti-wrinkle material.

The Effect of Cosmetic on Anti-Wrinkle of Acer mono Sap (우산고로쇠의 향장효과)

  • The purpose of this study was to research for anti-oxidation and anti-wrinkle effects of Acar mono Sap (AM). To cosmetic effect of AM, safety effect (MTT assay), anti-wrinkle effect (elastase, MMP-1 inhibition assay) and anti-oxidant effect (DPPH assay) were measured. When water extract of AM was used for cell viability, it was over 100% at 6% (6 ml/100 ml in phosphate buffer) concentration. AM showed 45.7% elastase inhibition and 23.7% MMP-1 inhibition at 50% (50 ml/100 ml in phosphate buffer) concentration so that it had good anti-wrinkle characteristic. And AM showed 68.9% antioxidation capacity at 50% concentration by using a DPPH assay. Consequently, AM can be used as natural materials or additives for human skin owing to their beneficial biologic functions, including the anti-wrinkle effect, for cosmetic compositions.

Adenine, new anti-wrinkle agent.

  • It has been known that adenine is a very important material in living cells. Because, adenine is a member of nucleotide base, so it takes part in DNA, RNA and ATP synthesis. There are many reports that adenine participated in ingredients, especially DNA, RNA, NADH and ATP, affect on the cell. As well adenosine, conjugated adenine to glycoside, was known to anti-wrinkle compound. But there is no report whether adenine shows a good effect on the skin, especially anti-wrinkle. So, in this study, we tested whether adenine affects cell proliferation, collagen synthesis, collagenase synthesis inhibition in human dermal fibroblasts. In addition, we performed clinical study with adenine cream. Cell proliferation effect was tested by MTT assay. Collagen and collagenase synthesis were measured by Immunoassay with ELISA kit. Clinical study was performed by IECK according to KFDA Functional Cosmetic method. The results of cell proliferation show that 10$^{-6}$ ~10$^{-8}$ % of adenine increases cell proliferation about 50 % compare with non-treated control. At 10$^{-7}$ ~10$^{-10}$ %, adenine increases type I collagen synthesis about 50%, decreases type I collagenase about 22% compare with non-treated control. The results of clinical study show that 0.05% adenine treated group reduces wrinkle significantly compare with placebo treated group. Therefore adenine may be a new anti-wrinkle candidate, through increases cell proliferation and collagen synthesis dramatically. And it decreases collagenase synthesis. So adenine could be used as a new anti-wrinkle agent.

The Anti-wrinkle Effects and Whitening Effects of Galla Rhois (오배자의 항 주름 효과 및 미백 효과에 미치는 실험적 연구)

  • Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search Galla Rhois (GR) inhibitory activity against aging process of skin in vitro. GR was screened for their inhibitory activity against elastase. GR (final concentration 1 mg/ml) appeared over 40% of inhibition of elastase activity. So we investigated anti wrinkle effects of GR look through MMP-1 inhibition activity. Also Extracts of GR showed higher anti-tyrosinase activity than arbutin (final concentration 1 mg/ml). These results suggest that GR could be potential sources of anti-aging and whitening effects for the skin. Objective : Galla Rhois extracts, Elastase (pancreatic from porcine pancreas), N-Succinyl-(Ala)3-p-nitroanilide, Ursolic acid, mushroom tyrosinase, L-tyrosine, arbutin, Amicon Ultra, Hand Hold UV lamp, MMP-1 primary antibody, MMP-1 secondary antibody Result : 1. GR appeared over 40% of elastase inhibitory activity. 2. GR had MMP-1 inhibition activity. 3. GR showed higher anti-tyrosinase activity than arbutin. Conclusion : According to above results, it is supposed that Galla Rhois could be strong potential sources of anti-aging and whitening effects for the skin.

  • Cosmetics have mild activity on the human body, and intended to be used for cleaning, beautifying the human body, and also keeping healthy skin or hair, promoting attractiveness or altering the appearance. Functional cosmetics, in other words cosmeceuticals, are restricted for following functions: 1) Whiten the skin tone by preventing deposition of melanin pigmentation or lightening of the color of melanin of skin, 2) alleviate or improve wrinkles of the skin, and 3) protect the skin from the ultra violet rays from the sun. According to the functions of the functional cosmetics, skin whitening products, anti-wrinkle products, and suntan & sunscreen products are manufactured. In order to manufacture and import the functional cosmetics in Korea, the approval process in KFDA is necessary. The review process in KFDA is performed based on The Korea Food and Drug Administration Notification 2007-44, "The Regulation of Reviewing the Functional Cosmetics" (June 29, 2007). Only after the approval of KFDA, functional cosmetics are allowed to advertise to the consumers for their functionality.

Anti-wrinkle Activity of Acanthopanax senticosus Extract in Ultraviolet B (UVB)-induced Photoaging (가시오가피 추출물의 광노화에 의한 주름형성 억제 효과)

  • This study was performed to investigate anti-wrinkle effects of Acanthopanax senticosus (AS) on ultraviolet B (UVB)-induced photoaging with wrinkle formation. AS extract showed higher DPPH radical scavenging activity (3 ${\mu}g/mL$ as $IC_{50}$) and collagenase inhibition (1.52 mg/mL as $IC_{50}$) than those of ascorbic acid (50 ${\mu}g/mL$ and 2.17 mg/mL, respectively). Cell proliferation and type I pN collagen synthesis were increased by 11.4% and 96.4%, respectively, compared with non treatment control. In vivo, SKH-1 hairless mice were administrated AS 400 mg/kg for 10 weeks with UVB irradiation three times a week. After 10 weeks, a visual assessment and replica assay were performed on each mouse. According to visual assessment of close-up photos and skin replica, oral administration of A. senticosus affected on inhibition of wrinkle formation caused by UVB irradiation on the skin of mice as compared to the vehicle treated control mice. These results indicated that A. senticosus could protect skin wrinkle formation caused by collagen synthesis of fibroblast cells and photo-irradiation of UVB in hairless mice.
