THE EFFECTS OF IRRADIATION ON THE PERIODONTAL TISSUES OF RATS WITH THE LOW CALCIUM DIET (방사선조사가 저칼슘식이 백서 치주조직에 미치는 영향에 관한 연구)

  • The purpose of this study was to investigate the changes of periodontal tissues in the irradiated mandibular bone in rats which were fed normal diet and low calcium diet In order to carry out this experiment, 64 seven-week old Sprague-Dawley strain rats weighing about 150gms were selected and equally divided into one experimental group of 32 rats and one control group with the remainder. The experimental group and the control group were then subdivided into two groups when the rats reached the age of 10 weeks, 16 rats were allotted for each subdivided group was composed of 16 rats and exposed to irradiation. The two groups were irradiated a single dose of 20Gy on the only jaw area and irradiated with a cobalt-60 teletherapy unit The rats in the control and experimental groups were serially dissected by fours on the 3rd, the 7th, the 14th, and the 21st day after irradiation. After each dissection, both sides of the dead rat mandibular bodies were removed and fixed with 10% neutral formalin. The specimens sectioned and observed in histopathological. histochemical. and immunocellular chemical methods. The obtained results were as follows: 1. In the mandibles of rats with low calcium diet the increased number of fibroblasts of periodontal ligaments. many small capillaries and irregular arrangement of loose collagen fibers were detected and the partial resorption of dentin and cementum could be found by the microscopic studies. 2. In the group of irradiated rats, degenerated periodontal tissues led to the condition of irregular arrangement of collagen fibers and the decreased number of fibroblasts. But this condition was somewhat restored after 21 days of experiment. 3. Periodontal tissues of the irradiated rat group with low calcium diet were destroyed earlier than those of the irradiated rat group with normal diet. Soon this condition was restored and then high cellularity and dense collagen fibers were observed. 4. Many periodontal cells bearing tumor necrosis factor could be clearly observed in the nonirradiated group of rats with normal diet, whereas could not be observed on the 7th day and reappeared on 14th day in the irradiated group of rats with normal diet. A few of them could be observed in the group of rats with low calcium diet, but they could be clearly observed in the both groups after 21 days of experiment.

Performance Analysis of Laboratory and Field Luminance for Phosphorescent Line Marking and Preliminary Study of Luminance Analysis Using Digital Images (축광노면표시의 실내 및 현장 휘도 성능분석과 디지털이미지를 이용한 휘도분석 사전연구)

  • OBJECTIVES : Visibility at night can be improved by using retroreflection for short distances and phosphorescent line markings for long distances. In this study, we analyzed the characteristics of the phosphorescent line marking through a laboratory luminance test. Field performance analysis was performed through tests conducted on the road. We also examined the luminance measurement methods using the digital image obtained during the phosphorescent visibility evaluation. METHODS : In this study, the laboratory luminance test of the phosphorescent line marking was conducted using seven specimens to characterize the luminance changes according to the type of the glass beads, the thickness of the phosphorescent line marking, and the brightness and irradiation time of the light source. Phosphorescent and general line markings were made at 150 m to investigate the field luminance performance. A preliminary review of the luminance measurement methods was made using a digital image from a digital single-lens reflex (DSLR) camera. The measured luminance ratio of the general and the phosphorescent line markings was compared with the calculated luminance ratio using luminance analysis. RESULTS : Through the laboratory luminance test, it was seen that the change in luminance, which corresponds to the brightness of the light source, appears large but the influence of the thickness and irradiation time is low. The field performance test of the phosphorescent line marking conducted on the road involved measuring the luminance on the day the marking was made and 7 days after the marking was made. The luminance was found to be $190mcd/m^2$ at 30 min after sunset and approximately $10-12mcd/m^2$ 4h after sunset. The results of the luminance test were captured using a digital image for each time group. The luminance ratio of the phosphorescent line marking, when compared to that of the general line marking, showed a similar trend within a 13% maximum error. Additionally, when this luminance ratio is compared to the direct field measurement, it could be confirmed that the luminance ratio, as captured in the digital image, showed a similar tendency. CONCLUSIONS : 1) The change in luminance corresponding to the brightness of the light source is significant in comparison with that corresponding to the thickness and the irradiation time. In addition, the results of the field test for the phosphorescent line marking satisfied the phosphorescent fire protection standard. 2) We examined the validity of the luminance measurement method using a digital image and we concluded that the change in the luminance ratio shows a similar tendency in both the cases. The results can form the basis for luminance measurement methodology for the construction and maintenance of phosphorescent line markings.

The Combined Effects of Curcumin Administration and 630 nm LED Phototherapy against DNCB-induced Atopic Dermatitis-like Skin Lesions in BALB/c Mice (BALB/c 마우스에서 DNCB-유도 아토피 피부염 유사병변에 대한 Curcumin 투여와 630 nm LED 광치료의 병용 효과)

  • Atopic dermatitis (AD) is a chronic inflammatory skin disease. It is characterized by eczematous lesions, skin dryness, and pruritus. The existing treatment drugs for AD have side effects, especially if the drugs are taken for extended periods. Therefore, new alternative therapies are necessary. The aim of this study was to investigate the combined effects of curcumin administration and LED irradiation on AD. AD-like lesions were induced in BALB/c mice by repeated application of 1-chloro-2,4-dinitrobenzene (DNCB) to the shaved skin of the ear and neck. Thirty male BALB/c mice were divided into five groups: vehicle, DNCB, curcumin, LED, and curcumin+LED groups. Curcumin (0.1 g/kg/day) was administrated repeatedly during a period of 14 days (experimental period) and 630 nm LED irradiation ($5J/cm^2/day$) was performed in the acryl box once a day for 10 days, after inducing AD-like lesions via DNCB application. The severity of AD-like lesions was evaluated during the experimental period, using a modified SCORAD index. Both ear and neck skin tissues were examined histologically for epidermal thickness, mast cell, eosinophil counting, and dermal collagen density. Epidermal cell proliferation and apoptosis were detected using immunohistochemistry and TUNEL, respectively. These were all reduced in SCORAD index, epidermal thickness, collagen density, number of mast cell and eosinophil in dermis, and number of proliferating cell and apoptotic cell in epidermis by curcumin administration and 630 nm LED irradiation. Moreover, all parameters were significantly lower in the curcumin+LED group compared with the curcumin group and LED group. These results suggest that the combined therapy of curcumin and LED is more effective than a single treatment. We recommend that this can be a feasible alternative therapy to manage AD.

  • Total 28 patients with resectable, locally advanced gastric cancer were entered in our prospective randomized study from June 15, 1988 to Sep. 15, 1990 in Yeungnam University Hospital. This study consisted of curative resection, IORT, external irradiation and combination of chemotherapy. Twenty-four of 28 patients were treated with single dose of 1500 cGy with 9 MeV election intraoperatively. External irradiation of $4300\sim4500$ cGy with 180 cGy per fraction, 5 days per week was started within 4th weeks of postoperative days. Various chemotherapy with or without external irradiation were added for reducing hematogenous and/or peritoneal dissemination and determination of complication of each arm. Duration of follow up was $4\sim31$ months. No serious complication related with radiation were reported compare to resection and chemotherapy only group. Although our follow up period is too short to draw any conclusion, IORT appears to improve local control, hopely further survival. Continuous follow up should be needed for evaluation of real therapeutic gain such as complication vs. improved survival.

The Effect of Radiation Therapy on the Healing Ability of Subsequent Surgical Wounds (방사선조사가 피부의 창상치유에 미치는 실험적 연구)

  • This study was undertaken to find the effect of radiation therapy on the healing ability of surgical wounds and on this basis, to find the proper time interval between the radiation therapy and surgery. Two hundred and fifty-two mice were used and a single dose of 2000 cGy was given in each instances to the hind limb of mice. Incisional wounds were produced after varying intervals in the previously irradiated areas and then they were followed up at regular intervals by the measurement of tensile strength. The wounds which received surgery immediately, 1 or 2 weeks after irradiation revealed marked delay and the wounds which received surgery 12, 16 or 20 weeks after irradiation demonstrated slight delay in wound healing in terms of tensile strength measurement. But the wounds which received irradiation 4 or 8 weeks before surgery did not differ much in the wound healing process from that of the control group. Histopathologic studies of the wounds demonstrated epithelization in most instances as quickly as in the control wounds. The appearance of fibroblasts and collagen fibers has delayed momently and appeared to have close correlation with the tensile strength healing curves.

  • Effects of varying gamma-irradiation doses and polyethylene package on the quality and chemical components of Jonathan apples were investigated. The results obtained are as follows: 1. The doses of 50 to 100 krad were effective in the control of fruit rot and physiological disorders. 2. Gamma-irradiation to Jonathan apples rather accelerated shrivelling at the early stage of storage period. 3. Combined treatments of radiation and polyethylene package enabled to control the shrivelling of apples. 4. The rate of freight loss was lower in the irradiated and/or packing lots than control 5. Gamma-irradiation and polyethylene package did not appear to influence on the sugar content of apples.

Morphological Changes of Hair Structure by UV Irradiation (자외선 조사에 의한 모발구조의 형태학적 변화)

  • In this study, we have investigated the effects of ultraviolet (UV) irradiation on the external and internal structure of human hair. For changes in morphological and chemical structure of UV-exposed hair, we utilized several microscopic techniques such as scanning electron microscope (SEM), transmission electron microscope (TEM), confocal laser scanning microscope (CLSM), and etc. The SEM showed the scales of UV-exposed hair appeared to be rough and bulkier because of chemical oxidation during UV irradiation. Small bulgy area of UV_exposed hair surface was appeared as a result of the decomposition in cysteic acid. In the TEM picture the UV-exposed hair showed the cleaved aperture of hair and destruction of melanin granules. Three dimensional topographical images were obtained by using CLSM. In the optical single section, high fluorescent intensity appeared in virgin hair. In the case of UV-exposed hair, low fluorescence intensity appeared. This means the aromatic amino acids in virgin hair were more abundant than UV-exposed hair.

The Relationship between Radiation-Induced Apoptosis and the Expression of Cytokines in the Rat's Liver (백서 간에서 방사선조사에 의한 Apoptosis와 Cytokine 발현과의 관계)

  • Purpose : To determine the role of cytokines in the apoptosis of rat's liver following irradiation. Materials and Methods : Sprague-Dawley rats were irradiated to entire body with a single dose of 8 Gy. The rats were divided Into 5 groups according to the sacrlfice day after irradiation. The liver and blood after 1, 3, 5, 7, and 14 days irradiation were sampled for evaluation of mechanism of apoptosis and role of cytokine in relation to radiation-induced tissue damage. The study was composed of microscopic evaluation of liver tissue, in situ detection method for apoptosis, immunohistochemical stain of IL-1, IL-4, IL-6 and TNF, bioassay and radioimmunoassay of IL-6 in liver tissue and blood. Results : Radiation-induced liver damage was noted from first day of radiation, and most severe parenchymal damage associated with infiltration of chronic inflammatory cells was seen in the groups of 5 days after radiation. A number of apoptosis were observed 1 day after radiation on both light microscope and in situ method. Afterwards, the number of apoptosis was gradually diminished. On immunohistochemical study, IL-1 and TNF were expressed 1, 3 days after radiation, but not expressed after that. IL-4 was not expressed in the entire groups. IL-6 was expressed with strong positivity in 1, 3 days after radiation. Bioassay and RIA of IL-6 in liver tissue and blood showed the highest value in 1 day after radiation, and the value is diminished after then. Conclusion. Apoptosis seemed to be the important mechanism of radiation-induced liver damage, and is possibly induced by the release of cytokines, such as IL-1, IL-6, TNF in view the simultaneously increased appearance of pooptosis and cytokines.

Effects of total body irradiation-based conditioning on allogeneic stem cell transplantation for pediatric acute leukemia: a single-institution study

  • Purpose: To evaluate the effects of total body irradiation (TBI), as a conditioning regimen prior to allogeneic stem cell transplantation (allo-SCT), in pediatric acute leukemia patients. Materials and Methods: From January 2001 to December 2011, 28 patients, aged less than 18 years, were treated with TBI-based conditioning for allo-SCT in our institution. Of the 28 patients, 21 patients were diagnosed with acute lymphoblastic leukemia (ALL, 75%) and 7 were diagnosed with acute myeloid leukemia (AML, 25%). TBI was completed 4 days or 1 day before stem cell infusion. Patients underwent radiation therapy with bilateral parallel opposing fields and 6-MV X-rays. The Kaplan-Meier method was used to calculate survival outcomes. Results: The 2-year event-free survival and overall survival rates were 66% and 56%, respectively (71.4% and 60.0% in AML patients vs. 64.3% and 52.4% in ALL patients, respectively). Treatment related mortality rate were 25%. Acute and chronic graft-versus-host disease was a major complication; other complications included endocrine dysfunction and pulmonary complications. Common complications from TBI were nausea (89%) and cataracts (7.1%). Conclusion: The efficacy and toxicity data in this study of TBI-based conditioning to pediatric acute leukemia patients were comparable with previous studies. However, clinicians need to focus on the acute and chronic complications related to allo-SCT.

Photo- and Sonic Degradation of Endosulfans(α, β, and sulfate) in Aqueous Solution (엔도설판류의 광 및 초음파분해)

  • Endosulfan-${\alpha}$ endosulfan-${\beta}$ and endosulfan-sulfate, which are classified as pesticides, were degraded by use of UV energy and ultrasonic irradiation. The degradation residuals were analysed by gas chromatography with an electron capture detector and TOC (total oragnic carbon) analysis. The reactions were conducted in a quartz annular reactor equipped with a low pressure mercury multilamp (8Wx2) and a sonic generator. All the aqueous solutions were concentrated as 10 mg/L initially. Endosulfans were degraded each to result in 48.2% (${\alpha}$), 50.0% (${\beta}$) and 76.5% (sulfate) of removal efficiency by UV energy, and 66.9% (${\alpha}$), 55.8% (${\beta}$) and 72.7% (sulfate) by ultrasonic irradiation, respectively. In contrast to the results of the single-component solutions, degradation of the endosulfan-sulfate was greatly suppressed to result in the lowest degradation rate and removal efficiency in the three-component solutions. This finding suggests that there should be a reversible reaction with a substantially low equilibrium constant between endosulfan-${\alpha}$ or -${\beta}$ and -sulfate in the coexistence of the three endosulfans. TOC data showed the endosulfans were decomposed by 20%~40% toward complete mineralization, producing a quantity of intermediates induced by the radical reactions. We found that all the decay reactions considered in this study nicely fell into pseudo first-order rate.
