Large-scale purification and single-dose oral-toxicity study of human thioredoxin and epidermal growth factor introduced into two different genetically modified soybean varieties

  • Thioredoxin (TRX) protein is an antioxidant responsible for reducing other proteins by exchanging cysteine thiol-disulfide and is also known for its anti-allergic and anti-aging properties. On the other hand, epidermal growth factor (EGF) is an important material used in the cosmetics industry and an essential protein necessary for dermal wound healing facilitated by the proliferation and migration of keratinocytes. EGF also assists in the formation of granulation tissues and stimulates the motility of fibroblasts. Hence, genetically modified soybeans were developed to overexpress these industrially important proteins for mass production. A single-dose oral-toxicity-based study was conducted to evaluate the potential toxic effects of TRX and EGF proteins, as safety assessments are necessary for the commercial use of seed-specific protein-expressing transgenic soybeans. To achieve this rationale, TRX and EGF proteins were mass purified from recombinant E. coli. The single-dose oral-toxicity tests of the TRX and EGF proteins were carried out in six-week old male and female Institute of Cancer Research (ICR) mice. The initial evaluation of the single-dose TRF and EGF treatments was based on monitoring the toxicity signatures and mortality rates among the mice, and the resultant mortality rates did not show any specific clinical symptoms related to the proteins. Furthermore, no significant differences were observed in the weights between the treatment and control groups of male and female ICR mice. After 14 days of treatment, no differences were observed in the autopsy reports between the various treatment and control groups. These results suggest that the minimum lethal dose of TRX and EGF proteins is higher than the allowed 2,000 mg·kg-1 limit.

  • In this study, we investigated fatigue crack growth behavior of laser welded sheet metal due to a single overload. Fatigue specimens were made using butt joint of cold rolled sheet metal that was welded by $CO_2$ laser. The fatigue crack propagation tests were performed in such a way that fatigue loading was parallel to the weld line while crack propagation was perpendicular to the weld line. Single overload was applied when fatigue crack tip was arrived near the weld line. The distances between the crack tip and the weld line at which a single overload was applied were 6, 4 and 2mm. The effect of specimen thickness and overload ratio on the fatigue behavior was determined. The plastic zone size of crack tip due to the single overload was determined from the finite element analysis. For investigating fatigue crack growth behavior, we used different thickness specimen 0.9mm and 2.0mm, and variable overload ratio applied fatigue crack propagation test. Also we used finite element analysis for investigating the plastic zone size of crack tip when single overload applied

A Growth-Stimulating Protein in Cow's Milk

  • Growth-stimulating effects of cow's milk was examined using Vero cell cultre. Medium containing whole cow's milk stimulated cell growth about the same degree as that containing fetal bovine serum. The growth-stimulating factor in cow's milk was purified using hydrophobic (phenyl-sepharose) and gel filtration (Sephadex G-100) column chromatographies. It appeared that the factor is a highly hydrophobic protein, since the major growth-stimulating activity was found in the fractions eluted with 50% ethylene glycol from the phenyl-sepharose column during the purification. The purified factor showed a single band on the polyacrylamide gel electrophoresis in the presence of 1% (w/v) SDS. The factor has been found to have a relatively high molecular weight in the range of about Mr=100,000-150,000. In the presence of the purified factor (5%, w/v) in the culture medium, the incorporation of [3H]-thymidine into the cells was increased approximately 2,400-fold over that in the presence of 5% (w/v) fetal bovine serum. It seems that the growth-stimulating factor purified in this study is one of the major growth factors in the cow's milk.

  • This work has been investigated the ralationship between single surface crack length and crack depth have influence on the fatigue life. The simulation based on experimental results of 2.25 Cr-1Mo steel at various crack configuration ratios has enabled successful prediction of fatigue life at room temperature. The effect of crack depth should be considered for predicting fatigue crack growth rates as well as that of surface crack length. It is also shwn that the crack growth mechanisms are in good agreement with expreimental data according to the interaction of crack length and crack depth.

  • An experimental investigation of the fatigue through crack growth behavior under simple stepped variable loading condition has been performed using Al7075-T651. Experiments were carried out by using cantilever bending type specimens, with chevron notches on a small electro-magnetic test machine. Tensile overloads have a retarding effect on the fatigue crack growth rates, therefore tensile overloads were used for the beneficial effect on the fatigue life. While in most cases compressive overloads have only a vanishing effect on crack growth rates, some experiments with single edge crack tension specimens reveal a marked growth retardation. The stress ratios used in this investigations varies from R=0.32 to 0.81, from R=0.04 to 0.76, from R=-0.15 to 0.73, and from R=-0.33 to 0.68 and the peak load for each case was not varied. The crack growth and crack closure were measured by Kikukawa's compliance method with a strain gauge mounted on the backside of each specimens. The results obtained are as follows. When the stepped variable load was applied, the smaller the stress ration was, the larger the delayed retardation of the crack growth rate was. The fatigue crack growh rate data obtained for through cracks were plotted well against the effective stress intensity factor range from 4.0 to 20.0MP{a^{SQRT}m}. It was found that the effective stress intensity factor range ratio was related well to the opening stress intensity factor, the maximum stress intensity factor, and crack length.

Single Image-Based 3D Tree and Growth Models Reconstruction

  • In this paper, we present a new, easy-to-generate system that is capable of creating virtual 3D tree models and simulating a variety of growth processes of a tree from a single, real tree image. We not only construct various tree models with the same trunk through our proposed digital image matting method and skeleton-based abstraction of branches, but we also animate the visual growth of the constructed 3D tree model through usage of the branch age information combined with a scaling factor. To control the simulation of a tree growth process, we consider tree-growing attributes, such as branching orders, branch width, tree size, and branch self-bending effect, at the same time. Other invisible branches and leaves are automatically attached to the tree by employing parametric branch libraries under the conventional procedural assumption of structure having a local self-similarity. Simulations with a real image confirm that our system makes it possible to achieve realistic tree models and growth processes with ease.

  • Aircraft structures and engineering structures are always subject to variable amplitude loads. Variable amplitude loads include some kind of loading history; for example, constant amplitude load, single peak overload and block overload etc. Crack growth under variable amplitude loading exhibits retardation effect. In this study, the 4 point bending fatigue test was performed by hydrolic servo fatigue testing machine on 7075-T6 Al-alloy. The retardation effect of overload ratio and numbers of overload cycle was quantitatively studied. 1) Change of retardation effect against increment of overload ratio is more evident when the multiple overload is applied than single overload is done. 2) The number of overload cycle is very important factor about the crack growth retardation effect when the overload ratio is more than 1.75; that is not when the overload ratio is less than 1.75. 3) Overload affected zone size increased gradually by increment of crack growth retardation effect. 4) Crack driving force is more greatly reduced when the crack tip branched off two direction than it sloped to one direction.

  • Constant .DELTA.K fatigue crack growth rate experiments were performed by applying an intermediate single and multiple overload for structural steel, SM45C. The purpose of the present study is to investigate the influence of multiple overloads at various stress intensity factor ranges and the effect of statistical variability of crack retardation behavior. The normalized delayed load cycle, delayed crack length and the minimum crack growth rate are increased with increasing baseline stress intensity factor range when the overload ratio and the number of overload application were constant. The crack retardation under low baseline stress intensity factor range increases by increasing the number of overload application, but the minimum crack growth rate decreases by increasing the number of overload application. A strong linear correlation exists between the minimum crack growth rate and the number of overload applications. And, it was observed that the variability in the crack growth retardation behavior are presented, the probability distribution functions of delayed load cycle, delayed crack length and crack growth life are 2-parameter Weibull. The coefficient of variation of delayed load cycle and delayed crack length for the number of 10 overload applications data are 14.8 and 9.2%, respectively.

The specimens were either 0.9 or 2.0 mm thick. The size of the plastic zone at the crack tip due to the single applied overload was also determined using finite element analysis.
