Assessment of the Changes in the Microbial Community in Alkaline Soils using Biolog Ecoplate and DGGE (Biolog Ecoplate와 DGGE 방법을 이용한 알칼리화 토양의 미생물군집 변화 평가)

  • Soil microbial community analysis of farmland soil sprayed with lye in order to use fertilizer in Nigeria was performed. As a control, two kinds of soils not sprayed with lye, located in Eungo and Lagos with general practice in agriculture was selected. Soil sprayed with lye was pH 8.25 through alkalization reaction, while the other soil samples were pH 6.22 and 5.94. Substrate utilization and species diversity index of soil sprayed with lye were low than that of the other soils with the analysis of Biolog ecoplate. As a result of principal component analysis, the relationship between three samples was low. Microbial community analysis was performed by DGGE and most of them were soil uncultured bacterium. Especially, Uncultured Acidobacteria and Uncultured Methylocystis sp., which had been isolated from the rhizosphere of soybean grown in that site were discovered in the soil sprayed with lye.

Cloning and expression of new laccase gene (soncotA) from Bacillus sonorensis KCTC13918 in E. coli (Bacillus sonorensis KCTC13918로부터 새로운 laccase유전자 (soncotA)의 클로닝과 대장균에서의 발현)

  • A new putative laccase gene (soncotA) which show 78% homology with that from Bacillus licheniformis (liccotA) was isolated from draft genome sequence of Bacillus sonorensis KCTC 13918. A 1,545 bp of PCR product corresponding 514 amino acids was cloned into NdeI-NotI site of pET21c and expressed as soluble form in E. coli. About 59 kDa size of recombinant laccase was purified into homogenity by Ni-NTA column and laccase activity was confirmed by zymography. The enzymatic properties of recombinant laccase were characterized. The specific activity of B. sonorensis laccase was 0.033 fold lower than that of Bacillus licheniformis laccase. The finding of new laccase gene broadened the enzymatic diversity of Bacillus species laccases.

  • The word \"permaculture\" is a contraction of \"permanent agriculture\" or \"permanent culture\". Permaculture principles are designed to support (or ensure) the survival of humanity, the earth and all on it, and, to improve our standard of living, Permaculture can be defined as : a design system for creating sustainable human environments that can be (relatively) easily constructed and maintained. The principles of permaculture designs are relative location every element (such as house , rice paddy, road, esc) is placed in relation to each other such that each assists the other : each element performs multiple functions ; each function is supported by many elements : energy efficient planning ; using biological resources rather than fossil fuels : energy cycling on site (both fuel and human energy) : Using and accelerating natural plant succession to establish favourable sites and soils ; poly-culture and diversity of beneficial species for a productive, interactive system ; use of edge and natural patterns for best effect. These principles can be used for any permaculture design, in any climate, and at any scale. There has been very little scientific examination of Permaculture, and as such, it must be viewed as a design philosophy.viewed as a design philosophy.

3D-QSAR Study of Competitive Inhibitor for Acethylcholine Esterase (AChE) Nerve Agent Toxicity

  • The cholinesterase-inhibiting organophosphorous (OP) compounds known as nerve agents are highly toxic. The principal toxic mechanism of OP compounds is the inhibition of acethylcholine esterase (AChE) by phosphorylation of its catalytic site. The reversible competitive inhibition of AChE may prevent the subsequent OP intoxication. In this study, three-dimensional quantitative structure-activity relationship (3D-QSAR) was performed to investigate the relationship between the 29 compounds with structural diversity and their bioactivities against AChE. In particular, predictive models were constructed using the comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA). The results indicate reasonable model for CoMFA ($q^{2}=0.453,\;r^{2}=0.697$) and CoMSIA ($q^{2}=0.518,\;r^{2}=0.696$). The presence of steric and hydophobic group at naphtyl moiety of the model may lead to the design of improved competitive inhibitors for organophosphorous intoxication.

Genetic Analysis of the Envelope Region of Hepatitis G Vrius isolated from Korean Hepatitis Patients (한국인 간염환자에서 분리한 G형 감염바이러스(HGV)의 외피영역의 유전적 다양성 분석)

  • The genetic of a recently described virus, hepatitis G virus(HGV) was investigated. HGV envelope 1 (E1) nucleotide sequences isolated from six Korean hepatitis b virus-positive patients by using a reverse transcription-poly-merase chain reaction procedure, were analysed and compared to the seven previously reported HGV isolates. Sequence homology among the Korean isolates was 88-97% whereas among the isolates from different geographic areas was 80-92%, indicating geographical divergence of HGV. Nucleotide substitutions spread uniformly throughiut the E1 fragment. Furthermore, compared to the prototype HGV sequence, frameshift mutations were observed in most of the Korean isolating that a different translating initiation site for the polyprotein exists in the Korean type HGV.

However, recently it comes the argument of skeptic of web 2.0 problem. Thus, In this research I will try to give an right comprehension and will try to have more accurate prediction of web 2.0 with the study of open service of web which is the key of web 2.0 success in a future.

  • Pleomorphic adenoma is a benign salivary gland tumor with histologic diversity. The majority of these tumor occurs in the parotid gland. The authors experienced the patients, who complained the tumor-like soft tissue mass on the palatal area. After careful analysis of clinical, radiological and histopathological findings, we diagnosed it as pleomorphic adenoma in the palatal area, and obtained characteristic features were as follows: 1. Main clinical symptom was a painless, slow growing, soft tissue mass with normal intact overlying mucosa on the palatal area. 2. In the radiographic examminations, well encapsulated homogeneous soft tissue mass was shown in the lesion site, and cortical thinning on the palate was also observed. 3. In histopathologic examminations, proliferated cellular components in the hyaline stroma were observed as double layered duct-like structure and densely solid sheet appearance.

  • Visible light communication(VLC) is one type of short-range, optical, and wireless communication system utilizing light emitting diode(LED) and laser diode(LD) as optical source. In a VLC system, visible light is used as a transmission medium and used to illuminate. Using VLC has a lot of advantages: it is harmless to human body; it transmits with high power, and it has excellent security, a high data rate, and a license free frequency band. With such a unique blend of communication and illumination in one system, the most common application would be an indoor environment. We aim at reviewing key issues in VLC network such as : FOV(field of view), priority MAC, cooperative MAC, link switching, LED-ID technique, cell site diversity, and link recovery.

Study on herbs vegetation in the vicinity of onsan industrial complex (溫山工團 周邊 海松林의 草本植生에 關한 調査)

  • In order to estimate effects of air pollution for herbs in the plnus thunbergii forests arrounding onsan industrial complex, this study was surveyed. number of species of herb also had a difference over 10 species by distance at the source of air pollution, and species diversity and evennes were also increasing at remote sites in industrial complex as woody plants. At the vicinity of industrial complex, $SDR_4$ of calamagrostis arundinatios arundinacea., miscanthus sinensis and cymbopogon tortilis var. goeringii were very high, and those of lianas as cocculus trilobus, lonicera japonica and paederia scandens, and phytolacca americana were low, but because of being higher than other sites, it was able to inferred that lianas had a tolerance to air pollution. The composition of biological spectra of herbs showed to type of $H-D_1-R_{2-3}-1$ at near site in the source of air pollution and $H-D_1-R_{1-3}-t$ at the other sites, and the propotion of lians forms were relatively high at the vicinity of industril complex.

Phylogenetic and Recombination Analysis of Apple Stem Grooving Virus Isolates from Pears in Korea

  • The apple stem grooving virus (ASGV) is one of the most harmful latent viruses infecting pear orchards worldwide. To examine the genetic diversity of ASGV in Korean pear orchards, the complete coat protein (CP) gene of five ASGV isolates collected from various regions were identified. The five Korean ASGV isolates showed 88-96% nucleotide identity with the 11 isolates worldwide occurring elsewhere in the world. Phylogenetic analysis of five isolates, as well as the previously sequenced isolates, indicated that the ASGV clusters had no correlation with the host or geographical regions of origin. Recombination analysis showed that one of the five Korean isolates is a recombinant, with a recombination site in the CP gene region (nt 532-708). This study is the first report of natural recombination within the CP gene of ASGV isolates from pears grown in Korea.
