고온 및 저온처리에 었어서 가잠 Virus 다각체병의 유발에 대하여 (studies on the induction of nuclear and cytoplasmic polyhedrosis viruses by high and low temperature treatment in the silkworm, Bombsx mori L.)

  • 가잠의 Virus다각체의 저온 및 고온처리에 대한 유발현상을 조사하기 위하여 5령기잠을 공시하고 저온(5$^{\circ}C$, 24hrs.), 고온(Hot water bath 에 4$0^{\circ}C$, 5min으로 침지 및 Oven 내에 4$0^{\circ}C$, 30min.으로 단독 및 복합처리하여 핵다각체병(N) 중장형 다각체병(C), 병발형(NC) 유발률을 조사하고 동시에 핵다각체병(N)과 중장형다각체병(C)대하여 Giemsa solution 염색법에 의한 염색성을 조사하였다. 1. 냉장(5$^{\circ}C$, 24 시간) 처리와 고온(Hot water bath, 4$0^{\circ}C$, 5분간) 처리를 한 핵복다각체병(N)은 중장형다각체병(C)보다 그 유발률이 적고 또 복합처리에 있어서도 중장형다각테병(C)의 유발율은 핵다각체병(N)보다 많다. 2. 냉장처리후 고온(4$0^{\circ}C$, 30분간 건열) 처리, 반대로 고온처리후 냉장처리한 Virus다각체의 유발률도 중장형다각체병(C) 보다 핵형다각예병(N)의 유발현상이 현저하게 많이 나타났다. 3. Giemsa solution 염색법에 의한 핵다각체병(N)의 다각체는 염색이 되지 않으나 중장형다각예병(C)의 다각체는 잘 염색되어 염색성에 의하여 구별할 수 있었다.

Evolution of a Productive Bivoltine Hybrid of Silkworm Bombyx mori L.(SD7${\times}$SD12) with Shorter Larval Duration

  • With an objective of evolving quantitatively and qualitatively superior bivolitine silkworm hybrid of Bombyxmori L. for tropics with shorter larval duration without compromising on productivity traits, a breeding programme was initiated at this institute during 1997 by utilizing breeding resource material from the institutes Germplasm collection. The breed SD7 is characterized with plain larvae spinning white oval cocoons and SD12 is characterized by sex-limited larval markings spinning white dumb-bell cocoons. After fixation, these breeds along with other newly evolved breeds were subjected for hybrid evaluation in the laboratory. Based on the hybrid studies, the hybrid SD7${\times}$SD12 was selected and evaluated for one year comprising three major sea- sons (6 trials) of tropics viz., Pre-monsoon (characterized by high temperature, low humidity and without any rain fall), Monsoon (characterized by moderate temperature, heavy rain fall with high humidity) and post-monsoon season (characterized by low temperature and low humidity with frequent rain fall) to know the fluctuations in the larval duration. The evaluation studies indicated that the hybrid is having shorter larval duration with productive merits. The hybrid SD7${\times}$SD12 recorded survival percent of 92.74, shell weight of 46.7 cg, shell ratio % of 24.0 and filament length of 1,200 meters with a shorter larval duration of 24 hrs; when compared to productive hybrid CSR2${\times}$CSR5(control). The breeding methodologies and hybrids usefulness for tropics were discussed.

Molecular characterization and inhibition analysis of the acetylcholinesterase gene from the silkworm maggot, Exorista sorbillans

  • Several organophosphorus (OP) insecticides can selectively kill the silkworm maggot, Exorista sorbillans (Es) (Diptera: Tachinidae), while not obviously affecting the host (Bombyx mori) larvae, but the mechanism is not yet clear. In this study, the cDNA encoding an acetylcholinesterase (AChE) from the field Es was isolated. One point mutation (Gly353Ala) was identified. The Es-353G AChE and Es-353A AChE were expressed in baculovirus-insect cell system, respectively. The inhibition results showed that for eserine and Chlorpyrifos, Es-353A AChE was significantly less sensitive than Es-353G AChE. Meanwhile, comparison of the I50 values of eserine, dichlorvos, Chlorpyrifos and omethoate of recombinant Es AChEs with its host (Bombyx mori) AChEs indicated that, both Es AChEs are more sensitive than B. mori AChEs. The results give an insight of the mechanism that some OP insecticides can selectively kills Es while without distinct effect on its host, B. mori.

Molecular Cloning of a cDNA Encoding Putative Apolipophorin from the Silkworm, Bombyx mori

  • ApolipophorinIII (apoLp-III) is a protypical exchangeable apolipoprotein that is abundant in hemolymph of many insect species. Its function lies in the stabilization of low-density lipophorin particles (LDLp) crossing the hemocoel in phases of high energy consumption to deliver lipids from the fat body to the flight muscle cells. But, recent studies with naive Galleria mellonella-apoLp-III gave first indication of an unexpected role of that protein in insect immune activation. In this research, we cloned a cDNA encoding putative apoLp-III from the silkworm, Bombyx mori injected with E. coli and characterized its role. We constructed a cDNA library using whole bodies of B. mori larvae injected with E. coli, carried out the differential screening, and selected the up-regulated clones. Among these clones, we focused on a cDNA showing a high sequence similarity to the apolipophorinIII from other insects and analyzed the nucleotide and deduced amino acid sequences. The pupative B. mori Jam123 apoLp-III cDNA contained 1,131 bp encoding 186 amino acid residues. Phylogenetic analysis revealed that the nucleotide and amino acid sequences of the B. mori apoLp-III cDNA formed a highly inclusive subgroup with Bombycidae. But, it was interesting that B. mori Jam123 is closer to B. mandarina than B. mori P50 and B. mori N4. Northern blot analysis showed a signal in the fat body, posterior silkgland and midgut.

Breeding of New Productive Bivoltine Hybrid, CSR12 $\times$ CSR6 of Silkworm Bombyx mori L.

  • With an objective of evolving quantitatively and qualitatively superior bivoltine breeds/hybrids of silkworm Bombyx mori L. for tropical conditions, breeding work was initiated in Central Sericultural Research and Training Institute, Mysore during 1992 by utilizing two Japanese hybrids namely BNl8$\times$BCS25 and Shunrei$\times$Shogetsu along with Indian evolved breed, KA. The breed CSR12 which is characterized with sex-limited larval marking and white oval cocoons was evolved from the Japanese hybrid BNl8 ${\times}$ BCS25 by crossing with KA, while the breed CSR6 which is characterized with normal marking (marked larvae) and white dumbbell cocoons was extracted from the Japanese commercial hybrid Shunrei${\times}$Shogetsu through continuous inbreeding coupled with selection. After fixation, these breeds along with other newly evolved breeds were subjected to hybrid study under optimum environmental conditions in the laboratory for expression of full potential of the genotypes. These hybrids were evaluated by Multiple Trait Evaluation Index (Mano et al., 1993). The hybrid CSR12${\times}$CSR6 was selected based multiple trait evaluation index value. The hybrid CSR12$\times$CSR6 recorded survival of 96.0%, shell weight of 50.0 cg, shell ratio of 24.3%, raw silk percentage of 19.6, filament length of 1,216 m, boil off loss of 22.4% and renditta of 5.1. On the other hand, the control hybrid (KA ${\times}$ NB4D2) has recorded survival of 90.6%, shell weight of 42.1 cg, shell ratio of 20.4%, raw silk percentage of 15.9, filament length of 999 m, boil off loss of 24.8% and renditta of 6.3. The hybrid CSR12$\times$CSR6 was authorized during 1997 by Central Silk Board, Government of India for commercial exploitation during favourable months based on national level race authorization test.

  • 누에의 5령 60시간째 합성효약 홀몬(JHA)을 체중 1g당 1$\mu\textrm{g}$, 10$\mu\textrm{g}$을 경피투여한 후 체액 중 단백질 및 아미노산의 변동을 조사하였던 바 그 결과는 다음과 같다. 1. 5령유충기간은 대조구보다 JHA-1구가 1일, JHA-10구가 4일 연장되었다. 2. 견종 및 견층종은 합성효약 홀몬 투여구가 대조구보다 무거웠고, 견층비율은 JHA-10구가 낮았다. 3. 체액단백질의 농도는 합성효약홀몬투여구가 대조구에 비해 현저하게 증가하였다. 4. 체액 중 전 아미노산양은 JHA-10구가 대조구에 비해 약 2배 정도 많았으며, 특히 glycine와 arginine의 증가 정도는 현저하다.

실내 인공사료육에 의한 뽕나무하늘소(Apriona germari Hope) 유충의 발육 (Larval Development of Mulberry Longicorn Beetle, Apriona germari Hope, on the Artificial Diet)

  • 실내 인공사료육에 의한 뽕나무하늘소(Apriona germari Hope) 유충의 발육특성을 조사하기 위하여 야외 뽕밭에서 채집된 뽕나무하늘소 부화유충을 14L:10D 광조건의 $25^{\circ}C$ 항온기에서 애누에용 인공사료와 뽕나무 가지 분말을 동량비로 석은 인공사료에 의하여 실내사육한 결과, 유충의 1령에서 12령까지의 두폭 범위는 0.12~0.69 cm 내에 있었고 각 영기의 성장비는 1령과 2령간이 가장 컸다. 체중의 증가에 있어서 1령에 비해 8령의 경우는 약 176배, 12령의 경우와는 약 354배의 성장도 차이를 보였다. 각 영별 경과일구는 영이 진전될수록 경과일수가 길어졌으며 1령에서서 9령까지의 경과일수는 186.03일, 12령까지의 경과일수는 304.58일이었다. 인공사료육에 의한 1세대 유충 생존율은 3령기에 걸쳐 약 45%로 매우 낮았으나 그 이후 안정되어 8령까지의 유충 생존율은 40.8%이었다. 용화율은 32.4% 이었으며, 용화 시기는 7령에서부터 11령에 걸쳐 나타났지만, 8령과 9령째가 대부분이었다.

바이러스성 건화병의 고온에 의한 발병역제 효과 (Inhibitory Effect of High Temperature on the Multiplication of Flacherie Virus in the Silkworm, Bombyx mori L.)

  • 고도(37$^{\circ}C$)처리에 의한 바이러스성 건연병의 발병억제 효과를 검정한 결과는 다음과 같다. 1. 고온처리 시기는 5령기잠 또는 4.5령기잠이 가장 알맞았다. 2. 고온처리 기간은 12-24시간이 적절했다. 3. 2령기잠에 바이러스를 접종하고 5령기잠 또는 4,5령기잠에 각각 12 및 24시간 고온처리 했을 때, 생존율은 고온 무처리 대비 지수로 33.8-39.5%가 높았고, 1만두 수견량 24.2-32.0%은 높았다. 4. 3령기잠에 바이러스를 접종하고 위와 같은 방법으로 고온처리 했을 때, 생존율은 무처리 대비 지수로 53.6-70.0%, 1만두 수견량은 45.8-54.2%가 높았다. 5. 4령기잠에 바이러스 접종 후 5령기잠에 12 및 24시간 고온처리 했을 때는 무처리 대비 지수로 생존율은 49.7-68.9%, 1만기 수견량은 44.3-69.0%이 높았다.

  • Bacillus thuringiensis를 한국의 토양에서 분리하여 특성을 연구했다. 8균주를 분리하여 HL-24, HL-25, HL-33, HL-34, HL-35, HL-38, HL-39와 HL-40으로 명명했으며. HL-24와 HL-25는 부정형의 crystal를 가졌고, HL-33과 HL-35는 이중피라미드형, HL-34. HL-38, HL-39와 HL-40은 둥근형의 crystal를 가진 것을 위상차현미경으로 관찰했다. 분리균들의 생화학적인 특성은 이미 알려진 균주들과 유사했으나. 특이한 차이점을 가지고 있었다. HL-25, HL-33과 HL-34는 cephalothin, colistin과 penicillin G에 내성을, HL-33과 HL-35균주는 chloramphenicol에 내성을 HL-39와 HL-40은 penicillin G에 내성을 나타냈다. 균주HL-24, HL-25, HL-33과 HL-34는 누에 유충에 대한 치사성은 100.85.55. 및 70%를 각각 나타냈으며 HL-24, HL-25, HL-38, HL-39와 HL-40의 모기 유충에 대한 치사성은 10. 20. 100. 100 및 90%였다. HL-24와 HL-25균주는 누에와 모기 유충에 모두 치사성은 나타냈으나. 모기에는 살충효과가 10~20%로 매우 낮았다.

Manifestation of Hybrid Vigour and Combining Ability in Polyvoltine $\times$ Bivoltine Hybrids of Silkworm, Bombyx mori L.

  • The general and specific combining abilities and hybrid vigour of seven polyvoltine breeds [BL24, BL62, BL6S, BL67, BL68, BL69, and Pure Mysore (PM)] with five bivoltine breeds (NB4D2, CSR2, CSR5, CSR18 and CSR19) were evaluated in a line ${times}$ tester design by analysing six quantitative traits viz., fecundity, yield 110,000 larvae both by number and weight, cocoon weight, shell weight and cocoon shell ratio. ANOVA estimates showed significant effects for all the traits indicating the role of both additive and non- additive gene actions. Estimation of GCA revealed that the breed BL67 exhibited significant positive effects toy all the six traits followed by BL68 for four important economic traits viz., fecundity, cocoon weight, shell weight and cocoon shell ratio. Among testers, CSR5 showed significant GCA effects for five traits whereas CSR2 exhibited for shell weight and shell ratio. Three hybrids viz., BL67${times}$ NB4D2, BL67 ${times}$CSR5 and BL67${times}$ CSR19 excelled in four quantitative traits for SCA. Majority of the hybrids expressed positive hybrid vigour for cocoon yield/10,000 larvae by weight, cocoon weight and shell weight. Four hybrids namely PM ${times}$ NB4D2, BL67 ${times}$NB4D2, BL67${times}$ CSR5 and BL67${times}$ CSR19 manifesting positive hybrid vigour for five economic traits were adjudicated as best Heterotic hybrids and recommended for commercial exploitation to increase the silk yield.
