Interspecific crosses between rapeseed (Brassica napus L.) and chinese cabbage (B. campestris var. pekinensis Makino) and turnip (B. rapa L.) were made in order to examine the cross possibility and morphological phenotype of $F_1$ hybrid. The growth of pollen tube of cross between rapeseed and chinese cabbage was more rapid than cross between rapeseed and turnip. Silique formation and seed setting in silique were relatively high in the cross between rapeseed and chinese cabbage. The percentage of silique set from interspecific cross between rapeseed and chinese cabbage was 90.6% and higher 23.3% than the percentage of silique from interspecific cross rapeseed and turnip. The average number of seed per silique obtained from the cross rapeseed and chinese cabbage, and rapeseed and turnip reached 15.5 and 11.6, respectively. The morphological phenotypes of $F_1$ hybrid plants obtained from seeds in the cross between rapeseed and chinese cabbage resembled rapeseed mainly, but leaf length and leaf width were increased. The size, shape and lobation of leaves of $F_1$ hybrid plants from interspecific cross between rapeseed and turnip were intermediated between their parent species, but color of leaves was dark-green.
To develop new varieties of rapeseed (Brassica napus L.), the seeds of three varieties 'Naehan', 'Tammi', and 'Halla' were treated with proton ion beams and gamma rays (0~2,000 Gy), and then the characteristics of the mutants induced were examined up to $M_5$ generation to select the lines with fixed useful traits. In $M_5$ generation, we had selected several lines that were highly fixed for some useful traits such as plant height, maturity and flower size; one line with both earlier maturity and shorter stem than wild type, one line with only earlier maturity, two lines with shorter stem, one line with large flower, and one line with chlorophyll mutation. Among them, NP600-1-1-198-2 (induced from variety 'Naehan' was treated with proton ion beams 600 Gy) was superior for its distinction from the original variety, uniformity and stability. The unique characteristics of NP600-1-1-198-2 were dark green leaves, green stem, yellow flower, and black seed coat. Its flowering date was April 14, eight days earlier than its original variety, while seed maturity date was June 16 (five days earlier) and plant height 105 cm (shorter by 10 cm). NP600-1-1-198-2 has 52 silique per panicle, 6.2 cm silique length, 23 seeds per silique, 4.2 g per 1000 seeds and 45.9% oil content. The seed oil contained 67.8% of oleic acid, 16.7% of linoleic acid and 7.3% of linolenic acid but no erucic acid.
A total of 447 accessions consisting of seven Brassica spp.; Brassica carinata (34), B. juncea (199), B. rapa subsp. dichotoma (18), B. rapa. subsp. oleifera (14), B. rapa subsp. rapa (36), B. rapa subsp. trilocularis (56) and B. alba subsp. alba (90) were studied for their morphological characters and fatty acid compositions. There was a wide variation for morphological traits, oil content and fatty acid composition among Brassica species. Seed number/silique and yield/plant were varied from 4.2 (B. alba) to 25.1 (B. rapa subsp. trilocularis) and from 170.7 g (B. rapa subsp. oleifera) to 351.9 g (B. juncea L. Czern.), respectively. Among Brassica species, B. rapa subsp. trilocularis exhibited the highest oil (29.2%), stearic (20.4%) and erucic acid (45.3%) content. B. carinata had the highest content of palmitic (5.2%), oleic (21.2%) and linolenic acid (11.1%). B. rapa subsp. dichotoma and B. rapa subsp. oleifera exhibited the highest content of linoleic (8.1%) and behenic (26.9%) acid, respectively. B. rapa subsp. trilocularis exhibited the highest (45.3%) erucic acid content and significant positive relationship was observed between oleic acid and linoleic acid. This variation of agronomic and fatty acid compositions in Brassica species can be utilized to develop new varieties.

The cultivation area of rapeseed (Brassica napus L.) has been increased for oil production and landscaping purpose in Korea. However, as the color of rapeseed flower is very simple, diversified flower color is necessary to improve landscape effect. Interspecific and intergeneric crosses between rapeseed (Brassica napus) and three Cruciferae crops were performed in order to grow diverse flower color of rapeseed. The silique formation rate of interspecific cross rapeseed with cabbage (B. oleracea L) was relatively high (65.8%) and higher than intergeneric cross with rapeseed and radish (Raphanus sativus L.), rapeseed and Orychophragmus, respectively. During silique developing period after artificial pollination, there were many siliques without seeds due to the failure of fertilization. The average number of seed per silique obtained from cross rapeseed and cabbage, rapeseed and radish, rapeseed and O. violaceus were 0.12, 0.4 and 0.12, respectively. The phenotypes of $F_1$ hybrid plants from cross rapeseed and Cruciferae crops were mostly similar to maternal line, but leaf length and leaf width were increased. The interspecific cross of rapeseed and cabbage generated ivory color of flower which is the medium color of parents, and intergeneric cross of rapeseed and O. violaceus created entities with larger flowers which seems to enhance landscape effect. The fatty acid composition of most hybrid seeds intermediated of the two parents for oleic acid, linoleic acid and linolenic acid, content. Whereas hybrid of rapeseed and radish produced less erucic acid than radish parent.

This study was carried out to understand the effect of sowing date on growth characteristics, flower duration of a total of 6 domestic rapeseed cultivars, "Sunmang", "Tammiyuchae", "Tamlayuchae", "Naehanyuchae", "Yongsanyuchae" and "Hallayuchae" with different sowing dates (25 Sept., 5 Oct., 15 Oct., 25 Oct., 5 Nov.) in Muan, Jeollanamdo, Republic of Korea. The results obtained were summarized as the follows: The number of seedling stand after overwintering was decreased in all 6 cultivars. It changed little until 5 Oct., and then showed continuously largely decrease as the sowing date being delayed. As the sowing date was being delayed, plant height was decreased while Silique length and thousand seed weight were increased. Seed yield in all 6 cultivars decreased almost linearly with late sowing date. In correlation analysis between the delaying sowing date and growth characteristics, it was inversely correlated with plant height ($r=-0.769^{**}$), No. of branches/plant ($r=-0.760^{**}$), No. of siliqua/panicle ($r=-0.631^{**}$) and seed yield ($r=-0.946^{**}$), while showed a positive correlation with silique length (r=0.635), seed diameter ($r=0.629^{**}$) and thousand grain weight ($r=0.422^*$). No. of seeds/silique and seed set percentage were not significantly correlated with the delaying sowing date. The flower duration was long in order of Sunmang, Tammiyuchae, Yongsanyuchae, Naehanyuchae, Hallayuchae and Tamlayuchae. Varietal variation of flowering date was larger with early sowing date than with delaying sowing date. The range of flower duration across all cultivars was from 1 day of Tamlayuchae to 14 days of Sunmang according to the sowing date, and the end flowering date was able to be extended from 2 to 9 days, compared to normal date of end flowering.
The Arabidopsis gene AtLEC (At3g15356) gene encodes a putative 30-kDa protein with a legume lectin-like domain. Likely to classic legume lectin family of genes, AtLEC is expressed in rosette leaves, primary inflorescences, and roots, as observed in Northern blot analysis. The accumulation of AtLEC transcript is induced very rapidly, within 30 min, by chitin, a fungal wall-derived oligosaccharide elictor of the plant defense response. Transgenic Arabidopsis carrying an AtLEC promoter-driven ${\beta}$-glucuronidase (GUS) construct exhibited GUS activity in the leaf veins, secondary inflorescences, carpel heads, and silique receptacles, in which no expression could be seen in Northern blot analysis. This observation suggests that AtLEC expression is induced transiently and locally during developmental processes in the absence of an external signal such as chitin. In addition, mechanically wounded sites showed strong GUS activity, indicating that the AtLEC promoter responds to jasmonate. Indeed, methyl jasmonate and ethylene exposure induced AtLEC expression within 3-6 h. Thus, the gene appears to play a role in the jasmonate-/ethylene-responsive, in addition to the chitin-elicited, defense responses. However, chitin-induced AtLEC expression was also observed in jasmonate-insensitive (coi1) and ethylene-insensitive (etr1-1) Arabidopsis mutants. Thus, it appears that chitin promotes AtLEC expression via a jasmonate- and/or ethylene-independent pathway.
Two naturalized species, Chaerophyllum tainturieri Hook. (Umbelliferae) and Barbarea verna (Mill.) Asch. (Cruciferae) were newly reported from South Korea. C. tainturieri., 'Jeon-Ho-A-Jae-Bi', was found at World Cup park in Sangam-dong, Mapo-gu, in Seoul, and the genus Chaerophyllum is reported for the first time from South Korea in the present paper. It is distinguished from the genus Sphallerocarpus by having cylindrical fruit, and each furrow with one vitta. B. verna, 'Bom-Na-Do-Naeng-I', was found on a riverside area of Suyeong River, Geumsa-dong, in Busan. It can be distinguished from B. orthoceras by silique 4-8 mm long , seeds 1-2 mm long, and basal rosette leaves with a 4-11 large pairs of lateral lobes.
Background: The patatin-related phospholipase AIII family (pPLAIIIs) genes alter cell elongation and cell wall composition in Arabidopsis and rice plant, suggesting diverse commercial purposes of the economically important medicinal ginseng plant. Herein, we show the functional characterization of a ginseng pPLAIII gene for the first time and discuss its potential applications. Methods: pPLAIIIs were identified from ginseng expressed sequence tag clones and further confirmed by search against ginseng database and polymerase chain reaction. A clone showing the highest homology with pPLAIIIβ was shown to be overexpressed in Arabidopsis using Agrobacterium. Quantitative polymerase chain reaction was performed to analyze ginseng pPLAIIIβ expression. Phenotypes were observed using a low-vacuum scanning electron microscope. Lignin was stained using phloroglucinol and quantified using acetyl bromide. Results: The PgpPLAIIIβ transcripts were observed in all organs of 2-year-old ginseng. Overexpression of ginseng pPLAIIIβ (PgpPLAIIIβ-OE) in Arabidopsis resulted in small and stunted plants. It shortened the trichomes and decreased trichome number, indicating defects in cell polarity. Furthermore, OE lines exhibited enlarged seeds with less number per silique. The YUCCA9 gene was downregulated in the OE lines, which is reported to be associated with lignification. Accordingly, lignin was stained less in the OE lines, and the expression of two transcription factors related to lignin biosynthesis was also decreased significantly. Conclusion: Overexpression of pPLAIIIβ retarded cell elongation in all the tested organs except seeds, which were longer and thicker than those of the controls. Shorter root length is related to auxinresponsive genes, and its stunted phenotype showed decreased lignin content.
A plant-specific B3 domain and AP2 domain-containing transcription factor, RAV1 acts as a negative regulator of growth in many plant species and its transcription was down-regulated by BR and ABA. In this study, we found that RAV1-overexpressing transgenic plants showed abnormally developed ovules, resulting in reduced seed size, weight, and number in a silique. Interestingly, the endogenous expression of RAV1 fluctuated during seed development; it remained low during the early stage of seed development and sharply increased in the seed maturation stage. In plants, seed development is a complex process that requires coordinated growth of the embryo, endosperm, and maternal integuments. Among many genes that are associated with endosperm proliferation and embryo development, three genes consisting of SHB1, MINI3, and IKU2 form a small unit positively regulating this process, and their expression was regulated by BR and ABA. Using the floral stage-specific RNAs, we found that the expression of MINI3 and IKU2, the two downstream genes of the SHB1-MINI3-IKU2 cascade in the seed development pathway, were particularly reduced in the RAV1-overexpressing transgenic plants. We further determined that RAV1 directly binds to the promoter of MINI3 and IKU2, resulting in their repression. Direct treatment with brassinolide (BL) improved seed development of RAV1-overexpressing plants, but treatment with ABA severely worsened it. Overall, these results suggest that RAV1 is an additional negative player in the early stages of seed development, during which ABA and BR signaling are coordinated.

Although the evolution of Arabidopsis thaliana and humans diverged approximately 1.6 billion years ago, recent studies have demonstrated that protein function and cellular processes involved in disease response remain remarkably conserved. Particularly, γ-secretase, a multisubunit protein complex that participates in intramembrane proteolysis (RIP) regulation, is also known to mediate the cleavage of more than 80 substrates including the amyloid precursor protein (APP) and the Notch receptor. Although the genes (PS1/2, APH-1, PEN-2, and NCT) coding for the γ-secretase complex components are present in plant genomes, their function remains largely uncharacterized. Given that the deposition of 42 amino acid long amyloid-β peptides (hAβ42) is thought to be one of the main causes of Alzheimer's disease, we aimed to examine the physiological effects of hAβ42 peptides on plants. Interestingly, we found that Arabidopsis protoplast death increased after 24 h of exposure to 3 or 5 µM hAβ42 peptides. Furthermore, transgenic Arabidopsis plants overexpressing the hAβ42 gene exhibited changes in primary root length and silique phyllotaxy. Taken together, our results demonstrate that hAβ42 peptides, a metazoan protein, significantly affect Arabidopsis protoplast viability and plant morphology.
