멜라노마 세포(B16F10)에서의 개망초 추출물을 이용한 미백효과에 관한 연구 (A Study on the Whitening Effect of Erigeron annuus (L.) Pers. Ethanol Extract on Melanoma Cell (B16F10))

  • 개망초의 항산화능 측정으로 전자공여능을 측정한 결과 농도 의존적으로 활성이 증가하여 $500{\mu}g/ml$에서 82%의 높은 전자공여능을 나타내었다. $ABTS^+$ 라디칼 소거능 측정 결과 농도가 증가함에 따라 활성이 증가하였으며, $500{\mu}g/ml$에서 87% 이상의 활성을 나타내었다. Tyrosinase 저해 활성 측정결과 개망초 에탄올 추출물이 $100{\mu}g/ml$에서 69%의 효과를 나타내었다. 농도가 증가함에 따라 억제활성이 증가하는 것을 확인할 수 있었고, $100{\mu}g/ml$에서 32.2%의 효과를 나타내었다. 개망초 추출물에 대한 수렴효과를 측정한 결과 농도의존적으로 활성이 증가하였으며, $100{\mu}g/ml$에서 80%이상의 우수한 수렴효과를 확인할 수 있었다. 세포 생존율을 MTT 분석법을 통해 확인한 결과 농도 구간이 $100{\mu}g/ml$ 일 때 96%의 생존율을 보였으며, 따라서 관련 실험을 세포생존율이 100%에 가까운 25, 50, $100{\mu}g/ml$에서 진행하였다. B16F10 세포에 개망초 에탄올 추출물을 처리한 결과 MITF, TRP-2의 단백질 발현양이 감소하는 것을 확인할 수 있었으며, TRP-1과 tyrosinase 발현양이 크게 감소하는 것을 확인할 수 있었다. 상위 단계 유전자인 ERK와 CREB의 발현양은 미비했으나, 인산화된 ERK와 인산화된 CREB에서 농도가 증가함에 따라 단백질 발현양이 감소하는 것을 확인할 수 있었다. 개망초 에탄올 추출물을 25, 50, $100{\mu}g/ml$의 농도 별로 처리하였으며 MITF, TRP-1, TRP-2, tyrosinase의 mRNA 발현 측정 결과 농도가 증가함에 따라서 mRNA 발현을 억제하는 것을 확인할 수 있었다. 따라서 항산화 및 멜라닌 생합성에 관여하는 그 하위 유전자 및 상위 유전자의 발현을 저해하여 멜라닌 생성을 억제하는 것으로 추측되고 그에 따라 우리나라 전역에서 재배가 가능하고 국화과에 속하는 신귀화식물 개망초를 기능성 천연 미백소재로써 이용 가능성을 확인하였다.

LPS로 유도된 RAW 264.7 대식세포의 염증반응에서 MAPK 신호경로 조절을 통한 지칭개 에탄올 추출물의 항염증 효과 (Anti-inflammatory Effects of Hemistepta lyrata Bunge in LPS-stimulated RAW 264.7 Cells through Regulation of MAPK Signaling Pathway)

  • 본 연구에서는 LPS로 자극을 유도한 RAW 264.7 대식세포에서 지칭개 추출물의 항염증 효능을 알아보기 위해 이와 연관된 다양한 인자(NO, PGE2, IL-6, 및 TNF-α)와 MAPK 신호전달 경로에 대해서 조사하였다. 먼저HL 처리에 따른 세포 생존율에 대해 조사한 결과, HL을 농도별로 처리했을 때 저농도인 25 ㎍/mL에서부터 고농도인 100 ㎍/mL까지 모두 90% 이상의 생존율이 나타났으며, LPS를 처리한 RAW 264.7 대식세포에서도 90% 이상의 생존율을 나타내 실험에 사용된 HL의 농도가 RAW 264.7 대식세포에서 무독성임을 확인할 수 있었다. 이러한 무독성 조건에서 HL의 항염증 활성을 확인하기 위하여 염증 매개체(inflammatory mediators)로 잘 알려진 NO와 PGE2의 생성 변화를 확인한 결과, LPS로 염증반응이 유도된 RAW 264.7 대식세포에서 NO와 PGE2의 생성이 농도 의존적으로 억제됨을 확인하였다. HL이 NO와 PGE2를 억제하는 항염증 효과가 있음을 관찰하였고, 이에 전 염증성 사이토카인 분비에도 유의성 있는 효과를 나타낼 것이라 판단되어 전 염증성 사이토카인(IL-6와 TNF-α)에 어떠한 영향을 미치는지 조사하였다. HL은 LPS로 유도된 RAW 264.7 대식세포의 염증반응에서 IL-6의 생산을 유의적으로 억제함을 관찰할 수 있었다. 한편 다른 전 염증성 사이토카인인 TNF-α생산에는 아무 영향을 주지 않았다. 이러한 결과는 HL이 전 염증성 사이토카인 중 TNF-α조절에 관여하지 않고, IL-6 생성을 억제하여 염증 매개체의 생성을 억제한다고 추측할 수 있었다. HL이 NO, PGE2, 및 IL-6의 조절에 작용하는 메커니즘이 상위 시그널인 MAPK cascade의 억제를 통해 나타나는 효과인지 알아보기 위해 염증과 관련된 MAPK 시그널인 p38, ERK, 및 JNK의 발현 변화를 관찰하였다. HL은 p38과 ERK의 발현 활성화를 상당히 약화시켰지만 JNK는 p38과 ERK 보다 덜 민감하게 조절함을 관찰할 수 있었다. 이상의 결과를 종합해 보면 LPS로 유도된 RAW 264.7 대식세포에서 HL은 MAPK 신호경로인 JNK 발현에 유의적인 영향을 주지 못해서 JNK와 관련된 TNF-α생산에 영향을 주지 못한 것으로 판단된다. 다른 MAPK 신호경로인 p38과 ERK의 발현을 약화시킴으로써 그 다음 기작인 IL-6, NO, 및 PGE2의 생산을 억제시켜 염증반응을 억제한 것으로 추측되어 진다. 이상의 결과를 종합해 보면 HL이 항염 활성을 가지고 있음을 확인할 수 있었으며, 이를 기반으로 HL의 MAPK 신호경로를 통한 염증성 사이토카인과 염증 매개체와의 연관성에 대한 기초자료로 활용할 수 있는 근거 자료가 될 수 있을 것으로 생각된다. 또한, 다양한 경로를 통한 염증 조절 기전 연구는 추가적으로 이루어져야 할 것으로 사료된다.

데이터마이닝 기법을 활용한 비외감기업의 부실화 유형 분석 (The Pattern Analysis of Financial Distress for Non-audited Firms using Data Mining)

  • 본 연구에서는 데이터마이닝 기법의 일종인 자기조직화지도(Self-Organizing Map, SOM)를 이용하여 비외감기업의 부실화 유형을 구분하고자 한다. 자기조직화지도는 인공 신경망을 기초로 자율학습을 통해 입력된 값을 유사한 군집끼리 묶어내는 방법으로, 기존의 통계적 군집 분류 방법보다 성능이 뛰어나고, 고차원의 입력데이터를 저차원으로 시각화할 수 있다는 장점 때문에 다양한 분야에서 각광받고 있다. 본 연구에서는 기존 연구의 주요 분석대상이었던 외감기업에 비해 부실화 빈도는 높지만 데이터 수집의 어려움으로 인해 분석대상에서 다소 제외되었던 비외감기업의 부실화 유형에 대해 알아보고, 유형별 구체적인 사례도 소개하고자 한다. 재무자료수집이 가능한 100개의 비외감 부실기업에 대해 분석한 결과, 비외감기업의 부실화 유형은 다섯 가지로 구분되었다. 유형 1은 전체 집단의 약 12%를 차지하며, 수익성, 성장성 등 재무지표가 다른 유형에 비해 열등하였다. 유형 2는 전체 집단의 약 14%로, 유형 1보다는 덜 심각하지만 재무지표가 대체로 열등하였다. 유형 3은 성장성 지표가 열등한 그룹으로 기업간 경쟁이 극심한 가운데 지속적으로 성장하지 못하고 부실화된 경우로 약 30%의 기업이 포함되었다. 유형 4는 성장성은 탁월하나 부채경영 등 과감한 경영으로 인해 유동성 부족이나 현금부족 등의 이유로 부실화된 그룹으로 약 25%의 기업이 포함되었다. 유형 5는 거의 모든 재무지표가 우수한 건전기업으로, 단기적인 경영전략의 실수 또는 중소기업의 특성상 경영자의 개인적 사정으로 부실화 되었을 가능성이 큰 그룹으로 약 18%의 기업이 포함되었다. 본 연구 결과는 부실화 유형을 구분하는데 기존의 통계적 방법이 아닌 자기조직화지도를 이용하였다는 점에서 학문적 의의가 있고, 비외감기업의 재무지표만으로도 1차적인 부실화 징후를 발견할 수 있다는 점에서 실무적 의의가 있다고 할 수 있다.

방사선에 의한 폐 섬유화증에서 c-Jun N-terminal Kinase(JNK)의 역할 (The Role of c-Jun N-terminal Kinase in the Radiation-Induced Lung Fibrosis)

  • 서 론 : 폐암의 치료에 사용되는 방사선 조사는 흉곽 및 여러장기에 다양한 합병증을 발생시키며, 특히 방사선 섬유화증과 방사선 폐렴을 일으킨다. 방사선 조사의 초기 효과는 보통 염증 세포들의 침윤, 간질 및 폐포 부종, 상피세포의 탈락에 의한 방사선 폐렴이며 후기 효과는 방사선 섬유화증을 초래한다. 방사선 조사는 폐장의 염증 세포에서 TGF-beta의 단백 합성 및 활성도를 증가시키고, 생체외 실험에서 TGF-beta는 mitogen activated protein kinases(MAPKs)를 활성화시킨다는 것이 알려져 있다. c-Jun N-terminal kinase(JNK)는 MAPKs중의 하나로 핵단백질인 c-Jun을 인산화(phosphorylation) 시켜 전사(transcription)를 증가시키는데 생체외 실험에서 자외선 조사후 대식 세포에서 JNK의 활성이 증가되는 것으로 알려져 있다. 하지만 현재 까지 생체내에서 JNK가 방사선 조사에 의해 활성화되는지 그리고 이들 활성화가 방사선 섬유화증의 병인에 관계하는 지는 알려진 바 없다. 본 연구는 흉부에 방사선을 조사한 백서를 이용하여 방사선 섬유화증의 병인에 JNK가 신호 전달 체계에서 주요한 역할을 담당하는 지를 알아보고자 시행하였다. 대상 및 방법 : C57BL/6 백서의 전 흉부에 14 Gy의 $^{60}CO{\gamma}$-ray를 조사한 후 일정한 간격(1주, 4주, 8주)으로 폐장의 세포 분석을 위한 기관지 폐포 세척술, elastin의 합성 정도를 측정하기 위한 Verhoeff stain, collagen 합성 양을 알기 위한 hydroxyproline의 측정, JNK 활성도를 알기 위한 in vitro JNK assay를 시행하였다. 결 과 : 폐장의 기관지 폐포 세척 소견상 총세포수는 방사선 조사 4주, 8주후에 증가하는 소견을 보였다. 세포의 감별 분석상 림프구는 4주후 증가되는 경향을 보였다. Verhoeff 염색상 폐포 조직의 특이 변화 소견은 없었으며 hydroxyproline의 양은 방사선 조사전에 비해 4주, 8주후에 증가하는 소견을 보였다. 방사선 조사 후 시간이 경과함에 따라 4주 후에 c-Jun N-terminal kinase(JNK)의 활성도가 가장 높았으며 8주 후에는 4주 후와 비슷한 활성도를 보였다. 결 론 : 흉곽에 방사선 조사 후 hydroxyproline양의 증가와 함께 JNK 활성도의 증가 소견을 보여 방사선 폐섬유화증의 병인에 JNK가 관여될 것으로 사료된다.

하저 지반특성 규명을 위한 전기비저항 탐사 (DC Resistivity method to image the underground structure beneath river or lake bottom)

  • 수로는 약선대를 따라 형성되는 경우가 많으며, 특히 강을 따라 단층이 발달하는 경우가 많다. 교량과 같은 수상 구조물의 건설을 위하여 탐사의 필요성이 많았음에도 불구하고 육상과는 달리 수상 전기비저항 탐사가 활발하게 적용되지 못한 것은 현장탐사와 해석의 어려움 때문이었다고 볼 수 있다. 그러나 무엇보다도 중요한 것은 양질의 자료 획득이며, 양질의 자료 획득이 육상탐사보다 어려운 것은 영상화하고자 하는 지하구조가 물로 덮여 있다는 점에서 연유한다. 이 연구에서는 이와 같은 점에 중점을 두고 수치 모델링과 현장탐사 사례 분석을 통하여 수상 전기비저항 탐사 자료의 특성, 현장 탐사와 해석 방법에 대하여 논하였다. 수상 전기비저항 탐사는 전극이 물 표면에 혹은 물 바닥에 설치되어 있는가에 따라 탐사자료의 획득방법, 고려사항, 해석 방법이 달라지므로, 전극을 물 표면에 띄운 경우와 바닥에 설치한 경우로 나누어 논의를 전개하였다. 이를 통하여 하상 전기비저항 탐사는 수 층 하부 지반을 정확하게 영상화할 수 있음을 보였다. 또한 수상 전기비저항 탐사에서 전극을 물 바닥에 설치하는 것이 물 표면에 설치하는 것보다 훨씬 더 분해능이 높은 영상을 획득할 수 있으나, 육상탐사에 비하여 지하구조에 대한 민감도는 훨씬 낮고, 유동전위 등에 의하여 전기잡음이 높을 가능성이 높기 때문에 육상탐사보다 훨씬 높은 S/N 비를 갖는 현장 탐사자료의 획득이 필수적이며 가능한 한 높은 분해능을 갖는 전극배열을 선택하여야 하는 것으로 나타났다 전극을 물 바닥에 설치하는 방법은 고분해능 영상을 제공할 수 있기 때문에 지하의 정밀 영상을 획득하기 위한 정밀탐사에 적합하다. 이에 반해 물 표면에 전극을 띄워서 자료를 획득하는 방법은, 특히 이동하면 연속적으로 탐사하는 스트리머 전기비저항 탐사는 매우 빠른 시간에 넓은 지역을 탐사할 수 있기 때문에 개략탐사에 많은 활용이 기대된다.향상을 위해 좀더 적극적인 치료가 필요하다.과 TIMP-2의 발현의 정도는 차이가 있었으며, 각 장기의 특정한 세포에서만 발현이 되었다. TIMP-1의 경우 간과 신장에서는 방사선에 의해 발현이 증가되었으나 폐에서는 발현이 증가되지 않았다. TIMP-2의 경우 폐에서는 방사선에 의해 발현이 증가하였으나 간과 폐에서는 방사선에 의한 발현의 변화는 불규칙적이었다. 방사선 조사 후 경과 시간과 방사선량에 따른 발현의 변화도 일정하지 않았다.EX> 발현의 감소가 관찰되어, Captopril이 조기 폐손상을 억제하는 방사선보호제로서 기전의 일부에 $TNF\alpha$$TGF\beta1$이 관여함을 확인할 수 있었다.주의 정책으로 우리 민족의 정서와 문화를 보존하는 일에 등한시하여 왔다. 이 때문에 우리 민족 고유의 뿌리를 점차 잃어가고 있다. 이러한 시점에서 도시노점상을 정리하기 위한 목적으로 정부에서 도시 5일장을 개장한 것은 역사의 아이러니라 아니할 수 없다. 이렇게 정부주도로 개장된 5일장이 운영되어 온 지 2년이 되어가고 있다. 개장 초기에는 시에서의 지원도 적극적이고 소비자들의 호응도 좋았으나, 최근에 들어 활성화의 속도가 둔화되면서 도시 5일장의 개념을 재정립할 필요성이 제기되고 있다. 정부의 주도로 시작된 5일장이므로 정부의 적극적인 추진하에 풍물시장 번영회와 활성화 방안을 모색해야 한다. 쌍다리 풍물시장의 5일장을 활성화하기 위하여 도시 5일장의 개념을 ${\ulcorner}$국민들의 생활수준이 향상되고 여가를 즐길 수 있는 여건이 형성되고 있으므로 전통문화(향토문화)를 유지하고 시민들의 정서함양에 기여할 수 있는 여가공간 조성${\lrcorner}$으로 규정해야 한다. 이러한 개념 하에 5일장의 주체를 명확히 하기 위해 시청 지역경제과를 중심으로 지자제의 실시에

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The strip was illuminated at 45˚c through optical fiber and the intensity of reflected light was determined at rectanguLat angle using a photodiode. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms.   • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70${\pm}$1.32mmHg/min)compared to CF dialyzers(4.32${\pm}$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms.
