  • The characteristic and efficiency of natural shell sand in the coast of Cheju-Do and Jin-Do were studied to apply as agricultural lime for the soil acid adjustment. 1. The alkalinity of shell sand from Cheju coast was higher than that of Jindo and more weathered into fine particles. The particle size distributions of Cheju shell sand were condensed finer particles than 32 mesh in Cheju shell sands and in more coarser particles than 32 mesh in Jindo one. 2. The effect of Cheju shell sand on increasing soil pH value in the upland condition was low at the beginning but more gradually increased after 8 weeks from the treatment than ground lime, and Jindo was very dull during the period of treatment. The commercial lime crushed from Jindo was approximately equal to the ground lime. 3. In the submerged condition, the shell sand of Cheju reacted with soil acid more quickly than ground lime after 2 days, and Jindo was very slow. 4. The relative efficiency of various particle size fraction of the shell sand was superior to the ground lime. The lime particles between 9 to 14 mesh and 20 mesh had more remarkable difference of pH value than other fine particles. The efficiency among finer sizes than 20 mesh particle was approximately equal to each others. 5. The shell sand from the Cheju would be applied directly as agricultural lime without any treatment, and Jindo also can be expected to be effective as agricultural lime with crushing procedure.

Selective determination of mercury (II) ion in aqueous solution by chemiluminescence method (화학발광법에 의한 수용액 중의 선택적 수은(II) 이온 정량)

  • A selective determination method of mercury (II) ion in aqueous solution by luminol-based chemiluminescence system (luminol CL system) has been developed. Determination of metal ions such as copper (II), iron (III), chromium (III) ion in solution by the luminol CL system using its catalytic role in the reaction of luminol and hydrogen peroxide has been reported by several groups. In this study, the catalytic activity of mercury (II) ion in the reaction of luminol and hydrogen peroxide was observed by the enhanced CL intensity of the luminol CL system. Based on this phenomenon, experimental conditions of the luminol CL system were investigated and optimized to determine mercury (II) ion in aqueous solution. While mercury (II) ion in mixed sample solution containing mercury (I) and (II) ions highly enhanced the CL intensity of the luminol CL system, the mercury (I) ion could not enhanced the CL intensity. Thus selective determination of the mercury (II) ions in a mixture containing mercury (I) and (II) ions could be achieved. Each concentration of mercury (I) and (II) ions in aqueous solution can be obtained from the results of the CL method that give the concentration of only mercury (II) ion and the inductively coupled plasma (ICP) method that give the total concentration of mercury ions. On the optimized conditions, the calibration curve of mercury (II) ion was linear over the range from $1.25{\times}10^{-5}$ to $2.50{\times}10^{-3}M$ with correlation coefficient of 0.991. The detection limit of mercury (II) ion in aqueous solution was calculated to be $1.25{\times}10^{-7}M$.

Enhancement of Anticancer Activity of Acer mono by High Pressure Extraction Process (고로쇠 수피 초고압 추출물의 항암활성 증진)

  • We investigated a method to improve anticancer activities of Acer mono by ultra high pressure extraction process. The extract yields by ultra high pressure were 9.49% and 9.87% for 5 min and 15 min processing time, respectively, which were relatively higher than 3$\sim$4% of conventional extraction processes due to their resid bark structure. The extract for 15 minutes extraction (HPE15) showed higher potent scavenging effect as 94.56% than the control, BHA as 93.24%. On SOD-like test, HPE15 also showed the highest activity as 38.6% at 1.0 mg/mL concentration. The cytotoxicity of HPE15 on normal human lung and kidney cell were below 23.54% in adding 1.0 mg/mL. Generally, human cancer cell growth stomach adenocarcinoma (AGS), lung adenocarcinoma (A549), breast adenocarcinoma (MCF-7), colon adenocarcinoma (Caco-2) and liver adenocarcinoma (Hep3B) were inhibited up to 75% with higher selectivity of above 4.0. High antioxidant activity of HPE15 resulted in high anticancer activity, and its activity was also due to higher yields of Acer mono by ultra high pressure extraction process. It was also proved by HPLC comparison analysis.

Expression of RUNX3 in Human Gastric Cancer (위암에서 RUNX3 발현의 임상적 의의)

  • Purpose: RUNX3, a novel tumor suppressor, is frequently inactivated in gastric cancer. In the present study, we examined the pattern of RUNX3 expression in gastric cancer cells from gastric cancer specimens and the impact of its alteration on clinical outcome. Materials and Methods: A total of 124 samples of both gastric cancer and normal tissue were obtained from 124 patients who underwent curative gastrectomy at the Seoul Medical Center from January 2001 to December 2005. RUNX3 expression was determined by immunohistochemical staining, and the results were analyzed. Statistical analysis wabased on clinicopathological findings and differences in survival rates. Results: The mean age of the patients was 61 years, and the male:female ratio was 1.9:1. The expression rate of RUNX3 was 59.7% (74/124). The expression rate was higher in differentiated gastric cancers (nucleus: 9.1%, cytoplasm: 57.6%) than in the undifferentiated types (nucleus: 5.2%, cytoplasm: 46.6%) (P=0.133). The 5-year survival rates according to RUNX3 expression determined from cancer tissue were 88.9% for the nucleus $\pm$ cytoplasm(+) group of patients, 76.1% for the cytoplasm only (+) group of patients, and 65.3% for the RUNX3 negative expression group of patients (P=0.626). Only UICC TNM staging showed statistical significance related to the survival rate, as determined by multivariate analysis. Conclusion: The RUNX3 expression rate was higher in differentiated gastric cancer than in the undifferentiated types without significance. Although RUNX3 expression predicted better survival, based on multivariate analysis, the finding was not statistically significant. More cases should be further evaluated.

Effects of Steam Flaking on In situ DM Digestibility and Aflatoxin and Ochratoxin Contents during Storage of Corns (옥수수의 steam flake 처리가 in situ 건물소화율 및 저장기간별 aflatoxin과 ochratoxin 함량에 미치는 영향)

  • This study was conducted to investigate the effects of steam flaking of corn grains on in situ dry matter degradability in the rumen and contents of mycotoxins (aflatoxin, ochratoxin) during storage. Yellow dent corns imported from USA and India were flaked, and stored for 8 weeks under the standard temperature and pressure (STP; $25^{\circ}C$/ 1 atm.). Experimental treatments were composed of four corn grains (untreated-USA corn, USCW; steam flaked-USA corn, USCF; untreated-India corn, IDCW; steam flaked-India corn, IDCF) with 4 replications according to 9 storage periods (0, 1, 2, 3, 4, 5, 6, 7, 8 week). Two ruminally cannulated Holstein bulls were used for in situ trial. Pathogen contamination trial was performed by comparing the mycotoxin contents in corns during storage periods. Dry matter disappearance rate in the rumen was about 3.0 to 44.1% higher (P<0.05) for USCW than IDCW, but was not difference between USCF and IDCF. With steam flaking of corn, dry matter degradability in the rumen was significantly (P<0.05) increased in corn from India, but was not affected in corn from USA. Aflatoxin content was very low level in corns from USA and steam flaked corns, but was higher than the tolerance limit of domestic aflatoxin content regulation when IDCW was stored over 6 week under STP. Ochratoxin content was low level in all treatments. From above results, it is reasonable that the corn imported from India might be flaked for enhance the ruminal DM degradability and safe from aflatoxin pathogen.

Movement of Cold Water Mass in the Northern East China Sea in Summer (하계 동중국해 북부 해역에서 저층 냉수괴의 거동)

  • The Yellow Sea Cold Water (YSCW) is formed by cold and dry wind in the previous winter, and is known to spread southward along the central trough of the Yellow Sea in summer. Water characteristics of the YSCW and its movement in the northern East China Sea (ECS) are investigated by analyzing CTD (conductivity-Temperature-Depth) data collected from summertime hydrographic surveys between 2003 and 2009. By water mass analysis, we newly define the North Western Cold Water (NWCW) as a cold water mass observed in the study area. It is characterized by temperature below $13.2^{\circ}C$, salinity of 32.6~33.7 psu, and density (${\sigma}_t$) of 24.7~25.5. The NWCW appears to flow southward at about a speed less than 2 cm/s according to the geostrophic calculation. The newly defined NWCW shows an interannual variation in the range of temperature and occupied area, which is in close relation with the sea surface temperature (SST) over the Yellow Sea and the East China Sea in the previous winter season. The winter SST is determined by winter air temperature, which shows a high correlation with the winter-mean Arctic Oscillation (AO) index. The negative winter-mean AO causes the low winter SST over the Yellow Sea and the East China Sea, resulting in the summertime expansion and lower temperature of the NWCW in the study area. This study shows a dynamic relation among the winter-mean AO index, SST, and NWCW, which helps to predict the movement of NWCW in the northern ECS in summer.

Anticancer and Immune Activities of the Extracts from Amorpha fruticosa L. (족제비싸리 부위별 추출물의 항암 및 면역활성)

  • This study was performed to experiment about useful biological activities of the parts of the extracts from Amorpha fruticosa LINNE. Experimental studies were progressed through the anticancer activities and immune activities such as cell cytotoxicity, inhibition activities of cell growth, cell growth of human B and T cell, productivity of cytokines and natural killer cell activities. The cell cytotoxicity using human Lung normal cell (HEL299) was showed cytotoxicity of below 22% by extracts of Amorpha fruticosa L. in 1.0 g/l concentration. The anticancer activities were increased in over 70% by roots extracts of Amorpha fruticosa L. in A549 and AGS cells. The immune cell growth using human immune B and T cells was increased against control by barks extracts of Amorpha fruticosa L. The secretion of cytokines $(IL-6,\;TNF-{\alpha})$ from human immune B and T cells was showed secretion of the amount of cytokines by roots extracts of Amorpha fruticosa L. Also NK cell growth was increased against control all of the extracts of Amorpha fruticosa L. From the results, the roots and barks extracts of Amorpha fruticosa L. were showed useful biological activities.

Effects of Pine Needle Extract Oil on Blood Glucose and Serum Insulin Levels in db/db Mice (2형 당뇨 마우스에서 솔잎 추출용액의 경구투여가 혈당조절에 미치는 영향)

  • This study was performed to investigate the hypoglycemic effect of pine needle extract oil against type 2 diabetes. Six-week-old male C57BL/Ks(db/db) mice were divided into four groups : negative control, pine needle extract oil low dose, high dose and positive control groups, which fed daily for 6 weeks with corn oil, pine needle extract oil 112.5 mg/kg, 450 mg/kg or metformin (150 mg/kg ), respectively. The oral administration of the pine needle extract oil resulted in the significant and dose-dependent decreases of blood glucose levels in comparison with corn oil treatment. The levels of HbAlc showed a tendency of the decrease by the high dose treatment of the pine needle extract oil and were positively correlated with blood glucose levels (r=0.5046, p=0.0023) . However, the levels of serum insulin and C-peptide were not affected by pine needle extract oil or metformin treatments. The levels of serum leptin, which is related with the insulin sensitivity, showed a tendency of the increases by pine needle extract oil treatment and were negatively correlated to blood glucose levels (r=-0.4754, p=0.0052). In conclusion, these results suggest that the pine needle extract oil have a potential for the oral anti-hyperglycemic agent and the mode of action may be related with the improvement of the insulin sensitivity through blood leptin.

Studies on Pollutant Concentrations in Ground Water and Their Behavior in Soils , Cheju Island. I. Changes in Nitrate-nitrogen Concentration in Ground-water near Hotel complexes and Business district During the Period form 1987 to 1992 (제주도 지하수중 오염물질의 농도와 토양중 그의 행동에 관한 연구 1. 호텔 및 상가주변에서의 지하수중 $NO_3-N$의 농도 변화 (1987-1992))

  • This study was conducted to investigate the degree of ground water pollution used for drinking water in Cheju Island. Samples were collected monthly from 31 wells of 10 divided areas and were analyzed for $NO_3-N$, pH, $SO_4$, Cl and hardness from 1987 to 1992. $NO_3-N$ concentrations in the samples, with the exception of sample No.23, did not exceed the standard concentration of drinking-water(10mg/l). $NO_3-N$ concentrations at area 10, unpolluted area, ranged from <1.0 mg/l to just over 1.2 mg/l. However, samples from the business district of the area 8 showed the concentrations ranging from 5 mg/l in 1987 to 8.l mg/l in 1992 with a mean of 6.8 mg/l, about 5 times higher than those from the area 10. $NO_3-N$ concentration in sample No. 23 increased from 4 mg/l in 1987 to 12.6 mg/l in 1991. Average rate of increase in $NO_3-N$ in samples of No. 1, 2, 4, and 8 at area 8 was about 1.2mg/l per year in the study period. The ground water at area 1, 2, 3, 4, 5, 6, and 7 showed in the range of pH 7 to 8.1, being similar to area 10. However, pH at area 8 was in the range of 6.6 to 7.3, being lower than that in the other areas. Hardness at area 1, 2, 3, 4, 5, and 6 were in the range of 30 to 80 mg/l, being higher than that at area 10. Those at area 8 were the highest among all the other areas tested. The results of this study suggest that $NO_3-N$ contamination in ground water could be a problem at hotel complexes and business district in Cheju Island.

  • Fenoxycarb, pyriproxyfen and methoprene are juvenile hormone mimic insecticide. These insecticides have been widely used for mosquito, fly, scale insects, and Lepidoptera. The purpose of this study was to develop a simultaneous determination procedure of fenoxycarb, pyriproxyfen and methoprene residues in crops using HPLC-UVD/MS. These insecticide residues were extracted with acetone from representative samples of four raw products which comprised brown rice, apple, green pepper, and Chinese cabbage. The extract was diluted with saline water, and then n-hexane/dichloromethane partition was followed to recover these insecticides from the aqueous phase. Florisil column chromatography was additionally employed for final clean up of the extract. The analytes were quantitated by HPLC-UVD/MS, using a $C_{18}$ column. The crops were fortified with each insecticide at 3 levels per crop. Mean recovery ratios were ranged from 80.0 to 104.3% in four representative agricultural commodities. The coefficients of variation were less than 4.8%. Quantitative limit of fenoxycarb, pyriproxyfen, and methoprene was 0.04 mg/kg in crop samples. A HPLC-UVD/MS with selected-ion monitoring was also provided to confirm the suspected residues. The proposed simultaneous analysis method was reproducible and sensitive enough to determine the residues of fenoxycarb, pyriproxyfen and methoprene in the agricultural commodities.
