버크셔 품종의 돼지 성장과 육질관련 후보유전자의 단일염기 다형성에 관한 연구 (Investigation of Single Nucleotide Polymorphisms in Porcine Candidate Gene for Growth and Meat Quality Traits in the Berkshire Breed)

  • 4개의 후보 유전자를 분석해본 결과, 돼지의 주요 염색체 부위 및 유전자들이 주요 경제성 요인들과 관계가 있는 것으로 확인됐다. 양돈업계에서 DNA 기술을 이용한 염색체 정보를 활용하기 위해 본 연구에서는 4개의 후보 유전자에서 생성된 중합효소연쇄반응(PCR) 생성물을 비교 재 서열 함으로써 단일염기변이(SNP) 표지들을 개발했다. 또한 이들 4개의 SNP에 대해 PCR 제한효소 절편길이 다형 성(RFLP)분석을 전개한 후, 이를 대한민국 내 버크셔 종 돼지 개체군의 유전자형을 분석하는데 활용했다. 본 연구는 유용한 단일염기변이를 식별하고 돼지개체군 내 경제적으로 중요한 특성들과 SNP의 연관성을 확인하는 데 그 목적이 있다.

부산일대 곰솔림과 소나무림의 생육 밀도에 관한 연구 (A Study for Growth Density on the Pinus thunbergii and Pinus densiflora Communities in area of Busan, Korea)

  • 기후변화로 인한 곰솔림과 소나무림의 세력은 약해질 것으로 예상되고 있으나, 곰솔과 소나무에 대한 식재요구도는 여전히 높은 실정이다. 이에 본 연구는 한반도 난온대림지역에 속하는 부산일대, 남동부지역에 주요 조경수목인 소나무림과 곰솔림을 대상으로 자연림의 생육밀도를 파악한 후 난온대림에 적합한 생태적 식재밀도를 제시하고자 하였다. 연구는 한반도 남동부에 해당하는 부산광역시를 중심으로 소나무림 22개소, 곰솔림 60개소를 선정하여 $100m^2$ 방형구를 설치하여 조사구 내 출현하는 교목, 아교목층 수종의 흉고직경 및 개체수를 조사한 후, 회귀분석을 통해 평균흉고직경급별 적정 개체수 산정을 위한 회귀식을 도출하였다. 회귀분석 결과, 흉고직경의 생장에 따른 교목층 생육개체수 예측모형의 결정계수는 곰솔림 0.700, 소나무림 0.533으로 단일 요인에 의한 설명력으로는 매우 높은 것으로 분석되었다. 예측 회귀모형은 곰솔림의 경우 $Y=31.176e^{-0.055x}$, 소나무림의 경우 $Y=38.351e^{-0.059x}$(X=흉고직경, Y=개체수/$100m^2$)였다. 남동부지역의 곰솔림과 소나무림의 생육밀도는 중부지방보다 다소 높았다.

  • FGFs는 Xenopus의 초기 배발생에서 중배엽 형성, 전후축패턴형성, scatter factor로서 낭배기의 기관형성에 관여하는 등 다양한 기능을 가지고 있는 것으로 알려져 있다 그 중 bFGF는 배양 분리편으로부터 다양한 기관을 유도해낼 수 있으며 그 효과는 처리시간 및 농도 의존적이라고 알려져 왔다. 본 연구는 Xenopus의 예정표피역을 분리하여 bFGF와 HGF을 단독 및 복합처리 하였을 때 기관분화 및 유도효과를 검토하기 위하여 실시하였다. 단독처리 및 복합 처리된 배양액에 동물극 분리편을 정상배가 st. 43에 이를 때까지 2$0^{\circ}C$에서 3일간 배양하여 조직학적 및 면역조직화학적 방법으로 조직의 분화양상을 확인하였다. 성장인자는 bFGF를 0, 0.5, 1. 10. 50 ng/ml의 농도와 HGF를 0, 1, 10, 50, 100ng/ml의 농도로 조합하여 처리한 결과 bFGF 단독처리 때보다 HGF와의 혼합처리에서 기관분화율의 상승효과가 관찰되었다. 분화된 기관은 1 ng/ml의 bFGF 와 10ng/ml의 HGF, 10ng/ml의 bFGF와 1ng/ml의 HGF의 농도에서 매우 다양한 것으로 나타났다. 눈은 1과 10ng/ml의 bFGF ,그리고 1과 10 ng/ml의 HGF 농도조합에서 높은 비율로 분화하였다. 또한 분리편 배양에 의해 유도된 눈과 정상 배의 눈에서 RPE65를 인식하는 단일클론 항체 40All, 25F5를 사용하여 AP 반응이 강하게 나타나 눈의 유도를 확인할 수 있었다.

Prostate Cancer Risk in Relation to a Single Nucleotide Polymorphism in the Insulin-like Growth Factor-binding Protein-3 (IGFBP3) Gene: a Meta-analysis

  • Insulin-like growth factor-binding protein-3 (IGFBP3) has been identified as a putative tumor suppressor with multifunctional roles in the IGF axis. Recently, there have been a growing body of studies investigating the relation between the IGFBP3 A-202C polymorphism, circulating IGFBP3 and prostate cancer risk, but their outcomes varied leading to controversy. Hence, it is necessary to perform a meta-analysis covering all eligible studies to shed a light on the association of IGFBP3 A-202C and cancer risk. Finally, we included a total of 11 relevant articles between 2003 and 2010 covering 14 case-control studies including 9,238 cases and 8,741 controls for our analysis. Our results showed that A-202C was a marginal risk factor of prostate cancer (allele contrast: OR=1.08, 95% CI :1.01-1.16; dominant model: OR=1.11, 95% CI :1.01-1.22; heterozygote codominant model: OR=1.11, 95% CI :1.03-1.18; homozygote contrast: OR=1.19, 95% CI :1.03-1.37). Stratification analysis revealed that sample size and control source were two major heterogeneous meta-factors especially in the recessive model (source: Population-based control group :p=0.30,I2=16.7%, Hospital-based control group: p=0.20, I2=30.3%; sample size: Small: p=0.22,I2= 32.8%, Medium: p=0.09,I2=48%, Large p=0.60,I2=0.0%); However, contrary to previous findings, no significance was found in racial subgroups. No significant publication bias was found in our analysis. Considering the robustness of the results and the discrepancy among some studies, there might be some unsolved confounding factors, and further more critical large studies are needed for confirmation.

귀비탕(歸脾湯)이 Estradiol Valerate로 유도된 흰쥐의 다낭성 난소에 미치는 영향 (Effects of Gwibi-Tang on the Estradiol Valerate-induced Polycystic Ovaries in Rats)

  • Purpose: This study was designed to investigate the effects of Gwibi-Tang(GBT) on the polycystic ovary(PCO) induced by estradiol valerate(EV) in rats. Methods: PCO was induced by single intramuscular injection with EV(4mg) in female rats. Normal group(n=8) were injected with sesame oil and orally administrated distilled water for eight weeks. PCO control group(n=8) were injected with EV and orally administrated distilled water for eight weeks. GBT treated group(n=8) were injected with EV and orally administrated GBT for eight weeks. Then we measured weight of body, ovaries, adrenal glands, and uterus of rats. The histopathology changes of ovaries were also evaluated. The expression of nerve growth factor(NGF) was analyzed in the central nervous system, adrenal glands and ovaries by immunohistochemistry. And also CRF expression in median eminance of Rats were analyzed. Results: 1. The weight(g) of rats in GBT treated group($275{\pm}14$) was significantly increased(p<0.01) compared with control group($253{\pm}8$), 2. The weight(mg) of ovaries in GBT treated group($75.8{\pm}16.7$) was significantly increased(p<0.001) compared with control group($37.4{\pm}6.7$). 3. The number of mature follicles in GBT treated group($3.6{\pm}1.2$) was significantly increased(p<0.01) compared with control group($1.5{\pm}1.5$. 4. The number of atretic follicles in GBT treated group($8.0{\pm}3.1$) was significantly decreased(p<0.01) compared with control group($18.6{\pm}6.0$). 5. The number of cystic follicles in GBT treated group($0.5{\pm}0.5$) was significantly increased(p<0.01) compared with control group($2.3{\pm}1.3$). 6. The number of corpora lutea in GBT treated group($6.1{\pm}3.9$) was significantly increased(p<0.01) compared with control group($1.6{\pm}2.3$). 7. The expression of NGF-immunoreactive cells in the ovarian granulosa cells in GBT treated group was lesser observed than control group. Conclusion: From the above results, we concluded that Gwibi-Tang has inhibitory effect on the development of EV-induced polycystic ovary. And it's effect may be related with decreased NGF activities in the ovary.

태양 전지용 $CuInSe_2$ 단결정 박막 성장과 태양 전지로의 응용 (Growth of $CuInSe_2$ single crystal thin film for solar cell development and its solar cell application)

  • The stoichiometric mixture of evaporating materials for the $CuInSe_2$ single crystal thin film was prepared from horizontal furnace. Using extrapolation method of X-ray diffraction patterns for the polycrystal $CuInSe_2$, it was found tetragonal structure whose lattice constant $a_0$ and $c_0$ were $5.783\;{\AA}$ and $11.621\;{\AA}$, respectively. To obtain the $CuInSe_2$ single crystal thin film, $CuInSe_2$ mixed crystal was deposited on throughly etched GaAs(100) by the HWE(Hot Wall Epitaxy) system. The source and substrate temperature were $620^{\circ}C$ and $410^{\circ}C$ respectively. The crystalline structure of $CuInSe_2$ single crystal thin film was investigated by the double crystal X-ray diffraction(DCXD). Hall effect on this sample was measured by the method of Van der Pauw and studied on carrier density and mobility depending on temperature. From Hall data, the mobility was likely to be decreased by impurity scattering in the temperature range 30 K to 100 K and by lattice scattering in the temperature range 100 K to 293 K. The temperature dependence of the energy band gap of the $CuInSe_2$ obtained from the absorption spectra was well described by the Varshni's relation, $E_g(T)=1.1851\;eV-(8.99{\times}10^{-4}\;eV/K)T^2/(T+153\;K)$. The open-circuit voltage, short current density, fill factor, and conversion efficiency of $n-CdS/p-CuGaSe_2$ heterojunction solar cells under $80\;mW/cm^2$ illumination were found to be 0.51V, $29.3\;mA/cm^2$, 0.76 and 14.3 %, respectively.

The source and substrate temperature were $600^{\circ}C$ and $440^{\circ}C$ respectively. The crystalline structure of $CaAl_2Se_4$: Co single crystal thin film was investigated by the double crystal X-ray diffraction (DCXD). Hall effect on this sample was measured by the method of Van der Pauw and studied on carrier density and mobility depending on temperature. From Hall data, the mobility was likely to be decreased by impurity scattering in the temperature range 30 K to 100 K and by lattice scattering in the temperature range 100 K to 293 K. The temperature dependence of the energy band gap of the $CaAl_2Se_4$: Co obtained from the absorption spectra was well described by the Varshni's relation, $E_g(T)=3.8239eV-(4.9823{\times}10^{-3}eV/K)T_2/(T+559K)$. The open-circuit voltage, short current density, fill factor, and conversion efficiency of $p-Si/p-CaAl_2Se_4$: Co heterojunction solar cells under $80mW/cm^2$ illumination were found to be 0.42 V, $25.3mA/cm^2$, 0.75 and 9.96%, respectively.

한국 성인 남녀의 미혼에 대한 인식: 척도개발을 중심으로 (A Development of Awareness Scale of Korean Men and Women on Singleness)

  • 본 연구의 목적은 한국 성인 남녀를 대상으로 미혼에 대한 인식을 알아보는 척도를 개발하는 것이었다. 이를 위해 연구 1에서는 한국 성인 미혼 및 기혼 남녀 490명(남: 244명, 여: 246명, 평균연령 = 35.32세)을 대상으로, 혼자 살 때 필요한 조건, 좋은 점(혹은 유리한 점)과 나쁜 점, 혼자 사는 것을 방해하는 요인을 물어보는 개방형 질문을 실시하였다. 내용분석 결과, 각 질문 내용은 경제적 측면, 관계적 측면, 성격적 측면, 자아관련 측면, 가족적 측면, 사회제도 측면으로 구분되었다. 연구 2에서는 연구 1에서 얻은 자료를 근거로 혼자 살 때 필요한 조건, 혼자 살 때 좋은 점, 혼자 살 때 어려운 점에 대한 문항을 구성한 후, 한국 성인 총 800명(남: 400명, 여: 400명)을 대상으로 탐색적 요인분석을 실시하였다. 그 결과, 혼자 살 때 필요한 조건은 5개요인(24문항, 총설명량: 61.7%), 혼자 살 때 좋은 점은 6개요인(29문항, 총설명량: 61.4%), 혼자 살 때 어려운 점은 6개요인(31문항, 총 설명량: 64.75%)이 추출되었다. 이 요인 구조의 적합성을 검증하기 위해 400명(남: 200명, 여: 200명)을 대상으로 확인적 요인분석을 실시한 결과, 각 인식차원에 대한 척도 모형이 적합한 것으로 나타났다. 이러한 결과를 한국 사회의 미혼 및 결혼 관련 변인들과 현상의 측면에서 논의하였다.

Targeted Editing of Myostatin Gene in Sheep by Transcription Activator-like Effector Nucleases

  • Myostatin (MSTN) is a secreted growth factor expressed in skeletal muscle and adipose tissue that negatively regulates skeletal muscle mass. Gene knockout of MSTN can result in increasing muscle mass in sheep. The objectives were to investigate whether myostatin gene can be edited in sheep by transcription activator-like effector nucleases (TALENs) in tandem with single-stranded DNA oligonucleotides (ssODNs). We designed a pair of TALENs to target a highly conserved sequence in the coding region of the sheep MSTN gene. The activity of the TALENs was verified by using luciferase single-strand annealing reporter assay in HEK 293T cell line. Co-transfection of TALENs and ssODNs oligonucleotides induced precise gene editing of myostatin gene in sheep primary fibroblasts. MSTN gene-edited cells were successfully used as nuclear donors for generating cloned embryos. TALENs combined with ssDNA oligonucleotides provide a useful approach for precise gene modification in livestock animals.

A Whole Genome Association Study to Detect Single Nucleotide Polymorphisms for Blood Components (Immunity) in a Cross between Korean Native Pig and Yorkshire

  • The purpose of this study was to detect significant SNPs for blood components that were related to immunity using high single nucleotide polymorphism (SNP) density panels in a Korean native pig (KNP)${\times}$Yorkshire (YK) cross population. A reciprocal design of KNP${\times}$YK produced 249 $F_2$ individuals that were genotyped for a total of 46,865 available SNPs in the Illumina porcine 60K beadchip. To perform whole genome association analysis (WGA), phenotypes were regressed on each SNP under a simple linear regression model after adjustment for sex and slaughter age. To set up a significance threshold, 0.1% point-wise p value from F distribution was used for each SNP test. Among the significant SNPs for a trait, the best set of SNP markers were determined using a stepwise regression procedure with the rates of inclusion and exclusion of each SNP out of the model at 0.001 level. A total of 54 SNPs were detected; 10, 6, 4, 4, 5, 4, 5, 10, and 6 SNPs for neutrophil, lymphocyte, monocyte, eosinophil, basophil, atypical lymph, immuno-globulin, insulin, and insulin-like growth factor-I, respectively. Each set of significant SNPs per trait explained 24 to 42% of phenotypic variance. Several pleiotropic SNPs were detected on SSCs 4, 13, 14 and 15.
