Construction of a Transgenic Tobacco Expressing a Polydnaviral Cystatin (폴리드나바이러스 유래 시스타틴 유전자 발현 형질전환 담배 제작)

  • CpBV (Cotesia plutellae bracovirus) is a polydnavirus and encodes a cystatin (CpBV-CST1) gene. Its overexpression suppresses insect immunity and alters insect developmental processes. This study aimed to construct a genetically modified (GM) tobacco to further explore the physiological function of the viral cystatin and to apply to control insect pests. To this end, the transgenic tobacco lines were screened in expression of the target gene and assessed in insecticidal activity. A recombinant vector (pBI121-CST) was prepared and used to transform a bacterium, Agrobacterium tumefasciens. The transformed bacteria were used to generate transgenic tobacco lines, which were induced to grow callus and resulted in about 92% of shoot regeneration. The regenerated plants were screened by PCR analysis to confirm the insertion of the target gene in the plant genome. In addition, the expression of the target gene was assessed in the regenerated plants by quantitative real-time PCR (qRT-PCR). The qRT-PCR analysis showed that the transgenic line plant expressed the target gene about 17 times more than the control tobacco, indicating a stable insertion and expression of the target gene in the transgenic tobacco line. The insecticidal activity was then analyzed using the screened transgenic tobacco lines against the teneral 1st instar larvae of the oriental tobacco budworm, Helicoverpa assulta. Though there was a variation in the insecticidal efficacy among transgenic lines, T9 and T12 lines exhibited more than 95% mortality at 7 days after feeding treatment. These results suggest that CpBV-CST1 is a useful genetic resource to be used to generate GM crop against insect pests.

  • These experiments were carried out to define the effects of 2.4-D, NAA and Benzyladenine on the differentiation and growth of the organs and the induction of callus from the potato meristem. The results were summarized as follows ; 1. The differentiation and growth of the shoots from the potato meristem was promoted in increased concentration of Benzyladenine but the callus was not induced on the M.S. medium containing Benzyladenine. 2. On the M.S. medium containing NAA 0.5 mg/l or higher concentration of NAA, the shoots were not initiated but the callus were induced from potato meristem. The growth of callus was promoted in increased concentration of NAA. 3. The roots were initiated from 50% of potato meristems planted on the M.S. medium containing more than 0.1 mg/l of NAA but the roots were pot initiated on the medium containing 2.4-D. 4. The shoot growth was significantly increased by increasing of 2.4-D concentration up to 0.1 mg/l, but the shoots were not initiated on the medium containing 2.4-D more than 1.0 mg/l. 5. For the induction and growth of the callus from potato meristem, NAA was more effective than 2.4-D and the most effective medium was M.S. medium supplemented with 2.0 mg/l of NAA. 6. The M.S. mediums supplemented with BA 2.0 mg/l and NAA 0.1 mg/l or BA 1.0 mg/l and 2.4-D 0.1 mg/l showed good results for entire plant regeneration from potato meristem.

Comparison of Above-ground Growth Characteristics Between Naturally Regenerated and Planted Stands of Pinus densiflora for. erecta Uyeki in Gangwon Province (천연갱신과 인공조림된 강원지역 금강소나무의 지상부 생장특성 비교)

  • This study was undertaken to compare early growth characteristics, such as height, diameter at root collar (DRC), and the dry weights of stem, branches, needles and roots between naturally regenerated and planted stands of Pinus densiflora for. erecta in Dae-gi and Bo-gwang site, Gangwon province, Republic of Korea. Average DRC growth was similar between the naturally regenerated and the planted stands in Dae-gi area, however, difference was statistically significant (P<0.01) between the naturally regenerated and the planted stands in Bo-gwang area, 4.66 cm and 2.61 cm, respectively. Average height of the naturally regenerated stands was significantly greater (P<0.01) than that of planted stands in both Dae-gi and Bo-gwang areas. The composition ratio of the dry weights for each part of measured trees was the highest in the stem in the naturally regenerated stands, but in the planted stands, the needle was the highest in both sites. The root/shoot ratio in the Dae-gi and Bo-gwang areas in the planted stands were much greater than those in the naturally regenerated stands, 0.25-0.30 and 0.17-0.18, respectively. The ratio of needle to stem biomass (N/S), branch to stem biomass (B/S), and needle to branch biomass (N/B) was significantly greater in the planted stands than those in the naturally regenerated stands in both sites. The results of this study represent that the regeneration methods, natural and artificial regenerations, can have an influence on the early growth of P densiflora for. erecta. This results not only provide the need for a correct planting method, but also will be useful in studies on the environmental adaptability of trees.
