Recently, patterned magnetic films and elements attract a wide interest due to their technological potentials in ultrahigh-density magnetic recording and spintronic devices. Among those patterned magnetic structures, magnetic anti-dot patterning induces a strong shape anisotropy in the film, which can control the magnetic properties such as coercivity, permeability, magnetization reversal process, and magneto-resistance. While majority of the previous works have been concentrated on anti-dot arrays with a single magnetic layer, there has been little work on multilayered anti-dot arrays. In this work, we report on study of the magnetic properties of bilayered anti-dot system consisting of upper perforated Co layer of 40 nm and lower continuous Ni layer of 5 nm thick, fabricated by photolithography and wet-etching processes. The magnetic hysteresis (M-H) loops were measured with a superconducting-quantum-interference-device (SQUID) magnetometer (Quantum Design: MPMS). For comparison, investigations on continuous Co thin film and single-layer Co anti-dot arrays were also performed. The magnetic-domain configuration has been measured by using a magnetic force microscope (PSIA: XE-100) equipped with magnetic tips (Nanosensors). An external electromagnet was employed while obtaining the MFM images. The MFM images revealed well-defined periodic domain networks which arise owing to the anisotropies such as magnetic uniaxial anisotropy, configurational anisotropy, etc. The inclusion of holes in a uniform magnetic film and the insertion of a uniform thin Ni layer, drastically affected the coercivity as compared with single Co anti-dot array, without severely affecting the saturation magnetization ($M_s$). The observed changes in the magnetic properties are closely related to the patterning that hinders the domain-wall motion as well as to the magneto-anisotropic bilayer structure.

This paper proposes a distributed space-frequency block code (SFBC) for relay-assisted single carrier frequency-domain equalization (SC-FDE). The proposed technique achieves spatial diversity gain over fast fading channels without the complexity of multiple antennas. The mobile equipment of the proposed system has a very simple transmitter structure with constant amplitude transmit sequences, which is desirable especially for uplink communications. In order to obtain spatial diversity, the transmit sequence of relay is efficiently generated in the time domain, which is equivalent to the SFBC. Further, efficient implementation of relay and destination structures is also presented. Extensive simulation results show that the proposed system significantly outperforms the distributed space-time block code (D-STBC) SC-FDE over fast fading channels.

$BK_{Ca}$ channels were suggested to contain one or more domains of the ‘regulator of K+ conductance’(RCK) in their cytosolic carboxyl termini (Jiang et al.2001). It was also shown that the RCK domain in mammalian $BK_{Ca}$ channels might sense the intracellular $Ca^{2+}$ with a low affinity (Xia et al. 2002). We aligned the amino acid sequence of the $\alpha$-subunit of rat $BK_{Ca}$ channels (rSlo) with known RCK domains and identified a second region exhibiting about 50% homology. This putative domain, RCK2, contains the characteristic amino acids conserved in other RCK domains. We wondered whether this second domain is involved in the domain-domain interaction and the gating response to intracellular $Ca^{2+}$ for rSlo channel, as revealed in the structure of RCK domain of E. coli channel (Jiang et al.2001). In order to examine the possibility, site-directed mutations were introduced into the RCK2 domain of rSlo channel and the mutant channels were expressed in Xenopus oocytes for functional studies. One of such mutation, G772D, in the putative nucleotide-binding domain resulted in the enhanced $Ca^{2+}$ sensitivity and the channel gating of rSlo channel. These results suggest that this region of $BK_{Ca}$ channels is important for the channel gating and may form an independent domain in the cytosolic region of $BK_{Ca}$ channels. In order to obtain the mechanistic insights of these results, G772 residue was randomly mutagenized by site-directed mutagenesis and total 17 different mutant channels were constructed. We are currently investigating these mutant channels by electrophysiological techniques.ical techniques.
Structural system identification (SSI) is an inverse problem of difficult solution. Currently, difficulties lie in the development of algorithms which can cater to large size problems. In this paper, a parameter estimation technique based on evolutionary strategy is presented to overcome some of the difficulties encountered in using the traditional system identification methods in terms of convergence. In this paper, a non-traditional form of system identification technique employing evolutionary algorithms is proposed. In order to improve the convergence characteristics, it is proposed to employ immune algorithms which are proved to be built with superior diversification mechanism than the conventional evolutionary algorithms and are being used for several practical complex optimisation problems. In order to reduce the number of design variables, domain decomposition methods are used, where the identification process of the entire structure is carried out in multiple stages rather than in single step. The domain decomposition based methods also help in limiting the number of sensors to be employed during dynamic testing of the structure to be identified, as the process of system identification is carried out in multiple stages. A fifteen storey framed structure, truss bridge and 40 m tall microwave tower are considered as a numerical examples to demonstrate the effectiveness of the domain decomposition based structural system identification technique using immune algorithm.

A novel methodology, referred to as Auxiliary Domain Method (ADM), allowing for a very efficient solution of nonlinear reliability problems is presented. The target nonlinear failure domain is first populated by samples generated with the help of a Markov Chain. Based on these samples an auxiliary failure domain (AFD), corresponding to an auxiliary reliability problem, is introduced. The criteria for selecting the AFD are discussed. The emphasis in this paper is on the selection of the auxiliary linear failure domain in the case where the original nonlinear reliability problem involves multiple objectives rather than a single objective. Each reliability objective is assumed to correspond to a particular response quantity not exceeding a corresponding threshold. Once the AFD has been specified the method proceeds with a modified subset simulation procedure where the first step involves the direct simulation of samples in the AFD, rather than standard Monte Carlo simulation as required in standard subset simulation. While the method is applicable to general nonlinear reliability problems herein the focus is on the calculation of the probability of failure of nonlinear dynamical systems subjected to Gaussian random excitations. The method is demonstrated through such a numerical example involving two reliability objectives and a very large number of random variables. It is found that ADM is very efficient and offers drastic improvements over standard subset simulation, especially when one deals with low probability failure events.

In this paper, we propose a high-resolution time-frequency domain reflectometry technique as a methodology of detection and estimation of faults on a wire. This method adopts the time-frequency cross correlation characteristics of the observed signal in both time and frequency domains simultaneously. The accuracy of the proposed method is verified with experiments using a RG type coaxial cable and comparing it with traditional time domain as well as frequency domain reflectometry methods. It is clearly shown here that the proposed algorithm produces excellent results compared to the conventional methods for single as well as multiple fault cables.

CY-007 and CY-049 pteridine glycosyltransferases (PGTs) that differ in sugar donor specificity to catalyze either glucose or xylose transfer to tetrahydrobiopterin were studied here to uncover the structural determinants necessary for the specificity. The importance of the C-terminal domain and its residues 218 and 258 that are different between the two PGTs was assessed via structure-guided domain swapping or single and dual amino acid substitutions. Catalytic activity and selectivity were altered in all the mutants (2 chimeric and 6 substitution) to accept both UDP-glucose and UDP-xylose. In addition, the wild type activities were improved 1.6-4.2 fold in 4 substitution mutants and activity was observed towards another substrate UDP-N-acetylglucosamine in all the substitution mutants from CY-007 PGT. The results strongly support essential role of the C-terminal domain and the two residues for catalysis as well as sugar donor specificity, bringing insight into the structural features of the PGTs.
We have investigated the magnetization reversal behavior in ferromagnetic $Co_{0.5}Fe_{0.5}$ alloy films using the magneto-optical Kerr microscope capable of the direct observation of time-resolved domain patterns. Interestingly enough, as the sample thickness increases the magnetization reversal behavior becomes changed from a single domain wall motion to the random nucleations of domains. Also, from the stochastic analysis of the domain jump sizes during the domain wall motion, it was found that the magnetization reversal behavior in the samples shows the critical scaling behavior with the critical exponent of ${\tau}{\sim}1.33$.

Microstrip patch antennas are analyzed by a multiresolution wavelet method. The spectral Green's dyad of the structure is obtained and its joint spatial-spectral domain representations are presented. Based on the joint spatial-spectral domain representation, we show that the spectral-domain wavelets are useful in the analysis of this problem. We obtain the matrix equations of the integral equations of this Green's dyad by using the method of moment(MoM), and efficiently solve the problem using the spectral domain wavelet transform concepts in conjuction with the conjugate gradient method. The results for a single-layered square patch are compared with those of conventional MoM and CG-FFT.
Cyanobacteriochromes (CBCRs) are phytochrome-related photoreceptor proteins in cyanobacteria and cover a wide spectral range from ultraviolet to far-red. A single GAF domain that they contain can bind bilin(s) autocatalytically via heterologous recombination and then fluoresce, with potential applications as biomarkers and biosensors. Here, we report that a novel red/green CBCR GAF domain, SPI1085g2 from Spirulina subsalsa, covalently binds both phycocyanobilin (PCB) and phycoerythrobilin (PEB). The PCB-binding GAF domain exhibited canonical red/green photoconversion with weak fluorescence emission. However, the PEB-binding GAF domain, SPI1085g2-PEB, exhibited an intense orange fluorescence (λabs.max = 520 nm, λfluor.max = 555 nm), with a fluorescence quantum yield close to 1.0. The fluorescence of SPI1085g2-PEB was selectively and instantaneously quenched by copper ions in a concentration-dependent manner and exhibited reversibility upon treatment with the metal chelator EDTA. This study identified a novel PEB-binding cyanobacteriochrome-based fluorescent protein with the highest quantum yield reported to date and suggests its potential as a biosensor for the rapid detection of copper ions.
